具体实施方式
以下本文将通过具体的实施例来描述发明。如未特别指明之处,可根据本领域技术 人员所熟悉的酿酒技术和基因重组技术手册以及本文所引用的参考文献中所列的方法来实施。另外,实施例中所使用的材料和设备均可从市场上购买。
实施例1絮凝性质优化的酵母的获得
根据我们设计的基因序列(SEQ ID No:1,其编码的蛋白变体的氨基酸序列为SEQ IDNo:2),通过商业途径委托广东暨大基因药物工程研究中心有限公司合成我们发现的絮凝蛋白变体基因并构建入酿酒酵母中。克隆过程参照《分子克隆实验指南》以及所用商品化试剂的操作指南进行,简要过程如下:
通过DNA自动合成仪,合成絮凝蛋白变体基因的核酸片段,用T4多核苷酸激酶(购自TaKaRa公司)将这些核酸片段的5’端进行磷酸化,然后等摩尔比混合这些核酸片段后于75℃变性5分钟,然后退火降温至12℃,加入T4DNA连接酶(购自TaKaRa公司)连接12小时。然后,取1μL上述连接产物在50μL反应体积中进行PCR扩增,其中正向引物如序列表的SEQID No:3所示(引入了EcoR I内切酶位点)、反向引物如序列表的SEQ ID No:4所示(引入了Xba I内切酶位点),反应条件为:以94℃变性2分钟,然后以94℃变性30秒、63℃退火60秒并72℃延伸90秒进行30个循环,最后以72℃延伸12分钟并降温至4℃。
琼脂糖凝胶电泳上述PCR产物,回收约4.5kb大小的片段,用EcoR I和Xba I双酶切该片段,并与经这两个内切酶酶切的pYESS2/CT质粒(可购自Invitrogen公司)用T4DNA连接酶进行连接,电转化法将pYESS-fln质粒转入酿酒酵母(Saccharomyces cerevisiae)QN732株(购自广西易多收生物科技有限公司,其为非凝絮性酵母,但是发酵产生的酒中有淡淡巧克力香味)中,涂布在选择性培养基上培养,取生长出的酵母提取总DNA后,用上述正向引物和反向引物PCR扩增,琼脂糖凝胶电泳发现有约4.5kb大小的片段,表明已经将如序列表的SEQ ID No:1所示的基因导入了酿酒酵母中了。同时,将pYESS2/CT空质粒和克隆有野生型絮凝蛋白基因的pYESS2/CT质粒电转化入该非凝絮性酵母,分别作为阴性对照和阳性对照。
我们将20mL絮凝检测液(50mmol/L柠檬酸钠,5mmol/LEDTA,20mmol/L CaCl2,pH 4.5;并分别加入以下终浓度的物质:不加入(无糖),1M葡萄糖)置于50mL摇瓶中,分别加入上述各酵母细胞1克(湿重),以30℃、100r/min振荡培养2小时。然后,取5mL细胞悬浮液至10mL试管中,立刻在液面取样200μL,垂直静置5分钟,然后在液面取样200μL,分别测定垂直静置前后的OD600值,计算絮凝率(絮凝率=(1-垂直静置后的OD600值/垂直静置前的OD600值)*100%)。结果如表1所示,酿酒酵母QN732基本不具有絮凝性质;克隆了野生型絮凝蛋白基因后,无论在无糖还是在含糖环境下,酵母都将具有絮凝性质;本发明的絮凝蛋白变体基因转入的酵母在含糖环境下不具有絮凝性质,将保持悬浮状态发酵,而在无糖环境下将具有絮凝性质,容易沉淀到发酵罐底部,便于发酵终止后收集。

实施例2非蒸馏酒的制备
将500克大米磨碎,加700克水浸泡1小时,然后煮熟,加2公斤水和α淀粉酶(可购自南宁赛华发酵制品有限公司)10U/克大米原料,于55℃搅拌30分钟,然后升温至95℃继续搅拌30分钟,使大米液化。然后加入乳酸调节pH至4.2,于58℃加入糖化酶(可购自南宁赛华发酵制品有限公司)150U/克大米原料,保温90分钟,去除糟渣后得到糖化液。
将上述糖化液装入灭菌的发酵罐,加入实施例1的克隆有絮凝蛋白变体基因的酵母2克(干重)和发酵助剂Fermaid K(可购自上海杰兔工贸有限公司)5克,于28℃发酵48小时,然后加入0.5L 15°Bx的甘蔗汁(新榨甘蔗汁用水稀释到15°Bx,然后高温灭菌,即成),温度控制在18~22℃,密封发酵9~12天。当残糖<4克/L时,发酵结束,由于此时酵母基本沉积在罐底,因此将上清液部分简单过滤即可取得澄清的酒体,经巴氏灭菌,加水稀释至含酒精12%(体积比)即可装瓶。多批次酿造后,经检测,各批次酒理化指标稳定(见 表2),符合国家酒类的标准;该酒香味中带有甘蔗汁的清香和巧克力的清香,口感丰满。
