9. Verfahren nach Anspruch 7 oder 8, dadurch gekennzeichnet, dass man die Reduktion bei niederen bis leicht erhöhten Temperaturen, z. B. bei Temperaturen zwischen 0 und 100 0C durchführt.
10. Verfahren nach einem der Ansprüche 7 bis 9, dadurch gekennzeichnet, dass man die Umsetzung bei der Siedetemperatur des verwendeten Lösungsmittels durchführt.
11. Verfahren nach einem der Ansprüche 7 bis 10, dadurch gekennzeichnet, dass man als Hydrid Boran/Tetrahydrofuran und als Lösungsmittel Tetrahydrofuran verwendet.
12. Verfahren nach einem der Ansprüche 7 bis 11, dadurch gekennzeichnet, dass man bei der Reduktion ein Rohprodukt eines a-Acyloxy-acetamids der allgemeinen Formel I verwendet.
13. Verfahren nach einem der Ansprüche 7 und 9-12, dadurch gekennzeichnet, dass die Aufarbeitung des Endproduktes der Formel II über eine wässrige Base, z. B. über Wasser/Ammoniak, erfolgt.

Erfindungsgemäss erhält man die neuen a-Acyloxyacetamide der obigen allgemeinen Formel I durch Umsetzung eines 4-Amino-benzaldehyds der allgemeinen Formel III

Zur erfindungsgemässen Herstellung eines Phenyläthanolamins der allgemeinen Formel II wird ein so erhaltenes a Acyloxy-acetamid der allgemeinen Formel l, welches auch als Rohprodukt weiter umgesetzt werden kann, mit einem Hydrid, mit einem Metallhydrid oder einem komplexen Metallhydrid wie Boran/Tetrahydrofuran oder Lithiumaluminiumhydrid, reduziert.
Die Umsetzung eines 4-Amino-benzaldehyds der allgemeinen Formel III mit einem Isonitril der allgemeinen Formel IV und mit einer Carbonsäure der allgemeinen Formel V wird zweckmässigerweise in einem geeigneten Lösungsmittel wie Methylenchlorid Chloroform, Benzol, Äther, Tetrahydrofuran oder Dioxan bei niederen Temperaturen, z.B. bei Temperaturen zwischen -20 und 75 "C, zweckmässigerweise jedoch bei Raumtemperatur, durchgeführt. Die Umsetzung wird jedoch vorzugsweise in der Weise durchgeführt, dass unter Rühren und bei Raumtemperatur entweder gleichzeitig aus je einem Tropftrichter eine Verbindung der allgemeinen Formel IV und eine Carbonsäure der allgemeinen Formel V zu einer Lösung eines Aldehyds der allgemeinen Formel III oder zu einer Lösung eines Isonitrils der allgemeinen
Formel IV und eines Aldehyds der allgemeinen Formel III eine Carbonsäure der allgemeinen Formel V innerhalb mehrerer Stunden, z. B. 5-10 Stunden, zugetropft und anschliessend noch 10 bis 165 Stunden gerührt wird.
Die Reduktion eines erhaltenen a-Acyloxy-acetamids der allgemeinen Formel I mit einem Hydrid, einem Metallhydrid oder einem komplexen Metallhydrid wird vorzugsweise in einem geeigneten Lösungsmittel wie Äther, Tetrahydrofuran oder Dioxan bei niederen bis leicht erhöhten Temperaturen, z.B. bei Temperaturen zwischen 0 und 100 "C, zweckmässi gerweisejedoch bei der Siedetemperatur des Reaktionsgemisches durchgeführt. Besonders vorteilhaft wird jedoch die Reduktion mit Boran/Tetrahydrofuran bei der Siedetemperatur des als Lösungsmittel verwendeten Tetrahydrofurans durchgeführt.
Die Isolierung des gewünschten Endproduktes der allgemeinen Formel II erfolgt zweckmässigerweise über wässrige Basen, z. B. über Wasser/Ammoniak, wobei eine gegebenenfalls noch vorhandene a-Acyloxygruppe gespalten wird.
Ein so erhaltenes Phenyläthanolamin der allgemeinen Formel II lässt sich gewünschtenfalls anschliessend mit einer anorganischen oder organischen Säure in sein physiologisch verträgliches Säureadditionssalz mit der betreffenden Säure überführen. Als Säuren kommen hierfür beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Milchsäure, Weinsäure, Zitronensäure, Fumarsäure oder Maleinsäure ind Betracht.
Die als Ausgangsstoffe verwendeten Verbindungen der allgemeinen Formeln IV und V sind literaturbekannt.
Einen als Ausgangsstoff verwendeten Aldehyd der allgemeinen Formel III erhält man z. B. durch Reduktion eines entsprechenden 4-Amino-benzoesäure-halogenids oder -esters und anschliessende Oxidation des gegebenenfalls erhaltenen Benzylalkohols mit Braunstein.
Aus der Literatur ist bereits ein Verfahren zur Herstellung der Phenyläthanolamine der allgemeinen Formel II bekannt, das dadurch gekennzeichnet, dass ein Glykolsäureamid der allgemeinen Formel

in der Rl, R2 und R3 wie eingangs definiert sind, mit einem komplexen Metallhydrid reduziert wird.
Dieses Verfahren weist jedoch den Nachteil auf, dass die hierfür als Ausgangsstoffe erforderlichen Glykolsäureamide sich nur schwierig herstellen lassen.
Die gemäss dem erfindungsgemässen neuen Verfahren erhaltenen guten Ausbeuten konnten von einem Fachmann nicht vorhergesehen werden, das aus der Literatur bekannt ist (siehe J. Amer. chem. Soc. 67, 1499-1500 [1945]), dass sich die Passerini-Reaktion mit sterisch gehinderten und mit a,I3-ungesättigten Carbonylverbindungen nicht durchführen lässt. Hierbei wird die Inreaktivität der a,ss-ungesättigten Carbonylverbindungen, z. B. von Crotonaldehyd, gegenüber der Passerini-Reaktion durch die Neutralisation des elektronischen Zentrums der Carbonylfunktion aufgrund der mesomeren Grenzstrukturen

Der Fachmann musste daher erwarten, dass das elektronische Zentrum der Carbonylfunktion eines 4-Aminobenzaldehyds der allgemeinen Formel III aufgrund seiner mesomeren Grenzstruktur der allgemeinen Formel

desaktiviert und somit der Passerini-Reaktion nicht zugänglich ist. Überraschenderweise ist dies nicht der Fall.
Die nachfolgenden Beispiele sollen die Erfindung näher erläutern:
Beispiel 1 a-Acetoxy-a-(4-amino-3,5-dichlor-phenyl) N-tert.butyl-acetamid
Zu einer Lösung von 3,8 g (0,02 Mol) 4-Amino-3,5-dichlor-benzaldehyd in 50 ml absolutem Methylenchlorid tropft man unter Rühren bei Raumtemperatur 3,32 g (0,04 Mol) tert.Butylisonitril und 4,8 g (0,08 Mol) Eisessig aus zwei getrennten Tropftrichtern mit gleicher Geschwindigkeit während 8 Stunden. Anschliessend wird die erhaltene Lösung zur Hälfte eingeengt und abgekühlt. Der hierbei ausgefallene nicht umgesetzte 4-Amino-3,5-dichlor-benzaldehyd wird abgesaugt und das Filtrat erneut zur Hälfte eingeengt.
Zu der erhaltenen Mischung gibt man das gleiche Volumen an Diisopropyläther hinzu, wobei das gewünschte Produkt auskristallisiert, welches abgesaugt und mit Diisopropyl äther gewaschen wird.
Schmelzpunkt: 175-176 C.
Beispiel 2 a-Acetoxy-u-(4-amino-3,5-dichlor-phenyi)-
N-tert.butyl-acetamid
Zu einer Lösung von 30 g (0,16 Mol) 4-Amino-3,5-dichlor-benzaldehyd in 280 ml Methylenchlorid werden 45 ml tert.Butylisocyanid gegeben und anschliessend unter Rühren und bei Raumtemperatur ca. 40 ml Eisessig während 16 Stunden getropft. Anschliessend rührt man noch 4 Stunden und engt im Vakuum solange ein, bis die ersten Kristalle ausfallen. Nach dem Abkühlen auf 0 "C wird der ausgefallene nicht umgesetzte 4-Amino-3,5-dichlorbenzaldehyd abgesaugt. Dieser kann direkt erneut umgesetzt werden. Das Filtrat wird mit n-Hexan versetzt, wobei das gewünschte Pro dukt ausgefällt wird. Der Niederschlag wird abgesaugt, mit n-Hexan gewaschen und getrocknet.
Schmelzpunkt: 175-176 C.
Beispiel 3 1 -(4-Amino-3,5-dichlor-phenyl)-2-tert.butylamino-äthanol
Zu einer Lösung von 4,01 g a-Acetoxy-a-(4-amino-3,5 dichlorphenyl)-N-tert.butyl-acetamid in 50 ml absolutem Tetrahydrofuran werden 100 ml einer Imolaren Lösung von Boran in Tetrahydrofuran gegeben. Nach 2stündigem Erhitzen unter Rückfluss wird eingeengt, 100 ml Wasser zugegeben und mit Salzsäure bis pH 2 angesäuert. Die wässrige Phase wird mit Äthylacetat gewaschen, anschliessend mit Ammoniak alkalisch gestellt und mit Methylenchlorid extrahiert. Nach dem Einengen der organischen Phase erhält man das Hydrochlorid aus Isopropanol durch Zugabe von ätherischer Salzsäure.
Schmelzpunkt des Hydrochlorids: 174-175 "C.
Beispiel 4 a-Acetoxy-a-(4-amino-3-chlor-5-trifluormethyl-phenyl)- essigsäure-tert.butylamid
Zu einer Lösung von 4,47 g (20 mMol) 4-Amino-3-chlor5-trifluormethyl-benzaldehyd in 50 ml Methylenchlorid werden bei Raumtemperatur unter Rühren 3,32 g (40 mMol) tert.Butylisonitril und 4,8 g (80 mMol) Eisessig gleichzeitig während 5 Stunden zugetropft und anschliessend 65 Stunden bei Raumtemperatur gerührt. Nach Entfernen der Lösungsmittel im Vakuum wird der Rückstand in Äther gelöst, nacheinander mit Wasser und einer gesättigten Natriumbicarbonat-Lösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum zur Trockene eingedampft. Den Rückstand chromatographiert man über eine Kieselgelsäule mit Methylenchlorid als Elutionsmittel. Die Fraktionen, welche die gewünschte Verbindung enthalten, werden vereinigt, eingedampft und der Rückstand aus Äther kristallisiert.
Schmelzpunkt: 155-156 C.
Beispiel 5
1 -(4-Amino-3-chlor-5-trifluormethyl-phenyl)
2-tert.butylamino-äthanol-hydrochlorid
2 g (5,4 mMol) a-Acetoxy-a-(4-amino-3-chlor-5-trifluormethyl-phenyl)-essigsäure-tert.butylamid werden in 15 ml absolutem Tetrahydrofuran gelöst und unter Stickstoff zu 27 ml einer 1 molaren Lösung von Boran in Tetrahydrofuran auf einmal zugegeben. Nach 4stündigem Kochen am Rückfluss gibt man weitere 15 ml Imolare Boranlösung hinzu und erhitzt weitere 2 Stunden am Rückfluss. Danach wird der Überschuss an Boran mit Aceton zerstört, der Bor-Komplex mit Wasser zersetzt und anschliessend das Lösungsmittel im Vakuum abgedampft. Der wässrige Destillationsrückstand wird mit 2n Salzsäure bis pH 1 angesäuert, mit Ammoniak wieder alkalisch gemacht und mit Äther extrahiert.
Der Äther-Extrakt wird mit 0,5n Salzsäure extrahiert, die saure Lösung mit Äther gewaschen, mit Ammoniak alkalisch gemacht und erneut mit Äther extrahiert. Der Äther-Extrakt wird mit Wasser gewaschen, mit Magnesiumsulfat getrocknet und im Vakuum eingedampft. Der ölige Eindampfrückstand wird in Äther gelöst und mit ätherischer Salzsäure angesäuert. Die ausgefallenen Kristalle werden abgesaugt und mit Äther gewaschen.
Schmelzpunkt: 192-193 C.
Beispiel 6 a-Acetoxy-a-(4-amino-3,5-dichlor-phenyl)
N-tert.butylacetamid
Zu einer Lösung von 14,1 g (0,08 Mol) 4-Amino-3,5dichlorbenzaldehyd in 60 ml Methylenchlorid werden bei Raumtemperatur und unter Rühren 3,6 g (0,06 Mol) Essigsäure und 2,5 g (0,03 Mol) tert.Butyl-isonitril gegeben. Anschliessend wird die Reaktionsmischung 3 Stunden unter Rückfluss erhitzt. Nachdem die obige Operation (Zugabe von Essigsäure, tert.Butyl-isonitril und Erhitzen unter Rückfluss) 4mal wiederholt wurde, giesst man die Reaktionsmischung in kalte 2n Natronlauge und trennt die organische Phase ab. Nach Waschen mit Wasser und Trocknen der organischen Phase über Natriumsulfat wird im Vakuum eingeengt und der Rückstand aus Methylenchlorid/Hexan kristallisiert.
Schmelzpunkt: 175-176"C.
Beispiel 7 a-(4-Amino-3,5-dichlor-phenyl)-a-benzoyloxy
N-tert.butylacetamid
Hergestellt aus 4-Amino-3 ,5-dichlor-benzaldehyd, tert. Butylisonitril und Benzoesäure in Chloroform und bei Rückflusstemperatur analog Beispiel 4.
Schmelzpunkt: 189-192"C (Sintern ab 185"C).
Beispiel 8 a-(4-Amino-3,5-dichlor-phenyl)-a-valeroyloxy
N-tert.butylacetamid
Hergestellt aus 4-Amino-3,5-dichlor-benzaldehyd, tert. Butylisonitril und Valeriansäure analog Beispiel 4.
Schmelzpunkt: 95-98 C.
Beispiel 9 1 -(4-Amino-3,5-dichlor-phenyl)-2-tert.butylamino-äthanol
Hergestellt aus a-(4-Amino-3,5-dichlor-phenyl)-a- benzoyloxy-N-tert.butyl-acetamid und Lithiumaluminiumhydrid in Tetrahydrofuran analog Beispiel 3.
Schmelzpunkt des Hydrochlorids: 174-175 "C.

Hergestellt aus 4-Amino-3 , 5-dichlor-benzaldehyd, tert. Butylisonitril und Valeriansäure analog den Beispielen 2 und 5 ohne Isolierung des gebildeten a-(4-Amino-3,5-dichlor phenyl)-N-tert.-butyl-a-valeroyloxy-acetamids.

Example 1 a-Acetoxy-a- (4-amino-3,5-dichlorophenyl) N-tert-butyl-acetamide
To a solution of 3.8 g (0.02 mol) of 4-amino-3,5-dichlorobenzaldehyde in 50 ml of absolute methylene chloride, 3.32 g (0.04 mol) of tert-butylisonitrile and 4.8 g (0.08 mol) glacial acetic acid from two separate dropping funnels at the same speed for 8 hours. The solution obtained is then concentrated in half and cooled. The precipitated unreacted 4-amino-3,5-dichlorobenzaldehyde is filtered off with suction and the filtrate is again evaporated in half.
The same volume of diisopropyl ether is added to the mixture obtained, the desired product crystallizing out, which is filtered off with suction and washed with diisopropyl ether.
Melting point: 175-176 C.
Example 2 a-acetoxy-u- (4-amino-3,5-dichlorophenyi) -
N-tert-butyl acetamide
45 ml of tert-butyl isocyanide are added to a solution of 30 g (0.16 mol) of 4-amino-3,5-dichlorobenzaldehyde in 280 ml of methylene chloride, and about 40 ml of glacial acetic acid are then added dropwise with stirring at room temperature for 16 hours . The mixture is then stirred for a further 4 hours and concentrated in vacuo until the first crystals fail. After cooling to 0 ° C., the precipitated unreacted 4-amino-3,5-dichlorobenzaldehyde is filtered off with suction. This can be reacted directly. The filtrate is mixed with n-hexane, the desired product being precipitated out. The precipitate becomes suction filtered, washed with n-hexane and dried.
Melting point: 175-176 C.
Example 3 1 - (4-Amino-3,5-dichlorophenyl) -2-tert.butylaminoethanol
100 ml of an imolar solution of borane in tetrahydrofuran are added to a solution of 4.01 g of a-acetoxy-a- (4-amino-3,5 dichlorophenyl) -N-tert.butyl-acetamide in 50 ml of absolute tetrahydrofuran. After heating under reflux for 2 hours, the mixture is concentrated, 100 ml of water are added and the mixture is acidified to pH 2 with hydrochloric acid. The aqueous phase is washed with ethyl acetate, then made alkaline with ammonia and extracted with methylene chloride. After concentrating the organic phase, the hydrochloride is obtained from isopropanol by adding ethereal hydrochloric acid.
Melting point of the hydrochloride: 174-175 "C.
Example 4 a-Acetoxy-a- (4-amino-3-chloro-5-trifluoromethyl-phenyl) acetic acid tert-butylamide
To a solution of 4.47 g (20 mmol) of 4-amino-3-chloro-5-trifluoromethyl-benzaldehyde in 50 ml of methylene chloride, 3.32 g (40 mmol) of tert-butylisonitrile and 4.8 g (80 mmol) of glacial acetic acid was added dropwise at the same time for 5 hours and then stirred at room temperature for 65 hours. After removing the solvents in vacuo, the residue is dissolved in ether, washed successively with water and a saturated sodium bicarbonate solution, dried over magnesium sulfate and evaporated to dryness in vacuo. The residue is chromatographed on a silica gel column using methylene chloride as the eluent. The fractions containing the desired compound are combined, evaporated and the residue is crystallized from ether.
Melting point: 155-156 C.
Example 5
1 - (4-amino-3-chloro-5-trifluoromethyl-phenyl)
2-tert.butylaminoethanol hydrochloride
2 g (5.4 mmol) of a-acetoxy-a- (4-amino-3-chloro-5-trifluoromethyl-phenyl) -acetic acid-tert.butylamide are dissolved in 15 ml of absolute tetrahydrofuran and under nitrogen to 27 ml of a 1st molar solution of borane in tetrahydrofuran added at once. After refluxing for 4 hours, a further 15 ml of imolar borane solution are added and the mixture is refluxed for a further 2 hours. The excess borane is then destroyed with acetone, the boron complex is decomposed with water and the solvent is then evaporated off in vacuo. The aqueous distillation residue is acidified with 2N hydrochloric acid to pH 1, made alkaline again with ammonia and extracted with ether.
The ether extract is extracted with 0.5N hydrochloric acid, the acidic solution washed with ether, made alkaline with ammonia and extracted again with ether. The ether extract is washed with water, dried with magnesium sulfate and evaporated in vacuo. The oily evaporation residue is dissolved in ether and acidified with ethereal hydrochloric acid. The precipitated crystals are filtered off and washed with ether.
Melting point: 192-193 C.
Example 6 a-acetoxy-a- (4-amino-3,5-dichlorophenyl)
N-tert-butyl acetamide
To a solution of 14.1 g (0.08 mol) of 4-amino-3,5-dichlorobenzaldehyde in 60 ml of methylene chloride, 3.6 g (0.06 mol) of acetic acid and 2.5 g (0.05 03 mol) of tert-butyl isonitrile. The reaction mixture is then heated under reflux for 3 hours. After the above operation (addition of acetic acid, tert-butyl-isonitrile and heating under reflux) was repeated 4 times, the reaction mixture is poured into cold 2N sodium hydroxide solution and the organic phase is separated off. After washing with water and drying the organic phase over sodium sulfate, the mixture is concentrated in vacuo and the residue is crystallized from methylene chloride / hexane.
Melting point: 175-176 "C.
Example 7 a- (4-Amino-3,5-dichlorophenyl) -a-benzoyloxy
N-tert-butyl acetamide
Made from 4-amino-3, 5-dichlorobenzaldehyde, tert. Butylisonitrile and benzoic acid in chloroform and at the reflux temperature as in Example 4.
Melting point: 189-192 "C (sintering from 185" C).
Example 8 a- (4-Amino-3,5-dichlorophenyl) -a-valeroyloxy
N-tert-butyl acetamide
Made from 4-amino-3,5-dichlorobenzaldehyde, tert. Butylisonitrile and valeric acid analogous to Example 4.
Melting point: 95-98 C.
Example 9 1 - (4-Amino-3,5-dichlorophenyl) -2-tert-butylaminoethanol
Prepared from a- (4-amino-3,5-dichlorophenyl) a-benzoyloxy-N-tert.butyl-acetamide and lithium aluminum hydride in tetrahydrofuran analogously to Example 3.
Melting point of the hydrochloride: 174-175 "C.
Example 10
1 - (4-Amino-3,5-dichlorophenyl) -2-tert-butylaminoethanol
Made from 4-amino-3, 5-dichlorobenzaldehyde, tert. Butylisonitrile and valeric acid analogous to Examples 2 and 5 without isolation of the a- (4-amino-3,5-dichlorophenyl) -N-tert.-butyl-a-valeroyloxyacetamide formed.
Melting point of the hydrochloride: 174-175 "C.
