Procédé de préparation de nouveaux thiodérivés de colchicéines La présente invention a pour objet un procédé de préparation de composés nouveaux de la série de la colchicéine qui résultent du remplacement du groupe - OCH3 du cycle tropolonique C de la formule I ci-dessous par un groupe - S - alcoyle, substitué ou non, et qui correspondent à la formule générale II ci-dessous, où R représente un radical osidique acylé ou non et R' représente un radical alcoyle substitué ou non.

La formule générale I est écrite en conformité avec les formules admises pour la colchicine (R = CH3), la déméthylcolchicine (R = H ; cf. Santavy et Reichstein, Helv. Chim.
Acta. 1950, 33, 1606) et le colchi- coside (R = C6H11O5) dont la préparation a fait l'objet du brevet suisse No 315880 et dont la filiation avec la déméthylcolchicine (R = H) a été prouvée (brevet suisse No 316152). En accord avec les travaux de nomenclature de Horowitz et Ullyot (J. Am. Chem. Soc., 1952, 74, 597), on peut attribuer aux alcoyl-thiocolchicéines la formule II.
Les nouveaux composés obtenus trouvent leur application dans le domaine biologique et industriel de la colchicine, notamment dans les modifications de la mitose et la création de polypoïdes soit par épandage de solutions aqueuses ou de suspensions des produits sur des sols cultivés, soit par traitement préalable des semences avec les produits purs ou dilués dans un solvant ou sur un support.
Au point de vue physiologique, certains de ces nouveaux composés présentent une toxicité nettement inférieure à celle des dérivés colchiciques antérieurement utilisés.

On peut effectuer la réaction soit en présence d'un catalyseur acide, soit en milieu alcalin. Lorsqu'on fait agir un alcoylmercaptan, substitué ou non, dans les mêmes conditions que pour les composés colchiciques, sur des composés isocolchiciques, la réaction ne procède pas par simple remplacement du groupe - OR" par le groupe -S-alcoyle ; il y a simultanément transposition et, dans le produit final, le groupe cétonique et le groupe alcoylthio reprennent les positions indiquées dans la formule II.
Si l'on opère en présence d'un catalyseur acide, de préférence l'acide paratoluène-sulfonique, l'acide benzène-sulfonique, un hydracide, le trifluorure de bore, etc., la réaction entre le composé colchicique ou isocolchicique et le mercaptan peut se faire en présence d'un solvant ou sans solvant, à une température voisine de la température ordinaire ou un peu supérieure, de préférence entre 15 et 50 . Dans le cas du méthylmercaptan, il convient d'opérer, au laboratoire, en tube scellé et, industriellement, en autoclave. Selon la réactivité du composé tropolonique et la température de réaction, le contact avec le mer- captan et le catalyseur peut être maintenu pendant un laps de temps s'échelonnant entre plusieurs heures et plusieurs jours.
Lorsqu'on utilise un solvant, on le choisit de préférence de manière à mettre en solution le composé dé départ. La réaction terminée, on peut récupérer le mer- captan en excès, généralement par distillation, après refroidissement du mélange réactionnel. On peut utiliser à nouveau le mercaptan récupéré. Le solvant éventuel peut être séparé suivant les procédés habituels. Le thiodérivé résultant peut être purifié par cristallisation ; on l'obtient avec un bon rendement. Lorsqu'on met en #uvre des composés de la série de l'isocolchicéine, la réaction est conduite dans ces mêmes conditions, mais le rendement est en général inférieur, et il faut parfois dans ce cas recourir à la chromatographie pour purifier le mélange réactionnel obtenu.
La réaction peut également être effectuée en milieu alcalin. On fait réagir pour cela une solution aqueuse du sel de sodium, de potassium ou de lithium d'un alcoylmercaptan, substitué ou non sur le composé colchicique ou isocolchicique, en absence ou présence d'un solvant organique tel que le méthanol, l'éthanol, le tétrahydrofuran, le dioxane, etc. Ce mode d'exécution du procédé présente un certain avantage sur celui de la catalyse acide dans le cas où l'on utilise le méthylmercaptan. En effet, les sels alcalins du méthylmercaptan ne sont pas volatils et l'on peut opérer en vase ouvert. Le sel alcalin du mercaptan peut être préparé à part et introduit au moment de la réaction.
On peut encore préparer extemporanément une solution aqueuse ou alcoolique de ce sel en faisant réagir le mercaptan sur une solution titrée d'un alcali ou encore sur l'éthylate ou le méthylate de sodium, et utiliser directement cette solution. Après réaction, on peut acidifier, diluer par l'eau et extraire le thiodérivé formé par un solvant non miscible à l'eau et le purifier par cristallisation fractionnée ou chromatographie. Les rendements sont excellents et permettent l'obtention de composés très purs.
Ce mode d'exécution est particulièrement avantageux en, ce qu'il permet d'éviter l'hydrolyse de la liaison osidique qui peut se produire lorsque la réaction entre le composé colchici- que ou isocolchicique et l'alcoylmercaptan est effectuée en présence d'un catalyseur acide.
Les exemples suivants illustrent l'invention. Les points de fusion sont des points de fusion lente. Les pouvoirs rotatoires ont été déterminés dans le chloroforme, à 0,5 %, s'il n'est pas précisé autrement. Dans un tube à sceller, on introduit 3 g de tétracétate de colchicoside et 0,4 g d'acide p-toluène- sulfonique monohydraté, refroidit à - 80n C, et introduit ensuite 12 g de méthylmercaptan. On scelle le tube et laisse monter sa température à -f- 20n C. On laisse réagir pendant cinq jours au cours desquels

un dépôt apparaît. Après distillation du méthylmer- captan, on reprend le résidu par 100 cm3 d'acétate d'éthyle ou de chloroforme, lave la solution au bicarbonate de soude, puis à l'eau, sèche sur sulfate de sodium et distille le solvant.
On obtient un résidu d'aspect résineux, pesant 2,82 g, qui ne cristallise pas dans les solvants usuels.
On peut transformer le produit obtenu, par hydrolyse en déméthyl-thiocolchicine ou par saponification en thiocolchicoside, par exemple comme suit: 2,7 g de tétracétate de thiocolchicoside sont dissous dans 42 cm3 d'éthanol et additionnés de 7 cm3 de soude normale. On abandonne vingt-quatre heures à 20 C, neutralise et extrait avec un mélange chloroforme-éthanol (8 : 2). L'extrait est lavé à l'eau, séché et évaporé à sec. Le résidu est constitué par le thio- colchicoside. Après l'avoir trituré dans 10 volumes d'éther, on l'essore et obtient, après séchage, 1,2 g de thiocolchicoside, sous forme de poudre refusant de cristalliser dans les solvants usuels.
On dissout dans 30 cm3 de méthanol 1,5 g de tétracétate de thiocolchicoside brut, préparé comme indiqué ci-dessus, et ajoute 120 cm3 d'acide chlorhydrique 0,2 N. On évapore le méthanol et chauffe au bain-marie pendant deux heures au cours desquelles un précipité noir se forme. On refroidit, ajoute de la soude jusqu'à ce que le pH soit supérieur à 10 et extrait la solution alcaline par le chloroforme pour la débarrasser des substances non phénoliques. La solution sodique rouge est acidifiée par de l'acide chlorhydrique et épuisée au chloroforme. Après séchage et évaporation à sec, celui-ci laisse un résidu de 480 mg de déméthyl-thiocolchicine brute, correspondant à un rendement de 58 % de la théorie.
Par recristallisation dans 2 à 3 cm3 de diméthylform- amide, auxquels on ajoute 15 cm3 d'acétate d'éthyle, on obtient 420 mg de déméthyl-thiocolchicine fondant à 305 C. Le mélange avec le produit purissime obtenu par une autre voie, F. 308 C, fond à 306 C, ce qui prouve l'identité des deux substances. Exemple 2 Thiocolchicoside (formule Il, R = C6H11O5, R' = - CH3) à partir du colchicoside : 400 mg de colchicoside sont introduits dans une solution de 400 mg de méthylmercaptide de sodium dans 2 cm3 d'eau sous vive agitation. Après seize heures d'abandon à 23 C, on acidifie la solution orangée par l'acide acétique et l'extrait à plusieurs reprises par le mélange chloroforme-éthanol 1 : 1. Les extraits réunis sont séchés sur carbonate de potassium puis distillés à sec.
Le thiocolchicoside brut ainsi obtenu est purifié par recristallisations successives en éthanol et acétate d'éthyle. Obtenu avec un rendement de 50 %, il se présente sous forme de petits carrés ou de prismes octogonaux jaune clair fondant à 215 C, [α]## = - 550 10 (c = 0,5 %, eau). Exemple 3 Obtention de l'éthyl-thiocolchicoside : Formule II, R' = C2H5 ; R = C6H11O5 On dissout 1,084 g de colchicoside obtenu selon le brevet N 315880, de la titulaire, dans 3 cm3 d'eau distillée, ajoute 504 mg d'éthylmercaptide de sodium et abandonne 24 heures à la température ambiante. On acidifie par l'acide acétique à basse température et extrait par 10 fois 10 cm3 du mélange chloroforme-éthanol (1 : 1). Les extraits rassemblés sont lavés à la soude N saturée de chlorure de sodium, séchés sur sulfate de magnésium et évaporés à sec sous vide. On reprend dans un mélange constitué de méthanol, d'éther, et de chloroforme (8 : 4 : 1), essore et recristallise en méthanol pour obtenir 1,09 g d'éthylthiocolchicoside, F = 250 - 252 C, [α]## = -260 3 (c = 0,5, éthanol).
Ce produit, qui n'a pas encore été décrit, se présente sous forme de petits cristaux jaunes, solubles dans l'eau et l'alcool, peu solubles dans le benzène, le chloroforme et le méthanol froid, insolubles dans l'éther.
