Soweit die Herstellung der Ausgangsprodukte nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu an sich bekannten Verfahren herstellbar.
Die Verbindungen der Formel I und ihre Säureadditionssalze sind in der Literatur bisher nicht beschrieben worden.
Sie weisen im Tierversuch interessante pharmakologische Eigenschaften auf und können daher als Heilmittel verwendet werden.
Sie zeichnen sich durch ausgeprägte antihypertensive Eigenschaften aus, wie sich bei Tierversuchen an der wachen, spontan hypertonen Ratte bei Dosen von ungefähr 0,001 bis 1 mg/kg s.c. und am narkotisierten, normotonen Hund [Methode nach D. Chu, A. Hofmann und E. Stürmer, Naunyn-Schmiedberg's Arch. Pharmacol. (1975), Supl. 287, R 18] bei Dosen von ungefähr 0,001 bis 10 mg/kg i.v. zeigt.
Aufgrund ihrer blutdrucksenkenden Wirkung können sie als Antihypertensiva z.B. in der Hochdrucktherapie Anwen dungfinden.
Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Verabreichung und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer täglichen Dosis von ca. 0,002 bis 10 mg/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 0,2 bis 300 mg. So enthalten z.B. für orale Applikationen die Teildosen etwa 0,05 bis 150 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Die neuen Verbindungen können als Arzneimittel allein oder in entsprechenden Arzneiformen für orale, enterale oder parenterale Verabreichung verwendet werden.
In dem nachfolgenden Beispiel, welches die Erfindung näher erläutert, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel I, I -Dimethyl-3-(12-Methoxy-6-methyl-9-ergolen-8ss- ylmethyl)-harnstoff a) 1, l-Dimethyl-3-(l -acetyl-6-methyl-2,3P-dihydro-9- ergolen-8p-ylmethyl)-harnstoff
Zu einer Suspension von 13 g (35,8 mM) 1,1-Dimethyl-3- (6-methyl-2,3 p-dihydro-9-ergolen-8p-ylmethyl)-harnstoff- hydrochlorid in 300 ml absolutem Methylenchlorid werden 14 ml Triäthylamin gegeben. Nachdem die Substanz gelöst ist, wird bei 10-15" eine Lösung von 3,6 ml (50,7 mM) Acetylchlorid in 20 ml abs. Methylenchlorid zugetropft und 1 Stunde bei Raumtemperatur gerührt. Anschliessend wird auf Wasser/Eis gegossen, mit 20 ml gesättigter KHCO3-Lösung versetzt und 2mal mit Methylenchlorid, das 5% Isopropanol enthält, extrahiert.
Die vereinigten organischen Phasen werden mit Natriumsulfat getrocknet, filtriert und am Rotationsverdampfer eingedampft. Nach Trocknen am Hochvakuum resultieren 13 g (98% der Theorie) der Titelverbindung in Form eines gelb-braunen Schaumes. Aus einer Lösung von 2-Butanon/Wasser/Methanol (5:3:2) kristallisiert das Hydrogenfumarat der Titelverbindung. Smp.: Zers. ab 135".

b) I, I -Dimethyl-3 -( I -acetyl-6-methyl- 12-nitro-2,3 ss- dihydro-9-ergolen-8ss-ylmethyl)-harnstoff
Zu einer unter Argon stehenden Lösung von 13 g (35 mM) der unter a) erhaltenen rohen Base in 80 ml Schwefelsäure (100%-ig) werden bei 10 innerhalb 3 Minuten 1,45 ml (35 mM) Salpetersäure (100%ig) zugetropft. Nach 20 Minuten Rühren bei Raumtemperatur wird auf Eis gegossen, bei einer Temperatur von 5-10" mit 10 N Natriumhydroxyd auf pH8 gestellt und mit Methylenchlorid, das 10% Isopropanol enthält, extrahiert. Die organischen Phasen werden vereinigt, mit Natriumsulfat getrocknet, filtriert und am Rotationsverdampfer getrocknet. Nach Trocknen am Hochvakuum resultieren 13,5 g (93% der Theorie) der Titelverbindung, die ohne Reinigung reduziert werden.
c) 1,1 -Dimethyl-3-( 1 -acetyl- 1 2-amino-6-methyl-2,3 - dihydro-9-ergolen-8ss-ylmethyl)-harnstoff
Eine Lösung von 13,5 g (32,7 mM) l,1-Dimethyl-3-(lacetyl-6-methyl- 12-nitro-2,3ss-dihydro-9-ergolen-8ss- ylmethyl)-harnstoff in 300 ml Essigsäureäthylester und 100 ml Methanol wird in Anwesenheit von 1,5 g Palladium auf Kohle (10%ig) unter Normalbedingungen bis zur Aufnahme der berechneten Menge Wasserstoff hydriert. Nach dem Filtrieren und Eindampfen am Rotationsverdampfer wird das Rohprodukt an 300 g Silicagel H mit Methylenchlorid enthaltend 15% Methanol und 0,5% wässerigen Ammoniak chromatographiert. Es resultieren 9 g (72% der Theorie) der Titelverbindung.
d) I,l-Dimethyl-3-(12-hydroxy-6-methyl-9-ergolen-8ss- ylmethyl)-harnstoff
Eine Lösung von 4,5 g (11,7 mM) des unter c) erhaltenen Produktes in 100 ml 1 N HCI wird unter Rühren während 17 Stunden auf 95" erwärmt. Anschliessend wird unter Eiskühlung mit konzentrierter Ammoniaklösung auf pH 7 gestellt und 4mal mit Methylenchlorid, das 10% Isopropanol enthält, extrahiert. Die vereinigten organischen Phasen werden mit Natriumsulfat getrocknet, filtriert und am Rotationsverdampfer eingedampft. Der Eindampfrückstand wird in 10 ml Äthanol gelöst. Nach Zugabe von 1 ml konzentrierter Ammoniaklösung wird während 1 Stunde Luft durch die Lösung geblasen. Nach dem Eindampfen wird das Rohprodukt mit Methylenchlorid enthaltend 10% Methanol und 0,2% Ammoniaklösung an 100 g Silicagel H chromatographiert.
Es resultieren 2,0 g der reinen Titelverbindung als beiger Rückstand. Zur Herstellung des Hydrochlorides wird die erhaltene Base (5,9 mM) in Methylenchlorid/Methanol (1:1) gelöst, mit 2,94 ml (5,9 mM) 2 N HCI versetzt und am Rotationsverdampfer bis zur beginnenden Kristallisation eingeengt. Es resultieren 2 g (45% der Theorie) des Hydrochlorides der Titelverbindung als beiges Kristallisat. Smp.: Zers. ab 220". [o]2o0 = + 126,2 [c = 0,435 in Äthanol/Wasser (1:l)J.
e) I,I-Dimethyl-3-(I2-methoxy-6-methyl-9-ergolen-8ss- ylmethyl)-harnstoff
Zu einer auf15" gekühlten Lösung von 5 ml 0,08 N methanolischer Tetrafluorborsäure in 150 ml Methanol, die 2,55 g (7,5 mM) der unter d) erhaltenen Base enthält, werden unter Argon vorsichtig 20 ml (ca. 71,5 mM) einer ca. 1,5%gen ätherischen Diazomethan-Lösung zugegossen. Anschliessend wird 20 Stunden unter Argon bei Raumtemperatur gerührt.
Zur Aufarbeitung wird der Kolbeninhalt am Wasserstrahlvakuum um ca. 20% eingeengt und der Rest am Rotationsverdampfer eingedampft. Nach Zugabe von 50 ml Äthanol und 1 ml konz. Ammoniak-Lösung wird nochmals eingedampft und der Rückstand an 120 g Silicagel H mit Methylenchlorid enthaltend 10% Methanol und 0,2% konz. Ammoniaklösung chromatographiert. Es resultieren 1,2 g (45% der Theorie) der Titelverbindung als Base, die aus Aceton kristallisieren.
