L'invention concerne des dérives du glycérol intéressants pour leur activité anti-tumorale, une méthode de préparation de ces dérivés et des compositions thérapeutiques les contenant.

et, pour l'obtention des composés Ib ou le dans lesquels Ri représente l'hydrogène, l'hydrogénolyse du groupe protecteur -SO2CH2Ò.
Dans certains cas, le réactif Z peut être également le solvant de la réaction, et la définition «un excès stœchiométrique» n'a alors plus de signification.
Comme la réaction est identique pour l'obtention des composés la, Ib et le, elle sera illustrée uniquement pour le composé la par le schéma réactionnel I (page 5).
Les dérivés du glycérol et, plus particulièrement, les dérivés de la phosphocholine, sont déjà décrits, par exemple, dans le brevet EP 130 527; un de ces composés efficaces pour le traitement du cancer: l'oc-tadécylamino-3-tétradécyl-0-1-propanediol-1,2-phosphocholine-0-2, et un composé de référence: Et-18-OCH3 (méthoxy-PAF, Andreesen, 1988), ont été retenus pour la comparaison avec les composés de l'invention. Les résultats ont montré que les composés de l'invention ont une activité anti-tumorale plus grande, comme le prouvent les résultats du test pharmacologique ci-joint.
L'invention concerne finalement des compositions thérapeutiques contenant un des composés de l'invention, comme ingrédient actif, en mélange avec des diluents et/ou supports appropriés.
Les différents composés de départ IIa, IIb et Ile peuvent être préparés selon les schémas réaction-nels II, III, IV et V (pages 6 à 9).
Le composé de départ lia peut être préparé selon le schéma réactionnel II : la particularité de ces réactions réside dans l'étape Sa -» 4a : le mécanisme comprend 2 substitutions de type SN2, avec la migration des groupes -OR3 et -NR1R2 comme le décrit K. Suzuki, K. Okano dans Svnthesis 723 (Sept. 1983).
Le composé de départ IIb peut être préparé:
- selon le schéma réactionnel III : le composé IIb peut comprendre un groupe protecteur lorsque le radical Ri, du composé final Ib, est l'hydrogène; une déprotection par hydrogénolyse est ensuite conduite sur le composé final;
- selon le schéma réactionnel IV, voie A ou B, spécifiquement lorsque R3 représente le radical -CONH-alcoyl ou -CON-dialcoyl; le composé de départ 6b du schéma réactionnel IV est identique au composé 2a du schéma réactionnel II.
En ce qui concerne le composé Ile (schéma réactionnel V), on se référera au composé IIb, premier alinea.
Ces étapes sont décrites dans les exemples préparatifs suivants.

18 g (30 mmol) du composé 2§ sont dissouts dans 100 ml d'éther anhydre et 50 ml de dichlorométhane. 6,84 g (60 mmol) de chlorure de méthanesulfonyle dans 50 ml de dichlorométhane sont alors ajoutés sous agitation et le mélange est ainsi porté au reflux pendant 5 heures. Après addition d'eau, la phase organique est décantée, séchée et évaporée. Le produit brut obtenu, est chromatographié (éluant: chloroforme), pour obtenir 16,7 g du composé 3a (80%).

Le composé ,1b : phényl-2-hydroxy-5-dioxane-1,3, est préparé selon Verkaade P.E. et Van Roon J.D. (Ree. Trav. Chim. Pays-Bas, 61. 831,1942). mp = 80°C.
10 g de sels de sodium du compose Ib, obtenu par réaction avec de l'hydrure de sodium dans du dimé-thylformamide, est traité avec 16 g de iodure de méthyle. Le mélange est agité à 50°C pendant 5 heures, puis la diméthylformamide est éliminée sous vide. Le résidu ainsi obtenu est dissout dans du dichlorométhane, lavé et séché. Benzvloxv-3-méthoxv-2-propanol (3b)
4,2 g du composé 2b sont dissouts dans 10 mi de tétrahydrofuranne à 0°C. Puis on ajoute doucement et sous agitation, une solution de BH3 dans du tétrahydrofuranne (1 M, 30 ml). L'agitation est ainsi maintenue pendant 48 heures à température ambiante. Le mélange est ensuite refroidi à 0°C, plongé brusquement dans de l'eau froide et extrait avec de l'éther. Le solvant est éliminé et le produit brut obtenu est chromatographié (éluant: éther de pétrole/éther successivement à 80:20 puis 70:30 par volume), pour obtenir 2,6 g (62%) du composé 3b (composé visqueux).

On ajoute, sous agitation, 6,84 g (60 mmol) de chlorure de méthanesulfonyl dans 50 ml de dichlorométhane, à une solution de 5,88 g (30 mmol) du composé 3b et 10 ml de triéthylamine dans 100 ml d'éther anhydre et 50 ml de dichlorométhane; le mélange est ainsi porté au reflux pendant 5 heures. Après addition d'eau, la phase organique est décantée, séchée et évaporée. Le produit brut obtenu est chromatographié (éluant: éther de pétrole/éther 80:20 par volume), pour donner 6 g (74%) du composé 4b (composé visqueux).

2 g (5 mmol) de méthoxy-3-(N-méthyl-octadécylamino)-2-propanol lia et 3 ml de triéthylamine, sont dis-souts dans 20 ml de benzène anhydre, et le mélange est refroidi à 5°C sous circulation d'azote. 1 g (7 mmol) de chloro-2-oxo-2-dioxaphospholane-1,3,2 dans 4 ml de benzène, est ajouté sous agitation, et l'agitation est maintenue toute la nuit. Le sel d'amine est filtré et lavé avec du benzène. Le filtrat est évaporé sous pression réduite jusqu'à siccité. Le résidu est dissout dans 20 ml de cyanure de méthyl anhydre et transféré dans un réacteur. Le mélange est agité pendant 15 heures à température ambiante. Après évaporation du solvant, 30 ml de pyridine anhydre sont ajoutés au résidu obtenu, et le mélange est alors agité pendant 24 heures, à 80°C et sous azote. Une solution de chloro-2-oxo-2-dioxaphospholane (2 g, 14 mmol) dans du benzène (4 ml), est ajoutée, sous circulation d'azote, à une solution refroidie (5°C) et agitée de (N-méthyl-octadécylamino)-3-méthyl-carbamoyloxy-2-propane-ol-1 Mb (2,9 g, 7 mmol) et de 3 ml de triéthylamine dans du benzène anhydre (20 ml). Le mélange est agité pendant 8 heures à température ambiante, puis filtré. Le filtrat est évaporé sous pression réduite. Le résidu est dissout dans CH3CN anhydre (50 ml) et transféré dans un réacteur. 30 ml de CH3CN saturé par NMe3 gazeux sont ajoutés et le mélange est ainsi chauffé à 65°C pendant 24 heures. Le solvant est ensuite évaporé et le résidu obtenu est chromatographié sur silicagel (éluant CHCl3/MeOH 90:10 puis 70:30 et 30:70, puis méthanol) pour obtenir 1,74 g (43%) du composé souhaité.

TOXICITÉ
La toxicité des composés de l'invention, a été déterminée per os sur la souris, par des méthodes usuelles. Leurs DL50 sont supérieures à 650 mg/kg.
PHARMACOLOGIE
Les composés selon l'invention, ont été étudiés pour leur habilité à inhiber la prolifération de cellules tumorales in vitro.
Ils inhibent la prolifération des cellules tumorales HL.60 et A.427 après 24 heures.
HL.60: cellule de leucémie promyélocytique
A.427: cellule de carcinome pulmonaire
Ils montrent un effet cytostatique à la dose de 0,02 mM qui n'est pas une dose toxique pour les deux lignées de cellules tumorales humaines. La lignée de cellules de carcinome pulmonaire semble plus sensible que la lignée de cellule de leucémie promyélocytique.
L'effet des composés selon l'invention, sur la prolifération à long terme, est décrit plus précisément comme suit.
Tous les exemples de l'invention ont été testés et comparés avec deux composés voisins, retenus dans l'état de la technique: - l'octadécyl-0-1 méthylglycéro-0-2 phosphocholine-3 (EM8-OCH3 ou méth-oxy-PAF, Andreesen, 1988).
- l'octadécyl-3 tétradécyl-0-1 propane-diol-1,2-phosphocholine-0-2 [composé (D); Brevet EP 130 527].
Pour cette étude, une lignée de cellules d'adénocarcinome de côlon, appelée aussi HT.29, a été utilisée; ce sont des cellules capables d'adhérer à un substrat. Les cellules HT.29 sont cultivées dans un milieu Me Coy, complété avec du sérum bovin fœtal 10% (SBF; Gibco). Le milieu de croissance contient 100 U/ml de pénicilline et 100 ng/ml de streptomycine (Flow Labs).
Les composés de l'invention ainsi que les composés (D) et Et-18-OCH3, sont dissouts dans une solution contenant 60% d'éthanol et 40% de tampon salin de phosphate. Les dilutions en série sont préparées dans le même tampon phosphate.
La dose testée est de 0,02 mM. Le temps de traitement dure 24 heures à 37°C.
L'effet des composés de l'invention sur la prolifération des cellules à long terme et sur leur survie, a été évalué en étudiant l'efficacité de clonage et la morphologie des colonies de cellules HT.29. Pour mener cette étude, 5.102 de cellules HT.29, préalablement traitées par les différents composés, pendant 24 heures, sont déposées dans des fioles de culture tissulaire de 25 cm2.
Ces cultures cellulaires sont alors incubées à 37°C pendant 15 jours. A la fin de ce temps d'incubation, les cultures cellulaires sont rinsées deux fois avec le tampon phosphate, fixées avec de l'éthanol à 70% pendant 30 minutes et teintées pendant le même temps avec du Giemsa 10% (Sigma Chemicals).
Les résultats correspondent à la valeur de l'«efficacité de clonage relative», calculée comme suit:

Nombre, de cellules recouvertes et sont rassemblés dans les tableaux suivants.
Il a été montré que les colonies formées, après traitement par les composés de l'invention, ont perdu leur profil régulier, ont une réaction plus faible à la tache de Giemsa et leur taille est plus petite que les colonies non traitées.

POSOLOGIE
En thérapie humaine, les composés de l'invention sont administrés de préférence par voie I.V. La posologie usuelle est de 2,5 à 5 mg/dm2 par jour, 3 à 6 jours par mois en perfusion lente.
Les symboles statistiques se réfèrent à la comparaison entre chaque composé et le composé Et-18-OCH3. Les différents symboles: NS, *, ", et "* signifient que le résultat est respectivement non significatif, significatif, très significatif et hautement significatif.

2. Procédé de préparation des composés selon la revendication 1, comprenant la réaction, dans un solvant aprotique, en présence d'une base organique, à une température comprise entre 0 et 80°C, et sous circulation d'azote, respectivement des composés de formules IIa, IIb et Ile
