Procédé de préparation de dérivés trisubstitués de 1'étiocholane La présente invention a pour objet un procédé de préparation de la 3a-acétoxy-17-hydroxy-étiocho- lane-11-one sous ses deux formes stéréoisomères, la 3a-acétoxy-17a-hydroxy-étiocholane-11-one de for mule II et la 3a-acétoxy-17p-hydroxy-étiocholane-11- one de formule III.
Ces composés possèdent des pro priétés physiologiques et pharmacodynamiques inté ressantes et peuvent servir en outre de produits intermédiaires pour la préparation des dérivés lla- ou 11(3-hydroxylé.s correspondants.
Le procédé selon l'invention est caractérisé en ce qu'on soumet le 20- dihydroperoxyde de la 3a-acétoxy-pregnane-11,20- dionc à l'action de la chaleur ou des rayons ultra violets ou à l'action conjointe de ces deux formes d'énergie au sein d'un alcanol inférieur, isole du mélange réactionnel la 3a-acétoxy-pregnane-11,20- dione par cristallisation,
élimine les fractions aci des puis les fractions cétoniques réagissant avec le réactif T de Girard et isole la 3a-acétoxy- 17a-hydroxy-étiocholane-11-one et la 3a-acétoxy- 17(3-hydroxy-étiocholane-11-one par chromatogra phie.

Dans le brevet français No 1175415, la titulaire a décrit la préparation du 20-dihydroperoxyde de la 3a-acétoxy-pregnane-11,20-dione de formule I, par l'action de l'eau oxygénée anhydre au sein d'un sol- vaut organique sur la 3a-acétoxy-pregnane-11,20- dione.
Il a été trouvé maintenant qu'en présence de cer tains solvants, tels que le méthanol, l'action de la chaleur ou des rayons ultraviolets produit une dé composition du dihydroperoxyde, ladite décomposi tion étant d'autant plus rapide qu'on se rapproche du point d'ébullition du solvant utilisé, le processus étant accéléré par l'apport simultané des deux formes d'énergie.
Les produits formés au cours de la réac tion sont principalement la 3a-acétoxy-pregnane-11, 20-dione transformable à nouveau en dihydroper- oxyde de départ ; d'autre part, il se forme une frac tion acide et un mélange de 3u.-acétoxy-17a-hydroxy- étiocholane-11-one de formule II, de 3a-acétoxy-17[3- hydroxy-étiocholane-11-one de formule III et de 3a- acétoxy-étiocholane-11-one. Ces trois derniers com posés sont facilement séparables par chromatogra phie.
La réaction peut être suivie par le dosage de l'oxygène actif du dihydroperoxyde introduit ; elle est terminée lorsque celui-ci a disparu.
Le mode préférentiel d'exécution du procédé selon la présente invention consiste à porter à l'ébul lition et sous irradiation ultraviolette, un mélange de dihydroperoxyde de formule I, et de méthanol jus qu'à disparition du produit de formule I. Par refroi dissement, une partie de la 3a-acétoxy-pregnane- 11,20-dione formée cristallise.
Après évaporation à sec, on reprend les eaux mères de cristallisation par un solvant neutre, non miscible à l'eau, les débar rasse de la fraction acide formée par lavage au bi carbonate de sodium et soumet la fraction neutre obtenue à un traitement au réactif T de Girard, ce qui permet de récupérer une nouvelle fraction de 3a- acétoxy-pregnaner11,20-dione cristallisée ; les com posés étiocholaniques possédant uniquement une fonction cétonique en II ne réagissant pas avec le réactif T, se comportent comme des composés non cétoniques. Il suffit de soumettre cette fraction n'ayant pas réagi avec le réactif T à une chromato graphie pour séparer les composés cherchés de for mules II et III.
L'exemple suivant illustre l'invention sans toute fois la limiter. On peut faire varier notamment la nature du solvant, la température, supprimer l'irra diation en chauffant seulement ou irradier sans chauffer, sans s'éloigner pour cela de l'idée de l'in vention. Les points de fusion sont des points de fu sion instantanée déterminés sur bloc de Maquenne. Exemple <I>Préparation de la</I> 3a-acétoxy-17-hydroxy-étioclzo- lane-11-one <I>sous ses deux formes</I> stéréoisomères <I>de formules 11 et III.</I>
50g de dihydroperoxyde de formule I, sont mis en suspension dans 1500 cm3 de méthanol. En irra diant à l'aide de d'eux lampes ultraviolettes Hanau S.80 et en agitant mécaniquement, on chauffe à l'ébullition à reflux jusqu'à ce que le papier iodo- amidonné ne décèle plus la présence d'oxygène actif, ce qui demande environ neuf heures. La solution est alors refroidie et concentrée sous vide à basse tem pérature au septième environ du volume de départ. On refroidit à - 10 C, laisse la cristallisation se poursuivre pendant environ une heure, essore les cris taux formés qu'on lave au méthanol et sèche.
On obtient ainsi 15 g de 3v.-acétoxy-pregnane-11,20- dione. Après évaporation à sec des eaux mères de cristallisation, on dissout le résidu dans l'éther et éli mine la fraction acide par lavage avec une solution aqueuse saturée de bicarbonate de sodium. L'extrait éthéré est lavé à l'eau, puis séché sur sulfate de ma gnésium et évaporé à sec sous vide. Le résidu obtenu est traité dans les conditions habituelles par un poids égal de réactif T de Girard.
On obtient ainsi, d'une part, une solution aqueuse des hydrazones T qui, par acidification, fournit à nouveau 10 g de 3a-acétoxy- pregnane-11,20-dione portant le total de récupéra tion de ce composé à 50 % en poids du dihydro- peroxyde de départ et, d'autre part, un extrait éthéré renfermant les produits qui ne se sont pas combinés au réactif T. L'extrait éthéré neutre est évaporé à sec puis repris par 600 cm, de cyclohexane. On filtre pour éliminer un insoluble et chromatographie sur alumine.
On procède ensuite à l'élution avec a) du benzène ; b) le mélange benzène-éther 1 %, 2 %, 5 %, 10 %, 20 % et 50 %.
L'éluat benzénique fournit, après évaporation du solvant et recristallisation en méthanol, la 3a-acé- toxy-étiocholanc-11-one, F. 1300 C, [a]D = -I- 740 (c = 0,5 %, acétone) déjà décrite dans le brevet suisse N 368164 de la titulaire.
Les premiers éluats benzène-éther jusqu'à 10 % d'éther fournissent, par évaporation à sec, 2 g de 3a acétoxy-17ci-hydroxy-étiocholane-11-one de formule II. Après recristallisation en méthanol, F. 172o C, [(X]20 = -I- 801, 3 (c = 0,5 %, acétone). Le pro duit se présente en aiguilles incolores, peu solubles en acétone et alcool, insolubles dans l'éther et l'eau. Il exerce une action cardiotonique nette.

Process for preparing trisubstituted derivatives of etiocholane The present invention relates to a process for preparing 3a-acetoxy-17-hydroxy-etiocholan-11-one in its two stereoisomeric forms, 3a-acetoxy-17a- hydroxy-etiocholan-11-one of formula II and 3a-acetoxy-17p-hydroxy-etiocholan-11-one of formula III.
These compounds have interesting physiological and pharmacodynamic properties and can also serve as intermediates for the preparation of the corresponding 11a- or 11 (3-hydroxylated) derivatives.
The process according to the invention is characterized in that the 20-dihydroperoxide of 3a-acetoxy-pregnane-11,20-dionc is subjected to the action of heat or of ultraviolet rays or to the joint action of these two forms of energy within a lower alkanol, isolates the 3a-acetoxy-pregnane-11,20-dione from the reaction mixture by crystallization,
removes the aci fractions from then the ketone fractions reacting with Girard's T reagent and isolates 3a-acetoxy-17a-hydroxy-etiocholan-11-one and 3a-acetoxy-17 (3-hydroxy-etiocholan-11-one by chromatography.

In French patent No. 1175415, the licensee described the preparation of 20-dihydroperoxide of 3a-acetoxy-pregnane-11,20-dione of formula I, by the action of anhydrous hydrogen peroxide in a sol - is organic on 3a-acetoxy-pregnane-11,20-dione.
It has now been found that in the presence of certain solvents, such as methanol, the action of heat or ultraviolet rays produces a decomposition of the dihydroperoxide, said decomposition being all the more rapid as one approaches the boiling point of the solvent used, the process being accelerated by the simultaneous supply of the two forms of energy.
The products formed during the reaction are mainly 3a-acetoxy-pregnane-11, 20-dione convertible again into the starting dihydroperoxide; on the other hand, an acid fraction is formed and a mixture of 3u.-acetoxy-17a-hydroxy-etiocholan-11-one of formula II, of 3a-acetoxy-17 [3-hydroxy-etiocholan-11-one of formula III and 3α-acetoxy-etiocholan-11-one. These last three compounds are easily separable by chromatography.
The reaction can be followed by assaying the active oxygen of the dihydroperoxide introduced; it is finished when it has disappeared.
The preferred embodiment of the process according to the present invention consists in bringing to the boiling point and under ultraviolet irradiation, a mixture of dihydroperoxide of formula I and of methanol until the product of formula I has disappeared. , part of the 3α-acetoxy-pregnane-11,20-dione formed crystallizes.
After evaporation to dryness, the mother liquors of crystallization are taken up in a neutral solvent, immiscible with water, the acid fraction formed by washing with sodium bicarbonate is removed from them and the neutral fraction obtained is subjected to treatment with the reagent. T from Girard, which makes it possible to recover a new fraction of crystallized 3α-acetoxy-pregnaner11,20-dione; the etiocholanic compounds possessing only a ketone function at II which does not react with the reagent T, behave like non-ketone compounds. It suffices to subject this fraction which has not reacted with the reagent T to chromatography in order to separate the sought compounds of formulas II and III.
The following example illustrates the invention without however limiting it. It is possible to vary the nature of the solvent and the temperature in particular, to eliminate the irradiation by heating only or to irradiate without heating, without departing for this from the idea of the invention. The melting points are instantaneous melting points determined on a Maquenne block. 50 g of dihydroperoxide of formula I are suspended in 1500 cm3 of methanol. By irradiating with the aid of Hanau S.80 ultraviolet lamps and stirring mechanically, it is heated to the reflux boiling point until the iodine-starched paper no longer detects the presence of active oxygen, which takes about nine hours. The solution is then cooled and concentrated under vacuum at low temperature to about seventh of the starting volume. Cooled to - 10 C, allowed crystallization to continue for about an hour, filtered cris rates formed, washed with methanol and dried.
In this way 15 g of 3v.-acetoxy-pregnane-11,20-dione are obtained. After evaporation to dryness of the mother liquors of crystallization, the residue is dissolved in ether and the acid fraction is removed by washing with a saturated aqueous solution of sodium bicarbonate. The ethereal extract is washed with water, then dried over magnesium sulfate and evaporated to dryness under vacuum. The residue obtained is treated under the usual conditions with an equal weight of Girard's T reagent.
There is thus obtained, on the one hand, an aqueous solution of the T hydrazones which, by acidification, again provides 10 g of 3a-acetoxy-pregnane-11,20-dione, bringing the total recovery of this compound to 50% by weight of the starting dihydro-peroxide and, on the other hand, an ethereal extract containing the products which have not combined with the reagent T. The neutral ethereal extract is evaporated to dryness and then taken up in 600 cm 3 of cyclohexane. It is filtered to remove an insoluble material and chromatography on alumina.
The elution is then carried out with a) benzene; b) 1%, 2%, 5%, 10%, 20% and 50% benzene-ether mixture.
The benzene eluate gives, after evaporation of the solvent and recrystallization from methanol, 3a-acetotoxy-etiocholanc-11-one, M.p. 1300 C, [a] D = -I- 740 (c = 0.5%, acetone) already described in Swiss patent N 368164 of the holder.
The first benzene-ether eluates up to 10% ether yield, by evaporation to dryness, 2 g of 3a-acetoxy-17ci-hydroxy-etiocholan-11-one of formula II. After recrystallization from methanol, mp 172o C, [(X] 20 = -I- 801.3 (c = 0.5%, acetone). The product is in colorless needles, sparingly soluble in acetone and alcohol, insoluble. in ether and water, it exerts a distinct cardiotonic action.
