Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So hemmen sie kardioselektiv ss
Rezeptoren, wie sich im Tierversuch, z. B. bei i.v.

B. in

where R1, R2, R3 and Ph have the above meaning and Z is a hydrogenolysable radical, hydrogenolysed.
Residues which can be split off by hydrogenolysis are, for example, α-arylakyl radicals, such as benzyl radicals, or aryloxycarbonyl radicals, such as benzyloxycarbonyl radicals, which can be split off in the customary manner by hydrogenolysis, in particular by catalytically excited hydrogen, such as by hydrogen in the presence of a hydrogenation catalyst, for example palladium or platinum. Other radicals which can be split off by hydrogenolysis are, for example, haloethoxycarbonyl radicals, such as the 2,2,2-trichloroethoxycarbonyl radical or the 2-iodoethoxy or 2,2,2-bromoethoxycarbonyl radical, which are split off in the usual way, in particular by nascent hydrogen can.
Nascent hydrogen can be obtained by the action of metal or metal alloys on hydrogen-supplying agents, such as carboxylic acid, alcohols or water, zinc or zinc alloys in particular coming into consideration together with acetic acid. The hydrogenolysis of halogen-ethoxycarbonyl radicals can preferably be carried out by means of chromium-II compounds, such as chromium-II chloride or chromium-II-acetate. When carrying out the hydrogenolysis, care must be taken that other reducible groups, especially the urea group, are not attacked.
In the compounds obtained, in the context of the end products, substituents can be modified, introduced or split off in the usual way, or the compounds obtained can be converted into other end products in the usual way.
For example, in compounds obtained which contain a CC double or triple bond, the CC double or triple bond can be obtained by catalytic hydrogenation, such as by hydrogen in the presence of a hydrogenation catalyst, for example nickel, platinum or palladium, such as Raney nickel, platinum black or palladium on activated carbon, can be converted into a CC single bond. Care must be taken to ensure that other reducible groups, especially the urea group, are not attacked.
In compounds obtained which contain a C-C triple bond, this can furthermore only be hydrogenated to a C-C double bond and, if desired, stereospecifically to a C-C-cis or C-C-trans double bond. The hydrogenation of a C-C triple bond to a C-C double bond can be carried out, for example, by hydrogenation with 1 mol of hydrogen in the presence of a less active hydrogenation catalyst such as iron or palladium, for example Raney iron or palladium on barium sulfate, in particular at elevated temperature. The hydrogenation to a C-C-cis double bond can be carried out, for example, by means of 1 mol of hydrogen in the presence of a deactivated catalyst such as palladium on charcoal in the presence of quinoline, palladium on calcium carbonate in the presence of lead salts, or Raney nickel.
The hydrogenation to a C-C trans double bond can be carried out, for example, using sodium in liquid ammonia, with short reaction times and no excess of reducing agent being used, especially with regard to the urea group, and with an ammonium halide, such as ammonium chloride, being added as a catalyst if necessary.
The reactions mentioned are carried out in the usual way in the presence or absence of diluents, condensation and / or catalytic agents, at a reduced, normal or elevated temperature, if appropriate in a closed vessel.
Depending on the process conditions and starting materials, the end products are obtained in free form or in the form of their acid addition salts, which is also included in the invention. For example, basic, neutral or mixed salts, optionally also hemi-, mono-, sesqui or polyhydrates thereof, can be obtained. The acid addition salts of the new compounds can be converted into the free compound in a manner known per se, e.g. B.
with basic agents such as alkalis or ion exchangers.
On the other hand, the free bases obtained can form salts with organic or inorganic acids. For the preparation of acid addition salts, in particular those acids are used which are suitable for the formation of therapeutically useful salts. Examples of such acids are:
Hydrogen halides, sulfuric acids, phosphoric acids, nitric acid, perchloric acid, aliphatic, alicyclic, aromatic or heterocyclic carboxylic or sulfonic acids, such as formic, acetic, propionic, succinic, glycolic, lactic, apple, wine, lemon, Ascorbic, maleic, hydroxymaleic or pyruvic acid; Phenylacetic, benzoic, p-aminobenzoic, anthranil, p-hydroxybenzoic, salicylic or p-aminosalicylic acid, emboxylic acid, methanesulphonic, ethanesulphonic, hydroxyethanesulphonic, ethylene sulphonic acid; Halobenzenesulfonic, toluenesulfonic, naphthalenesulfonic acid or sulfanilic acid; Methionine, tryptophan, lysine or arginine.
These or other salts of the new compounds, such as. B. the picrates, can also be used to purify the free bases obtained by converting the free bases into salts, separating them and in turn making the bases free from the salts. As a result of the close relationships between the new compounds in free form and in the form of their salts, in the preceding and in the following the free compounds are meaningfully and appropriately also the corresponding salts.
The invention also relates to those embodiments of the process in which a starting material is used in the form of a crude reaction mixture obtainable under the reaction conditions or a reaction component, if appropriate in the form of its salts.
If they have asymmetric carbon atoms, and depending on the choice of starting materials and procedures, the new compounds can be present as optical antipodes or racemates or, if they contain at least two asymmetric carbon atoms, also as isomer mixtures (racemic mixtures).
Mixtures of isomers (mixtures of racemates) obtained can be separated into the two stereoisomeric (diastereomeric) pure racemates in a known manner on the basis of the physico-chemical differences between the constituents, for example by chromatography and / or fractional crystallization.
Racemates obtained can be obtained by known methods, for example by recrystallization from an optically active solvent, with the aid of microorganisms or by reaction with an optically active acid which forms salts with the racemic compound and separation of the salts obtained in this way, e.g. B. due to their different solubilities, decompose into the diastereomers, from which the antipodes can be released by the action of suitable agents. Optically active acids that are particularly common are e.g. B. the D- and L-forms of tartaric acid, di-o-toluyltartaric acid, malic acid, mandelic acid,
Camphorsulfonic acid or quinic acid. It is advantageous to isolate the more effective of the two antipodes.
For carrying out the reactions according to the invention, it is expedient to use those starting materials which lead to the groups of end products mentioned especially at the beginning and especially to the end products specifically described or emphasized.
The starting materials are known or, if they are new, can be obtained by methods known per se.
The new compounds can be used as remedies, e.g. B. in
Form of pharmaceutical preparations, find use, which they or their salts in mixture with a z. B. contain suitable pharmaceutical, organic or inorganic, solid or liquid carrier material for enteral or parenteral administration.
The following examples illustrate the invention without, however, restricting it. The temperatures are given in degrees Celsius.
Example 1 7 g of 1-jp- (N'-methylureido) -phenoxy] -2-hydroxy- (3-isopropylbenzylamino) -propane in 70 ml of ethanol are added at 50 ° C. and normal pressure after adding 1 g of palladium carbon (10% mg) hydrogenated. When the uptake of hydrogen has ended, the catalyst is filtered off and the filtrate is evaporated in vacuo. The residue crystallizes after the addition of ether. The crystals are suction filtered and washed with ether. The 1 - [p- (N'-methylureido) -phenoxy-2-hydroxy-3-isopropylaminopropane of the formula is obtained in this way
