背景技术
慢性炎性肠疾病(或IBDs)特别地包括两种不同的病症:溃疡性结肠炎(UC)和Crohn’s病。这两种不同且相关的疾病的特征在于肠或多或少的弥漫性炎性病变,特别地应归于肠免疫系统机能亢进的状况,其原因未知。
它们的表现基本上在消化系统中,伴有腹泻、腹痛、体重减轻和组织炎症。

而且,C.Cherbut等人也证实FOSs实际上具有益生作用,即能够刺激结肠中有益菌的肠内生长,本发明示例性的有益菌如乳酸菌和丁酸菌,其导致结肠pH降低。
FOSs的保护机制不能被清楚地解释。
在专利申请US2004/0219157中已经提出FOSs刺激非特异性免疫学参数的动态平衡和刺激淋巴细胞亚群的生长。
也认为生长受FOSs刺激的乳酸菌为病原菌的拮抗剂,它们通过生成抗微生物物质和通过降低结肠的pH阻断所述病原菌生成。
乳酸菌也可以粘附到肠壁,从而防止这些相同的病原菌定居。
而且,FOSs也通过诱导乳酸和丁酸生成对结肠pH的降低起作用。
然而,肠内酸中毒效应并非只具有优点。
在国际专利申请WO04/026316中,实际上描述了特别由乳酸菌生长引起的酸中毒最终引起结肠粘膜糜烂,增加了溃疡性结肠炎的危险。
而且,乳酸在结肠中的累积也可以导致在血液中的过量释放,从而引起代谢性酸中毒。
菊糖和FOSs都具有在结肠中发酵太快的缺点,因此,可导致对结肠保护效应有害的微生物群失衡。
为了抗衡该有害作用,在所述的专利申请WO04/026316中,提议将FOSs与一种以在结肠中缓慢发酵为特征的多糖组合,本发明中多糖的实例如聚葡萄糖。
聚葡萄糖为在山梨醇和适当的酸性催化剂(比如柠檬酸)的存在下和在高温下通过葡萄糖的任意聚合合成的。
聚葡萄糖作为膨胀剂和作为低热值成分被广泛地用在营养品中。聚葡萄糖在小肠中既不能被消化也不能被吸收,相当多的部分发现于粪便中。
专利申请WO04/026316特别地教导使用聚葡萄糖预防由结肠的微生物群引起的失衡诱导的酸中毒效应,特别是由益生剂比如菊糖和FOSs诱导的那些。
因此,认为聚葡萄糖促进特定的菌群消耗乳酸,抗衡FOSs诱导的其过量生成。
发明内容
根据本申请公司的知识从所有上述的得出,不存在单一的多糖组合物,其能满足肠用有效保护性组合物的所有要求。
因此,本发明的目的是克服现有技术的缺点。
因此,本申请公司发现加入支链麦芽糊精有利地使有可能符合至今已据说不可调和的所有目标,其通过以许多调查研究为代价设想和开发了一种用于结肠的新的富含纤维的抗炎和/或镇痛组合物,其满足所有上述标准,即对结肠粘膜的有保护效应、中等降低结肠的pH、且有利于产生丙酸菌和丁酸菌,及以较少程度产生乳酸菌。
因此,本发明的一个目的是支链麦芽糊精,其具有15%至35%的1→6糖苷键,还原糖含量小于20%,多分子性指数小于5,数均分子量Mn至多等于4500g/mol,用于治疗人体或动物体的方法。
为了本发明的目的,术语“支链麦芽糊精”是指在申请公司所拥有的专利EP1.006.128中描述的麦芽糊精。
在专利EP1.006.128中描述的所有的支链麦芽糊精组合物适于制备根据本发明的肠用抗炎和/或镇痛组合物。
根据一个优选的变型,所述支链麦芽糊精的还原糖含量为2%至5%,数均分子量Mn为2000至3000g/mol。
根据AOAC方法No.2001-03(2001)测定,以干物质计,支链麦芽糊精的总纤维含量大于或等于50%。
本发明的一个目的是用于治疗人体或动物体的富含纤维的组合物,特征在于其包含作为活性成分的支链麦芽糊精。
本发明的肠用富含纤维的抗炎和/或镇痛组合物包括以干重计0.5%至20%,优选5%至10%的所述支链麦芽糊精,以便构成纤维的充足供应和对结肠的保护效应。
本发明的肠用抗炎和/或镇痛组合物中支链麦芽糊精的重量低于0.5%时,则供应的纤维不足以具有可检测效应。
这些支链麦芽糊精具有难消化的特征,其导致它们在被预防的小肠中同化。
他们提供了对代谢和肠平衡有益的难消化的纤维源。
实际上,它们的高含量的1-6糖苷键赋予它们益生性质,其为完全特异性的:实际上发生的是丁酸产生(butyrogenic)菌、乳酸菌或丙酸菌代谢这些高支链化合物。
这些支链麦芽糊精也促进双歧杆菌的生成,其对不想要的细菌有害,从而促进α-和β-糖苷酶活性的表达。
本发明的肠用抗炎和/或镇痛组合物使有可能以2至10倍,优选3至8倍刺激盲肠内含物和粪便中α-和β-糖苷酶的酶活性,如将在下文中举例说明的。
这导致其对消费者的健康完全有益的性质。
而且,结肠中的微生物对本发明的抗炎和/或镇痛组合物的支链麦芽糊精的消耗将导致盲肠、肠和粪便内含物的pH降低0.5至1个单位,从而反映出所述微生物均衡生长。
使用本发明的抗炎和/或镇痛组合物也使有可能增加盲肠中挥发性有机酸的产生,其中有机酸选自乙酸、丁酸和丙酸,优选丙酸和丁酸。
对结肠粘膜的保护效应特别地在给药TNBS后的动物中证实,且由显著的结果反映,如将在下文中举例说明的。
向动物继续正常饲喂,其明显地被保护而对抗由TNBS诱导的坏死性炎症,如由在肠上皮中测定的髓过氧化物酶(或MPO)活性降低证实。
实际上,该MPO活性反映了嗜中性粒细胞浸润进入吞噬体和细胞间隙,并使有可能定量与其直接相关的炎性过程。
因此,本发明的肠用抗炎和/或镇痛组合物可使肠上皮的髓过氧化物酶活性从5%降低至40%,优选从7%降低至35%。
该作用明显地反映出所述组合物抗肠炎的保护效应,使有可能设想制备肠用抗炎和/或镇痛组合物,其改善人类和动物患者的健康。
而且,发现本发明的支链麦芽糊精不会产生渗透性腹泻,即使以大剂量使用时。
当消耗低分子量的发酵性碳水化合物,例如乳果糖和果糖寡聚糖时,观察到渗透性腹泻现象。
该现象表现为大便含水量增加和粪便内含物渗透压增加,有可能该含水量增加达到出现腹泻的程度。出人意外的和意想不到的,本发明的支链麦芽糊精不会引起该现象,尽管它们为发酵性的。
在饮食中,本发明的肠用抗炎和/或镇痛组合物可以为即可使用的形式,或者饮料形式,比如果汁、汤或酸乳形式或加入早餐谷类食品中。
而且,所述的组合物可用于动物,更特别地用于猫、狗、猪、兔或对肠炎敏感的其它家畜、免疫力降低的动物。
该组合物也被提议用于补充患有IBDs的个体的饮食、以及用于患有肠易激综合征的个体、和患有旅行者腹泻和其病因学通常未知的腹痛个体。
就药用而言,本发明的肠用抗炎和镇痛组合物可以包括支链麦芽糊精和至少一种其它活性成分,二者比例取决于所考虑活性成分的性质。
其它活性成分优选地为肠用抗炎药。
在治疗IBDs中,例如,可提议两种类型的治疗:
-一种治疗,其使用源自水杨酸化合物的药物,例如柳氮磺吡啶或其衍生物,比如5-氨基水杨酸盐(5-ASA),
-一种治疗,其基于肾上腺皮质激素家族的药物,比如可的松或泼尼松龙。
本发明的一个实施方案涉及如上所述的包含支链麦芽糊精的组合物,特征在于其还包含至少一种选自柳氮磺吡啶和其衍生物或肾上腺皮质激素类的活性成分。
根据一个具体的实施方案,本发明涉及如上所述的组合物,其包含支链麦芽糊精,还包含选自柳氮磺吡啶和其衍生物的活性成分,特征在于支链麦芽糊精的重量与柳氮磺吡啶或其衍生物之一的重量之比为2至30。
本发明的一个具体的实施方案涉及如上所述组合物,其包含支链麦芽糊精,还包含选自肾上腺皮质激素类的活性成分,特征在于支链麦芽糊精的重量与肾上腺皮质激素类的重量之比为2至250。
典型地,本发明的组合物为液体、粉剂、糖浆、栓剂、片剂或锭剂的形式。
本发明的一个实施方案涉及用于治疗人体或动物体的试剂盒,其包括:
a)如上所述的第一组合物,包含支链麦芽糊精;和
b)第二组合物,包含肠用抗炎药。
本发明的一个实施方案为治疗或预防肠炎和/或平息肠痛的方法,其包括给药至个体足够的治疗量的支链麦芽糊精。

如上所述的包含支链麦芽糊精的组合物可有利地与包含肠用抗炎药的第二组合物联合给药至个体或动物。在治疗期间,所述两种组合物可以随时间同时给药或顺次给药。给药第二组合物的方法取决于所使用的肠用抗炎药。
本发明的一个具体的实施方案为治疗或预防肠炎和/或平息肠痛的方法,其包括随时间同时或顺次给药在如上所述的试剂盒中描述的两种组合物。
本发明的一个实施方案是支链麦芽糊精在制备治疗或预防肠炎和/或平息肠痛的组合物或试剂盒中的用途。
在可以治疗或预防的所述疾病和疼痛中,可提及的包括慢性炎症性肠病、肠易激综合征、旅行者腹泻或腹痛。在慢性炎性肠疾病中,可提及的包括溃疡性结肠炎和Crohn’s病。
至于本发明的组合物的镇痛作用,估计其与PPARγ和MOR受体的表达有关。
PPARγ(或过氧化物酶体增殖物活化受体γ)属于核受体家族部分。它们特别地被脂肪酸活化,并参与转录应答中代谢和营养信号的转导。它们在保持肠粘膜的完整性中起主要作用。
本领域技术人员已知PPARγ主要参与结肠炎症的调节。它们也在结肠癌的情况下表达,并且它们的活化抑制细胞生长和细胞分化。
MORs(或阿片样物质受体)发现于中枢和周围神经系统中,并可特别地存在于结肠中。MORs的主要功能是镇痛功能。另一个功能是抑制肠运动。MORs也参与肠炎的调节。
如将在下文中举例说明的,值得注意的是本发明组合物可使过氧化物酶体增殖物活化受体γ(PPARγ)的活性增加1.2至3倍,优选1.6至2倍。
类似地,本发明组合物可使μ阿片样物质受体(MORs)的数量增加1.2至10倍,优选2.5至7.5倍,更优选4至5倍。
最后,所述组合物特别适于应激个体,其中所述应激以肠水平显示自身。
阅读说明性的和非限定性的下述实施例将更清楚地理解本发明。
具体实施方式
实施例1
使用实验室大鼠来研究给药TNBS后在它们的食物中的组合物对保护它们的结肠粘膜的效果,所述组合物包含本发明的支链麦芽糊精(MDB)或葡萄糖(对照)与不溶性纤维(废玉米颗粒或纤维素)的组合。
混合本发明的不溶性纤维(本发明的实例为废玉米颗粒)和MDBs以便模拟从根据健康管理局推荐的饮食中的谷类制品中摄入的纤维。
而且,根据类胡萝卜素和多酚(特别是酚酸和阿魏酸)的富含来选择废玉米颗粒。
在该实施例中选择的本发明的支链麦芽糊精具有15%至35%的1→6糖苷键,还原糖含量为2%至5%,多分子性指数小于5,数均分子量Mn为2000至3000g/mol:

将Sprague Dawley来源的64只OFA大鼠分成8组,每组在它们的食物和饮料中接受其中组成为下述表I所示的特定饮食。
葡萄糖和支链麦芽糊精在营养摄入的饮料中以5%重量/重量的比例存在。纤维素和废谷粒在营养摄入的食品中以5%重量/重量的比例存在。

在隔离一周后(该隔离期间动物接受标准食物和饮用水),大鼠消耗根据在表1中描述的饮食的食物和饮料20天。
然后,将它们禁食24小时。
在第21天,通过直肠内注射在表1中规定的产品处理所述动物。
5组至8组动物接受直肠内注射500μl的在乙醇中稀释成40%GayLussac的TNBS,而1组至4组动物接受直肠内注射500μl的9‰的NaCl。
以10mg/kg体重每日注射TNBS。
已知该剂量产生严重的但可逆的炎症反应。
在注射后3天内监测动物体重的变化。
在第24天,通过CO2窒息处死动物。
称重动物,接着在尸体解剖后,取出结肠,排空,然后称重。
随后用肉眼观察,并给出Wallace得分。
也测定在肠上皮中的髓过氧化物酶(或MPO)活性。
该活性反映了嗜中性粒细胞浸润进入吞噬体和细胞间隙,并使有可能定量与其直接相关的炎性过程。

至于MPO活性的测定,需要根据下述方案制备结肠。
将结肠片段悬浮在6ml十六烷基三甲基溴化铵缓冲液(在50mM磷酸盐缓冲液中的0.5%的HTAB,pH7.0)中。因此,由此处理得片段用Polytron研磨和均质化10秒。使用来自Sonics and Materials Inc.,Danbury,Connecticut,USA的Vibra Cell 500 watts装置(转化器动力500W,在探针上消散的动力为30%-即150W/cm2,脉冲器在位置2-即第二种的66%)超声处理每种样品。
超声处理后,在再次在相同的条件下超声处理前,接受3次冷冻-融化周期。随后,在4℃,10000g样品离心。
在470nm和在30℃监测表观密度给出活性值(表示为吸光单位/分/克结肠)。在下表III和IV中给出了得到的所有结果(值表示为在每组8只动物上进行的量度结果的平均值±标准偏差)。

体重变化显示在第20天所有的动物具有同样的体重。在第21天,所有的动物体重减轻,因为它们在直肠内注射前禁食。
在第22天,已经用直肠内注射9‰NaCl的所有动物恢复了大量的体重。
组8是唯一接受炎症-引发给药TNBS的组,其体重变化(evolution)从第22天上升。
组5、6和7的动物的体重变化仅仅从第23和24天适当上升。
因为组8的动物体重曲线与没有接受TNBS的动物的相同,这表明这些动物从第21天开始进食。因此,这些动物被保护以对抗TNBS诱导的坏死性炎症。

该结果通过测定Wallace得分来证实。实际上,接受本发明组合物与废谷粒和TNBS的动物的Wallace得分为3.9,与之相比,其它接受TNBS的组得到的平均得分为5.8、5.9和6.5。
3.9的平均得分显示出组8动物的炎症水平明显较低。
首先,结肠重量测定的结果显示出接受TNBS注射的动物的结肠比没有用TNBS处理的动物的结肠更重。
这一现象特别地是由于侵入发炎结肠粘膜的水肿。
与没有接受TNBS的组的动物结肠的平均重量相比,组8的动物结肠的平均重量保持很高,但是其仍然是所有用TNBS处理的那些组动物中最低的重量。
至于MPO活性的测量,其对没有用TNBS处理的组的动物来说显然是低的,其再一次表明与用TNBS处理的其它组动物相比,组8显示出最小的MPO活性。
因此,组8的动物明显地被保护以对抗TNBS诱导的坏死性炎症。
实施例2
使用实验室大鼠来研究本发明的支链麦芽糊精(与实施例1的那些相同)和葡萄糖(对照)对在雄性Wistar大鼠中给药TNBS诱导的结肠刺激的作用,和对在有害光刺激回避调节测定中它们的认知行为的作用。
该测定使用对亮光环境反感的大鼠。原理为经历该环境的动物为在调节测定的情况下学习更慢的动物。
首先,大鼠在操作调节的情况下学习控制其有害光环境:动物学习压下活动杆(LA)以便获得30秒的黑暗期作为正增强。
所述装置也包括另外的杆,当其开动时,不可能获得光:非活动杆(LI)。
活动杆和非活动杆被压下的次数使有可能评价大鼠控制活动的水平。
通过比较在“光”期两个杆(LA对LI)中的每个被压下的次数来评价学习的获得(辨别两个杆)。
将48只雄性Wistar/AF SPF大鼠分成4组,其在它们的营养摄入中接受如下表V所示构成的饮食。

在第15天,动物被禁食48小时。
在第17天,麻醉大鼠,4组(组2至组4)中的两组接受以3mg/kg体重的比率的结肠内给药500μl的20%Gay Lussac的TNBS-乙醇(即每只大鼠1mg-该剂量被认为会诱导与微弱肠刺激有关的疼痛)。
从第17至第23天,动物继续接受用MDBs或用葡萄糖补充的饮食。
在第22天,进行认知测定:有害光刺激回避试验(ALSAT)。
下表VI和VII给出了将ALSAT测定应用到各组动物的结果。表VII给出了在该测定过程中压下的总数。

根据表VIII,仅接受本发明的MDB和有或没有TNBS的动物能够区分开LA和LI,更明显地压LA,因此,证实了所述产品对抗由TNBS诱导的疼痛的积极作用。

与接受葡萄糖的动物相比(对照动物),当动物接受本发明的MDB时,表IX的肉眼观察的得分较低。
该观测值与显微镜观察较好地相关,因为对组4给予1.9的得分,而对组2给予2.8。
因此,可以得出所有的动物显示出炎症,但严重程度不同。
当动物接受本发明的MDB时,肠病不太严重,从而证实了之前说明的肉眼观察的结果。
这些结果与学习测定结果有关,证实显示出学习较好的动物被保护对抗TNBS诱导的疼痛,证实了本发明组合物的镇痛性质。
实施例3
通过测定血触珠蛋白研究本发明的支链麦芽糊精(实施例1的那些)对抗小猪肠炎的保护效应。
触珠蛋白为一种由肝脏合成的血浆糖蛋白(α2-球蛋白),能结合血红蛋白。通过使用本领域技术人员可使用的诊断试剂盒的免疫学方法测定触珠蛋白的含量。
血触珠蛋白含量在在炎性综合症中增加,与病因无关。其动力学缓慢,因此,如果其水平高,这反映出炎症已经存在一段时间的事实。
相反,其水平在血液中降低反映出对炎症的保护效应。
在研究开始时,在一组128只称重7.2±1.04kg的断奶小猪中进行试验,将该组分成4批,每批32只动物,每批的活重和性别相同(16只阉割的雄性和16只雌性)。
如下进行试验处理(总共77天的时间):
第1批:用常规饮食饲喂对照动物,
第2批:动物接受占食物重量2%比例的本发明MDB,
第3批:医学处理动物,在测定期间(14天)用包含比例分别为1000和400mg/kg的两种抗生素(金霉素和螺旋霉素)的食物饲喂,然后在试验的剩余时期(15-77天)用常规饮食再次饲喂。
第4批:医学处理动物,在试验期间(14天)用包含比例分别为1000和400mg/kg的两种抗生素(金霉素和螺旋霉素)的食物饲喂,然后在试验的剩余时期(15-17天)接受比例为食物重量2%的本发明MDB。
在试验结束时,从每亚组的6只小猪采集血样,测定其中的触珠蛋白含量(用血液的mg/ml表示)。
下表XI给出了得到的结果。

结果表明用本发明的肠用富含纤维的抗炎和镇痛组合物饲喂的动物(第2批)的血液触珠蛋白含量显著地比接受常规饮食的动物的低,因而,其反映出血液炎症水平和全身性炎症水平比对照组低。
在第4批中,结果低于对照组,即使其不重要。然而,这一降低支持了预期的效果,即,抗炎状况较低。
实施例4
在实验室大鼠中研究本发明的支链麦芽糊精(实施例1相同的那些)对肠发酵的作用。
将40只Sprague Dawley来源的OFA大鼠分成4组,其在其摄食中接受下述饮食,下表XII给出了所述饮食的详细成分。
组4接受用果糖寡聚糖补充的摄食(RaftiloseP95,由Orafti公司销售)。

一周的隔离期后(在该隔离期内动物接受标准摄食和饮用水),大鼠消耗所述食物36天。
在第0天,动物被禁食24小时。随意饮水。在第1天,收集粪便。
给予动物在表XII中描述的饮食。
在第28天,动物被禁食24小时。随意饮水。
在第29天,再次收集粪便。
在第36天,处死动物。
进行器官的一般肉眼观察。结扎并取出盲肠。称重充满的盲肠、盲肠的内含物和空盲肠。
测定粪便和盲肠内含物的pH和固体。
也评价粪便的酶活性(α-糖苷酶和β-糖苷酶)。
研究挥发性脂肪酸在盲肠内含物(乙酸、丙酸、丁酸)中的分布。
下表XIII给出了有关充满的盲肠重量、空盲肠重量和盲肠内含物pH的数据(表示为每批10只动物的平均值±标准偏差)。

表XIII显示了与接受标准摄食或包含10%葡萄糖的食物的动物相比,接受10%的本发明MDB或10%RaftiloseP95的动物的充满的盲肠和空盲肠的重量明显地较高。
与对照批相比,接受10%的MDB的动物的充满的盲肠的重量增加46%,接受10%的RaftiloseP95的动物增加53%。
对于空盲肠的重量部分,接受MDB的批次改变30%,接受RaftiloseP95的批改变50%。
这些结果表明MDB和RaftiloseP95增加了盲肠的重量,因此,增加了盲肠的细菌量以及盲肠粘膜的重量,从而导致了针对炎症的物理保护。
该表也显示出对于接受MDB的批次的盲肠内含物的pH有明显地降低,从而反映出实质上的盲肠发酵活性。
这一pH的降低反映出酸性分子增加,有利于碱性分子减少,所述碱性分子实质上具有更大的攻击性。
对于所述部分,RaftiloseP95没有显示出这些性质,因为盲肠内含物的pH没有显著地降低。
表XIV给出了有关盲肠内含物的挥发性脂肪酸分布的数据。

该表显示出用10%的MDB补充的摄食使盲肠内含物的丙酸明显地增加。
对于接受RaftiloseP95的批,也获得该结果,但是以较不明显的方式。
对于盲肠内含物中乙酸的量没有出现显著性差异。

这些结果使不可能在接受MDB的动物中观察到粪便pH的明显降低,尽管观察到盲肠pH降低。
另一方面,接受RaftiloseP95的动物的粪便pH降低了。

在第29天,通过给药10%的MDB极大地增加了糖苷酶活性。这也是接受10%的RaftiloseP95动物的情形,但是以较不明显的方式。
实际上,与对照批相比,在接受MDB的批中,观察到α-糖苷酶和β-糖苷酶分别增加310%和298%,而对于RaftiloseP95批,观察到其分别仅增加172%和63%。
粪便中糖苷酶活性的增加导致结肠消化残存的多糖。
因此,该高糖苷酶活性可以导致某些多酚(在修复结肠炎中的重要参与者)的生物利用度降低以及氧化应激减低。
实施例5
在实验室大鼠中研究本发明的支链麦芽糊精(与实施例1的那些相同)对丁酸生成的作用。
将18只Fischer实验室大鼠分成3组,其在它们的摄食中接受下表XVIII给出的饮食。
组3接受用果糖寡聚糖补充的摄食(Actilight,由Beghin-Meiji公司销售)。

对于接受MDB的动物,盲肠丁酸的含量增加,因此,将其按动物中产生丁酸的糖(butyrogenic glucidic)基质来分类。
丁酸是细胞生长和分化的一个重要因素,从而证实了MDB抗结肠炎症的保护作用。
实施例6
在实验室小鼠中研究本发明的支链麦芽糊精(与实施例1的那些相同)和葡萄糖(对照)对参与肠炎和镇痛的各种细胞受体生成的作用。
将20只7周龄Balb/c小鼠分成2组。
一组接受由10%的葡萄糖的饮用水溶液组成的饮料,另一组接受10%的MDB的饮用水溶液。
动物接受该饮料的非限制供应和标准小鼠食物29天。
在第29天,处死小鼠,取出结肠并分析下述标志物:
-过氧化物酶体增殖物-活化的受体(PPARγs)
-μ阿片样物质受体(MOR).
为了评价本发明的支链麦芽糊精在炎症的生理调节中的作用,使用由Clontech Laboratories Inc.公司销售的NucleoSpinRNA II试剂盒来分离取出的结肠的总RNA。使用反转录酶将总RNA反转录成cDNA。
扩增反转录反应,用使用引物的实时PCR(Applied Biosystems)定量PPARγ和MOR。结果用每个体内β-肌动蛋白控制的mRNA分子的mRNA分子数表示。
下表XX给出了测定大鼠结肠粘膜中PPARγ和MOR后获得的结果的平均值。

将10%的本发明的MDB引入食物中29天观察到这两个因素相当大的增加:
-PPARγ增加1.72倍,
-MOR增加4.43倍。
用10%的MDB处理的批的结果比用10%的葡萄糖的对照批获得的结果显示出明显较大的量(对PPARγ,p<0.03,对MOR,p<0.04)。
因此,本发明的MDB有助于特别地通过保持肠粘膜的完整调节可能的炎症。
实际上,PPARγ分子数的增加显示出当动物消耗MDB时结肠具有较好的抗炎状况。
对于该部分,MOR疼痛受体数的增加与内脏对疼痛敏感性的降低同义。这些结果全部与实施例2的结果一致,其中MDB的镇痛作用已经被在动物认知行为上的作用证实。
Claims (14)
1.支链麦芽糊精,其具有15%至35%的1→6糖苷键,还原糖含量小于20%,多分子性指数小于5,以及数均分子量Mn至多等于4500g/mol,所述支链麦芽糊精用于人体或动物体的治疗方法中。
2.如权利要求1所述的支链麦芽糊精,特征在于所述的支链麦芽糊精的还原糖含量为2%至5%,数均分子量Mn为2000至3000g/mol,所述支链麦芽糊精用于人体或动物体的治疗方法中。
3.如权利要求1和2中任一项所述的支链麦芽糊精,特征在于根据AOAC方法No.2001-03测定,以干物质计,所述的支链麦芽糊精的总纤维含量大于50%,所述支链麦芽糊精用于人体或动物体的治疗方法中。
4.用于治疗人体或动物体的富含纤维的组合物,特征在于其包括作为活性成分的如权利要求1至3中任一项所述的支链麦芽糊精。
5.如权利要求4所述的组合物,特征在于其包含以干重计0.5%至20%,优选5%至10%的所述的支链麦芽糊精。
6.如权利要求4和5中任一项所述的组合物,特征在于其为饮料、汤或酸乳形式或加入到早餐谷类食品中。
7.如权利要求4或5中任一项所述的组合物,特征在于其还包含至少一种选自柳氮磺吡啶和其衍生物及肾上腺皮质激素类的活性成分。
8.如权利要求4至5中任一项所述的组合物,还包含选自柳氮磺吡啶和其衍生物的活性成分,特征在于支链麦芽糊精的重量与柳氮磺吡啶或其衍生物之一的重量之比为2至30。
9.如权利要求4和5中任一项所述的组合物,还包含选自肾上腺皮质激素类的活性成分,特征在于支链麦芽糊精的重量与肾上腺皮质激素类的重量之比为2至250。
10.如权利要求7至9中任一项所述的组合物,特征在于其为液体、粉剂、糖浆、栓剂、片剂或锭剂的形式。
11.用于治疗人体或动物体的试剂盒,其包括:
a)如权利要求4至10中任一项所述的第一组合物;和
b)第二组合物,包含肠用抗炎药。
12.如在权利要求1至3中任一项所述的支链麦芽糊精的用途,用于制备治疗或预防肠炎和/或平息肠痛的组合物或试剂盒。
13.如权利要求12所述的用途,用于制备治疗或预防慢性炎症性肠病、肠易激综合征、旅行性腹泻或腹痛的组合物或试剂盒。
14.如权利要求13所述的用途,特征在于所述慢性炎症性肠病选自溃疡性结肠炎和Crohn’s病。
