Gegebenenfalls substituierte einwertige Kohlenwasserstoffreste, in dem das erste, mit dem Stickstoffatom verbundene Kohlenstoffatom nicht einem aromatischen System angehört, R1 und R2, die gleich oder verschieden sein können, sind z. B. gegebenenfalls substituierte aliphatische, cycloaliphatische, cycloaliphatisch-aliphatische oder araliphatische Kohlenwasserstoffreste, wie z. B. Alkyl, Hydroxyalkyl, Alkenyl, Alkinyl, Cycloalkyl, Cycloalkenyl, Cycloalkylalkyl oder -alkenyl, Cycloalkenyl-alkyl oder -alkenyl, Aralkyl oder Aralkenyl, beispielsweise Phenyl-niederalkyl oder -niederalkenyl, und insbesondere niedere Reste der genannten Art, wobei mit niederen Resten solche bezeichnet werden, die bis zu 7 C-Atome enthalten.
Niedere Alkylreste sind beispielsweise Methyl, Äthyl, n-Propyl oder Isopropyl, oder geradkettiges oder verzweigtes Butyl, Pentyl oder Hexyl, die in beliebiger Stellung gebunden sein können.
Niedere Hydroxylalkylreste sind beispielsweise Reste, in denen die Alkylteile obige Bedeutungen haben, wie 2-Hydroxyäthyl, 3 -Hydroxy-n-propyl, 2,3-Di-hydroxy-n-propyl oder 3-Hydroxy-n-butyl.
Niedere Alkenylreste sind beispielsweise Allyl oder Methallyl.
Niedere Alkinylreste sind insbesondere Propargylreste.
Cycloalkyl- oder -alkenylreste sind beispielsweise gegebenenfalls niederalkylierte Cycloalkyl- oder -alkenylreste mit 3-7, insbesondere 5-7 Ringgliedern, wie gegebenenfalls niederalkyliertes Cyclopentyl, Cyclohexyl, Cycloheptyl, Cyclopentenyl, Cyclohexenyl, beispielsweise 1- oder 3-Cyclohexenyl, oder Cycloheptenyl.
Cycloalkyl-alkyl- oder -alkenylreste sind vor allem solche, in denen die Alkyl- bzw. Alkenylteile niedere Alkyl- bzw.
Alkenylreste sind, insbesondere die oben genannten, und in denen die Cycloalkylteile vorzugsweise die oben für Cycloalkylreste gegebene Bedeutung haben, wie 1- oder 2-Gyclopentyläthyl, 1-, 2- oder 3-Cyclohexyl-propyl, Cycloheptyl-methyl oder 1- oder 2-Cyclohexyl-äthyl.
Cycloalkenyl-alkyl- oder -alkenylreste sind vor allem solche, in denen die Alkyl- bzw. Alkenylteile niedere Alkylbzw. Alkenylreste sind, insbesondere die oben genannten, und in denen die Cycloalkenylteile vorzugsweise die oben für Cycloalkenylreste gegebene Bedeutung haben, wie 1- oder 2-Cyclopent-3 -enyl-äthyl, 1- oder 2 -Cyclohex-1 enyl -äthyl, Cyclohept-1-enyl-methyl oder 1- oder 2-Cyclohex-3-enyl äthyl.
Phenyl-niederalkylreste sind beispielsweise solche, in denen die Niederalkylteile obige Bedeutung haben, wie 1- oder 2-Phenyläthyl oder Benzyl, wobei die Phenylteile gegebenenfalls durch Niederalkyl, insbesondere die oben genannten, niedere Alkoxyreste, Halogenatome oder Trifluormethylreste substituiert sind, während Phenyl-niederalkenylreste beispielsweise solche sind, in denen die Niederalkenylreste obige Bedeutung haben, wie 1- oder 2-Phenyl-äthenyl oder Cinnamyl, wobei die Phenylteile gegebenenfalls wie oben für die Phenyl-niederalkylreste angegeben substituiert sind.
Niedere Alkoxyreste sind beispielsweise Methoxy, Äthoxy, Propoxy, Isopropoxy, Butoxy, Amyloxy oder Methylendioxy.
Halogenatome sind insbesondere Fluor, Chlor oder Brom.
Zweiwertige Kohlenwasserstoffreste, in denen die ersten, mit dem Stickstoffatom verbundenen Kohlenstoffatome nicht einem aromatischen System angehören, für die R1 und R2 zusammen stehen und die gegebenenfalls durch Heteroatome unterbrochen und/oder substituiert sein können, sind vorzugsweise niedere Alkylenreste, die geradkettig oder verzweigt sein können und vor allem 4-6 Kettenkohlenstoffatome bei ununterbrochener Kohlenstoffkette bzw. 4 oder 5 Kettenkohlenstoffatome bei durch Heteroatome unterbrochener Kohlenstoffkette aufweisen. Als Heteroatome kommen insbesondere Sauerstoff, Schwefel und Stickstoff in Betracht.
Beispiele für derartige Reste sind Butylen-(1,4), Pentylen-(1,5), Hexylen-(1,5), Hexylen-(2,5), Hexylen (1,6), Heptylen-(1,6), 3 -Oxapentylen-(1,5), 3 -Oxahexylen-(1,6), 3 -Thia-pentylen-(1,5 ), 2,4 -Dimethyl- 3 -thiapentylen-(1,5 ), 3-Aza-pentylen-(1,5), 3-Niederalkyl- 3 -azapentylen-(l,a), wie 3-Methyl-3-aza-pentylen-(1,5) oder 3-Azahexylen-(1,6).
Meta- oder para-Phenylenreste Ph können unsubstituiert oder ein-, zwei- oder mehrfach substituiert sein. Substituenten sind vor allem Niederalkyl, Niederalkenyl, Niederalkinyl, Cycloalkyl, Phenylniederalkyl und Niederalkoxy, insbesondere die oben genannten, Phenyl, Niederalkenyloxy, Niederalkinyloxy, Halogen, insbesondere die oben genannten, sowie substituiertes Niederalkyl, Phenoxy, Trifluormethyl und/oder Cyano.
Niederalkenyloxyreste sind beispielsweise Allyloxy oder Methallyloxy und Niederalkinyloxyreste sind vor allem Propargyloxy.
Substituierte Niederalkylreste sind vorzugsweise niedere Alkylreste, insbesondere die oben genannten, die durch
Hydroxy, Niederalkoxy, insbesondere die oben genannten, gegebenenenfalls substituiertes Mercapto, wie freies Mer capto oder Niederalkylmercapto, z. B. Methyl- oder Äthylmercapto, oder Halogen, insbesondere die oben genannten, substituiert sind. Derartige Reste sind z. B. Hydroxyniederalkyl, Niederalkyl-mercapto-niederalkyl und Halogen-niederalkyl, wie 2-Hydroxyäthyl, 2-Methoxyäthyl, 2-Mercaptoäthyl, Methylmercaptomethyl und 2,2 -D ichloräthyl.
Der aliphatische oder cycloaliphatische Rest R3 ist vorzugsweise ein aliphatischer oder cycloaliphatischer Kohlenwasserstoffrest, insbesondere einer der oben genannten, sowie Niederalkoxy -niederalkyl oder Halogenniederalkyl, wie die oben genannten, sowie durch Heteroatome unterbrochene Cycloalkylreste.
Durch Heteroatome unterbrochene Cycloalkylreste sind insbesondere durch Sauerstoff, Schwefel oder Stickstoff unterbrochene Cycloalkylleste, wie Tetrahydrofuryl, Tetrahydropyranyl, Tetrahydrothienyl, Tetrahydrothiapyranyl, Piperidyl oder Pyrrolidinyl.
Kondensationsprodukte von Verbindungen der Formel I mit Aldehyden oder Ketonen sind insbesondere Verbindungen der Formel II

Der Alkylidenrest X ist insbesondere ein niederer Alkylidenrest, wie ethyliden, n-Propyliden, Isopropyliden oder geradkettiges oder verzweigtes Butyliden oder Pentyliden, oder vor allem Methylen. Die genannten Alkylidenreste und insbesondere der Methylenrest können einen oder mehrere Substituenten tragen, wie cycloaliphatische Kohlenwasser stoffreste, insbesondere die oben genannten, oder aromatische Reste.
Aromatische Reste sind vor allem Phenylreste, die gegebenenfalls wie oben für die Phenylteile von Phenylniederalkylresten angegeben substituiert sind, oder heterocyclische aromatische Reste, wie Furyl, Thienyl, Pyrryl und vor allem Pyridyl.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So hemmen sie kardioselektiv ss -Re- zeptoren, wie sich im Tierversuch, z. B. bei i. v. Gabe von 0,1-1 mg/kg an der Pentobarbital-narkotisierten Katze im Isoproterenol-Tachykardie-Test, bei i. v. Gabe von mehr als 10 mg/kg an der narkotisierten Katze durch Hemmung der Isoproterenol-Vasodilatation, am isolierten Meerschweinchenherz nach Langendorff in einer Konzentration von etwa 0,3-3 y/ml durch Hemmung der Isoproterenol-Tachykardie (Blockade von cardialenss -Rezeptoren) sowie bei i. v. Gabe von etwa 5-30 mg/kg an der narkotisierten Katze durch Blockade von vasculären ss -Rezeptoren zeigt.
Die neuen Verbindungen können daher als kardioselek tivess-Rezeptorenblocker verwendet werden. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere pharmazeutisch wirksamer Verbindungen.
Besonders hervorzuheben sind Verbindungen der Formel I, worin R1 für Wasserstoff, Niederalkyl, Niederalkenyl, Cycloalkyl mit 4-7 Ringgliedern, oder Phenylniederalkyl steht, wobei der Phenylteil der Phenylniederalkylreste unsubstituiert oder durch Niederalkyl, Niederalkoxy, Halogen und/oder Trifluormethyl substituiert ist, R2 und R3, die gleich oder verschieden sind, jeweils für Niederalkyl, Niederalkenyl, Cycloalkyl mit 4-7 Ringgliedern oder Phenylniederalkyl stehen, wobei der Phenylteil der Phenylniederalkylreste gegebenenfalls wie bei R1 angegeben substituiert ist, und Ph ein para-Phenylenrest ist, und deren Kondensationsprodukte der Formel II, worin X für Äthyliden, Isopropyliden oder vor allem für Methylen steht, wobei der Methylenrest gegebenenfalls durch Phenyl oder Pyridyl substituiert ist.

Diese Umsetzung wird in der üblichen Weise durchgeführt. Bei Verwendung eines reaktionsfähigen Esters als Ausgangsmaterial wird vorzugsweise in Gegenwart eines basischen Kondensationsmittels und/oder mit einem Überschuss an Amin gearbeitet.

Diese Umsetzung wird in üblicher Weise, vorzugsweise in Gegenwart eines basischen Kondensationsmittels und/oder mit einem Überschuss an Amin durchgeführt. Geeignete basische Kondensationsmittel sind beispielsweise Alkalialkoholate, insbesondere Natrium- oder Kalium-alkoholate, oder auch Alkalicarbonate, wie Natrium- oder Kaliumcarbonat.
In erhaltenen Verbindungen kann man im Rahmen der Endstoffe in üblicher Weise Substituenten abwandeln, einführen oder abspalten oder erhaltene Verbindungen können in üblicher Weise in andere Endstoffe überführt werden.
So kann man in erhaltenen Verbindungen, welche eine C-C-Doppel- oder -Dreifach-Bindung enthalten, die C- Doppel- bzw. -Dreifach-Bindung durch katalytische Hydrierung, wie durch Wasserstoff in Gegenwart eines Hydrierungskatalysators, beispielsweise Nickel, Platin oder Palladium, wie Raney-Nickel, Platinschwarz oder Palladium auf Aktivkohle, in eine C-C-Einfachbindung überführt werden. Dabei muss darauf geachtet werden, dass andere reduzierbare Gruppen, vor allem die Harnstoffgruppe, nicht angegriffen werden.
In erhaltenen Verbindungen, welche eine C-C-Dreifachbindung enthalten, kann diese ferner lediglich zu einer C-C-Doppelbindung und wenn erwünscht stereospezifisch zu einer C-C-cis- oder C-C-trans-Doppelbindung hydriert werden. Die Hydrierung einer C-C-Dreifachbindung zu einer C-C-Doppelbindung kann beispielsweise durch Hydrierung mit 1 Mol Wasserstoff in Gegenwart eines weniger aktiven Hydrierungskatalysators, wie Eisen oder Palladium, beispielsweise Raney-Eisen oder Palladium auf Bariumsulfat, insbesondere bei erhöhter Temperatur, erfolgen. Die Hydrierung zu einer C-C-cis-Dpppelbindung kann beispielsweise mittels 1 Mol Wasserstoff in Gegenwart eines desaktivierten Katalysators, wie Palladium auf Tierkohle in Gegenwart von
Chinolin, Palladium auf Calziumcarbonat in Gegenwart von
Bleisalzen, oder auch Raney-Nickel, erfolgen.
Die Hydrierung zu einer C-C-trans-Doppelbindung kann beispielsweise mittels Natrium in flüssigem Ammoniak erfolgen, wobei insbesondere mit Rücksicht auf die Harnstoffgruppe kurze
Reaktionszeiten und kein Überschuss an Reduktionsmittel angewendet werden und wobei gegebenenfalls ein Ammo niumhalogenid, wie Ammoniumchlorid, als Katalysator zu gegeben wird.
Die genannten Reaktionen werden in üblicher Weise in
An- oder Abwesenheit von Verdünnungs-, Kondensations und/oder katalytischen Mitteln, bei erniedrigter, gewöhnli cher oder erhöhter Temperatur, gegebenenfalls im geschlos senen Gefäss durchgeführt.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säureadditions salze. So können beispielsweise basische, neutrale oder ge mischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesqui oder Polyhydrate davon, erhalten werden. Die Säureaddi tionssalze der neuen Verbindungen können in an sich be kannter Weise in die freie Verbindung übergeführt werden, z. B. mit basischen Mitteln, wie Alkalien oder Ionenaus tauschern. Anderseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Zur
Herstellung von Säureadditionssalzen werden insbesondere solche Säuren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind.
Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefel säuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, alipha tische, alicyclische, aromatische oder heterocyclische Car bon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-,
Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascor bin-, Malein-, Hydroxymalein- oder Brenztraubensäure;
Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl- oder p-Aminosalicylsäure, Em bonsäure, Methansulfon-, Äthansulfon-, Hydroxyäthan sulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluol sulfon-, Naphthalinsulfonsäure oder Sulfanilsäure; Methionin,
Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die
Basen frei macht. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer
Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmässig, gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die Erfindung betrifft auch diejenigen Ausführungs formen des Verfahrens, nach denen man einen Ausgangsstoff in Form eines unter den Reaktionsbedingungen erhältlichen rohen Reaktionsgemisches oder eine Reaktionskomponente gegebenenfalls in Form ihrer Salze einsetzt.
Die neuen Verbindungen können, sofern sie asymmetri sche Kohlenstoffatome aufweisen, und je nach der Wahl der
Ausgangsstoffe und Arbeitsweisen, als optische Antipoden oder Racemate oder, sofern sie mindestens zwei asymmetri sche Kohlenstoffatome enthalten, auch als Isomerengemische (Racematgemische) vorliegen.
Erhaltene Isomerengemische (Racematgemische) kön nen auf Grund der physikalisch-chemischen Unterschiede der
Bestandteile in bekannter Weise in die beiden stereoisomeren (diastereomeren) reinen Racemate aufgetrennt werden, 'beispielsweise durch Chromatographie und/oder fraktionierte
Kristallisation.
Erhaltene Racemate-lassen sich nach bekannten Metho den, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen oder 5 durch Umsetzen mit einer, mit der racemischen Verbindung
Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltenen Salze, z. B. auf Grund ihrer verschie denen Löslichkeiten, in die Diastereomeren, aus denen die Antipoden durchEinwirkung geeigneter Mittel freigesetzt werden können, zerlegen. Besonders gebräuchliche optisch aktive Säuren sind z. B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Äpfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden.
Zweckmässig verwendet man für die Durchführung der eIfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs besonders erwähnten Gruppen von Endstoffen und besonders zu den speziell beschriebenen oder hervorgehobenen Endstoffen führen.
Die Ausgangsstoffe sind bekannt oder können, falls sie neu sind, nach an sich bekannten Methoden erhalten werden.
Die neuen Verbindungen können als Heilmittel, z. B.
in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem z. B. für die enterale oder parenterale Applikation-geeigneten pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Die folgenden Beispiele erläutern die Erfindung ohne sie jedoch einzuschränken. Die Temperaturen sind in Celsiusgraden angegeben.

The epoxy used as the starting product can be produced as follows:
12 g of dimethylcarbamoyl chloride are added to a solution of 20 g of p-benzyloxy-aniline in 100 ml of pyridine and the mixture is left to stand for two days. Upon addition of water, the N, N-dimethyl-N '- (p-benzyloxyphenyl) urea precipitates, which melts at 155-158 after sublimation.
