Abstract

本发明是一种抗癌新药尖尾芋提取物的制备方法,它以植物尖尾芋(Alocasia eucullatta(Dour)Schott)为原料,制成可以治疗癌症的活性提取物。采用溶剂提取法、溶剂萃取法、树脂吸附法、正相柱层析法、反相柱层析法、凝胶层析法等工艺中的一种或者多种工艺,并结合浓缩干燥、冷冻干燥、喷雾干燥等常用干燥方法制备成尖尾芋提取物,将制成的尖尾芋提取物作为抗癌活性成分在癌症治疗中进行应用。

Description

一种抗癌新药尖尾芋提取物的制备方法
本发明涉及医药技术领域,确切地说它是抗癌新药尖尾芋提取物的制备方法。
尖尾芋(Alocasia eucullatta(Dour)Schott)为天南星科植物假海芋的根茎,生于山涧阴湿处,或树旁沟边、河边。分布于广东、广西、四川等地。该植物能清热、解毒、活血、消肿,治疔疮、痈肿、丹毒,跌打损伤,慢性气管炎、钩端螺旋体病等疾病。迄今为止,对尖尾芋的研究很少,多见于一般药效学报道。有关尖尾芋的抗癌药理活性很少报道,仅见侯学文从中分离得到一个凝集素,其对肺腺癌细胞株P18仅有很少杀伤能力。癌症是当今社会发病率高、死亡率高的一种顽痰,目前尚无治愈率较高的药物的报道。本发明人在寻找中草药抗癌活性成分过程中,发现尖尾芋提取物具有很好的抗肿瘤活性,并且从中获得多个具有抗癌活性的二苯乙烯、二苯乙烯苷、脑苷类化合物等多种骨架类型的化合物。
本发明的目的在于提供一种抗癌新药尖尾芋提取物的制备方法,其工艺简单实用,为尖尾芋的应用提供了广泛的途径。
本发明的目的是通过如下方案实现的:以尖尾芋(Alocasia eucullatta(Dour)Schott)为原料,采用溶剂提取法.溶剂萃取法.树脂吸附法,正相柱层析法、反相柱层析法等工艺中的一种或者多种工艺,并结舍浓缩干燥、冷冻干燥,喷雾干燥等常用干燥方法制备成尖尾芋总提取物,将制成的尖尾芋总提取物作为抗癌活性成分用于治疗多种癌症。
本发明的优点是:尖尾芋提取物工艺简单实用,抗癌疗效好,并且无毒副作用。为尖尾芋应用提供了广泛的途径。
下面结合实施例对本发明做进一步详细地描述。
实施例:
1.溶剂提取法/溶剂萃取法全用提取制备尖尾芋提取物
1.2kg用50%乙醇在90℃下回流,合并提取液,浓缩得到130g乙醇提取物。 将乙醇提取物混悬于水中,用乙酸乙酯萃取,经浓缩干燥即可得到尖尾芋提取物,收率为1.2%。
2.溶剂提取法/树脂吸附法合用提取制备尖尾芋提取物
尖尾芋1.0kg用80%乙醇在90℃下回流,合并提取液,浓缩得到13Og乙醇提取物。乙醇提取物上D-101型吸附树脂柱,先用水、5%乙醇/水洗脱以除去色素和杂质,而后用60%乙醇/水洗脱。其中60%乙醇/水洗脱部分减压浓缩除乙醇,再经冷冻干燥可得到尖尾芋提取物,收率为1.0%。
3.尖尾芋提取物的体外抑瘤实验
3.1细胞株、试剂及仪器
人宫颈癌Hela细胞株购自中科院细胞库;DMEM培养液,Gibco产品;胎牛血清,Gibco产品;胰蛋白酶,Gibco产品;DMSO溶液,sigma公司;96孔板,Corning公司;Revco 3000培养箱,Thermo公司;Multishkan MK3酶标仪,Thermo公司。
3.2肿瘤细胞培养方法
人宫颈癌Hela细胞株用含10%胎牛血清的DMEM培养液培养,培养条件为37℃,5%CO2浓度及饱和湿度。当细胞生长至对数生长期时,用25%胰酶消化细胞,洗涤吹散细胞稀释至106/ml,每孔100μL接种于96孔板,以备MTT实验。
3.3细胞增殖实验(MTT法)
尖尾芋提取物溶于DMSO溶液,取含1‰DMSO的尖尾芋提取物溶液,用含10%胎牛血清的DMEM溶液配成浓度分别为100、50、20、2μg/mL的尖尾芋提取物溶液,每孔100μL接种于96孔板,混匀,使尖尾芋提取物每孔终浓度分别达到50、25、10、1μg/mL,放入培养箱继续培养16、32、48h。每孔加入5mg/mLMTT溶液5μL,继续培养5h,测定各孔吸光度。以不加任何处理的培养细胞为空白对照,以单用培养细胞作为阴性对照。每个浓度梯度均设置12个平行组,分别计算细胞抑制率。按照以下公式计算抑制率:
抑制率=1-(实验组吸光度平均值/对照组吸光度平均值)×100%
结果见表1
表1尖尾芋提取物对Hela细胞体外增殖的抑制作用

结果表明:不同浓度,不同培养时间的尖尾芋提取物可显著抑制人宫颈癌Hela细胞的增殖,提示其可能有一定的抗癌效果。

Claims (2)

1.一种抗癌新药尖尾芋提取物的制备方法,其特征在于:以植物尖尾芋为原料,采用溶剂提取法和溶剂萃取法,并结合浓缩干燥、冷冻干燥、喷雾干燥常用干燥方法制备成尖尾芋提取物,将制成的尖尾芋提取物作为抗癌活性成分,使之在癌症治疗中进行应用;作为抗癌活性成分的尖尾芋提取物包括其中所含的各种类型的有机酸、二苯乙烯、二苯乙烯苷、甾体皂甙类化合物以及其他骨架类型的化合物;所采用的溶剂提取法中,选用乙醇,在室温下浸润提取或者用超声波振荡仪超声振荡提取,或者在加热状态下浸润提取或回流提取,提取次数可以是一次,也可以是多次;所采用的溶剂萃取法中,将乙醇提取物混悬于水中,用正丁醇或含水正丁醇直接进行萃取,或者经低极性溶媒脱脂后,再用正丁醇或含水正丁醇进行萃取,萃取次数可以是一次,也可以是多次。
2.一种如权利要求1所述的抗癌新药尖尾芋提取物的制备方法,其特征在于:植物原料尖尾芋可以是根茎,也可以是枝叶、果实等全草的任一部位,原料可以是新鲜采集的,也可以是经过炮制的。
