Die Erfindung betrifft ein neues Verfahren zur Herstellung von 1,3,4,5-Tetrahydro-2H-1,4-benzodiazepin-2-on-Deriva- ten.
Es ist bekannt, dass die Tetrahydro-benzodiazepin-2-on Derivate einerseits auch an sich wertvolle, tranquillant und sedativ wirkende Verbindungen sind und andererseits als wichtige Zwischenprodukte zur Herstellung der aus diesen Verbindungen z.B. durch Oxydation gewinnbaren Dihydrobenzodiazepin-2-on-Derivate verwendet werden können. Aus der Literatur sind verschiedene Verfahren zur Herstellung von Tetrahydro-benzodiazepin-2-on-Derivaten bekannt. So können nach der britischen Patentschrift Nr. 1 037 375 N-Benzhydrylglycin-Derivate durch Wärmebehandlung in die entsprechenden Tetrahydro- 1,4-benzodiazepin-2-on-Derivate übergeführt werden.
Nach der DAS 1199 776 werden durch die katalytische Hydrierung von Dihydro-1,4-benzodiazepin-2-on Derivaten die entsprechenden Tetrahydro-Verbindungen gewonnen. In der ungarischen Patentschrift 155 251 wird eine Methode zur Herstellung von 1,3 ,4,5-Tetrahydro-2H- 1,4- benzodiazepin-2-on-Verbindungen beschrieben, nach welcher von Benzhydrolderivaten ausgegangen wird und diese durch selektive N-Acylierung und nachfolgendes Hydrieren in das gewünschte Endprodukt übergeführt werden.
Es ist auch bekannt, dass bei der Herstellung von 1,4-Benzo diazepinen durch Ringschluss die Ringschlussreaktion bei pH Werten über 7,0 durchgeführt wird (G. A. Archer und R. H. Sternbach, Chem. Rev. 68, 747, 1968). Nach den Literaturangaben (vgl. A. Stempel und F. W. Landgraf, J. Org.
Chem. 27, 4675, 1962) kann ferner das selektive Entfernen der Carbobenzoxygruppe der mit Carbobenzoxy-aminosäuren acylierten 2-Amino-benzophenon-Derivate nur durch Behandlung mit Bromwasserstoffsäure durchgeführt werden; bei dem katalytischen Hydrieren solcher Verbindungen in saurem Medium in Gegenwart von Palladium-Aktivkohle wird gleichzeitig mit dem Abspalten des Carbobenzoxyrestes auch die Carbonylgruppe des Benzophenons reduziert.
Es wurde nun in überraschender Weise gefunden, dass bei der unter geeigneten Reaktionsbedingungen durchgeführten katalytischen Hydrierung von N-Glycylamido-benzophenonderivaten eine reduktive intramolekulare Cyclisierung stattfindet und Tetrahydro-benzodiazepin-2-on-Derivate gewonnen werden. So z.B. bei im Ring nitro-substituierten Derivaten erfolgt diese reduktive Cyclisierung in Gegenwart von Mine ralsäuren oder organischen Säuren mit Ausbeuten über 80%; die im Ring durch ein Halogenatom substituierten 2-Glycylamido-benzophenon-Derivate können in Gegenwart von Essigsäure und die im Ring unsubstituierten entsprechenden Verbindungen auch im neutralen Medium reduktiv intramolekular cyclisiert werden.
Die Erfindung ist also ein Verfahren zur Herstellung von 1,3,4,5-Tetrahydro-2H- 1 ,4-benzodiazepin-2-on-Derivaten der allgemeinen Formel I

In den erfindungsgemäss herstellbaren Tetrahydro-1,4benzodiazepin-Derivaten der allgemeinen Formel I bedeutet R vorzugsweise Wasserstoff. X kann sowohl ein Halogenatom oder eine Aminogruppe, als auch Wasserstoff vertreten; die Verbindungen der Formel I, in welchen X eine Aminogruppe vertritt, sind neue, in der Literatur bisher nicht beschriebene Produkte.
In den Verbindungen der allgemeinen Formel II ist die durch genannte katalytische Hydrierung abspaltbare Schutzgruppe Y vorzüglich eine Carbobenzoxygruppe, kann aber auch irgendeine andere Schutzgruppe vom Urethan-Typ sein.
Steht in solchen Verbindungen an der Stelle von X eine Nitrogruppe, dann wird diese bei der katalytischen Hydrierung ebenfalls reduziert.
Bei einer besonders vorteilhaften Ausführungsweise des erfindungsgemässen Verfahrens werden die Verbindungen der allgemeinen Formel II - besonders im Fall X =NO3 - in äthanolischer Lösung in Gegenwart von Essigsäure und Palladium-Aktivkohle hydriert, wodurch das Abspalten der Schutzgruppe und der Ringschluss in einem Schritt durchgeführt werden.
Das Isolieren des Endproduktes kann in einer sehr einfachen Weise geschehen: Nach dem Abfiltrieren des Katalysators wird das Filtrat unter vermindertem Druck zur Trockne verdampft, und der Rückstand z.B. aus Benzol umkristallisiert; im allgemeinen kann schon durch ein einmaliges Umkristallisieren ein chromatographisch reines Produkt erhalten werden.
Bei Verbindungen der allgemeinen Formel II, welche an der Stelle von X' eine Nitrogruppe enthalten, kann die erfindungsgemässe reduktive Cyclisierung sowohl in reiner Essigsäure-oder in verdünnten Mineralsäuren, als auch in Gegenwart von Mineralsäuren in organischen Lösungsmitteln mit gleich gutem Erfolg durchgeführt werden.
Das erfindungsgemässe Verfahren wird durch die nachstehenden Beispiele näher veranschaulicht. Die in den Beispielen angegebenen Schmelzpunkte wurden im Tottoli'schen Apparat bestimmt. Die dünnschichtchromatographischen Untersuchungen wurden an Kieselgel G nach Stahl , im System Essigester-Pyridin-Eisessig-Wasser 30 : 2,5 : 0,75 1,4 durchgeführt; die Entwicklung erfolgte mit Chlor und Tolidin. Die Struktur der erhaltenen Verbindungen wurde auch durch IR Spektroskopie nachgeprüft.
Beispiel 1 1,3 ,4,5-Tetrahydro-5 -phenyl-7-amino-
2H- 1 ,4-benzodiazepin-2-on a) 4,6 g Palladium-Aktivkohle (10%) werden in 50 ml Äthanol vorhydriert, dann werden der Suspension 17,0 g (39 mMol) 2-(N-Carbobenzoxyglycyl)-amino-5-nitro-benzophenon in 380 ml Äthanol und 9,2 ml Eisessig zugesetzt. Das Hydrieren wird bei atmosphärischem Druck, durch Einleiten von Wasserstoff in das Reaktionsgemisch, über Nacht durchgeführt, dann wird das Reaktionsgemisch abfiltriert und das Filtrat unter vermindertem Druck zur Trockne verdampft. Der Rückstand wird in 500 ml Chloroform gelöst, die Lösung mit 14%iger wässriger Ammoniaklösung, dann mit Wasser gewaschen, getrocknet und unter vermindertem Druck verdampft.
Es werden als Rückstand 9,8 g rohes 1,3,4,5-Tetrahydro-5 phenyl-7-amino-2H- 1 ,4-benzodiazepin-2-on erhalten. Nach Umkristallisieren aus Benzol erhält man 8,4 g reines Produkt (84,7% d. Th.); F. 182-1840C; Rf = 0,49.

Gefunden 71,3% 5,6% 16,4% b) Es wird nach Absatz a) gearbeitet, mit dem Unterschied, dass man 1,1 g 2-(N-Carbobenzoxyglycyl)-amino-5-nitro- benzophenon und 0,3 g Palladium-Aktivkohle in 40 ml Ätha not als Ausgangsstoffe verwendet und anstatt von Eisessig 0,5 ml cc. Salzsäure zusetzt. Es werden auf diese Weise 0;5 g (78% d. Th.) reines 1,3,4,5-Tetrahydro-5-phenyl-7-amino- 2H-1,4-benzodiazepin-2-on in der im Absatz a) angegebenen Qualität erhalten.
Beispiel 2 5-Phenyl-1,3,4,5-tetrahydro-2H-1,4- benzodiazepin-2-on
Zu einer vorhydrierten Suspension von 0,15 g Palladium Aktivkohle (10%) in 5 ml Methanol wird die Lösung von 1,0 g 2-Carbobenzoxyglycylamino-benzophenon in 20 ml Methanol zugesetzt. Das Gemisch wird in der im Beispiel 1 angegebenen Weise über Nacht hydriert, dann wird das Reaktionsgemisch abfiltriert und. das Filtrat zur Trockne verdampft. Der Rückstand wird in Chloroform gelöst, mit je 10 ml 0,4%iger Salzsäure, dann bis zur neutralen Reaktion mit Wasser gewaschen und getrocknet. Nach Verdampfen des Lösungsmittels werden 0,49 g (80,5% d. Th.) rohes 5-Phenyl-1,3,4,5-tetrahydro-2H
1,4-benzodiazepin-2-on erhalten. Ein für analytische Zwecke aus Isopropanol umkristallisiertes Muster dieses Produktes schmilzt bei 146-147 C; Rf = 0,10.
Beispiel 3
7-Chlor-5-phenyl-1-methyl-1,3,4,5-tetrahydro-
2H- 1 ,4-benzodiazepin-2-on
0,15 g Palladium-Aktivkohle (10%) und 0,15 g Platinoxyd werden in 5 ml Eisessig suspendiert, vorhydriert und dann werden zu dieser Katalysator-Suspension 1,1 g (2,4 mMol) 2 (Carbobenzoxyglycyl-N-methylamino)-5-chlor-benzophenon in 5 ml Eisessig zugesetzt. Das Gemisch wird bei Zimmertem peratur und atmosphärischem Druck 5 Stunden hydriert. Am Ende der zweiten Stunde wird noch eine gleiche Menge des obigen Katalysatorgemisches zugesetzt. Nach Beendigung der Hydrierung wird das Gemisch abfiltriert und das Filtrat zur Trockne verdampft. Der Rückstand wird in Chloroform gelöst und die Lösung zum Entfernen der Spuren von Essigsäure mit einer 8 %igen wässrigen Natriumbicarbonatlösung ausgeschüttelt.
Die abgetrennte Chloroform-Phase wird getrocknet und zur Trockne verdampft. Als Rückstand werden 0,65 g rohes
Produkt erhalten, dieses wird durch Behandlung mit 2N Salz säure in das Hydrochlorid übergeführt, abfiltriert und mit eine
8 %igen wässrigen Natriumbicarbonatlösung behandelt. Es werden auf diese Weise 0,55 g (0,79% d. Th.) 7-Chlor-1 methyl-5-phenyl- 1,3 ,4,5-tetrahydro-2H-1 ,4-benzodiazepin-2- on erhalten. Das aus Isopropanol umkristallisierte reine Produkt schmilzt bei 142-1450C; Rf = 0,15 (Hexan-Eisessig Chloroform 1:1: 8).
The invention relates to a new process for the preparation of 1,3,4,5-tetrahydro-2H-1,4-benzodiazepin-2-one derivatives.
It is known that the tetrahydro-benzodiazepin-2-one derivatives are, on the one hand, also valuable compounds with a tranquillant and sedative effect and, on the other hand, they are important intermediates for the preparation of e.g. Dihydrobenzodiazepin-2-one derivatives which can be obtained by oxidation can be used. Various processes for the preparation of tetrahydro-benzodiazepin-2-one derivatives are known from the literature. Thus, according to British Patent No. 1,037,375, N-benzhydrylglycine derivatives can be converted into the corresponding tetrahydro-1,4-benzodiazepin-2-one derivatives by heat treatment.
According to DAS 1199 776, the corresponding tetrahydro compounds are obtained by catalytic hydrogenation of dihydro-1,4-benzodiazepin-2-one derivatives. In the Hungarian patent specification 155 251 a method for the preparation of 1,3, 4,5-tetrahydro-2H-1,4-benzodiazepin-2-one compounds is described, according to which benzhydrol derivatives are used as a starting point and these are obtained by selective N-acylation and subsequent hydrogenation are converted into the desired end product.
It is also known that in the production of 1,4-benzodiazepines by ring closure, the ring closure reaction is carried out at pH values above 7.0 (G. A. Archer and R. H. Sternbach, Chem. Rev. 68, 747, 1968). According to the literature (cf. A. Stempel and F. W. Landgraf, J. Org.
Chem. 27, 4675, 1962) the selective removal of the carbobenzoxy group of the 2-amino-benzophenone derivatives acylated with carbobenzoxy-amino acids can also only be carried out by treatment with hydrobromic acid; During the catalytic hydrogenation of such compounds in an acidic medium in the presence of palladium-activated carbon, the carbonyl group of the benzophenone is also reduced at the same time as the carbobenzoxy radical is split off.
It has now been found, surprisingly, that in the catalytic hydrogenation of N-glycylamido-benzophenone derivatives, carried out under suitable reaction conditions, a reductive intramolecular cyclization takes place and tetrahydro-benzodiazepin-2-one derivatives are obtained. E.g. in nitro-substituted derivatives in the ring, this reductive cyclization takes place in the presence of mineral acids or organic acids with yields of over 80%; the 2-glycylamido-benzophenone derivatives substituted in the ring by a halogen atom can be reductively intramolecularly cyclized in the presence of acetic acid and the corresponding compounds unsubstituted in the ring in a neutral medium.
The invention is therefore a process for the preparation of 1,3,4,5-tetrahydro-2H-1, 4-benzodiazepin-2-one derivatives of the general formula I

In the tetrahydro-1,4benzodiazepine derivatives of the general formula I which can be prepared according to the invention, R preferably denotes hydrogen. X can represent a halogen atom or an amino group as well as hydrogen; the compounds of the formula I in which X represents an amino group are new products which have not yet been described in the literature.
In the compounds of general formula II, the protective group Y which can be split off by the aforementioned catalytic hydrogenation is preferably a carbobenzoxy group, but it can also be any other protective group of the urethane type.
If there is a nitro group in place of X in such compounds, then this is also reduced in the catalytic hydrogenation.
In a particularly advantageous embodiment of the process according to the invention, the compounds of the general formula II - especially when X = NO3 - are hydrogenated in an ethanolic solution in the presence of acetic acid and palladium-activated carbon, whereby the protective group is split off and the ring closure is carried out in one step.
The isolation of the end product can be done in a very simple way: after filtering off the catalyst, the filtrate is evaporated to dryness under reduced pressure, and the residue is e.g. recrystallized from benzene; In general, a single recrystallization can give a chromatographically pure product.
In the case of compounds of the general formula II which contain a nitro group instead of X ', the reductive cyclization according to the invention can be carried out with equal success either in pure acetic acid or in dilute mineral acids, or in the presence of mineral acids in organic solvents.
The process according to the invention is illustrated in more detail by the examples below. The melting points given in the examples were determined in a Tottoli apparatus. The thin-layer chromatographic investigations were carried out on silica gel G according to Stahl, in the system ethyl acetate-pyridine-glacial acetic acid-water 30: 2.5: 0.75 1.4; the development took place with chlorine and tolidine. The structure of the compounds obtained was also checked by IR spectroscopy.
Example 1 1,3,4,5-tetrahydro-5-phenyl-7-amino-
2H- 1, 4-benzodiazepin-2-one a) 4.6 g of palladium-activated carbon (10%) are pre-hydrogenated in 50 ml of ethanol, then 17.0 g (39 mmol) of 2- (N-carbobenzoxyglycyl) are added to the suspension -amino-5-nitro-benzophenone in 380 ml of ethanol and 9.2 ml of glacial acetic acid were added. 9.8 g of crude 1,3,4,5-tetrahydro-5-phenyl-7-amino-2H-1,4-benzodiazepin-2-one are obtained as residue. After recrystallization from benzene, 8.4 g of pure product are obtained (84.7% of theory); F. 182-1840C; Rf = 0.49.

The solution of 1.0 g of 2-carbobenzoxyglycylamino-benzophenone in 20 ml of methanol is added to a pre-hydrogenated suspension of 0.15 g of palladium activated carbon (10%) in 5 ml of methanol. The mixture is hydrogenated overnight in the manner indicated in Example 1, then the reaction mixture is filtered off and. the filtrate evaporated to dryness. The residue is dissolved in chloroform, washed with 10 ml of 0.4% hydrochloric acid each time, then with water until it reacts neutral and dried. After evaporation of the solvent, 0.49 g (80.5% of theory) of crude 5-phenyl-1,3,4,5-tetrahydro-2H are obtained
1,4-benzodiazepin-2-one obtained. A sample of this product recrystallized from isopropanol for analytical purposes melts at 146-147 C; Rf = 0.10.
Example 3
7-chloro-5-phenyl-1-methyl-1,3,4,5-tetrahydro-
2H-1,4-benzodiazepin-2-one
0.15 g of palladium-activated carbon (10%) and 0.15 g of platinum oxide are suspended in 5 ml of glacial acetic acid, pre-hydrogenated and then 1.1 g (2.4 mmol) of 2 (carbobenzoxyglycyl-N-methylamino) are added to this catalyst suspension ) -5-chloro-benzophenone added in 5 ml of glacial acetic acid. The mixture is hydrogenated at room temperature and atmospheric pressure for 5 hours. At the end of the second hour, an equal amount of the above catalyst mixture is added. After the hydrogenation has ended, the mixture is filtered off and the filtrate is evaporated to dryness. The residue is dissolved in chloroform and the solution extracted by shaking with an 8% aqueous sodium bicarbonate solution to remove the traces of acetic acid.
