技术领域
本发明涉及一种复合物的冷冻干燥体,其用于向细胞中导入核酸、寡核酸或其衍生物,所述复合物含有核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物;其制备方法和含有所述冷冻干燥体的核酸、寡核酸或其衍生物的导入用制剂、试剂和试剂盒。
背景技术
现在,通过向细胞中导入目的基因、反义寡核酸、其衍生物并表现该基因或功能,由此治疗先天性遗传病、癌细胞或AIDS等的基因疗法、反义疗法正得到实用化,与此同时,还正研究作为用于向细胞中导入基因(DNA)、反义寡核酸、其衍生物的携带者即各种载体。
正在研究这种载体的其中一个、即一种安全性方面没有问题、效率高、无免疫原性且容易制备的非病毒性载体,其为阳离子性聚合物、阳离子性脂质体、阳离子性脂质等阳离子性物质。
在使用这些阳离子性物质的方法中,由于DNA和阳离子性物质的复合物带正电荷,因此由于与血细胞和白蛋白等血液成分相互作用而凝集,从而成为导入细胞的障碍。为了解决这个问题,现在正对下述方法进行研究:将核酸和阳离子性聚合物的复合物用透明质酸衍生物包覆(专利文献1:日本特开2005-176830号);或者用具有含有羧基的侧链和含有糖残基的侧链的聚乙二醇(PEG)包覆(专利文献2:日本特开2003-231748号);或者用具有游离羧酸侧基的PEG来防止复合物的凝集(非专利文献1:J.Biomater.Sci.Polymer Edn.,Vol.14,No.6,pp.515-531(2003))这样的方法。
这样制备的DNA与阳离子性物质的复合物呈现出低凝集性,而且在细胞中的基因表达也良好。但是,这种复合物由于是不均匀的悬浮液,因此保存性低,需要在制备后立刻使用,而且若以高浓度制备,就会发生凝集,因此存在浓度很难调整,处理也复杂这样的缺点。而且,制备方面再现性也不好。
另一方面,在导入基因等物质时使用上述这样的DNA与阳离子性物质的复合物时,对复合物大小(粒径)的控制也很重要。这是因为给药到组织内或血流中时,随后的复合物的扩散、向细胞的传递、吸收的效率对其药理效果有很大影响。但是,一般来说,通过混合阳离子性聚合物这样的离子性高分子进行复合物化时,容易导致高分子的凝集,形成非常大的颗粒或纤维状的复合物。为了防止这样的情况,需要将进行混合的溶液的浓度调整到极低。但是,由于用于基因导入等的制剂需要某种程度以上的浓度,因此存在无法避免形成大的凝集块这样的问题。而且,还考虑了在暂时混合稀的溶液、形成小复合物后,进行浓缩的方法,但由于复合物颗粒迅速凝集,因此并不是适当的浓缩手段。
因此,为了解决这些问题,还研究了可以容易地输送生物制剂并提高储藏稳定性的一般方法即冷冻干燥法。但是,在对核酸、寡核酸或其衍生物和聚阳离子性物质的复合物进行冷冻干燥而得的物质中,确认了由于冷冻干燥而破坏了复合物的结构,因此即使用于基因导入和寡核酸的导入中,也几乎不会表现出作为基因或反义的功能(非专利文献2:J.Pharm.Sci.,vol90,pp1445-1455(2001))。
作为解决这些问题的方法,提出了添加高浓度的单糖和二糖等再进行冷冻干燥的方法(非专利文献3:Biochim BiophysActa.2000 Sep 29;1468(1-2):127-138.)。但是,这里所需要的糖的量,按重量比为DNA的500倍至1000倍,我们认为再水化后的溶液是比生理条件高出许多的高渗透压,因此并不实用。而且,单糖和二糖自身也没有对基因表达有利的效果。而且,为了减轻再水化后的渗透压,尝试利用中性多糖中的葡聚糖(非专利文献4:J.Pharm.Sci.,vol94,pp1226-1236(2005))。但是,高分子量的葡聚糖极大地抑制基因表达,而且在使用低分子量(分子量3000左右)的葡聚糖时,为了防止由于冷冻干燥而导致的凝集,需要按重量比添加DNA的100倍以上的、相当高浓度的葡聚糖(非专利文献4:J.Pharm.Sci.,vol94,pp1226-1236(2005))。这种冷冻干燥体在体内利用时,为了获得需要的DNA浓度,需要在冷冻干燥后用少量的水或溶剂再水化,进而再浓缩到高浓度。如果这样处理的话,则再水化后的葡聚糖浓度会超过10%,而且冷冻干燥时的DNA浓度、冷却温度等受到限制,在其实用方面的应用也困难。

发明内容
发明要解决的问题
本申请的发明人为了要解决上述缺点,进行了积极的研究,结果发现,用包含核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物的复合物的冷冻干燥体向细胞进行导入,被导入的基因或寡核酸或其衍生物良好地表现其功能,从而完成本发明。
用于解决问题的方法
本发明涉及一种复合物的冷冻干燥体,所述复合物含有核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物。而且还涉及含有该冷冻干燥体的核酸、寡核酸或其衍生物的导入用制剂和试剂以及试剂盒。而且,还涉及该冷冻干燥体的制备方法,该方法包括通过混合核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物,以形成复合物的工序;以及随后进行冷冻干燥的工序。本发明还涉及向细胞中导入基因、寡核酸或其衍生物的方法,所述方法使用该冷冻干燥体。
发明的效果
本发明的冷冻干燥体容易调节浓度,处理简便,保存性好。而且,本发明的冷冻干燥体由于含有阴离子性聚合物,因此即使在使用时通过用溶剂再水化而形成悬浮液或稀释液时,也可以获得任意浓度的含有极微小尺寸的复合物的稳定的分散体。而且,在导入基因时,不引起凝集,可向细胞有效导入核酸、寡核酸或其衍生物,而且由局部给药,静脉内给药等各种给药法表现出良好的功能表现能力。
用于实施发明的最佳方式
本发明的冷冻干燥体是含有核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物的复合物的冷冻干燥体。本复合物中,核酸、寡核酸或其衍生物与阳离子性聚合物或阳离子性脂质或含有其的集合体通过离子键形成复合物,阳离子性聚合物或阳离子性脂质再与阴离子性聚合物形成离子键。它们根据混合比、混合顺序等可以形成主要由阴离子性聚合物覆盖的复合物。
作为可以在本发明的冷冻干燥体中使用的核酸、寡核酸或其衍生物,可以使用通过基因治疗或反义治疗导入的任意的核酸或寡核酸等,具体可以例举各种DNA和RNA(单链和双链)(质粒DNA、双链寡RNA、mRNA、tRNA、rRNA、cDNA等);正义或反义寡核苷酸(包括重组体)和其衍生物;核糖核酸酶或它们的混合物等各种核酸、寡核酸或其衍生物。而且,这些核酸的碱基部分和糖部分如果需要可以进行修饰或取代。为核酸的话,可以优选使用质粒DNA,为反义核酸的话,可以优选使用寡DNA、其衍生物硫代寡聚物(S-oligo)、RNA干涉用双链RNA、核糖核酸酶RNA等,其中尤为优选使用质粒DNA。
作为可以在本发明的冷冻干燥体中使用的阳离子性聚合物,可以使用1个分子中具有多个、优选5个以上能在水中与DNA形成复合物的官能团且带正电荷的分子量为1000~300万左右的天然来源或合成的高分子,作为这种官能团,可以例举例如任选取代的氨基或铵基或其盐(这些基团可以被例如碳原子数为1~6的烷基、苯基等进行单取代或多取代)、亚氨基、咪唑基、胍基等有机氨基。作为这种阳离子性聚合物,可以例举例如、带正电荷的蛋白质或多肽;带正电荷的树枝状聚合物(Dendrimer);带正电荷的合成聚合物;和带正电荷的多糖类衍生物、或它们的盐、以及上述物质的组合。
作为可以在本发明的冷冻干燥体中作为阳离子性聚合物使用的带正电荷的蛋白质、带正电荷的多肽的分子量优选为1000~50万左右。作为这种蛋白质、多肽,具体可以例示鱼精蛋白、组蛋白、HelΔ1、明胶等蛋白质和多肽,另外还可以例示含有带正电荷的氨基酸残基的聚氨基酸。作为这种含有带正电荷的氨基酸残基的聚氨基酸,具体可以例示聚-L-赖氨酸、聚精氨酸、聚鸟氨酸等。作为这些蛋白质和多肽的盐,可以例举盐酸盐、硫酸盐、磷酸盐、硼酸盐等。
可以作为阳离子性聚合物使用的具有上述官能团的带正电荷的树枝状聚合物(Dendrimer)是在支化的分子链的末端或内部具有任选取代的氨基或铵基或其盐(这些基团可以被例如碳原子数为1~6的烷基、苯基等进行单取代或多取代)的树枝状聚合物,其分子量优选为1000~50万左右。作为树枝状聚合物,具体可以例举聚酰胺胺树枝状聚合物、聚赖氨酸树枝状聚合物等。而且,作为树枝状聚合物的盐,可以例举盐酸盐、硫酸盐、磷酸盐、硼酸盐等。
可以作为阳离子性聚合物使用的带正电荷的合成聚合物是上述那样的1个分子中具有多个、优选5个以上能在水中与DNA形成复合物的官能团的合成聚合物,其分子量优选为1000~300万。作为合成聚合物,具体可以例举聚乙烯亚胺(包括直链状聚乙烯亚胺、或聚支化型乙烯亚胺)、甲基丙烯酸2-二甲胺基乙酯的聚合物或共聚物、甲基丙烯酸2-三甲氨基乙酯的聚合物或共聚物等。合成聚合物的其中一例聚乙烯亚胺的分子量优选为1000~50万左右,更优选5000~20万左右,最优选为1万~10万左右。而且,作为聚乙烯亚胺的盐,可以例举盐酸盐、硫酸盐、磷酸盐、硼酸盐等。
可以作为阳离子性聚合物使用的带正电荷的多糖类衍生物为1个分子中具有多个、优选5个以上能在水中与DNA形成复合物的官能团,且分子量优选为1000~300万,更优选5000~500000的多糖类衍生物。作为这种多糖类,具体可以例举壳聚糖、导入了上述官能团的葡聚糖衍生物等。这些物质中,壳聚糖的分子量优选为1000~50万左右,更优选5000~20万左右,最优选为1万~10万左右。作为壳聚糖的盐,可以例举盐酸盐、醋酸盐等。而且,葡聚糖衍生物的分子量优选为3000~100万。作为这种葡聚糖衍生物,具体可以例举二乙氨乙基-葡聚糖等。
上述的阳离子性聚合物,即使是通常不带正电荷的,只要通过导入氨基等官能团而带正电荷,就可以使用,而且根据需要还可以再用糖链、寡肽、抗体等进行修饰。
作为可以在本发明的冷冻干燥体中使用的阳离子性脂质(包括阳离子性胆固醇衍生物),可以例举DC-Chol(3β-(N-(N′,N′-二甲氨基乙烷)氨甲酰基)胆固醇)、DDAB(N,N-二硬脂基-N,N-二甲基溴化铵)、DMRI(N-(1,2-二肉豆蔻基氧丙-3-基)-N,N-二甲基-N-羟乙基溴化铵)、DODAC(N,N-二油酰基-N,N-二甲基氯化铵)、DOGS(双十七烷基酰胺基甘氨酰亚精胺)、DOSPA(N-(1-(2,3-二油酰氧基)丙基)-N-(2-(精胺甲酰胺)乙基)-N,N-二甲基三氟醋酸铵)、DOTAP(N-(1-(2,3-二油酰氧基)丙基)-N,N,N-三甲基氯化铵)、或DOTMA(N-(1-(2,3-二油酰氧基)丙基)-N,N,N-三甲基氯化铵)、以及上述物质的组合。
而且,作为含有阳离子性脂质的集合体,可以使用混合了上述阳离子性脂质(例如DOSPA)和例如DOPE(二油酰磷脂酰乙醇胺)、胆固醇等中性物质的物质。例如,作为含有阳离子性脂质的集合体,优选可以例举脂质体Lipofectamine(上述DOSPA和DOPE的3:1w/w混合体脂质体)、脂质体Lipofectin(上述DOTMA和DOPE的1:1w/w混合体脂质体)、或它们的混合物等。
在本发明的冷冻干燥体中,作为阳离子性聚合物,优选可以使用聚乙烯亚胺;鱼精蛋白;HelΔ1;聚酰胺胺树枝状聚合物(Dendrimer)、聚赖氨酸树枝状聚合物等树枝状聚合物;壳聚糖;甲基丙烯酸2-二甲胺基乙酯的聚合物或共聚物;甲基丙烯酸2-三甲氨基乙酯的聚合物或共聚物等,特别优选使用聚乙烯亚胺、聚酰胺胺树枝状聚合物、聚赖氨酸树枝状聚合物、壳聚糖。而且,作为阳离子性脂质或含有其的集合体,可以优选使用脂质体Lipofectamine(上述DOSPA和DOPE的3:1w/w混合体脂质体)。
作为在本发明的冷冻干燥体中使用的阴离子性聚合物,可以使用在分子中含有阴离子性基、带负电荷的、分子量为500~400万左右的天然来源或合成的高分子,该高分子为1个分子中具有多个、优选5个以上在水中能与聚阳离子形成复合物的官能团,作为这种官能团,可以例举例如羧基、-OSO3H基、-SO3H基、磷酸基、和它们的盐。作为这种阴离子性聚合物,还包括两性离子性聚合物。
本发明的冷冻干燥体中,作为阴离子性聚合物,可以使用:具有选自于羧基、-OSO3H基、-SO3H基、磷酸基、和它们的盐中的官能团的多糖类或其衍生物;含有带负电荷的侧链的氨基酸残基的聚氨基酸;带有羧基侧链的PEG衍生物;具有选自于羧基、-OSO3H基、-SO3H基、磷酸基、和它们的盐中的官能团的合成高分子;具有选自于羧基、-OSO3H基、-SO3H基、磷酸基和它们的盐中的官能团、以及任选取代的氨基或铵基或其盐(这些基团可以被例如碳原子数为1~6的烷基、苯基等单取代或多取代)的高分子;以及上述物质的组合。
可以作为在本发明的冷冻干燥体中的阴离子性聚合物使用的具有上述那样的官能团的多糖类或其衍生物,可以优选使用葡糖胺聚糖。这种葡糖胺聚糖的分子量优选为1000~400万、更优选为4000~300万。作为这种葡糖胺聚糖,具体可以例举例如透明质酸、软骨素、硫酸软骨素、硫酸角质素、肝素、硫酸皮肤素(Dermatan Sulfate)等。其中可以优选使用透明质酸。透明质酸也可以以其盐或带负电荷的衍生物的形式使用。其分子量可以在5,000以上,优选在10,000以上,更优选10万~300万。作为透明质酸的盐,可以例示钠盐、钾盐、铵盐等。而且,作为透明质酸的衍生物,可以例举例如通过向透明质酸中导入聚乙二醇、肽、糖、蛋白质、碘酸、抗体或其一部分等而获得的物质,也包括导入了精胺、亚精胺等并具有带正电荷部分的两离子性的衍生物。
所谓可以在本发明的冷冻干燥体中作为阴离子性聚合物使用的、含有具有带负电荷侧链的氨基酸残基的聚氨基酸,是包括具有羧基、-O-SO3H基、-SO3H基、磷酸基、和它们的盐等基团作为侧链的氨基酸残基、且优选分子量为500~100万的聚氨基酸。作为这种聚氨基酸,具体可以例示聚谷氨酸、聚天冬氨酸等。
所谓可以在本发明的冷冻干燥体中作为阴离子性聚合物使用的、带有羧基侧链的PEG衍生物,是指每1分子PEG中具有多个、优选5个以上羧基侧链、且分子量为500以上、优选2,000以上、更优选4,000~40,000的PEG衍生物。带有羧基侧链的PEG衍生物还可以以其盐或带负电荷的衍生物的形式使用。作为它们的盐,可以例示钠盐、钾盐、铵盐等。作为这种PEG衍生物,具体可以例示记载于非专利文献1(J.Biomater.Sci.Polymer Edn.Vol.14,pp 515-531(2003))中的PEG衍生物。
所谓可以在本发明的冷冻干燥体中作为阴离子性聚合物使用的、具有选自于羧基、-OSO3H基、-SO3H基、磷酸基、和它们的盐中的官能团的合成高分子是指每1分子中具有多个、优选5个以上选自于羧基、-O-SO3H基、-SO3H基、磷酸基、和它们的盐中的官能团、且分子量优选为500~400万的聚合物或共聚物。作为这种聚合物或共聚物,具体可以例示分子量为1000~300万的丙烯酸或甲基丙烯酸的聚合物或共聚物、或聚乙烯醇的硫酸酯、琥珀酰亚胺化聚-L-赖氨酸等。
所谓可以在本发明的冷冻干燥体中作为阴离子性聚合物使用的、具有选自于羧基,-OSO3H基,-SO3H基,磷酸基和它们的盐中的官能团、以及任选取代的氨基或铵基或其盐(这些基团可以被例如碳原子数为1~6的烷基、苯基等单取代或多取代)的高分子是指每1分子具有多个、优选5个以上选自于羧基、-OSO3H基、-SO3H基、磷酸基和它们的盐中的官能团,以及如上所述的任选取代的氨基或铵基或其盐,且分子量为500以上,优选2,000以上、更优选4,000~40,000的高分子。作为这种高分子,优选可以例举,带有羧基侧链和其当量以下的上述氨基或铵基或其盐的PEG衍生物,具体可以例示可以用记载于非专利文献5(Macromol.Biosci.Vol.2,pp 251-256(2002))中的方法制备的PEG衍生物。
可以在本发明的冷冻干燥体中使用的阴离子性聚合物,即使通常不带负电荷,但只要通过导入羧基等官能团而带负电荷就可以使用,而且根据需要还可以再用糖链、寡肽、抗体等进行修饰。
本发明的冷冻干燥体中,作为阴离子性聚合物,可优选使用透明质酸、带有羧基侧链的PEG衍生物、聚丙烯酸等阴离子性聚合物或它们的盐,可以特别优选使用透明质酸、带有羧基侧链的PEG衍生物或它们的盐等。
而且,可以通过使用对欲导入核酸的目的细胞具有特异性粘着能力的阴离子性聚合物作为阴离子性聚合物,对目的细胞特异性进行核酸导入。例如在使用透明质酸作为阴离子性聚合物时,可以以具有与透明质酸特异性结合的CD44等细胞表面分子的细胞作为靶标。而且,通过使用导入了RGD肽的阴离子性聚合物,可以以多种肿瘤细胞作为靶标,而且通过使用导入了半乳糖侧链的阴离子性聚合物,可以以肝细胞或肝来源的细胞作为靶标。
本发明的冷冻干燥体中,作为阳离子性聚合物或阳离子性脂质或含有其的集合体与阴离子性聚合物的组合,可以优选例举聚乙烯亚胺和透明质酸;聚乙烯亚胺和带有羧基侧链的PEG衍生物;含有DOSPA的集合体(例如脂质体Lipofectamine(DOSPA和DOPE的3:1w/w混合体脂质体))和透明质酸;含有DOSPA的集合体(例如脂质体Lipofectamine)和带有羧基侧链的PEG衍生物。
本发明的冷冻干燥体中使用的核酸、寡核酸或其衍生物与阳离子性聚合物或阳离子性脂质或含有其的集合体的各带电基团的摩尔比(负电荷:正电荷比)根据目的细胞/核酸等/阳离子性聚合物等的种类而不同,可以是1:0.8~1:100,优选为1:1~1:50,更优选为1:1.2~1:30。这里所称核酸、寡核酸或其衍生物与阳离子性聚合物或阳离子性脂质或含有其的集合体的配合比为各带电基团的摩尔比,具体是指核酸、寡核酸或其衍生物的磷酸阴离子的负电荷:阳离子性聚合物或阳离子性脂质或含有其的集合体的正电荷或可以带正电的官能团的摩尔比。
本发明的冷冻干燥体中使用的核酸、寡核酸或其衍生物与阴离子性聚合物的各带电基团的摩尔比(负电荷:负电荷比)根据目的细胞/核酸等/阴离子性聚合物的种类而不同,可以为1:0.2~1:1000,优选为1:0.2~1:100,更优选1:1~1:60。这里所称核酸、寡核酸或其衍生物与阴离子性聚合物的配合比是各带电基团的摩尔比,具体指核酸、寡核酸或其衍生物的磷酸阴离子的负电荷:阴离子性聚合物的负电荷或可以带负电荷的官能团的摩尔比。
例如使用透明质酸作为阴离子性聚合物时,核酸与透明质酸的配合比可以是1:0.2~1:1000,优选为1:0.2~1:100,更优选1:1~1:60。
例如在使用带有羧基侧链的PEG衍生物作为阴离子性聚合物时,核酸与带有羧基侧链的PEG衍生物的配合比可以为1:0.2~1:1000,优选1:0.2~1:100,更优选1:1~1:60。
尤其是,在使用聚乙烯亚胺作为阳离子性聚合物,使用透明质酸作为阴离子性聚合物时,核酸:聚乙烯亚胺:透明质酸的配合比为1:2~60:1~240,优选1:4~24:1~160,更优选1:7~20:2~60,尤为优选1:8~14:2~32。
尤其是,在使用聚乙烯亚胺作为阳离子性聚合物,使用带有羧基侧链的PEG衍生物作为阴离子性聚合物时,核酸:聚乙烯亚胺:带有羧基侧链的PEG衍生物的配合比为1:2~60:1~240,优选1:4~24:2~160,更优选1:7~20:2~60,尤为优选1:8~14:4~32。
尤其是,在使用脂质体Lipofectamine(DOSPA与DOPE的3:1w/w混合体脂质体)作为含有阳离子性脂质的集合体,使用透明质酸作为阴离子性聚合物时,核酸:脂质体Lipofectamine:透明质酸的配合比为1:1~50:0.2~240、优选1:1.2~48:0.2~160,更优选1:1.5~30:0.5~60,尤为优选1:1.8~16:1~32。
尤其是,在使用脂质体Lipofectamine作为含有阳离子性脂质的集合体,使用带有羧基侧链的PEG衍生物作为阴离子性聚合物时,核酸:脂质体Lipofectamine:带有羧基侧链的PEG衍生物的配合比为1:1~50:0.1~160、优选1:1.2~48:0.2~160,更优选1:1.5~30:0.5~60,尤为优选1:1.8~16:2~32。
本发明的冷冻干燥体中所含的核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物的优选配合比如上所述,由于最适条件根据导入核酸等的细胞的数量和种类而变化,因此配合比可以由本领域技术人员根据所使用的细胞、核酸等的种类适当确定。
本发明的冷冻干燥体可以通过以下工序制备:通过将上述的核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物按上述的配合比进行混合,以形成复合物的工序;以及随后通过对其进行冷冻干燥的工序。作为混合的顺序,优选为以下顺序:按[1]核酸、寡核酸或其衍生物;[2]阳离子性聚合物或阳离子性脂质或含有其的集合体、[3]阴离子性聚合物的顺序、或、按[1]核酸、寡核酸或其衍生物;[2]阴离子性聚合物、[3]阳离子性聚合物或阳离子性脂质或含有其的集合体的顺序。核酸、寡核酸或其衍生物通过与阳离子性聚合物或阳离子性脂质或含有其的集合体用离子键结合,进而阳离子性聚合物或阳离子性脂质或含有其的集合体与阴离子性聚合物形成离子键的复合物。或者,根据各成分的配合组成,使这种复合物结构的外壳形成主要被阴离子性聚合物包覆、具有负的表面电位的形态。
接着,对获得的复合物进行冷冻干燥。冷冻干燥可以在通常的冷冻干燥条件下进行,例如可以通过在减压下(优选5~100mmHg、更优选10mmHg)、外气温度-78℃~60℃、优选-30℃~40℃下干燥来进行。干燥所需时间,根据减压度、溶剂的量而不同,通常在1小时~2天时间内完成。
这样制备的本发明的冷冻干燥体可以在对人或动物的各种基因治疗、反义治疗、或用于制备导入、控制、敲入、敲出了特定基因的实验动物或细胞。具体的是,可以通过在即将使用前将本发明的冷冻干燥体悬浮或溶解于水、生理盐水、缓冲液、葡萄糖溶液、培养基液等溶剂中形成再水化物之后再使用。再水化时,用例如核酸、寡核酸或其衍生物的100~10000倍(重量比)的溶剂悬浮或稀释冷冻干燥体。由于可以使用与冷冻干燥前不同的量、不同种类的溶剂,因此可以容易地制备以往困难的较高浓度的悬浮液或溶液(例如1ml中含1mg的DNA的液体)。
这样经再水化的本发明的冷冻干燥体在向细胞导入核酸等时,具体可以使用以下方法:例如在孔中通过用经水合的本发明的冷冻干燥体来处理被取出到体外的目的细胞而导入基因或反义核酸后,再将该细胞送回到生物体内,使目的基因表达的间接体内(ex vivo)法,或直接体内(in vivo)法、原位(in situ)法等基因或反义核酸的直接导入法等向生物体细胞导入核酸、寡核酸或其衍生物时通常使用的任意方法。
而且,本发明的冷冻干燥体还可以通过不进行再水化,而直接与欲导入核酸等的细胞相接触进行给药;或者对欲进行核酸导入的动物通过皮下移植进行给药、通过移植等手段给予到欲导入核酸等的目标组织内、组织表面或附近进行给药。
本发明的冷冻干燥体对细胞的适用量根据上述的导入方法、疾病的种类等而不同,例如根据核酸、寡核酸或其衍生物的量,在间接体内法、原位法中,每个直径为1~2cm的孔中可以给药0.2~10μg/104~7个细胞,在直接体内法中,根据给药部位而差异很大,在向肿瘤内局部给药时,可以给药例如5~1000μg/cm3肿瘤,在向膀胱等器官给药时可以给药例如0.1μg~100mg/器官、全身给药时可以给药例如0.1ng~10mg/Kg体重。
作为对生物体直接给药的直接体内法,还可以使用将经水合的本发明的冷冻干燥体水合物注射于静脉、皮下或肌肉、腹腔、肿瘤内、肿瘤附近等;使之被吸入于鼻腔、口腔、肺等;直接注入于膀胱内、直肠内;直接给药于病变部组织或附近的血管内;或者将其负载到凝胶状物、海绵等多孔体、无纺布等上而留置等基因治疗技术中的任意一种方法。
而且,在不对本发明的冷冻干燥体水化物再水化时,可以通过如上所述的间接体内法、原位法、或直接体内法使用上述量的冷冻干燥体。
本发明的冷冻干燥体中,通常的核酸、寡核酸或其衍生物与阳离子性聚合物或阳离子性脂质或含有其的集合体的复合物所带的正电荷,在被阴离子性聚合物中和时,其中和作用即使在向生物体、细胞给药后仍然保持,由此可抑制复合物对血清蛋白质、血细胞、细胞外基质等所产生的凝集等相互作用,而且由于抑制了酶对核酸、寡核酸或其衍生物的分解作用,核酸能被吸收到细胞中,其表达效率也高。
如上所述,本发明的冷冻干燥体水合物,可以作为核酸、寡核酸或其衍生物的导入用制剂或试剂,或核酸、寡核酸或其衍生物的导入用试剂盒使用。
根据实施例对本发明进行更为具体的说明。而且,这些实施例只是说明本发明,因此本发明不受其任何限定。
实施例1
通过质粒(plasmid)/聚乙烯亚胺(PEI)/透明质酸(HA)
复合物的冷冻干燥体进行的基因表达
将冷冻干燥的由基因·PEI·HA构成的三组分的复合物与小鼠的黑色素瘤细胞来源的B16一起培养,确认了荧光素酶基因的表达。
荧光素酶质粒使用非专利文献6(Biomacromolecules Vol.7,pp1274-1279)所记载的相同的荧光素酶质粒。PEI使用Polyscience公司制的直链状PEI、Mw=25000的PEI。HA使用ナカライテスク株式会社的“微生物来源”的透明质酸。PBS采用溶解于经蒸馏的离子交换水的Roman Industries公司制的磷酸缓冲盐(Phosphate Buffered Salts)(片状)。之后的实施例也同样。
[操作步骤]
[1]在导入基因的2天前,在24孔多孔板上接种B16细胞,用EMEM培养基培养2晚。
[2]在导入前一天,将含有1.3μg荧光素酶质粒的水溶液2μl与2μl的PEI水溶液按+/-比(电荷摩尔比)为8的方式进行混合,数次吸管吹吸(pipetting)后,加入各种浓度的HA溶液4μl,充分搅拌后在-30℃下冷冻。然后,进行冷冻干燥,从而制备出本发明的冷冻干燥体。再改变HA和PEI的混合顺序,用同样的方法进行制备。
[3]去除培养的培养基,然后将500μl含有10% FBS和25U青霉素和25μg链霉素的EMEM加入孔中。
[4]在[1]中制备的冷冻干燥体中混合16μl PBS,恒温1小时后,加入到孔中。
[5]于37℃、5%CO2-95%空气下培养4小时。
[6]将培养基更换成新的含有10% FBS和25U青霉素以及25μg链霉素的EMEM,于37℃培养20小时。
[7]20小时的培养后,去除培养基,在各孔中各加入200μlPicaGene的细胞溶解液。放置20分钟左右之后,剥离细胞,并回收到微型管(microtube)中。
[8]离心分离(15000rpm,1分钟)后,用上清进行荧光素酶测试。荧光素酶测试按照Pica Gene公司的荧光试剂盒的方法进行。
而且,在蛋白质测定时,直接采用该细胞溶解液。蛋白质测定用Bio-Rad公司的蛋白质测定试剂盒(Protein assay kit)进行。
为了比较,对不添加HA的、冷冻干燥的、不进行冷冻干燥的也研究基因表达。
[结果]
结果示于下图。这里,图中()中的值是PEI的阳离子、HA的阴离子相对于质粒的阴离子之比,具体是PEI、HA相对于DNA的电荷的摩尔比。
冷冻干燥的质粒/PEI二元复合物表达达到冷冻干燥前的1000分之1以下,基本观察不到表达,与之相对,添加了HA的对象可看到高表达,按质粒:PEI:HA=1:8:16(电荷)混合后再冷冻干燥的复合物,呈现比冷冻干燥前的质粒/PEI二元复合物高11%以上的表达效率。

实施例2
混合顺序的影响
在实施例1中的[2]中,在荧光素酶质粒中先加入HA,然后再加入PEI,再进行混合,然后进行冷冻干燥的对象进行相同评价。
[结果]
结果示于下图。这里,图中()中的值是PEI的阳离子、HA的阴离子相对于质粒的阴离子之比,具体是PEI、HA相对于DNA的电荷的摩尔比。
与冷冻干燥的质粒/PEI二元复合物几乎没有表达相比,在添加了HA的对象中可看到高表达,按质粒:PEI:HA=1:8:16(电荷)混合后再冷冻干燥的对象中,显示出比冷冻干燥前的质粒/PEI二元复合物高26%以上的表达效率。根据该结果,可知即使改变混合顺序,也可看到高表达。

实施例3
通过含有质粒/PEI/带有羧基侧链的PEG衍生物(以下PEG
-C)的冷冻干燥体进行的基因表达
实施例3中,使用通过非专利文献1(J.Biomater.Sci.Polymer Edn.Vol.14,pp 515-531(2003))中记载的方法合成的分子量为约1万且1分子中含有约18个羧基的PEG-C作为阴离子性聚合物。
将由冷冻干燥的基因·PEI·PEG-C构成的三组分的复合物与小鼠的黑色素瘤细胞来源的B16一起培养,确认荧光素酶基因的表达。
[操作步骤]
[1]在导入基因的2天前,在24孔多孔板上接种B16细胞,用EMEM培养基培养2晚。
[2]在导入前一天,将含有1.3μg荧光素酶质粒的水溶液12.5μl与12.5μl的PEI水溶液按+/-比(电荷摩尔比)为8的方式进行混合,数次吸管吹吸后,加入各种浓度的PEG-C溶液25μl,充分搅拌后在-30℃下冷冻。然后,进行冷冻干燥,从而制备出本发明的冷冻干燥体。
[3]去除培养的培养基,然后将500μl含有10% FBS和25U青霉素和25μg链霉素的EMEM加入孔中。
[4]在[2]中制备的冷冻干燥体中混合50μl PBS,恒温1小时后,加入到孔中。
[5]于37℃、5% CO2-95%空气下培养4小时。

结果示于下图。这里,图中()中的值是PEI的阳离子、PEG-C的阴离子相对于质粒的阴离子之比,具体是PEI、PEG-C相对于DNA的电荷的摩尔比。
冷冻干燥的质粒/PEI二元复合物中几乎没有观察到表达。而且,在加入了不具有电荷的PEG的对象中也几乎看不到表达。另一方面,加入了PEG-C的本发明的冷冻干燥体,可看到接近不进行冷冻干燥的原来的质粒/PEI二元复合物的近6成的高表达。

实施例4
通过质粒(plasmid)/脂质体Lipofectamine/HA冷冻干燥体
进行的基因表达
本实施例中,脂质体Lipofectamine使用Invitrogen公司制品。
将由冷冻干燥的基因·脂质体Lipofectamine·HA构成的三组分的冷冻干燥体与小鼠的黑色素瘤细胞来源的B16一起培养,确认了荧光素酶基因的表达。
[操作步骤]
[1]在导入基因的2天前,在24孔多孔板上接种B16细胞,用EMEM培养基培养2晚。
[2]在导入前一天,将含有1.3μg荧光素酶质粒的水溶液12.5μl与12.5μl的脂质体Lipofectamine水溶液按荧光素酶质粒与脂质体Lipofectamine的重量比最终为8的方式进行混合,数次吸管吹吸后,加入各种浓度的HA溶液25μl,培养30分钟后,于-30℃下冷冻。然后,进行冷冻干燥,从而制备出本发明的冷冻干燥体。
[3]去除培养的培养基,然后将500μl的含有25U青霉素和25μg链霉素的EMEM装入500μl孔中。
[4]在[2]中制备的冷冻干燥体中混合50μl PBS,恒温45分钟后,加入到孔中。
[5]于37℃、5% CO2-95%空气下培养4小时。
[6]更换成新的含有25U青霉素和25μg链霉素的1100μl的EMEM和400μl的FBS,于37℃下培养20小时。
[7]培养24小时后,去除培养基,用PBS清洗细胞一次,然后在各孔中各加入200μl的PicaGene公司的细胞溶解液。放置20分钟左右之后,剥离细胞,并回收到微试管(microtube)中。
[8]离心分离(15000rpm,1分钟)后,用上清进行荧光素酶测试。荧光素酶测试按照Pica Gene公司的荧光试剂盒的方法进行。
而且,在蛋白质测定时,直接采用该细胞溶解液。蛋白质测定用Bio-Rad公司的蛋白质测定试剂盒(Protein assay kit)。
为了比较,对不添加HA的对象,冷冻干燥的对象和不进行冷冻干燥的对象也研究基因表达。
而且,在含有80%FBS的EMEM培养基中进行[3]的细胞和DNA复合物的培养,对该培养产物也同时进行研究。此时,[6]中不添加FBS,只加入1500μl的含有25U青霉素和25μg链霉素的EMEM。
[结果]
结果示于下图。其中,图中()中的值是脂质体Lipofectamine、HA相对于质粒的重量比。
在不含FBS的培养基中进行基因导入的对象中,冷冻干燥的质粒/脂质体Lipofectamine二元复合物,表达达到冷冻干燥前的3000分之1以下,几乎没有观察到表达,与之相对,在添加了HA的对象物中可看到不进行冷冻干燥的原来的质粒/脂质体Lipofectamine二元复合物的约55%的表达。
80%血清存在下,冷冻干燥的质粒/脂质体Lipofectamine二元复合物的表达进一步降低,而冷冻干燥的质粒/脂质体Lipofectamine/HA三元复合物则呈现不进行冷冻干燥的原来的质粒/脂质体Lipofectamine二元复合物的约57%的高表达。

实施例5
通过质粒(plasmid)/脂质体Lipofectamine/PEG-C冷冻干
燥体进行的基因表达
用与质粒DNA的电荷比例为16倍的PEG-C代替HA来进行与实施例4同样的实验。具体而言,如下图记载的那样将电荷质量比例为质粒DNA的16倍的量的PEG-C溶于水中制成25μl并加入,从而代替实施例4中所加入的25μl HA溶液。
基因导入在不含血清的培养基中或含80%的血清的培养基中进行。
[结果]
结果示于下图。其中,图中()中的值是脂质体Lipofectamine、PEG-C相对于DNA的重量比。
冷冻干燥的DNA/脂质体Lipofectamine二元复合物几乎没有观察到表达,与之相对,加入了PEG-C的对象物呈现高表达,在80%血清存在下,质粒/脂质体Lipofectamine/PEG-C三元复合物呈现出比不进行冷冻干燥的原来的质粒/脂质体Lipofectamine二元复合物4倍以上的高表达效率。

实施例6
在冷冻干燥前后改变浓度的影响
在实施例5中的[2]中,用10倍量的溶剂制备低浓度的溶液,混合之后进行冷冻干燥,对[4]中与5同样加入50μl的PBS进行再水化的对象同样进行评价。
[结果]
结果示于下图。其中,图中()中的值是脂质体Lipofectamine、PEG-C相对于DNA的重量比。
冷冻干燥的DNA/脂质体Lipofectamine二元复合物,表达达到冷冻干燥前的1000分之1以下,几乎没有观察到表达,与之相对,加入了PEG-C的对象,冷冻干燥后浓缩的物质可看到高表达,在80%血清存在下,质粒/脂质体Lipofectamine/PEG-C三元冷冻干燥体呈现出比不进行冷冻干燥的原来的质粒/脂质体Lipofectamine二元复合物高接近30%的表达效率。
根据这些结果,确认通过进行冷冻干燥,可以制备可以容易地导入基因的任意浓度的复合物悬浮液·溶液。

实施例7
冷冻干燥的固体状的DNA复合物向生物体的给药
[操作步骤]
用400μl水稀释50μl编码荧光素酶的质粒的TE缓冲溶液(0.8mg/ml),在其中加入100μl透明质酸水溶液(5.8mg/ml),最后加入50μl PEI溶液(1.25mg/ml)。从混合三组分起30分种后于-30℃进行冷冻,然后进行冷冻干燥,获得了固体状的复合物。
将悬浮于100μl的培养基*1中的4.72×106的小鼠的黑色素瘤细胞来源的B16移植到5周龄的雄性ddY小鼠的皮下。肿瘤达到6~8mm时,麻醉下在肿瘤部分切出口子,将上述的固体状DNA/PEI/HA复合物埋入于肿瘤中,再进行缝合。
两天后,用乙醚使之死亡,取出肿瘤和皮,在1ml的细胞溶解液*2中均化。然后,于10000rpm、4℃离心20分钟,在上清液(5μl)中加入底物(Promega公司制)(20μl),用照度计测定30秒荧光素酶的发光量。
在1ml的BioRad的蛋白质定量试剂中加入20μl将各试样的上清液稀释到1/80的稀释物,20分钟后测定波长595nm下的吸光度,并定量总蛋白质量。
[结果]
固体状的DNA复合物在肿瘤内呈现出极高表达(结果示于下表)。

在培养板上通过冷冻干燥的DNA复合物向细胞进行基因
导入
[操作步骤]
用12.5μl的水稀释1.56μl编码荧光素酶的质粒的溶液(0.8mg/ml),在其中加入1.56μl的PEI溶液(1.25mg/ml),最后加入3.56μl的透明质酸水溶液(5.8mg/ml)。混合三组分之后加入到培养板的孔中,30分钟后,于-30℃冷冻,然后进行冷冻干燥。对于不加入透明质酸的对象物同样地进行冷冻干燥。而且,对于不进行冷冻干燥、采用新鲜悬浮液的对象物同时进行比较。
将悬浮于300μl培养基*1中的1.2×105的小鼠的黑色素瘤细胞来源的B16接种于将DNA复合物进行冷冻干燥的孔中。4小时后补充1ml的培养基,20小时后再替换成1ml新的培养基。此后24小时后,加入200μl的Promega的细胞溶解液,回收细胞,于15000rpm、4℃离心1分钟,在上清液(5μl)中加入底物(Promega公司制)(20μl),用照度计测定30秒荧光素酶的发光量。
在1ml的BioRad的蛋白质定量试剂中加入20μl将各试样的上清液稀释到1/5后的溶液,20分种后测定波长595nm下的吸光度,并定量总蛋白质量。

[结果]
没有加入透明质酸(HA)的对象如果进行冷冻干燥则几乎没有基因导入活性,加入透明质酸的对象物即使进行冷冻干燥,也呈现出与新鲜的悬浮液同样的高活性(结果示于下表)。

使用通过冷冻干燥浓缩的DNA复合物悬浮液进行体内基
因导入
[操作步骤]
用0、500、2000、或8000μl的水稀释62.5μl的编码荧光素酶的质粒的溶液(0.8mg/ml),在其中加入125μl透明质酸水溶液(5.8mg/ml),最后加入62.5μl(1.25mg/ml)的PEI溶液。混合三组分起30分钟后,于-30℃冷冻,然后进行冷冻干燥。
用250μl的5%葡萄糖将冷冻干燥的DNA复合物再溶剂化。
将悬浮于100μl的培养基*1中的4.72×106的小鼠的黑色素瘤细胞来源的B16移植到5周龄的雄性ddY小鼠的皮下。肿瘤达到6~8mm时,在小鼠的尾静脉给药再溶剂化的DNA复合物悬浮液。
24小时后,在乙醚麻醉下放血,取出肿瘤、肝脏、肺,在1ml的细胞溶解液*2中均化。然后,于10000rpm、4℃离心20分钟,在上清液(5μl)中加入底物(Promega公司制)(20μl),用照度计测定30秒荧光素酶的发光量。
在1ml的BioRad的蛋白质定量试剂中加入20μl将各试样的上清液稀释到1/80的稀释物,20分钟后测定波长595nm下的吸光度,并定量总蛋白质量。

实施例10
在培养板上进行冷冻干燥的siRNA复合物产生的基因表达
抑制效果
[操作步骤]
在25μl的Anti荧光素酶siRNA(Invitrogen公司制)的水溶液(21.28μg/ml)中加入25μl鱼精蛋白水溶液(78μg/ml),然后加入50μl透明质酸溶液(53.7μg/ml、或107.5μg/ml)。混合三组分之后加入到培养板的孔中,30分钟后,于-30℃冷冻,然后进行冷冻干燥。
将悬浮于100μl培养基*1的1.2×105的小鼠的黑色素瘤细胞来源的B16接种于培养板上,4小时后补充1ml的培养基,之后再加入25μl pDNA溶液(50μg/ml)和25μl PEI溶液(78μg/ml)的混合物。进而,在20小时后再替换成1ml新的培养基。
此后24小时后,加入200μl的Promega的细胞溶解液,回收细胞,于15000rpm、4℃离心1分钟,在上清液(5μl)中加入底物(Promega公司制)(20μl),用照度计测定30秒荧光素酶的发光量。
在1ml的BioRad的蛋白质定量试剂中加入20μl将各试样的上清液稀释到1/5的溶液,20分种后测定波长595nm下的吸光度,并定量总蛋白质量。

对不加入鱼精蛋白和透明质酸的也同样进行冷冻干燥。
[结果]
在加入鱼精蛋白(PRT)和透明质酸(HA)后进行冷冻干燥的板上培养的细胞,荧光素酶的表达受到显著抑制(参照下表)。

实施例11
改变浓度制备的DNA复合物再水化后的大小
[操作步骤]
用0、12.5、或200μl的水稀释1.5μl与实施例1中使用的荧光素酶质粒溶液(0.8mg/ml)相同的溶液,在其中加入3μl透明质酸水溶液(5.8mg/ml),最后加入1.5μl的PEI溶液(1.25mg/ml)。混合三组分起30分钟后,于-30℃冷冻,然后进行冷冻干燥。
用6μl水再水化冷冻干燥的DNA复合物,30分钟后,加入800μl的水,用马尔文(Malvern)公司的粒度分析仪(Zetaanalyzer)测定大小。
[结果]
生成的复合物颗粒中0~100nm的比例、和100~200nm的比例示于下表。成分后面的数值是用核酸碱基浓度表示的复合物制备时的最终DNA浓度。
不加入透明质酸(HA)的对象,再水化后几乎没有观察到微细的颗粒。另一方面,观察到加入透明质酸(HA)后进行冷冻干燥的对象,再水化后多数颗粒维持微小的尺寸。而且,再水化后的DNA浓度均相同,但发现越是在稀浓度条件下制备复合物,小颗粒的比例越多。

1.一种复合物的冷冻干燥体,所述复合物含有核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物。
2.根据权利要求1所述的冷冻干燥体,核酸、寡核酸或其衍生物与阳离子性聚合物或阳离子性脂质或含有其的集合体的各带电基团的摩尔比(负电荷:正电荷比)为1:0.8~1:100。
3.根据权利要求1或2所述的冷冻干燥体,核酸、寡核酸或其衍生物与阴离子性聚合物的各带电基团的摩尔比(负电荷:负电荷比)为1:0.2~1:1000。
4.根据权利要求1~3任一项所述的冷冻干燥体,阳离子性聚合物为带正电荷的分子量为1000~300万左右的天然来源或合成的高分子,且是一种1个分子中有多个能在水中与DNA形成复合物的官能团的高分子。
5.根据权利要求4所述的冷冻干燥体,阳离子性聚合物选自于带正电荷的蛋白质或多肽、带正电荷的树枝状聚合物、带正电荷的合成聚合物、带正电荷的多糖类衍生物、或它们的盐、以及上述物质的组合。
6.根据权利要求1~3任一项所述的冷冻干燥体,阳离子性脂质为DC-Chol(3β-(N-(N′,N′-二甲氨基乙烷)氨甲酰基)胆固醇)、DDAB(N,N-二硬脂基-N,N-二甲基溴化铵)、DMRI(N-(1,2-二肉豆蔻基氧丙-3-基)-N,N-二甲基-N-羟乙基溴化铵)、DODAC(N,N-二油酰基-N,N-二甲基氯化铵)、DOGS(双十七烷酰胺基甘氨酰亚精胺)、DOSPA(N-(1-(2,3-二油酰氧基)丙基)-N-(2-(精胺甲酰胺)乙基)-N,N-二甲基三氟醋酸铵)、DOTAP(N-(1-(2,3-二油酰氧基)丙基)-N,N,N-三甲基氯化铵)、或DOTMA(N-(1-(2,3-二油酰氧基)丙基)-N,N,N-三甲基氯化铵)。
7.根据权利要求1~6任一项所述的冷冻干燥体,阴离子性聚合物为分子中含有阴离子性基团,带负电荷的分子量为500~400万左右的天然来源或合成的高分子,且是一种1个分子中具有多个可以在水中与聚阳离子形成复合物的官能团的高分子。
8.根据权利要求7所述的冷冻干燥体,阴离子性聚合物选自于:具有从羧基、-OSO3H基、-SO3H基、磷酸基、和它们的盐中选择的官能团的多糖类或其衍生物;含有具有带负电荷的侧链的氨基酸残基的聚氨基酸;带有羧基侧链的PEG衍生物;从羧基、-OSO3H基、-SO3H基、磷酸基和它们的盐中选择的官能团的合成高分子;具有从羧基、-OSO3H基、-SO3H基、磷酸基和它们的盐中选择的官能团、以及任选取代的氨基或铵基或其盐的高分子、以及上述物质的组合。
9.根据权利要求1所述的冷冻干燥体,含有阳离子性脂质的集合体是包括DOSPA(N-(1-(2,3-二油酰氧基)丙基)-N-(2-(精胺甲酰胺)乙基)-N,N-二甲基三氟醋酸铵)的集合体,阴离子性聚合物是透明质酸。
10.根据权利要求1所述的冷冻干燥体,含有阳离子性脂质的集合体是包括DOSPA(N-(1-(2,3-二油酰氧基)丙基)-N-(2-(精胺甲酰胺)乙基)-N,N-二甲基三氟醋酸铵)的集合体,阴离子性聚合物是带有羧基侧链的PEG衍生物。
11.根据权利要求1所述的冷冻干燥体,阳离子性聚合物是聚乙烯亚胺;阴离子性聚合物是透明质酸。
12.根据权利要求1所述的冷冻干燥体,阳离子性聚合物是聚乙烯亚胺;阴离子性聚合物是带有羧基侧链的PEG衍生物。
13.根据权利要求11所述的冷冻干燥体,核酸、寡核酸或其衍生物:聚乙烯亚胺:透明质酸的配合比为1:4~24:1~160(各带电基团的摩尔比)。
14.根据权利要求12所述的冷冻干燥体,核酸、寡核酸或其衍生物:聚乙烯亚胺:带有羧基侧链的PEG衍生物的配合比为1:4~24:2~160(各带电基团的摩尔比)。
15.根据权利要求9所述的冷冻干燥体,核酸、寡核酸或其衍生物:含有DOSPA的集合体:透明质酸的配合比为1:1.2~48:0.2~160(各带电基团的摩尔比)。
16.根据权利要求10所述的冷冻干燥体,核酸、寡核酸或其衍生物:含有DO SPA的集合体:带有羧基侧链的PEG衍生物的配合比为1:1.2~48:0.2~160(各带电基团的摩尔比)。
17.一种权利要求1所述的冷冻干燥体的制备方法,其包括:通过混合核酸、寡核酸或其衍生物,阳离子性聚合物或阳离子性脂质或含有其的集合体,以及阴离子性聚合物,以形成复合物的工序;以及之后进行冷冻干燥的工序。
18.一种核酸、寡核酸或其衍生物的导入用制剂或试剂,其含有权利要求1~16任一项所述的冷冻干燥体。
19.一种核酸、寡核酸或其衍生物的导入用试剂盒,其含有权利要求1~16任一项所述的冷冻干燥体。
20.一种向细胞导入核酸、寡核酸或其衍生物的方法,其使用权利要求1所述的冷冻干燥体。
21.根据权利要求20所述的方法,其包括在向细胞导入核酸、寡核酸或其衍生物之前,将冷冻干燥体在溶剂中再水化的工序。
22.根据权利要求20所述的方法,其使用冷冻干燥体时不在溶剂中再水化。
