Abstract

Description
背景技术
切向流过滤(TFF)是一种分离方法,该方法在尺寸或分子重量差的基础上使用膜分离液体溶液或悬浮液中的组分。应用包括蛋白质和其它生物分子、例如核苷酸、抗原和单克隆抗体的浓缩、澄清和脱盐;缓冲液交换;工艺过程开发;膜选择研究;预色谱澄清以去除胶体微粒;小分子、例如葡萄糖和抗生素的去除热原;细胞培养物、溶菌产物、胶体悬浮液和病毒培养物的获取、洗涤或澄清;和样品制备。
在TFF中,使要过滤的溶液或悬浮液以错流的模式通过膜的表面。用于过滤的驱动力是横跨膜的压力,通常由蠕动泵产生。滤出液通过膜表面的速度也控制过滤速率,并且有助于防止膜的堵塞。因为TFF使通过膜表面的渗余物再循环,所以使膜的污垢最小化,保持了高的过滤速率,并且提高了产物回收率。
常规的TFF装置由多个元件组成,包括泵、进料溶液储罐、过滤组件和用于连接这些元件的导管。在使用中,将进料溶液从进料溶液储罐中导入过滤组件中,同时将来自过滤组件的渗余物从过滤组件再循环到进料溶液储罐中,直到得到需要的渗余物量。膜被夹在上部和底部歧管或支撑物之间,其中歧管或支撑物用于提供精确的抵抗装置内部水压的机械约束,并且还用来将过滤流分配穿过装置内的多个流动通道。这些歧管或支撑物通常由不锈钢制成,并且在每次使用前必须进行清洁和验证,尤其在生物制药和其它卫生应用中。这是昂贵和费时的方法。
期望提供一种在更换过滤介质时不需要上述清洁和验证步骤的过滤装置。
发明内容
根据本发明的教导,提供一种切向流过滤装置,其中在过滤元件和上部和底部支撑物或歧管之间设有衬垫。该衬垫结合歧管中预先存在的流动通道和入口以及出口端口。衬垫由廉价材料制成,因此在一次使用后可以任意处置,使得处理它们比清洁常规的歧管更成本有效。另外,衬垫可以被预杀菌。为了在操作条件下提供足够的强度和刚度,衬垫具有与支撑板邻接的肋的栅格图案,以有助于防止衬垫在夹紧力作用下扭转。

具体实施方式
首先参考图1,图中显示了根据本教导的过滤装置10的分解图。装置10包括上部支撑板12和隔开的底部支撑板13。支撑板12、13优选由不锈钢制备,并且足够坚硬和耐用,以提供抵抗内部水力操作压力例如50-60psi的组件的精确和有效的机械约束力。在支撑板12,13和组件的每层中设置孔28,用以容纳拉杆或螺纹销或螺栓14或其它夹紧装置以确保组件结合在一起。设置垫片15,并且它可以是弹性加载的。在支撑板12,13中不存在过滤流通道。
在装配状态的支撑板12之下设置的是可任意处理的衬垫16。衬垫16优选由适合使用的廉价材料制备,该材料在药物检验(优选是经官方批准的)上是可接受的。合适的构造材料包括塑料,例如聚苯乙烯,优选聚烯烃,例如聚丙烯、聚乙烯、它们的共聚物和混合物。聚砜由于其强度和刚度而尤其优选。衬垫16优选被浇铸成具有通道和开口。可选择地,并且次优选地,它可以通过铣削、钻孔和其它类似方法形成。
如图2最清楚地所示,衬垫16包括第一端口17A、5个子端口17C、第二端口17B和4个子端口17D。根据其在组件中的定位,端口17A用于引导进料或除去渗余物,并且端口17B用于除去渗透物,与常规一样同时防止滤出液与渗余物或进料的混合。端口17A以歧管布置的方式与5个子端口17C相连。端口17B以类似的方式与4个子端口17D相连。为了提供足够的间隔并且避免与其它部件干涉,端口17A和17B可以位于衬垫的相对侧。然而,在所示的应用中,当间隔充足或没有发生干涉时,它们可以设置在同一侧。每个端口17A、17B与流动路径或通道流体连通,该流动路径或通道与常规一样和各个孔流体连通,以容纳进料、渗余物或渗透物的流动,由此在装置中限定出过滤流的多个流动路径。
通道优选是锥形的,随着远离它们各自的端口而变窄,以使压力在每个子端口17C和17D处标准化。
回到图1,显示了安置在衬垫16之下的过滤元件20。过滤元件20可以是单个膜,并且优选是多个堆叠膜,例如堆叠的超滤或微滤膜,最优选以盒的形式设置。尽管显示了膜的单个盒子,但是本领域技术人员应当理解,可以使用多个盒子。合适的盒子是以名字出售的,并且可商购自MilliporeCorporation。
在过滤元件20之下设置的是第二衬垫22。优选第二衬垫22在结构上与第一衬垫16相同,但是如图所示,当装置处于装配状态时,衬垫22被相对于第一衬垫16的位置反转。这允许端口17A与装置的进料端口在其正常方向上相通,而当在反转位置上与渗余物端口相通。衬垫的端口17B与渗透物端口在两个方向上都相通。
优选衬垫l6、20的一侧包括多个相互接合(inter-engaging)的肋,在图3中显示的最清楚。肋给衬垫提供了加强的刚度,并且可以在模塑工艺中形成。肋定位在与支撑板接触的衬垫侧。肋从衬垫的一侧向另一侧延伸,除了被端口中断处。在图示的肋结构中,通过多个纵向和横向的肋形成栅格,具有9个横向肋25A-25I和9个纵向肋26A-26I。横向肋优选彼此平行,而纵向肋优选彼此平行并且垂直于横向肋。横向肋25B-25H优选等间距,但是横向肋25A和25B之间以及25A和衬垫的侧壁225之间的各自间隔较小,肋25H和251之间以及肋251和衬垫相对侧壁226之间的间隔也是这样。在侧壁处将肋更靠近地聚集一起给衬垫提供了额外的强度。纵向肋26A-26I全部等间隔,并且间隔优选与横向肋25B-25H的间隔相同或基本相同,使得在肋25B-25H和肋26A-26I之间限定的栅格包括多个正方形,在肋25H、侧壁226和肋26A-26I之间形成的栅格包括多个长方形,并且在肋25B、侧壁225和肋26A-26I之间形成的栅格包括多个长方形。在渗透物端口17B的周围形成了U形肋27A,就像渗余物端口17A周围的U形肋27B一样。
图示的复杂的肋结构给衬垫提供了强度和刚度。当装配时,具有施加在过滤元件20和衬垫上的明显的夹紧力,并且在衬垫的光滑侧和过滤元件20之间产生了密封。没有肋结构的情况下,衬垫将不能保持平坦,因此不能与过滤元件20完全密封。肋使得能够在本发明的过滤装置中有效地装配衬垫,在施加压力下处于密封结合状态,而不需要在支撑板上具有相应凹槽来与肋相配。因此,支撑板的与衬垫肋邻接的相应表面优选是平坦的,并且不需要特别设计来配合衬垫。
Claims (8)
1.一种过滤装置,包括:
上部板;
与所述上部板相隔开的底部板;
定位在所述上部板和所述底部板之间的过滤部件;
定位在所述上部板和所述过滤部件之间的可任意处理的第一衬垫,所述第一衬垫具有流体入口、流体出口、多个流体入口孔、和在所述第一衬垫面向所述上部板的一侧上的肋图案;
定位在所述底部板和所述过滤部件之间的可任意处理的第二衬垫,所述第二衬垫具有流体入口、流体出口、多个流体出口孔、和在所述第二衬垫面向所述底部板的一侧上的多个肋;
其中所述上部板和底部板没有流体流动路径,并且各自具有与相应衬垫上的所述多个肋相配的基本平坦的表面。
2.如权利要求1所述的过滤装置,其中所述第一和第二衬垫上的所述相应的肋状图案限定栅格图案。
3.如权利要求2所述的过滤装置,其中当所述栅格图案与相应上部板和底部板相配并承受压力时,所述栅格图案将所述衬垫保持在平坦结构中。
4.如权利要求1所述的过滤装置,其中在每个相应衬垫上的所述多个肋是相互接合的。
5.如所述权利要求1所述的过滤装置,其中所述上部板和底部板包括不锈钢。
6.如权利要求1所述的过滤装置,其中每个所述衬垫包括聚砜。
7.一种用于过滤装置的衬垫,其适于在支撑板和过滤元件之间密封,所述衬垫包括适于邻接所述过滤元件的平坦面和具有多个适于邻接所述支撑板的相互接合的肋的相对面,所述相互接合的肋为所述衬垫提供足够的强度,以便使所述衬垫能够在压力下在所述装置内有效地密封。
8.如权利要求7所述的衬垫,其中所述肋限定栅格图案。
