Die Erfindung bezieht sich auf das Verfahren zur Herstellung von Aminoderivaten von Benzophenonen der allgemeinen Formel:

R3 eine Methyl-, Benzyl-, p-Hydroxybenzyl-, eine gegebenenfalls weiter substituierte Indolyl-3-methyl-, Isopropyl-, Äthyl-, substituierte Äthyl-, Propyl-, Butyl- oder substituierte Butylgruppe,
R4 ein Wasserstoffatom oder ein Halogen darstellen und
X ein Anion bedeutet, das einer Säure gehört, die mit der NH2-Gruppe stabile Salze gibt.
Die Verbindungen der allgemeinen Formel I erhält man nach dem erfindungsgemässen Verfahren durch Solvolyse oder Hydrogenolyse der Verbindungen der allgemeinen Formel:

worin die Bezeichnungen R1, R2, R3 und R4 die gleiche Bedeutung wie in Formel I haben und Z eine Schutzgruppe für die Aminogruppe bedeutet.
Die Solvolyse wird mit Vorteil unter pH 7 mittels verschiedenen Solvolysemitteln durchgeführt. Wenn Z = Carbenzoxy (CbO) ist, erhält man die besten Resultate durch Verwendung 4%iger Bromwasserstoffsäure in Eisessig bei 0" C, aber auch 48% Bromwasserstoffsäure in Eisessig kann verwendet werden. Die Carbobenzoxygruppe kann auch hydrogenolytisch mit 10% Palladium auf Kohle als Katalysator gespalten werden. Die tertiäre Butoxygruppe (BoC) kann mittels Eisessig allein gespalten werden. Die erhaltenen optisch aktiven Aminobenzophenone der allgemeinen Formel I werden als Ammoniumsalze isoliert. Diese stellen vollständig stabile Verbindungen dar, sowohl in festem Zustand als auch in Lösung in organischen Lösungsmitteln und Wasser (unter pH3).
Oberhalb dieses pH cyclisieren diese Amine langsam zu den entsprechenden 1,3-Benzodiazepin-2-onen der allgemeinen Formel:

Besonders schnell cyclisieren freie Amine oberhalb pH 7.
So wurde z. B. für die Cyclisierungsgeschwindigkeit der Verbindungen der allgemeinen Formel I, in welcher R1 = Cl, R2 = R4 = H und R3 = CH3 zum entsprechenden 1,4-Benzodiazepin-2-on bei 37 C und pH 7,44 (was etwa dem physiologischen pH des Blutplasmas entspricht) ein k1-Wert von 4,335 x 104 sec-l gefunden. Dieser Wert wurde spectrophotometrisch bei 283 mu in einem Puffer-Dioxan (4:1) Gemisch gemessen, woraus eine Cyclisierungshalbwertzeit von 26,6 Minuten folgt. Wenn R1 = Cl, R2 = R4 = H und R3 = -CH(CH3)2 ist, wurde bei 37 C und pH 8,0 ein Wert für k von 2,77 x 10-4 sec" erhalten, was einer Halbwertzeit von 16,9 Minuten entspricht.
Diese quantitativen Resultate zeigen, dass die Verbindungen I-X sehr günstige hydrophobe Vorläufer der cyclischen 1,4-Benzodiazepin-2-one darstellen. Die Verbindungen I-X sind stabil unter den pH Bedingungen, welche im Magen vorherrschen und können daher per os verabreicht werden. Erst nach der Absorption im Magen-Darm Trakt gehen sie zu freien Aminen über, nämlich den lipoiden, schwer wasserlöslichen 1 ,4-Benzodiazepin-2-onen, welche dann leichter eine lipoide Zellmembran des Zentralnervensystems (ZNS) passieren. So wird durch Resorption eine verlängerte Wirkung der Titelverbindungen (I) gesichert, welche in vivo in die pharmakologisch gut bekannten 1 ,4-Benzo- diazepin-2-one übergehen. Da die zeitabhängige Wirkung von einigen Benzodiazepinen schon bekannt ist, erweist sich eine verlängerte Wirkung der Titelverbindungen als sehr wünschenswert.
Aus dem oben Beschriebenen geht hervor, dass eine tranquillisierende und hypnotische Wirkung der Verbindungen der allgemeinen Formel I auf ihrer Cyclisierung zu den 1,4 Benzodiazepin-2-onen in vivo beruht. Das gibt dieser Verbindungsklasse einen grossen Gebrauchswert, besonders wegen des sterischen Baues der Verbindungen der allgemeinen Formel I und III, welche eine bessere Interaktion mit dem Pharmakonrezeptor gewährleistet. Die früheren Vermutungen wurden experimentell bestätigt, da Experimente in vitro und in vivo zeigten, dass nur die Verbindungen mit der absoluten S-Konfiguration, d. h. die sich von L-Aminosäuren ableiten, sowohl eine biologische wie auch eine pharmakologische Wirkung besitzen. Demgegenüber zeigen Verbindungen mit der absoluten R-Konfiguration eine sehr niedrige oder keine Wirkung.
Je nach der Wahl der Ausgangsaminosäure lassen sich S- oder R-Konfigurationen in den Endprodukten erzielen.
Beispiel 1
Man löst 0,1 Mol (+)-S-5-Chlor-2-(carbobenzoxyalanyl) amino-benzophenon in 120 ml 4 M HBr/CH3COOH unter Kühlung mit Eiswasser. Nach 10 Minuten entfernt man das Eisbad und lässt das Reaktionsgemisch sich bis zur Zimmertemperatur erwärmen. Dann rührt man, bis die Blasenentwicklung aus der Lösung aufhört. Das Reaktionsgemisch wird im Rotaevaporator zur Trockene eingedampft und dann unter Zugabe von 3mal 200 ml Benzol und nachträgliches Eindampfen weiter getrocknet. Der ölige Rückstand wird durch Zugabe von Äther auskristallisiert. Das Rohprodukt kristallisiert man aus Äther-Methanol, gegebenenfalls unter Zugabe von etwas Cyclohexan, aus.
Auf dieselbe Weise werden auch andere Verbindungen der allgemeinen Formel I erhalten. Die wichtigen Konstanten dieser Verbindungen sind in Tabelle A angegeben.
Beispiel 2
Man löst 0,1 Mol (+)-S-5-Chlor-2-(N-methyl-N-carbo- benzoxy-alanyl)-amino-benzophenon in 150 ml 90 %dem Äthanol und setzt 0,2 bis 0,3 Mol HBr/CH3COOH und 10% Pd-C in einer Menge von 10% der eingesetzten Substanz hinzu.
Dann hydriert man in einem offenen System oder in einem geschlossenen System ohne bemerkenswerten Überdruck.
Den Reaktionsablauf verfolgt man durch Dünnschichtchromatographie bis zum Verschwinden des Flecks des Ausgangsmaterials (Äther-Chloroform als Eluierungsmittel). Nach Beendigung der Reaktion filtriert man den Katalysator ab und dampft das Reaktionsgemisch bis zur Trockene ein. Den Rückstand verarbeitet man in der gleichen Weise wie in Beispiel 1 beschrieben. Das reine Produkt schmilzt bei 165 bis 167 C und zeigt eine Drehung [a]57Ô = -60,2 C (c = 2,403 in MeOH). Wenn man von anderen N-Carbobenzoxysubstituierten Derivaten der allgemeinen Formel I ausgeht, erhält man entsprechende Produkte aus Tabelle A.
Beispiel 3
In 80 ml Eisessig löst man 0,05 Mol 5-Chloro-2-(N methyl-N-tert. -butoxycarbonylphenylalanyl)-amino-benzo- phenon und kühlt bis 0 C. Dann gibt man 10 ml 4M HBr/ CH3COOH zu und rührt bei Zimmertemperatur, bis die Gasentwicklung aufhört. Anschliessend wird das Lösungsmittel abgedampft und 2mal 50 ml Benzol zugesetzt und jedesmal wieder zur Trockene eingedampft. Das rohe Hydrobromid wird mit Äther umkristallisiert und weiter verarbeitet wie in Beispiel 1 beschrieben. Auf die gleiche Weise werden aus BoC-geschützten Ausgangsverbindungen auch andere in der Tabelle A angegebene Verbindungen hergestellt.
Beispiel 4
Wenn man nach irgendeiner Methode arbeitet, die unter den Beispielen 1, 2 oder 3 beschrieben ist, erhält man gleiche Resultate. Die Eigenschaften von verschiedenen Verbindungen sind in der Tabelle A angegeben.

Beispiel 5
Man löst 0,05 Mol des Hydrobromids einer Verbindung aus der Tabelle A in einem Gemisch von Wasser-Äthanol 100 ml:
100 ml, gibt 10%igeNaOH bis zum pH 8,5 zu und rührt bei Zimmertemperatur oder auf einem Wasserbad bis 40 C; die eintretende Cyclisierungsreaktion verfolgt man durch Dünnschichtchromatographie (Äther-CHC13 (1:1) als Eluierungsmittel). Nachdem die Reaktion beendigt ist, dampft man das Lösungsmittelgemisch in einem Rotaevaporator ein und kristallisiert den Rückstand aus dem geeigneten Lösungsmittel oder Lösungsmittelgemisch oder man reinigt die freie Base über das Hydrochlorid in einem Äthanol-Äther-Gemisch.
Wenn man die in der Tabelle A angegebenen Verbindungen cyclisiert, erhält man entsprechende (S)-1,4-Benzodiazepin2-one mit den charakteristischen Eigenschaften, die in der Tabelle B angegeben sind.

The compounds of the general formula I are obtained by the process according to the invention by solvolysis or hydrogenolysis of the compounds of the general formula:

From what has been described above it can be seen that a tranquilizing and hypnotic effect of the compounds of the general formula I is based on their cyclization to the 1,4 benzodiazepin-2-ones in vivo. This gives this class of compounds great utility, especially because of the steric structure of the compounds of the general formulas I and III, which ensures better interaction with the pharmaceutical receptor. The earlier assumptions have been confirmed experimentally, since experiments in vitro and in vivo showed that only the compounds with the absolute S configuration, i.e. H. which are derived from L-amino acids, have both a biological and a pharmacological effect. In contrast, compounds with the absolute R configuration show very little or no effect.
Depending on the choice of the starting amino acid, S or R configurations can be achieved in the end products.

0.1 mol of (+) - S-5-chloro-2- (carbobenzoxyalanyl) amino-benzophenone is dissolved in 120 ml of 4 M HBr / CH3COOH while cooling with ice water. Example 3
0.05 mol of 5-chloro-2- (N methyl-N-tert-butoxycarbonylphenylalanyl) -amino-benzophenone is dissolved in 80 ml of glacial acetic acid and the mixture is cooled to 0 ° C. 10 ml of 4M HBr / CH3COOH are then added and stir at room temperature until the evolution of gas ceases. The solvent is then evaporated off and 50 ml of benzene are added twice and each time it is evaporated to dryness again. The crude hydrobromide is recrystallized with ether and processed further as described in Example 1. Other compounds indicated in Table A are also prepared from BoC-protected starting compounds in the same way.

Example 5
0.05 mol of the hydrobromide of a compound from Table A is dissolved in a mixture of water-ethanol 100 ml:
100 ml, add 10% NaOH up to pH 8.5 and stir at room temperature or on a water bath up to 40 C; the cyclization reaction which occurs is followed by thin-layer chromatography (ether-CHCl3 (1: 1) as eluent). After the reaction has ended, the solvent mixture is evaporated in a rotary evaporator and the residue is crystallized from the suitable solvent or solvent mixture, or the free base is purified using the hydrochloride in an ethanol-ether mixture.
If the compounds given in Table A are cyclized, corresponding (S) -1,4-Benzodiazepin2-ones with the characteristic properties which are given in Table B are obtained.
