Abstract

技术领域
本发明涉及一种新的化妆品组合物,所述组合物包含来自鼓槌石斛兰(orchid Dendrobium chrysotoxum)的至少一种提取物作为活性剂以及至少一种化妆品上可接受的辅剂(excipient),所述组合物特别用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命。
背景技术
皮肤老化表现为皮肤各种内在的变化和可视迹象的不断出现,这些内在变化和可视迹象改变了皮肤的美观和质量。皮肤纹理更为粗糙,并出现许多皮肤缺陷(blemish)。
在细胞水平,老化作用导致生物机制不断劣化,例如结构蛋白(胶原、弹性蛋白)的合成放缓。
细胞凋亡是一种程序性细胞死亡,是在基因控制(genetic control)下通过片段化(fragmentation)从体内清除细胞的主动生物过程(activebiological process)。
该清除作用可以是自然的(组织中的细胞过剩),或由各种应激(stress)诱导。
细胞凋亡的生物级联是已知的,该生物级联启动许多效应子(例如半胱天冬氨酸蛋白酶(caspase)家族的蛋白酶)。
已知的细胞凋亡抑制剂(IAP)具有阻断半胱天冬氨酸蛋白酶原的激活和抑制成熟的半胱天冬氨酸蛋白酶的活性的功能,从而使得细胞凋亡现象放慢(Deveraux等,Genes Dev.13(1999),pp.239-252)。
在IAP中,应提及存活蛋白(survivin),它是一种双功能蛋白,能够同时控制细胞的凋亡并调节细胞的细胞周期。
对于可诱导这一细胞凋亡现象的应激,存活蛋白是角化细胞在该应激下得以存活以及对该应激具有抗性的调节剂。存活蛋白也调节角化细胞在表皮的更新和再生方面的能力。因此,能够减少表皮的细胞死亡(cellular capital),并保持有效的细胞更新(Dallaglio等,ExperimentalDermatology,18(5)464-72)。
存活蛋白的产生严格受控于在细胞凋亡期间释放的线粒体蛋白Smac/DIABLO(第二线粒体的半胱天冬氨酸蛋白酶激活剂或低等电点IAP直接结合蛋白)。
Smac/DIABLO这种蛋白以29kDa前体的形式合成,通过具有线粒体定位信号的N端域移位到线粒体中。然后,这一前体被切割,得到23kDa的成熟蛋白(Zhang等,Acta Biochim.Biophys.Sin.2007,39(2),108-116)。
已知UV辐射或自由基(如ROS)诱发皮肤中与皮肤老化有关的细胞应激。
在响应刺激(如UV辐射或活性氧(ROS))时,成熟的线粒体蛋白Smac/DIABLO被释放到细胞胞液中作为IAP的抑制剂。Smac/DIABLO以二聚体的形式与IAP结合,从而阻断半胱天冬氨酸蛋白酶的失活(Kurita-Ochiai T.等,J.Dent.Res.2010,89(7),689-694)。
类似地,在响应这些相同的刺激时,已证实减少了Smac/DIABLO的产生,并伴随有存活蛋白的过表达,使得细胞寿命增加(AzizMH.等,Photochem.Photobiol.2005,81(1),25-31)。
因此,为了更有效地对抗皮肤老化,特别有意义的是:能够在化妆品中含有一种或多种其它活性剂,该活性剂能够显著地减少Smac/DIABLO这种蛋白在表皮细胞中的表达和/或活性;并且能够使得IAP以上述方式抑制半胱天冬氨酸蛋白酶,从而改善对应激(由UV辐射或自由基诱发)的细胞反应性,并延长细胞寿命。
然而,迄今为止,对于用于化妆品应用的这种药剂尚无记载。
因此,特别令人意外地证实了兰花提取物、特别是鼓槌石斛兰的花的提取物对蛋白Smac/DIABLO的表达显示出上述抑制效果。
在兰花中,中医上已知若干属于石斛属的种。然而,所使用的是茎或叶的提取物(Gutierrez RMP,J.Med.Plant Res.,2010,4(8),592-638)。
具体而言,已经从鼓槌石斛的茎的提取物中分离出了毛兰素(erianin)(NG等,Life Science,2000,66(8),709-23),这种化合物的抗氧化活性已经在大鼠中得到了证实。
迄今为止,对于鼓槌石斛的花的提取物,尚未证实其特别的活性。
发明内容
本发明的主要目的在于提供新的化妆品组合物,旨在特别用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命;具体而言,上述活性通过抑制线粒体蛋白Smac/DIABLO的表达和/或活性而得到。
本发明的目的还在于提供使用该化妆品组合物的化妆护理方法。
本发明的目的还在于提供一种新的植物来源提取物,该提取物降低皮肤中、特别是表皮中线粒体蛋白Smac/DIABLO的表达和/或活性。
最后,本发明的目的在于特别是在化妆品工业中,通过工业规模可用的、简单而相对低成本的解决方案解决总体技术问题。
本发明的发明人等目前已经证实,用鼓槌石斛种的兰花的至少一种提取物对正常的人角化细胞(NHK)进行处理,诱发线粒体蛋白Smac/DIABLO的表达和/或活性显著降低。
对所述蛋白的表达和/或活性的这种抑制效果使得IAP在限制细胞凋亡和调控细胞的细胞周期方面具有更强的活性。
因此,根据第一方面,本发明涉及包含所述提取物的化妆品组合物;还涉及化妆护理方法或皮肤病学护理方法,该方法的特征在于包括将有效量的本发明所述的组合物施用到与之相关的、身体或面部的皮肤部位上。
根据第二方面,本发明涉及兰花提取物、特别是鼓槌石斛种的兰花提取物(本发明的提取物)以及该提取物在化妆品组合物或皮肤病学组合物中作为活性剂的用途;该提取物用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命,特别是在于该提取物抑制线粒体蛋白Smac/DIABLO的表达和/或活性。
具体实施方式
本发明的第一个主题涉及一种化妆品组合物,该组合物包含鼓槌石斛兰的花的至少一种提取物作为活性剂以及至少一种化妆品上可接受的辅剂。
该化妆品组合物特别旨在预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命。
本发明的提取物有利地抑制线粒体蛋白Smac/DIABLO的活性和/或表达。
该植物材料优选仅由鼓槌石斛兰的花组成。然而,本发明还涉及本发明的提取物与下述物质的组合:鼓槌石斛兰的其它部位的任何提取物,优选在与本发明的提取物相同的条件下制备;或从这些提取物之一中分离出来的化合物。
本发明的提取物可通过本领域技术人员已知的不同的提取方法来制备。
优选地,该提取物通过极性溶剂或极性溶剂混合物获得。
作为极性溶剂,溶剂或溶剂混合物优选选自:水;C1-C4醇,例如乙醇;C2-C6二醇,优选选自乙二醇(glycerol)、丁二醇和丙二醇;或它们的任意混合物。
根据优选的实施方式,该C1-C4醇为乙醇。
具体而言,该植物材料通过包含至少50%v/v乙醇直至100%v/v乙醇的溶剂提取;如果必要,余量由其它极性溶剂、特别是水组成。
提取可在加热回流下进行,或可在室温进行。
根据本发明优选的变型,通过在如前所述的极性溶剂或极性溶剂混合物中,将植物材料在室温下浸渍若干小时实现提取。
根据一种变型,所述提取物是可能成为溶液的干提取物。
在提取步骤本身之前,可将植物材料进行干燥和/或研磨和/或去脂(delipidated)。
提取物的去脂作用可通过非极性溶剂(例如C6-C7烷烃)实现,或者还可通过超临界状态的CO2实现,并且可能加入极性共溶剂(例如乙醇、甲醇或辛酸/癸酸甘油三酯(MygliolHüls AG))。
该提取还可任选包括至少一个对本发明的提取物进行脱色或纯化的补充步骤。
例如,提取物的脱色可为用非极性溶剂处理所述提取物,该非极性溶剂为用来将提取物去脂的非极性溶剂的类型。
该提取可通过将提取溶剂部分去除或全部去除的步骤完成。
在第一种情况中,通常将提取物进行浓缩,直至得到不含大量有机溶剂的水性浓缩物;在第二种情况中,得到干燥残余物。
或者,可将提取步骤的产物冻干(lyophilized)或粉碎(atomized)成粉末形式。
作为本发明所述的化妆品组合物中的活性剂,该提取物以干燥状态使用,或以处于化妆品上可接受的溶剂混合物或溶剂(可与用于提取的溶剂相同或不同)中的溶液或悬浮液进行使用。
该化妆品组合物包含有效量的提取物,该提取物用于获得所希望的效果、特别是对线粒体蛋白Smac/DIABLO的活性和/或表达的抑制作用。
因此,相对于组合物的总重量,该组合物优选包含0.0001wt%-1wt%、更优选0.001wt%-0.01wt%的干提取物。
在本发明的提取物与化妆品上可接受的其它活性剂组合使用的化妆品组合物中,也可得到或改善本发明的提取物的性质;所述其它活性剂为经纯化的物质和/或提取物、特别是植物提取物的形式,显示出与本发明所述的提取物相似和/或互补的化妆效果。
因此,该组合物可包含一种或多种其它植物提取物,优选包含:鼓槌石斛兰的其它部位的至少一种提取物;或其它兰花的至少一种提取物;或由前述提取物纯化得到的物质。
所述化妆品组合物还可优选包含:一种或多种刺激存活蛋白表达的其它活性剂,例如毛喉素;或含有这些活性剂的提取物,特别是毛喉鞘蕊花(Coleus forskolii)的提取物;或由选自普通念珠藻(Nostoccommune)、甄叔迦树(Buteafrondosa)、新绿藻(Neochloris oleabundans)、二形栅藻(Scenedesmus dimorphus)、姜黄(Curcuma longa)、番红花(Crocus sativus)、奥氏西非苏木(Daniellia oliveri)、Lepechiniacaulescens、Limnophila conferta的植物种获得的植物提取物;以及它们的任意混合物。
最后,该化妆品组合物可包含化妆品上可接受的其它活性剂,该活性剂选自如下物质组成的组:具有抗老化活性的物质;对皮肤具有脱色、美白或清洁活性的物质;具有减肥活性的物质;具有保湿(hydrating)活性的物质;具有镇定(calming)、舒缓(soothing)或放松(relaxing)活性的物质;具有刺激皮肤微循环以改善特别是面部肤色光泽的活性的物质;用于对油性皮肤进行护理、具有皮脂调节活性的物质;用于清洗或净化皮肤的物质;具有抗自由基活性的物质;以及它们的任意混合物。
优选地,本发明所述的组合物包含至少一种化妆品上可接受的辅剂,该辅剂可选自色素、着色剂、聚合物、表面活性剂、粘性剂(viscosityagents)、芳香剂(perfumes)、电解液、pH调节剂、抗氧化剂、防腐剂以及它们的任意混合物。
例如,所述化妆品组合物可为精华素(serum)、乳液(lotion)、霜(cream)、水凝胶(hydrogel),优选为面膜、水包油乳剂、无水组合物,或可为条状物(stick)形式、贴剂(patch)形式,或为唇膏、睫毛膏或粉底型化妆产品的形式。
本发明所述的提取物和组合物显示出特别希望的效果:预防或延缓皮肤老化迹象的出现;放慢或减弱皮肤老化迹象的影响;或延长细胞或组织的寿命。
因此,本发明的第二个主题涉及鼓槌石斛兰的花的至少一种提取物在化妆品组合物中作为活性剂的用途,该活性剂用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命。
优选地,该提取物或该组合物如上文或下文所定义。
优选地,包含所述提取物的组合物本身旨在预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命。
本发明的第三个主题涉及一种化妆护理方法,特别是用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命,该方法的特征在于包括将有效量的如上定义的化妆品组合物施用到身体或面部皮肤的至少一个部位。
优选地,将所述化妆品组合物施用到显示出下述迹象的、身体或面部的皮肤区域:可视的老化迹象,如存在皱纹或细小皱纹、或皮肤肤色失去光泽;其它迹象,如皮肤弹性和/或柔性消失、皮肤厚度减小、和/或皮肤干燥度或粗糙度增加。
本发明还涉及有效量的鼓槌石斛兰的花的至少一种提取物在制备用于预防或延缓皮肤内在和/或外在老化迹象的出现、或放慢皮肤老化迹象的影响的组合物方面的用途。该组合物尤其可通过局部施用到个体皮肤的至少一个显示出下述迹象的区域进行施用:可视的老化迹象,例如存在皱纹或细小皱纹、或皮肤肤色失去光泽;或其它迹象,例如皮肤弹性消失和/或柔性消失、皮肤厚度减小、和/或皮肤干燥度或粗糙度增加。该组合物尤其旨在用于施用到超过35岁、超过40岁、超过45岁、超过50岁、超过55岁乃至超过60岁的个体的皮肤上。该组合物可优选施用到成熟的皮肤上。特别地,该组合物旨在用于预防或延缓眼部和/或面部皮肤的老化迹象的出现。
本发明的第四个主题涉及抑制线粒体蛋白Smac/DIABLO的表达和/或活性的兰花提取物、特别是鼓槌石斛种的兰花提取物。
该提取物优选为如上定义的鼓槌石斛的花的提取物。
此外,该提取物优选通过如上所述的极性溶剂或极性溶剂混合物获得。
优选地,该提取物是可能成为溶液的干提取物。
根据随后关于提取物制备的实施例、证实提取物性质的试验以及使用该提取物的化妆品组合物的实施例所作出的解释性的描述,本发明的其它目的、特征和优点将会清楚地展现;上述实施例和试验均以阐明本发明的方式给出,并不限定本发明的范围。
除非另外指明,在实施例中,所有百分比均以重量百分比给出,温度为摄氏温度,压力为大气压力。
附图说明

实施例
实施例1:鼓槌石斛的花的提取物的制备
本发明的提取物的制备
根据本发明,制备四种提取物和作为对比的其它两种提取物。
提取物S-1、S-2、S-3和S-4的植物材料由鼓槌石斛兰的花组成。提取物R和G由鼓槌石斛兰的根或由皇后兰种(Grammatophyllum sp.)兰花的叶获得。
将干燥状态且经研磨的植物材料用含水酒精(90/10v/v乙醇/水)回流提取30分钟,以制备提取物S-1、R和G。然后去除提取溶剂,得到干提取物。
改变提取溶剂和温度条件,再进行上述提取以获得提取物S-2、S-3和S-4。测试了两个提取温度:环境温度(AT)16小时;或加热(溶剂回流)至少30分钟。
下面总结了所制备的提取物和它们相关的提取产率,用干提取物相对于处于干燥状态且经磨碎的起始植物材料的百分比(wt/wt)表示。
对于如下实施例2的化妆活性试验,为各提取物制备了原液(stocksolution),其中干提取物以每mL溶剂12.5mg提取物的浓度溶解于DMSO中。
实施例2:正常的人角化细胞(NHK)中蛋白Smac/DIABLO的表达水平的免疫酶分析
2.1阳性对照和提取物溶液的制备
毛喉素是已知的、存活蛋白表达的激活剂(专利申请FR2932086的比较实施例1)。
对于下面的试验,将最低包含98wt%毛喉素的毛喉鞘蕊花提取物用作阳性对照。例如,SIGMA出售粉末形式的该提取物。
将该提取物粉末溶解于DMSO中,以获得约10-6M的毛喉素(摩尔质量=410.5)浓度。
将由此制备的阳性对照的溶液添加到培养基中,从而稀释至千分之一,即毛喉素的浓度等于约10-9M。
同时,制备在培养基中稀释到千分之一的溶剂对照(DMSO)。
所测试的各提取物(S-1到S-4、R、G)的原液在角化细胞培养基中稀释,以获得12.5μg/mL的提取物终浓度。
2.2NHK的处理
在无血清的完全培养基(KSFMc,Gibco)中,将所培养的NHK以每孔50,000个细胞的量接种到48孔微孔板中。
该培养的第一天对应于D0。
处理步骤开始于D1,在孵育(37℃,5%CO2)NHK 24小时之后。
去除培养基后,用新的KSFMc培养基进行替换,该培养基包含要测试的提取物或者阳性对照或溶剂对照(T-SO)。每种处理平行进行三次。
处理16小时(对应于细胞周期过程中存活蛋白的表达峰的时间段)后,将半块板上的NHK裂解以进行分析(根据Smac/DIABLO蛋白分析试剂盒(参见下文)中记载的方案,即时制备裂解缓冲液)。另外半块板用PBS冲洗,然后储存在-20℃,从而通过按BCA法(BC分析试剂盒,Uptima Interchim)进行的比色试验,采用570nm处的吸光度测定来分析其中的总蛋白。
2.3分析方法
用Duoset Smac/DIABLO ELISA试剂盒(R&D Systems)评价角化细胞产生的Smac/DIABLO的水平。
添加与发色团(这里是HRP(辣根过氧化物酶))偶联的链霉亲和素,使得能够在过氧化氢的存在下,通过添加HRP的底物(四甲基联苯胺)评价HRP的酶活性。
一旦用硫酸阻断该酶促反应,即通过比色反应显示出Smac/DIABLO的存在(与其浓度成比例)。
吸光度在450nm的波长处测量。
在对试剂盒提供的Smac/DIABLO标准原液进行稀释的基础上,生成标准范围。
2.4NHK裂解物中Smac/DIABLO蛋白的分析结果
结果如图1所示。该结果以每μg总蛋白中的Smac/DIABLO蛋白(pg)表示(pg/μg总蛋白)。
相对于未经处理的基准水平(TSO),计算产生该蛋白的抑制百分比。阳性对照为毛喉素(FSK)。
结论
在降低NHK中的线粒体蛋白Smac/DIABLO的表达方面,所测试的四种本发明的提取物中的每一种都显示出显著的活性(S-1:-39%;S-2:-54%;S-3:-39%;S-4:-29%),所具有的效力可高于阳性对照的效力。
相反,鼓槌石斛的根提取物和皇后兰种的叶提取物对Smac/DIABLO蛋白的表达不具有显著的抑制作用。与DMSO指标组(DMSO indicator)的蛋白浓度相比,鼓槌石斛的根提取物的蛋白浓度差异不超过-9%,而皇后兰种的叶提取物的蛋白浓度差异不超过-13%。
将本发明的提取物(可通过使用极性溶剂的提取方法获得)作为化妆品组合物中的抗老化活性剂进行配制,特别用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或延长细胞或组织的寿命。
实施例3:用于面部的抗老化霜
通过重现实施例1的方法,获得在这一化妆品组合物中用作活性剂的、鼓槌石斛的花的提取物(提取物S-1)。
将干提取物以2%w/w溶解于60/40v/v甘油/水的混合物中。
这一提取物溶液被作为活性剂用于制备下述化妆品组合物(%,以相对于最终组合物的重量表示):

qs:足以溶解A相的化合物的量
B相
C相
氢氧化钠 0.04
水 qs 100
qs 100:足以将最终组合物补至100%的量
将A相的辅剂分散于水中,然后加热到80℃,随后将包括鼓槌石斛提取物的水-乙二醇溶液在内的所有其它化合物溶解。
将B相的化合物加热到85℃形成均相。
在B相中通过Ystral混合器将A相乳化。
最后,通过0.04%w/w的氢氧化钠溶液(C相)将由此获得的水包油乳液中和,然后冷却。
所获得的组合物为用于施用到整个面部或部分面部上的抗老化霜。
Claims (18)
1.一种化妆品组合物,所述组合物包含鼓槌石斛兰的花的至少一种提取物作为活性剂以及至少一种化妆品上可接受的辅剂。
2.如权利要求1所述的化妆品组合物,其特征在于,所述提取物抑制线粒体蛋白Smac/DIABLO的表达和/或活性。
3.如权利要求1或2所述的化妆品组合物,其特征在于,所述提取物通过极性溶剂或极性溶剂混合物获得,所述极性溶剂优选选自:水;C1-C4醇,优选乙醇;C2-C6二醇,优选选自乙二醇、丁二醇和丙二醇;或它们的任意混合物。
4.如权利要求1-3中任一项所述的化妆品组合物,其特征在于,所述提取物通过包含至少50%v/v的乙醇直至100%v/v的乙醇的溶剂获得;如果必要,余量由其它极性溶剂、特别是水组成。
5.如权利要求1-4中任一项所述的化妆品组合物,其特征在于,相对于所述组合物的总重量,所述化妆品组合物包含0.0001wt%-1wt%的干提取物,优选包含0.001wt%-0.01wt%的干提取物。
6.如权利要求1-5中任一项所述的化妆品组合物,其特征在于,所述化妆品组合物包含鼓槌石斛兰的花的提取物与其它化妆品上可接受的活性剂的组合,所述化妆品上可接受的活性剂为经纯化的物质和/或提取物、特别是植物提取物的形式。
7.如权利要求1-6中任一项所述的化妆品组合物,其特征在于,所述化妆品组合物包含:鼓槌石斛兰的其它部分的至少一种提取物;或其它兰花的至少一种提取物;或由这些提取物纯化得到的物质。
8.如权利要求1-7中任一项所述的化妆品组合物,其特征在于,所述化妆品组合物包含:一种或多种刺激存活蛋白的表达和/或活性的其它活性剂,例如毛喉素;或包含这些活性剂的提取物,特别是毛喉鞘蕊花的提取物;或由选自普通念珠藻、甄叔迦树、新绿藻、二形栅藻、姜黄、番红花、奥氏西非苏木、Lepechinia caulescens、Limnophila conferta的植物种获得的植物提取物;以及它们的任意混合物。
9.如权利要求1-7中任一项所述的化妆品组合物,其特征在于,所述可接受的化妆品辅剂选自:色素;着色剂;聚合物;表面活性剂;流变剂;芳香剂;电解质;pH调节剂;抗氧化剂;防腐剂;以及它们的混合物中的任一种。
10.鼓槌石斛兰的花的至少一种提取物在化妆品组合物中作为活性剂的用途,所述活性剂用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命。
11.如权利要求10所述的用途,其特征在于,所述组合物本身用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命。
12.如权利要求10或11所述的用途,其特征在于,所述提取物或所述组合物如权利要求2-8中的任一项所定义。
13.一种化妆护理方法,所述方法特别用于预防或延缓皮肤老化迹象的出现、放慢或减弱皮肤老化迹象的影响、或是延长细胞或组织的寿命;所述方法包括将有效量的、权利要求1-9中任一项所述的化妆品组合物施用到身体或面部皮肤的至少一个部位。
14.如权利要求13所述的化妆品护理方法,其特征在于,将所述化妆品组合物施用至显示出下述迹象的身体或面部的皮肤区域:可视的老化迹象,例如存在皱纹或细小皱纹,或皮肤肤色失去光泽;其它迹象,例如皮肤弹性消失和/或柔性消失、皮肤厚度减小、和/或皮肤干燥度或粗糙度增加。
15.有效量的、鼓槌石斛兰的花的至少一种提取物在制备用于预防或延缓皮肤内在和/或外在老化迹象的出现、或减慢皮肤老化迹象的影响的组合物方面的用途,特别是通过局部施用到个体皮肤的至少一个显示出下述迹象的区域:可视的老化迹象,例如存在皱纹或细小皱纹、或皮肤肤色失去光泽;或其它迹象,例如皮肤的弹性消失和/或柔性消失、皮肤厚度减小、和/或皮肤干燥度或粗糙度增加。
16.如权利要求15所述的用途,其中,所述个体超过35岁。
17.兰花的花的提取物、特别是鼓槌石斛种的兰花的花的提取物,所述提取物抑制线粒体蛋白Smac/DIABLO的表达和/或活性。
18.如权利要求17所述的兰花的花的提取物,其特征在于,所述提取物通过极性溶剂或极性溶剂混合物获得。
