REVENDICATIONS
1. Procédé de préparation de bromo-2-alpha-ergocryptine ou de son mésylate, caractérisé en ce qu'on fait réagir l'alpha ergocryptine avec la 1,3-dibromo -5,5- diméthyl -2,4-imidazolinedione dans un dissolvant inerte, à une température de O à 55 " C, et effectue une concentration du mélange jusqu'à dessiccation sous vide, une dissolution du résidu dans un dissolvant halogéné de poids moléculaire bas, un lavage avec une solution aqueuse d'une base minérale et ensuite avec de l'eau, séchage, traitement avec charbon actif et concentrationjusqu'à petit volume, séparation du produit par chromatographie sur colonne en éluant avec le même dissolvant halogéné comme employé pour la dissolution et en ajoutant de l'étha noljusqu'à atteindre une concentration de 5% en volume,
enfin on effectue une concentration jusqu'à dessiccation des fractions riches en produit et une recristallisation finale, et on produit éventuellement de mésylate correspondant par acidification avec l'acide méthane-sulfonique dans un dissolvant inerte et évaporation jusqu'à cristallisation finale.
2. Procédé selon la revendication 1, caractérisé en ce qu'on emploie comme dissolvant inerte un éther éthylique, de préférence le dioxanne et qu'on fait réagir l'alpha-ergocryptine avec la 1,3-dibrome-5,5-diméthyl-2,4-imidazolinedione en présence de 2,2'-azo-bis- (2-butyronitrile), qu'on emploie comme dissolvant halogéné le chlorure de méthylène qu'on exécute le lavage avec une solution de l'hydroxyde de potassium et en ce que l'acidification éventuelle se fait dans une cétone de poids moléculaire bas comme l'acétone ou méthyléthyl-cétone, un alcool de poids moléculaire bas comme le méthanol, éthanol, n-propanol ou isopropanol, un hydrocarbure chloré comme le chlorure de méthylène ou chloroforme, ou bien des mélanges de ces dissolvants.
La bromo-2-alpha-ergocryptine est utilisée comme inhibiteur de la secrétion de prolactine et comme inhibiteur de l'hormone de croissance. La bromo-2-alpha-ergocryptine a la formule structurale suivante:

La bromo-2-alpha-ergocryptine peut être obtenue en forme de base ou de sel avec l'acide méthanesulfonique (mésylate).
D'après cette réaction, on traite l'alpha-ergocyrptine (II) par bromation avec la 1,3-dibromo-5,5-diméthyl-2,4-imida- zolidenedione (dibromantin), en faisant réagir les deux composés sous contrôle sans la formation typique des produits résineux très colorés, qu'habituellement on trouve dans les réactions de bromation comme sous-produits. Ainsi donc, la purification de la bromo-2-alpha-ergocryptine est simplifiée énormément par l'absence des produits résineux ci-dessus mentionnés; cette purification peut être effectuée par les méthodes conventionnelles, c'est-à-dire, extraction, chromatographie sur colonne ou cristallisation.
La réaction est effectuée dans les dissolvants organiques inertes, comme les dérivés halogénés de poids moléculaire bas: trichloroéthyléne, chloroforme et chlorure de méthylène; les nitryles de poids moléculaire bas: acétonitrile, propionitrile et butyronitrile; les éthers de chaîne ouverte: éther diéthylique et éther diisopropylique; ou les éthers cycliques: tétrahydrofurane et dioxane. Ces derniers sont les plus appropriés dans les conditions de la présente invention pour procurer la solubilisation des réactifs. La réaction est convenablement effectuée à une température entre 0 et 55 C, encore que la température parfaite est 40 C. A des températures supérieures à 55 "C, la réaction a lieu aussi mais la formation de produits secondaires est augmentée.
La bromation de l'alpha-ergocryptine (II) par la dibromantin peut être favorisée avec une amorce de radicals, de préférence la 2,2'-azo-bis- (2-butyronitrile), encore que sa présence n'est pas indispensable pour la formation de la bromo2-alpha-ergocryptine.
Comme illustration de l'invention, on décrira un exemple de réalisation du procédé indiqué et pouvant être réalisé à l'échelon industriel.
Exemple
Bromo-2-alpha-ergocryptine (I):
On dissout 2,3 g d'alpha-ergocryptine (II) et 0,12 g de 2,2'-azo-bis- (2-butyronitrile) dans le dioxanne à 40 C. On y ajoute lentement 0,6 g de dibromantin dissous dans 25 ml de dioxanne. On maintient la température à 40 C pendant 60 minutes. On concentre le mélange de réaction jusqu'à dessication sous vide et on dissout le résidu dans 70 ml de chlorure de méthylène, on lave avec l'hydroxyde de sodium IN et de l'eau, on sèche sur sulfate de sodium anhydre, on traite avec charbon actif et on concentre à 1/7 de son volume.
La solution obtenue est déposée sur une colonne de verre de 2 cm diamètre contenant 30 g de Silicagel G; on élue avec chlorure de méthylène, en ajoutant lentement des quantités grossissantes d'éthanoljusqu'à atteindre 5% en volume.
Les fractions riches en bromo-2-alpha-ergocryptine sont recueillies et concentrées jusqu'à dessiccation, en obtenant 1,6 g d'un produit qui, par recristallisation dans méthyl-éthyl-cétone; éther diisopropylique rend la bromo-2-alpha-ergocryptine (I) d'un degré élévé de pureté. P.F.: 214-7 "C (d);

CLAIMS
1. Process for the preparation of bromo-2-alpha-ergocryptine or its mesylate, characterized in that the alpha ergocryptine is reacted with 1,3-dibromo -5,5- dimethyl -2,4-imidazolinedione in an inert solvent, at a temperature of 0 to 55 "C, and performs a concentration of the mixture until drying under vacuum, dissolution of the residue in a halogenated solvent of low molecular weight, washing with an aqueous solution of a base mineral and then with water, drying, treatment with activated carbon and concentration until small volume, separation of the product by column chromatography eluting with the same halogenated solvent as used for the dissolution and adding etha nol to reach a concentration of 5% by volume,

Method according to claim 1, characterized in that an ethyl ether, preferably dioxane, is used as inert solvent and that alpha-ergocryptine is reacted with 1,3-dibroma-5,5-dimethyl -2,4-imidazolinedione in the presence of 2,2'-azo-bis- (2-butyronitrile), which is used as the halogenated solvent methylene chloride which is washed with a solution of potassium hydroxide and in that the possible acidification takes place in a low molecular weight ketone such as acetone or methyl ethyl ketone, a low molecular weight alcohol such as methanol, ethanol, n-propanol or isopropanol, a chlorinated hydrocarbon such as chloride methylene or chloroform, or mixtures of these solvents.
Bromo-2-alpha-ergocryptine is used as an inhibitor of prolactin secretion and as a growth hormone inhibitor. Bromo-2-alpha-ergocryptine has the following structural formula:

The process according to the invention is defined in claim 1 and the reaction is as follows:

Bromo-2-alpha-ergocryptine can be obtained as a base or as a salt with methanesulfonic acid (mesylate).
According to this reaction, the alpha-ergocyrptin (II) is treated by bromination with 1,3-dibromo-5,5-dimethyl-2,4-imida-zolidenedione (dibromantin), by reacting the two compounds under control without the typical formation of highly colored resinous products, which are usually found in bromination reactions as by-products. Thus, the purification of bromo-2-alpha-ergocryptine is greatly simplified by the absence of the resinous products mentioned above; this purification can be carried out by conventional methods, that is to say, extraction, column chromatography or crystallization.
The reaction is carried out in inert organic solvents, such as halogenated derivatives of low molecular weight: trichlorethylene, chloroform and methylene chloride; low molecular weight nitryles: acetonitrile, propionitrile and butyronitrile; open chain ethers: diethyl ether and diisopropyl ether; or cyclic ethers: tetrahydrofuran and dioxane. These are most suitable under the conditions of the present invention for providing solubilization of the reagents. The reaction is suitably carried out at a temperature between 0 and 55 C, although the perfect temperature is 40 C. At temperatures above 55 "C, the reaction also takes place but the formation of side products is increased.
The bromination of alpha-ergocryptine (II) by dibromantin can be favored with a primer of radicals, preferably 2,2'-azo-bis- (2-butyronitrile), although its presence is not essential for the formation of bromo2-alpha-ergocryptine.
As an illustration of the invention, an embodiment of the process indicated and which can be carried out on an industrial scale will be described.
Example
Bromo-2-alpha-ergocryptine (I):
2.3 g of alpha-ergocryptine (II) and 0.12 g of 2,2'-azo-bis- (2-butyronitrile) are dissolved in dioxane at 40 C. 0.6 g of dibromantin dissolved in 25 ml of dioxane. The temperature is maintained at 40 ° C. for 60 minutes. The reaction mixture is concentrated until drying in vacuo and the residue is dissolved in 70 ml of methylene chloride, washed with IN sodium hydroxide and water, dried over anhydrous sodium sulfate, treated with activated carbon and concentrated to 1/7 of its volume.
The solution obtained is deposited on a 2 cm diameter glass column containing 30 g of Silicagel G; eluted with methylene chloride, slowly adding magnifying amounts of ethanol up to 5% by volume.
The fractions rich in bromo-2-alpha-ergocryptine are collected and concentrated until drying, obtaining 1.6 g of a product which, by recrystallization from methyl ethyl ketone; diisopropyl ether makes bromo-2-alpha-ergocryptine (I) of a high degree of purity. M.p .: 214-7 "C (d);
