Gegenstand der Erfindung ist ein neues Verfahren zur Herstellung von N-Cyclohexyl-N-methyl-(2-amino-3,5-di- brombenzyi)-amin der Bromhexin der Formel

und dessen Säureadditionssalze, insbesondere das Hydrochlorid.
Bromhexin ist eine bronchospasrnodische Substanz, die eine ausserordentlich effektive hustenlindernde Wirkung hat, während die Dosierung und Toxizität sehr gering sind.
Andere Verfahren zur Herstellung von Bromhexin sind z.B. in den finnischen Patenten 40631, 40803, 40804 und 41962 beschrieben.
Es wurde ein Verfahren gefunden, das sich völlig von den in den obengenannten Patenten beschriebenen Verfahren unterscheidet. Als Ausgangsmaterial dient dabei 2,4-Dibrom -6-[(N-methyl-N-cyclohexyl)-aminomethyl]-benzoesäure, die eine neue Substanz ist und folgende Formel II hat

Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man die Substanz der Formel II mit Stickstoffwasserstoffsäure (NH3 in Gegenwart einer starken Säure reagieren lässt (gemäss der sogenannten Schmidt'schen Reaktion), wobei die Carboxylgruppe in eine Aminogruppe umgewandelt wird. Diese Reaktion wurde bisher nicht zur Herstellung von Verbindungen dieser Art angewandt.
Aus der Literatur ist bekannt, dass Säuren, Aldehyde und Ketone in saurer Lösung unter Einwirkung von Stickstoffwasserstoffsäure abgebaut werden. Freie Säuren ergeben dabei Amine; Aldehyde ergeben Nitrile und Ketone ergeben Amide.
Im vorliegenden Falle ist die Reaktionsgleichung die folgende:

Dabei entsteht Stickstoffwasserstoffsäure im statu nascendi. Als Lösungsmittel wird ein inertes organisches Lösungsmittel, z.B. Chloroform, verwendet.
Die Temperatur ist auf 35 bis 100 C zu halten, wobei die Reaktion rasch verläuft. Die gebildete Base kann nach bekannten Methoden in entsprechende Säureadditionssalze umgewandelt werden. Die Umsetzung gelingt unerwartet gut (die Ausbeute beträgt 40 bis 60%) und es ist überraschend, dass sie überhaupt trotz sterischer Hinderung gelingt
Das Ausgangsmaterial II wurde wie folgt aus 2,4-Dibrom -6-methyl-benzoesäure (Schmelzpunkt 162 bis 164 C) her gestellt. Dabei wurde die Methylgruppe bromiert und das Reaktionsprodukt ohne Isolierung des reinen Produktes mit Methylecyclohexylamin umgesetzt.
Dabei entsteht die bisher nicht bekannte 2,4-Dibrom-6-[(N-methyl-N-cyclohexyl)-ami nomethyl]-benzoesäure, die bei 277 bis 280"C schmilzt.
Beispiel
20,25 g 2,4-Dibrom-6-[(N-methyl-N-cyclohexyl)-amino- methyl]-benzoesäure, 100 ccm konzentrierte Schwefelsäure und 200 ccrn Chloroform werden bei 35"C gerührt und 10 g Natriumazid wird bei dieser Temperatur während einer Stunde zugefügt. Danach wird die Mischung drei Stunden gekocht, dann gekühlt und auf Eis gegossen. Das Reaktionsprodukt wird mit Heptan extrahiert und das Heptan abdestilliert. Der Rückstand wird in Schwefelsäure gelöst und Salzsäure wird zugefügt, wobei N-Cyclohexyl-N-methyl-(2- -amino-3,5-dibrombenzyl)-amin . Hydrochlorid auskristallisiert. Die Ausbeute beträgt 11,3 g (55% der Theorie), Schmelzpunkt 240 bis 242"C.
