The new compounds have valuable pharmacological properties. So they inhibit cardioselective receptors, as shown in animal experiments, e.g. B. with i.v. Administration of 0.1-1 mg / kg to pentobarbital anesthetized cats in the isoproterenol tachycardia test, with iv. Administration of more than 10 mg / kg to the anesthetized cat by inhibiting isoproterenol vasodilation, to the isolated guinea pig heart according to Langendorff in a concentration of about 0.3-3 μg / ml by inhibiting the isoproterenol tachycardia (blockade of cardiac ss receptors ) and iv Administration of about 5-30 mg / kg to the anesthetized cat shows blockade of vascular ss receptors.

Administration of more than 10 mg / kg to the pentobarbital anesthetized cat inhibit isoproterenol vasodilation (0.5 y / kg iv.), And in concentrations of 0.3-3 y / ml on the isolated guinea pig heart (according to Langendorff) the isoproterenol Inhibit tachycardia (0.005 y / ml).
The new compounds are obtained by adding a compound of the formula II
EMI2.2
where Rs, R2, R3 and Ph have the above meanings and Z is an acyl radical which can be split off by hydrolysis, is hydrolyzed.
Cleavable acyl radicals are, for example, oxycarbonyl radicals, such as alkoxycarbonyl radicals, e.g. B. the tert. -Butoxycarbonyl radical, aralkoxycarbonyl radical, e.g. B. a carbobenzoxy radical, and in particular lower alkanoyl radicals or aryloyl radicals, e.g. B.
the acetyl radical or a benzoyl radical.
The hydrolysis is carried out in a customary manner by hydrolyzing agents and, for example, in the presence of basic agents or acidic agents. Such acidic agents are, for example, dilute inorganic acids such as sulfuric acid or a hydrohalic acid such as one of the above.
In the compounds obtained, in the context of the end products, substituents can be modified, introduced or split off in the usual way, or the compounds obtained can be converted into other end products in the usual way.
Thus, in compounds obtained which contain a CC double or triple bond, the CC double or triple bond can be achieved by catalytic hydrogenation, such as by hydrogen in the presence of a hydrogenation catalyst, for example nickel, platinum or palladium, such as Raney -Nickel, platinum black or palladium on activated carbon, can be converted into a CC single bond. Care must be taken to ensure that other reducible groups, especially the urea group, are not attacked.
In compounds obtained which contain a C-C triple bond, this can furthermore only be hydrogenated to a C-C double bond and, if desired, stereospecifically to a C-C-cis or C-C-trans double bond. The hydrogenation of a C-C triple bond to a C-C double bond can be carried out, for example, by hydrogenation with 1 mol of hydrogen in the presence of a less active hydrogenation catalyst such as iron or palladium, for example Raney iron or palladium on barium sulfate, in particular at elevated temperature. The hydrogenation to a C-Ccis double bond can be carried out, for example, using 1 mol of hydrogen in the presence of a deactivated catalyst, such as palladium on charcoal in the presence of quinoline, palladium on calcium carbonate in the presence of lead salts, or Raney nickel.
The hydrogenation to a C -C trans double bond can be carried out, for example, by means of sodium in liquid ammonia, with short reaction times and no excess of reducing agent being used, especially with regard to the urea group, and with an ammonium halide, such as ammonium chloride, being added as a catalyst if necessary.
The reactions mentioned are carried out in the usual way in the presence or absence of diluents, condensation and / or catalytic agents, at a reduced, normal or elevated temperature, if appropriate in a closed vessel.
Depending on the process conditions and starting materials, the end products are obtained in free form or in the form of their acid addition salts, which is also included in the invention. For example, basic, neutral or mixed salts, optionally also hemi-, mono-, sesqui or polyhydrates thereof, can be obtained. The acid addition salts of the new compounds can be converted into the free compound in a manner known per se, e.g. B.
with basic agents such as alkalis or ion exchangers.
On the other hand, the free bases obtained can form salts with organic or inorganic acids. For the preparation of acid addition salts, those acids are used in particular which are suitable for the formation of therapeutically useful salts. Examples of such acids are:
Hydrohalic acid, sulfuric acids, phosphoric acids, nitric acid, perchloric acid, aliphatic, alicyclic, aromatic or heterocyclic carboxylic or sulfonic acids, such as formic, acetic, propionic, succinic, glycolic, lactic, apple, wine, lemon, Ascorbic, maleic, hydroxymaleic or pyruvic acid; Phenylacetic, benzoic, p-aminobenzoic, anthranil, p-hydroxybenzoic, salicylic or p-aminosalicylic acid, emboxylic acid, methanesulphonic, ethanesulphonic, hydroxyethanesulphonic, ethylene sulphonic acid; Halobenzenesulfonic, toluenesulfonic, naphthalenesulfonic acid or sulfanilic acid; Methionine, tryptophan, lysine or arginine.
These or other salts of the new compounds, such as. B. the picrates, can also be used to purify the free bases obtained by converting the free bases into salts, separating them and in turn making the bases free from the salts. As a result of the close relationships between the new compounds in free form and in the form of their salts, in the preceding and in the following the free compounds are meaningfully and appropriately also the corresponding salts.
The invention also relates to those embodiments of the process in which a starting material is used in the form of a crude reaction mixture obtainable under the reaction conditions or a reaction component, if appropriate in the form of its salts.
The new compounds can, if they have asymmetric carbon atoms, and depending on the choice of starting materials and working methods, exist as optical antipodes or racemates or, if they contain at least two asymmetric carbon atoms, also as isomer mixtures (racemic mixtures).
Mixtures of isomers (mixtures of racemates) obtained can be separated into the two stereoisomeric (diastereomeric) pure racemates in a known manner on the basis of the physico-chemical differences between the constituents, for example by chromatography and / or fractional crystallization.
Racemates obtained can be obtained by known methods, for example by recrystallization from an optically active solvent, with the aid of microorganisms or by reaction with an optically active acid which forms salts with the racemic compound and separation of the salts obtained in this way, eg. B. due to their different solubilities, decompose into the diastereomers, from which the antipodes can be released by the action of suitable agents. Optically active acids commonly used are e.g. B. the D- and L-forms of
Tartaric acid, di-o-toluyltartaric acid, malic acid, mandelic acid,
Camphorsulfonic acid or quinic acid. It is advantageous to isolate the more effective of the two antipodes.
For carrying out the reactions according to the invention, it is expedient to use those starting materials which lead to the groups of end products mentioned especially at the beginning and especially to the end products specifically described or emphasized.
The starting materials are known or, if they are new, can be obtained by methods known per se.
The new compounds can be used as remedies, e.g. B.
in the form of pharmaceutical preparations that use them or their salts in admixture with a z. B. suitable for enteral or parenteral administration pharmaceutical, organic or inorganic, solid or liquid carrier material.
The following examples illustrate the invention without, however, restricting it. The temperatures are given in degrees Celsius.
