技术领域
本发明涉及一种用于容纳和存储生物材料,特别是DNA材料的容器。 
背景技术
无论是在研究方面还是在其他众多领域内,例如在生物工程、卫生、环境、农业食品、鉴别、法律、犯罪侦查学方面,尤其在完成生物或生物工程样品库方面,DNA都需要长期存储。 
生物材料源自人类、动物或植物,并且主要包括:组织、细胞、比如细菌、真菌、单细胞藻的微生物、病毒、蛋白质和例如DNA、RNA的核酸。 
问题是如何能把这种生物材料存储为避免接触到使其品质下降的成分,如空气中的氧气、水和阳光。同时还要保护这种生物材料使其避免接触到任何污染元素。 
专利EP 1 075 515介绍了一种在密闭的、不氧化的金属密封盒里长期存储DNA的方式,金属盒的实施方式是由两个半球组成。 
DNA在中性气氛中以非常微弱的湿度被封装,以便存储在常温环境中,因而避免了采用致冷设备。 
即使使用这种密封盒的存储方式令人满意,这种方式很难工业化推广。 
事实上,最好能用简便的方法重新利用密封盒内的DNA,甚至能够多次取用DNA。 
另外,从生物角度来说,需要提供一个阶段来实现样品的多重性。 
发明内容
然而,由两个半球形成的密封盒不是最合适的容器。为此,本发明推荐一种用于容纳生物材料,尤其是DNA材料的容器,该容器能够被完全密闭封堵,并可以具有开口而不降低上述存储物的品质,该容器能存储并能多次连续取用重新组成的DNA。另外,必须对每个密封盒设置一个永久的识别标记,还包括对上述容器开口。

具体实施方式
图1示出圆柱形的容器10,最佳的实施方式是直径为7毫米,高度为18毫米,厚度为几个十分之一毫米。 
该容器由一端密闭的壳体12组成,壳体由不透气、防腐并且适宜气密封的材料制成。 
在这种情况下,材料还应该符合一个附加要求,即材料的可变形性及其可塑性,用以实现上述一端密闭的圆柱形容器。 
实际上,为了制作壳体,在工业生产中深度冲压是最恰当的解决办法,一方面可以遵照前面所述的必要的尺寸精度,另一方面还维持了成本与大批量应用相一致。事实上,根据本发明,制造壳体采用的最佳不绣钢牌号为商业上已知的基准304L,或者冶金上已知的牌号Z2CN18-10。 
容器10内增加有同样是圆柱形的插入件14,并且该插入件以轻微摩擦放置到壳体12中。 
优选地,该插入件由玻璃制造,这对插入件容纳生物材料的能力和稳定性极有益处。 
所图示的插入件的高度低于壳体的高度。 
容器10还包括有塞子16,其用来强力堵塞住圆柱形壳体的开口端。 
该塞子16同样是圆柱形,其外部直径几乎相当于以所必需的摩擦强力进入圆柱形壳体内。 
出自同样的理由,塞子如同壳体一样是优选通过冲压式的机械变形获得的。 
该圆柱形塞子的高度很小,约为几毫米,在圆柱形壳体大致18毫米时它是3毫米。塞子的厚度同样是几个十分之一毫米,厚度最好等于或非常接近壳体的厚度,这里为0.25毫米,如将要进一步描述的,这是出于对焊接系数的考虑。 
塞子与壳体由同种材料制成,从而最终获得的容器壳体是单一材质的整体结构。 
图2上可以看出已经接收塞子的壳体呈现一种几何结构,即壳体和塞子的开口周边边缘是并列的。 
有利地,壳体和塞子在端部具有由冲压变形所形成的斜缘或外环槽,能够使塞子非常顺利地进入壳体。 
依照本发明的改进,可以将塞子设计得略带一点锥度,使其进入壳体内后不出现移动。 
图2简略示出了置放在插入件14内的生物材料18,该生物材料处于脱水状态。 
有利地,在容纳和存储上述生物材料的密闭容器内预先引入保护性气氛。 
图3示出最终以气密方式密封的容器。 
一种适当的方式是把塞子在壳体上焊接20。 
根据本发明,这种焊接方式是有益处的,事实上,在实施焊接之前生物材料已经被引入,以避免任何对该生物材料品质的降低或任何对该生物材料的损坏,必要时要限制甚至要避免任何加温现象。 
实际上本发明的方案在塞子和壳体的两个边缘上实施周边激光焊接。 
这种焊接不需要任何焊料,不改变材料的结构,材料一直保持着全部初始性质。 
两种材料有相同的或非常近似的厚度同样有好处,在焊接中能获得一致的效果。 
鉴于壳体管壁非常薄的厚度,该激光焊接需要非常小的功率,以使管壁的温度没有任何明显的提升和/或在管腔内及任何情况下,没有任何引起生物材料的变化和/或品质下降的可能性。 
采用YAG脉冲式激光的优势还在于温度的提升可以忽略不计。 
同样应该指出,焊接的唯一作用是密封并且不需要很强的机械连接强度就阻止了塞子在壳体内的移动。 
图4示出标记工序,该工序一般在引入生物材料之前就已完成,以用于流程追踪。 
另外,该标记应该与容器的寿命相等,要避免使用一些极易快速受损的相关物质,例如油漆标记、标签标记、印刷标记等。 
根据本发明,容器上的标记是通过形成标记22来完成。这个标记本身可以包含任何类型的数字参数、字母或条形码,或者还可用数据矩阵类型的矩阵码。 
有利地,通过用适当的激光束作用在材料上以改变材料表面状态的方法来做标记,这样,材料没有被刻出凹槽,而标记却是永久的。 
在此还可以得知,通过激光束来制作标记对容器产生一种完全可以忽略不计的能量消耗,这也包括对容器底部做标记的情况。 
另一种方式是采用一种RFID式的标签,即一种利用超高频的识别标签。在这种情况下,标签的作用是将一根天线机械地敷设和维持在容器的壁面上,该天线为有源元件。标签本身没有印记,从发明的角度把这种方式认定为永久性的。标签表面的任何损坏对有源识别元件不产生任何影响。 
图5示出的容器允许使用被容纳和存储的生物材料。 
这样,在塞子的底部预置一个入口24,尤其可以通过一个诸如菱形的钻孔工具进行钻孔切割。这个菱形的钻孔工具被标记定位在中央位置。 
鉴于塞子底部的厚度很薄,施加的压力要非常轻。 
还可以发现冲孔可以形成一个多翼片的切口,在材料形状记忆的作用下,这些翼片被卷压形成无凹凸的边缘,因此便于进入内部的入口,尤其便于移出那些通过上述入口进入内部的装置。事实上,滴管及其它的导管被支撑在翼片的形成线上。
生物材料尤其不会受到金属粒子的污染,因为没有机加工,只有对材料的冲压切割。 
同时还可以发现在这种操作中,由于插入件处于边缘部位,它也不会受到机械破坏。 
由于容器没有承受任何机械作用,只有塞子底部被菱形的钻头钻孔,标记同样保持完全清晰可读。 
因此,可以在这个入口24加上滴管、灌注器或类似的装置来添加任何适当液体,使脱水的生物材料置于溶液中。 
这种处于悬浮的生物材料可以根据需要通过相同的装置被全部或部分提取。 
当然,开口是最终性的,容器不再能进行非常长期的存储。 
然而,生物材料可以只被部分利用,并且为了不同的操作还需要存储数日。 
在这种情况下,容器还可以用来存储已经置于溶液中的生物材料,同时要特别注意在容器上安置一个堵塞头26,该堵塞头比如可由中性弹性材料制成。 
这个堵塞头可以是帽型,如图5所示,也可以是圆锥台形的,它能直接插入到塞子内并被径向弹力维持在这个位置上。 
应该指出,标记与容器息息相关,对开口的容器做记号是为了再次利用被打开容器的材料时不发生任何错误。 
还可以发现圆柱形容器也非常适应于自动化装置的操作,以用来定位在生物实验中常用的微片试验台上,尤其是众所周知的标准注册型号台SBS。 
同样,光学阅读机很容易识别刻制的标记,即便该标记有可能受损变质,即使比方在贴标的情况下,阅读机也不会出现阅读错误。 
所以,根据本发明,通过利用本容器,可以安排生物材料样品的可追踪性。 
 容器能具有在室温条件下存储的所有优点,主要是便捷了分析仪之间、实验室之间的交换,完成了生物工程数据,不仅如此,还容许使用自动化装置来完成将生物材料装入上述容器里的工作。 
这些阶段至少包括脱水工序、引入保护性气氛工序、焊接密封工序和标记工序。 
同样地,冲孔开口工作可以使用带有冲头的装置来实施,这样能调整行程并确保良好的引导。 
包括现有的自动化装置在内的所有自动分析装置都可以装配上这样的一种冲孔开口装置,避免了任何人工干预。 
图示中的插入件由玻璃制成,但也可以在这里设计置放一个由陶瓷制作的插入件,或任何其他已知或未知的并适宜存放生物材料的惰性材料制作的插入件。 
在某些应用中,插入件同样可以由相对于生物材料为惰性材料的球状物所替代,球状物事先已经吸附有这些生物材料。 
容器被设计用来存储室温的生物材料,但是对于某些生物材料有必要确保在低于或等于-20℃的温度中存储。根据本发明,容器内的存储温度可以降低至正几度。 

Claims (9)

1.一种用于在保护性气氛下,在室温中存储脱水性生物材料的容器(10),所述容器包括由气密材料制造的壳体(12),其特征在于:所述壳体(12)由一端密闭的圆柱形金属材料制成,并包括塞子(16),所述塞子用来和所述壳体气密封式连接,所述壳体(12)和所述塞子(16)的每一个都是由机械变形获得的整体结构。
2.根据权利要求1所述的用于在室温中存储脱水性生物材料的容器(10),其特征在于:所述塞子(16)由一端密闭的圆柱形金属材料制成,并且被设计为嵌套在圆柱形的所述壳体(12)内。
3.根据权利要求2所述的用于在室温中存储脱水性生物材料的容器(10),其特征在于:连接工序通过焊接(20)所述壳体(12)和所述塞子(16)的外部边缘来实施。
4.根据权利要求3所述的用于在室温中存储脱水性生物材料的容器(10),其特征在于:所述焊接(20)通过激光束来实施。
5.根据权利要求4所述的用于在室温中存储脱水性生物材料的容器(10),其特征在于:所述容器的直径为7毫米,高度为18毫米,厚度为几个百分之一毫米,以使其能被置放在孔板的孔内。
6.根据权利要求5所述的用于在室温中存储脱水性生物材料的容器(10),其特征在于:所述金属材料为不锈钢Z2CN18-10。
