具体实施方式
结合附图,通过对本发明较佳实施例的描述具体说明本发明。
【实施例1】表达新型抗生素的质粒的构建和新型抗生素制备
原始质粒为装载了大肠菌素变构多肽Ia’和immunity蛋白基因的pSELECTTM-1质粒(8.3kb)(购于Promega公司)。经双链寡聚核苷酸点突变技术(QuickChangeTM Kit,Strategene公司)将编码葡萄球菌信息素基因序列如:SEQ ID NO 1、3、5、7、9所示,分别插入到大肠菌素变构多肽Ia′基因的626号氨基酸位置之后,获得制备新型抗生素的一系列重组质粒,pBHC-SA1、pBHC-SA2、pBHC-SA3pBHC-SA4、pBHC-SE,如图1~图5所示,以及金黄色葡萄球菌信息素AgrDI的基因序列如SEQ ID NO 1所示连接在变构多肽Ia′基因的第1号氨基酸位置前而获得重组质粒pBHC-PA如图6所示。重组质粒转染入E.coli BL-21工程菌制备新型抗生素。
突变程序按Strategene QuickChange SiteDirected Mutagenesis Kit(catalog#200518)药箱手册进行:
1.准备点突变反应物:
5ul 10Xbuffer
2ul(10ng)装载了大肠菌素变构多肽和immunity蛋白基因的原始质粒pSELECTTM-1。
1.25ul(125ng)设计的5’-3’寡聚核苷酸引物(见SEQ ID NO24~34偶数序列)
1.25ul(125ng)设计的3’-5’寡聚核苷酸引物(见SEQ ID NO25~35奇数序列)
1ul dNTP
双蒸水50ul
1ul pfu
(除质粒、引物和双蒸水外,均为药箱所备试剂)
2.进行PCR扩增,扩增条件:变性95℃,35秒,退火53℃,70秒,延伸68℃,17分,共20个循环;
3.加入Dpn 1内切酶1ul消化母体DNA链后(37℃,1小时),取1ul反应物与XL1-Blue感受态细胞50ul冰孵30分钟,热冲击42℃,45秒,再置入冰中2分钟;
4.加入NZY培基0.5ml,220rpm,37℃摇菌1小时,取50-100ul反应物铺板(LB培基加1%琼脂,加50ug/ml氨苄青霉素,37℃过夜);
5.18小时后挑菌,提取质粒后测序确定突变成功;
6.将突变质粒100ng与制备的BL-21工程菌感受态细胞40ul冰孵5分钟,热冲击42℃,30秒,再置入冰中2分钟,加入SOC培基160ul,220rpm,37℃摇菌1小时后铺板(LB培基加1%琼脂,加50ug/ml氨苄青霉素,37℃过夜),挑取单克隆菌落大量增菌;
7.大量增菌,8-10升FB培基,250rpm,37℃,3-4小时;加入IPTG ,250rpm,28℃再生长4小时;4℃,6000g,20分钟离心沉淀菌体,取4℃,50mM硼酸缓冲液(pH9.0,2mMEDTA)80-100ml悬浮菌体,加入PMSF 50ug后超声破碎菌体(4℃,400W,1分钟,重复4-5次,间歇2-3分钟确保菌液温度),高速离心沉淀破碎的菌体(4℃,75,000g,90分钟),取上清加入硫酸链霉素500万单位沉淀DNA(4℃搅拌1小时),10000g,4℃,10分钟离心沉淀后,取上清装入分子量15,000透析袋于4℃,50mM硼酸缓冲液10升透析过夜后,再次10,000g,4℃,10分钟离心沉淀,取上清上样于CM离子交换柱,充分冲洗后,0.3M NaCl+50mM硼酸缓冲液洗脱即可得到所制备的新型抗生素。对应于以上6种重组质粒,可分别获得PMC-SA1、PMC-SA2、PMC-SA3、PMC-SA4、PMC-SE以及PMC-PA六种新型抗生素。
制备上述6种突变质粒的信息素基因所设计的寡聚核苷酸序列分别如下:

【实施例2】新型抗生素对青霉素敏感金葡菌的抑制作用
细菌为美国标准菌株,ATCC 25923青霉素敏感金葡菌,菌液2微升(105CFU/ml)加入1%胰蛋白胨,1%NaCl,0.5%酵母,0.5%葡萄糖,0.1%K2HPO4的培养液10毫升中,共准备6组,第一组加入0.3M NaCl+50mM硼酸缓冲液(即新型抗生素、野生型大肠菌素Ia和无质粒的BL-21工程菌所产蛋白的空白保存液,量与实验组中加入的新型抗生素液体量相同)作为对照,第二组加入100ng/ml青霉素G钠,第三组加入100ng/ml野生型大肠菌素Ia,第四组加入100ng/ml无质粒的BL-21工程菌所产蛋白,第五组加入100ng/ml大肠菌素Ia羧基端连接其它8肽的重组蛋白,第六组加入100ng/ml PMC-SA1新型抗生素。
上述各组液体分别置于100毫升三角烧瓶中,200rpm,37℃生长,每小时采样100微升加入96孔酶表板中经分光光度计(A 595nm)比色测试细菌生长浊度,画出细菌生长曲线来比较新型抗生素的抑菌效力,结果如图8所示,显示青霉素敏感金葡菌只能被青霉素和PMC-SA1所抑制。
【实施例3】新型抗生素PMC-SA1对耐甲氧西林金葡菌(ATCC BAA-42)杀菌效果的透射电镜观察(1%磷钨酸染色,放大25,000倍)。
培养液:1%胰蛋白胨,1%NaCl,0.5%酵母,0.5%葡萄糖,0.1%K2HPO4。
Con:对照组,菌加入前述培养液中和适量0.3M NaCl+50mM硼酸缓冲液中200rpm,37℃生长2小时后,菌形态仍然正常;
Oxa:菌加入前述培养液和苯唑西林500ug/ml中200rpm,37℃生长1.5小时后,菌的颜色和形态有变化,但菌体仍完整;
PMC:菌加入前述培养液中和PMC-SA1 10ug/ml处理半小时后,菌破裂,内容物泄漏。
如图9所示,新型抗生素与青霉素的杀菌机制完全不同,500ug/ml苯唑西林可损伤耐甲氧西林金葡菌,而10ug/ml PMC-SA1可有效杀伤耐甲氧西林金葡菌,苯唑西林分子(分子量450左右)和PMC-SA1分子(分子量70,000左右)的分子量相差160倍,本实施例中苯唑西林和PMC-SA1的使用浓度相差50倍,因此PMC-SA1在本实施例中表现出来的抗菌效力比苯唑西林强八千倍左右。
【实施例4】新型抗生素PMC-SA1的体外抗菌活性
一、实验材料
(1)药物
●PMC-SA1:规格:1.5mg/ml
●注射用头孢唑林钠:规格:0.5g/瓶,华北制药股份有限公司生产。
●注射用青霉素钠:规格:80万单位/瓶,华北制药股份有限公司生产。
●注射用氨苄西林钠:规格:1g/瓶,华北制药股份有限公司生产。
●注射用苯唑西林钠:规格:0.5g/瓶,华北制药股份有限生产。
●万古霉素:规格:2.5mg/ml,
●以上样品用无菌水溶解稀释,药物终浓度为128mg/L、64mg/L、32mg/L、16mg/L、8mg/L、4mg/L、2mg/L、1mg/L、0.5mg/L、0.25mg/L、0.125mg/L、0.06mg/L、0.03mg/L、0.015mg/L。
(2)细菌
临床分离菌株:所有菌种在收集分离的单位(华西医科大学检验科细菌室)均经鉴定。
金黄色葡萄菌28株(MRSA菌株10株,MSSA菌株18株)、表皮葡萄球菌10株(MRSA菌株5株,MSSE菌株5株)、肠球菌5株、大肠杆菌10株、鲍曼不动杆菌10株,共63株。
标准质控菌株:青霉素敏感金黄色葡萄球菌ATCC 25923、耐青霉素金黄色葡萄球菌ATCC29213,耐甲氧西林金黄色葡萄球菌ATCC BAA-42,耐万古霉素肠球菌ATCC 700802。
(3)培养基
●MH肉汤培养基:称取25g加100ml蒸馏水,加热溶解,分装,高压灭菌,116℃,20分钟。MH固体培养基:称取36g,加1000m蒸馏水,高压灭菌,116℃,20分钟,用于革兰氏阳性、阴性需氧菌的药敏试验。
●肠球菌用血培养基,即在MH培养基中加5-10%脱纤维兔血配置而成,5%CO2中37℃培养24小时。
二、试验方法
1、最低抑菌浓度MIC的测定
采用琼脂二倍稀释法测定PMC-SA1的最低抑菌浓度(MIC)。用多点接种仪(Deneley A400)将细菌接种于含不同药物浓度的琼脂平皿表面,每点含菌量为105CFU/ml,37℃孵育18-24小时观察结果,以无细菌生长平皿培养基中所含药物的最低浓度为药物对该菌的最低抑菌浓度(MIC值)。
三、结果

【实施例5】新型抗生素的对金葡菌、肠球菌和大肠杆菌感染动物体内保护试验
一.实验材料
(1).药物
使用药物同【实施例4】。
以上样品用无菌水溶解稀释,药物注射浓度为10mg/kg、5mg/kg、2.5mg/kg。
(2)细菌
耐甲氧西林金葡菌ATCC BAA-42、耐青霉素金葡菌ATCC 29213、耐万古霉素肠球菌ATCC700802、大肠杆菌(临床分离氨苄青霉素敏感株,32033)。
二.实验方法
昆明小鼠340只,雌雄各半,体重15~20克,随机分组为腹腔分别注射金黄色葡萄球菌ATCC BAA-42、ATCC29213、肠球菌ATCC 700802和大肠杆菌(临床分离氨苄青霉素敏感株,32033)四个大组,其中PMC-SA1、青霉素、氨苄西林、头孢唑林和万古霉素实验组分别为每组小鼠5只,对照组每组小鼠10只。腹腔注射致死剂量的细菌后,各药物试验组小鼠分别自静脉注射10mg、5mg、2.5mg/kg剂量的药物一次,每24小时观察结果,连续观察7-14天,以小鼠死亡为阳性结果。
三.结果
5mg、3mg和1mg药物剂量组结果显示,PMC-SA1组小鼠存活率在3个剂量组中与万古霉素组小鼠存活率始终保持了恒定的差距,因此在体内试验中PMC-SA1对抗耐甲氧西林金黄色葡萄球菌感染的治疗效果远优于万古霉素。这可能是PMC-SA1既杀菌又抑制细菌毒素分泌的双重效果所致。因而该药在临床应用时可能具有其他抗生素无法比拟的治疗效果,其他各抗生素小鼠死亡率>80%,已不具备可比性;对照组于48小时内全部死亡。

由实施例4和5的表1和表2结果可以看出:
1.金葡球菌(MRSA)体外抗菌活性比较:苯唑西林MIC90测定值比PMC-SA1测定值大8倍。PMC-SA1分子量(70,0000)是万古霉素分子量(1,400)的49倍,苯唑西林分子量(450)的155倍。按单位体积内相同药物分子数标定,则PMC-SA1对金葡球菌(MRSA)体外抗菌活性相当于万古霉素对MRSA体外抗菌活性的3倍,相当于苯唑西林对MRSA体外抗菌活性的1,240倍。
2.PMC-SA1感染小鼠体内保护实验中,最小剂药量(2.5mg/kg)条件下PMC-SA1组的死亡率最低(40%),其他各组死亡率分别为青霉素组60%,氨苄西林组100%,万古霉素60%,对照组100%。该结果显示PMC-SA1的保护率最高(60%);青霉素和万古霉素的保护率次之(40%)。
【实施例6】新型抗生素对表皮葡萄球菌生长的影响
细菌为购自中国菌种保存中心(国家医药管理局生物制品检定所,北京天坛)的表皮葡萄球菌标准菌株,编号26069,菌液3微升(106CFU/ml级菌量)加入1%蛋白胨,1%NaCl,0.5%酵母。0.5%葡萄糖,0.1%K2HPO4的培养液10ml中,共准备4组,第一组加入0.3MNaCl+50mM硼酸缓冲液(量与实验组中抗菌多肽液体量相同)作为对照,第二组加入2μg/ml青霉素G钠,余各实验组加入2μg/ml PMC-SA1和PMC-SE(保存液为0.3M NaCl+50mM硼酸缓冲液)。
上述各组液体置于100ml三角烧瓶中,200rpm,37℃生长,每小时采样100μl加入96孔酶标板中经分光光度计(A 595nm)比色测定细菌生长浊度,画出细菌生长曲线来比较抗菌多肽的抑菌效力,结果见图10,由图中可见,表皮葡萄球菌的生长可被抗金葡多肽部分抑制,可被抗表葡多肽有效的抑制。
【实施例7】几种新型抗生素抑菌效果的比较
以下试验比较了本发明制备的6种抗菌多肽的抑菌活性,细菌为美国标准菌株,耐甲氧西林金黄色葡萄球菌ATCC BAA-42,菌液5微升(106CFU/ml级菌量)加入1%蛋白胨,1%NaCl,0.5%酵母。0.5%葡萄糖,1%K2HPO4的培养液10ml中,共准备8组,第一组加入0.3MnaCl+50mM硼酸缓冲液(量与实验组中抗菌多肽液体量相同)作为对照,第二组加入5μg/ml青霉素G钠,余各组加入PMC-SA1、PMC-SA2、PMC-SA3、PMC-SA4、PMC-SE和PMC-PA(保存液为0.3M NaCl+50mM硼酸缓冲液)。
上述各组液体置于100ml三角烧瓶中,200rpm,37℃生长,每小时采样100μl加入96孔酶标板中经分光光度计(A 595nm)比色测试细菌生长浊度,画出细菌生长曲线来比较抗菌多肽的抑菌能力,结果见图11。
结果显示,耐甲氧西林金葡菌的生长几乎不能被苯唑西林抑制,PMC-PA的抑菌效果最差,抑制效果最好的是PMC-SA1,其次是PMC-SE和PMC-SA4,PMC-SA2和PMC-SA3再次之。
【实施例8】新型抗生素对绿脓杆菌生长的影响
细菌为购自中国微生物菌种保藏管理委员会普通微生物中心的绿脓杆菌标准菌株(ATCC27853),菌液5微升(106CFU/ml级菌量)加入MH培养液10ml中,共准备7组,第一组加入0.3MNaCl+50mM硼酸缓冲液(量与实验组中抗菌多肽液体量相同)作为对照,第二组加入5μg/ml野生型大肠菌素Ia,第三组加入5μg/ml PMC-SA1,第四组加入5μg/mlPMC-PA。
上述各组液体置于100ml三角烧瓶中,200rpm,37℃生长,每小时采样100μl加入96孔酶标板中经分光光度计(A 595nm)比色测定细菌生长浊度,画出细菌生长曲线来比较抗菌多肽的抑菌效力,结果见图12。
由图中可见,实验中绿脓杆菌的生长仅被PMC-PA完全抑制,而野生型大肠菌素Ia和PMC-SA1均不能有效抑制绿脓杆菌的生长。
【实施例9】新型抗生素的多重耐药绿脓杆菌感染动物体内保护试验
一.实验材料
(1)药物
PMC-PA、哌拉西林、阿米卡星、头孢他啶、左氧氟沙星。
(2)细菌
多重耐药绿脓杆菌(临床分离株13280,华西医科大学检验科细菌室)。
二.实验方法
昆明小鼠60只,雌雄各半,体重15~20克,随机分组,腹腔注射多重耐药绿脓杆菌,其中PMC-PA、哌拉西林、阿米卡星、头孢他啶和左氧氟沙星实验组分别为每组小鼠10只,对照组每组小鼠10只。腹腔注射致死剂量的细菌后,各药物试验组小鼠分别自静脉注射上述药物一次,每24小时观察结果,连续10天,以小鼠死亡为阳性结果。
三.结果
如图13所示小鼠生存曲线,腹腔注射致死剂量多重耐药绿脓杆菌后,(1),对照组在2天内全部死亡,(2),哌拉西林50mg/kg组在3天内全部死亡,(3),阿米卡星15mg/kg组在4天内全部死亡,(4),头孢他啶50mg/kg组的10天存活率为20%,(5),左氧氟沙星5mg/kg组的10天存活率为40%,(6)PMC-PA 5mg/kg组的10天存活率为100%。
结果显示,对多重耐药绿脓杆菌致死性感染,本发明的新型抗生素PMC-PA表现出了所试现用抗生素无法比拟的抗菌活性。
【实施例10】大肠菌素变构多肽的免疫效果观察
取大肠菌素Ia变构多肽、实施例1中PMC-SA1及野生型大肠菌素Ia、发明人前期发明的抗金葡菌多肽(ZL01128836.1)各一份免疫小鼠:上述各蛋白与佐剂混合后,基础量和追加量为腹腔注射每只50μg(0.5ml)1次。间隔两周,免疫5针。ELISA间接法检测小鼠血清效价(肖毅等,抗金葡菌多肽单克隆抗体制备及初步测定,陕西医学杂志,35(1):6-7,32(2006))。野生型大肠菌素Ia以及抗金葡菌多肽(ZL01128836.1)所免疫小鼠血清效价为10-4至10-5,而大肠菌素Ia变构多肽以及本发明PMC-SA1所免疫小鼠血清效价为10-2至10-3。
因此大肠菌素Ia变构多肽以及本发明的新型抗生素PMC-SA1造成宿主致敏反应的可能性要比野生型大肠菌素Ia以及抗金葡菌多肽造成宿主致敏反应的可能性低一至两个数量级。
