Gegenstand des Hauptpatentes ist ein Verfahren zur Herstellung von Bis[(4-Hydroxy-3-hydroxymethylphenyl) äthanol]diamin-derivaten der Formel A, worin n' 2 oder 3 bedeutet. Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Bis[(4-Hydroxy-3-hydroxymethylphenyl)äthanol]diamin-derivaten der Formel I, worin n eine ganze Zahl von 1 bis 4 bedeutet und die beiden Substituenten am Phenylenring in ortho- oder meta-Stellung zueinander angeordnet sind, sowie ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Bis[(4 Hydroxy-3-hydroxymethylphenyl)äthanol]diamin-derivaten der Formel I, indem man Verbindungen der Formel II, worin n obige Bedeutung besitzt und die beiden Substituenten am Phenylenring in ortho- oder meta-Stellung zueinander angeordnet sind und R1 je Methyl, Äthyl oder Benzyl bedeutet, mit ätherspaltenden Mitteln behandelt und die erhaltenen Verbindungen der Formel I in Form der freien Basen oder ihrer Säureadditionssalze gewinnt.
In den Verbindungen der Formel I bedeutet n vorzugsweise 2 oder 3.
Das erfindungsgemässe Verfahren kann nach an sich zur Ätherspaltung üblichen Methoden durchgeführt werden. So kann man beispielsweise die Verbindungen der Formel II mit Lewis-Säuren, z. B. mit Bortribromid oder Aluminiumchlorid, in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z. B. einem halogenierten Kohlenwasserstoff, wie Methylenchlorid oder Tetrachlorkohlenstoff oder einem aromatischen Kohlenwasserstoff, wie Toluol oder Benzol, bei -80 bis +70 reagieren lassen, oder Verbindungen der Formel II kurzzeitig mit starken Mineralsäuren, wie z. B.
Bromwasserstoff- oder Jodwasserstoffsäure, gegebenenfalls bei erhöhter Temperatur, z. B. bei etwa 20 bis 100 behandeln, oder auf die Verbindungen der Formel II Hydrochloride organischer Basen, wie Anilin oder Pyridin, bei erhöhter Temperatur einwirken lassen.
Die Verbindungen der Formel I können auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden und können gewünschtenfalls in ihre Säureadditionssalze überführt werden.
Die Verbindungen der Formel II können z. B. erhalten werden, indem man Verbindungen der Formel III, worin R und n obige Bedeutung besitzen und R2 für niederes Alkyl steht, reduziert.
Die Reduktion kann beispielsweise in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem Äther, wie Diäthyläther, Tetrahydrofuran, Dioxan, Dimethoxyäthan mit Lithiumaluminiumhydrid oder Aluminiumhydrid, ausgeführt werden.
Verbindungen der Formel III können z. B. erhalten werden, indem man Verbindungen der Formel IV, worin R1 obige Bedeutung besitzt und X für Chlor oder Brom steht, mit Verbindungen der Formel V, worin n obige Bedeutung besitzt, umsetzt.
Die Umsetzung kann z. B. so erfolgen, dass man Verbindungen der Formel V mit einer Verbindung der Formel IV in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem aromatischen Kohlenwasserstoff, wie Toluol oder Benzol, in einem cyclischen Äther, wie Dioxan, in einem chlorierten Kohlenwasserstoff, wie Chloroform, einem Diniederalkylamid, einer niederen aliphatischen Monocarbonsäure, wie Dimethylformamid oder in Aceton, vorteilhafterweise in Gegenwart eines basischen Kondensationsmittels, z. B. eines Alkalimetallkarbonates, wie Natrium- oder Kaliumkarbonat, oder einer organischen Base, wie Triäthylamin oder Pyridin, bei Temperaturen von etwa 50 bis 1500 umsetzt. Die Reaktionszeit kann 1 bis 10 Stunden betragen.
Die Umsetzung lässt sich jedoch auch ohne Lösungsmittel in der Schmelze durchführen, wobei die Reaktionstemperatur zwischen etwa 80 und 200 und die Reaktionszeit zwischen etwa 1/2 und 5 Stunden betragen können.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden. Sie besitzen insbesondere bronchospasmolytische Wirkungen, wie sich in Tierversuchen zeigt. So hemmen sie an der Katze in Dosen von etwa 0,0002 bis 0,01 mg/kg intravenös injiziert den durch Histamin induzierten Bronchospasmus.
Aufgrund ihrer bronchospasmolytischen Wirkungen können die Substanzen zur Behandlung von obstruktiven Atemwegserkrankungen, wie Bronchospasmen, z. B. Asthma bronchiale, verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von 0,05 bis 0,15 mg/kg Körpergewicht p. o.
appliziert erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 2 bis 10 mg.
Für orale Applikationen enthalten die Teildosen etwa 0,5 bis 5 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Zur Inhalation können Sprays, welche etwa 1% der Verbindungen der Formel I neben geeigneten Trägersubstanzen enthalten, verwendet werden.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In dem nachfolgenden Beispiel, das die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel
N,N' -Bis[2-hydroxy-2-(4-hydroxy-3 hydroxymethylphenyl)äthylj-2,2'-(o-phenylen)bis (äthylamin)
13,0 g N,N'-Bis[2-hydroxy-2-(3-hydroxymethyl-4 methoxyphenyl)-äthyl]-2,2' -(o-phenylen)bis(äthylamin) werden in 150 ml Methylenchlorid gelöst und auf -75" gekühlt. 15 g Brotribromid werden in Form einer einmolaren Lösung in Methylenchlorid so langsam unter Rühren zugetropft, dass die Temperatur nicht über 700 steigt. Man lässt das Gemisch langsam auf Zimmertemperatur kommen, destilliert nach 5 Stunden das Lösungsmittel ab und erhitzt den Rückstand eine Stunde lang mit 100 ml Äthanol am Rückfluss. Nach dem Abdestillieren des Äthanols löst man den Rückstand in 100 ml Wasser und bringt mit Ammoniak auf pH 8-9. Die Titelverbindung scheidet sich langsam ab.
Man kristallisiert sie nach dem Filtrieren und Trocknen aus Essig ester um. Smp. 67 bis 70".
Das Ausgangsmaterial kann wie folgt erhalten werden: a) 57,2 g 3-Carbomethoxy-4-methoxyphenacylbromid und 54,0 g N,N'-Bis(benzyl)-2 N,N'-Bis(benzyl)-2,2'-(o-phenylen)bis(äthylamin) werden in 250 ml Methyläthylketon 2 Stunden lang am Rückfluss erwärmt. Dann kühlt man die Mischung im Eisbad, filtriert ab und dampft das Filtrat ein. Das als Ö1 zurückbleibende N,N'-Dibenzyl-N,N'-bis(3 -carbomethoxy-4-methoxy- phenacyl)-2,2' -(o-phenylen)bis(äthylamin) wird ohne Reinigung weiterverarbeitet.
b) 58,3 g des vorstehend beschriebenen Produkts werden mit 12,0 g Lithiumaluminiumhydrid durch zweistündiges Er hitzen am Rückfluss in 1500 ml Tetrahydrofuran reduziert.
Man zersetzt mit wenig Wasser, filtriert ab und löst den Filterrückstand in 2N Salzsäure. Durch Zusatz von Kaliumhydrogencarbonat stellt man ein pH von 89 ein und extrahiert den Brei erschöpfend mit Isobutanol. Die vereinigten Tetrahydrofuran- und Isobutanolextrakte werden über Natriumsulfat getrocknet und eingedampft. Hierbei bleibt das N,N'-Dibenzyl N,N' -bis[2-hydroxy-2- (4-methoxy-3 -hydroxymethylphenyl) äthyl]-2,2'-(o-phenylen)bis(äthylamin) als beinahe farb loses Harz zurück, das direkt weiterverarbeitet wird.
c) 30 g des vorstehend beschriebenen Produktes werden in 150 ml Äthanol gelöst und nach Zusatz von 1,0 g Palladiumkohle bei 50 und 4 atü Wasserstoffdruck bis zur Beendigung der Wasserstoffaufnahme hydriert. Man filtriert den Katalysator ab, dampft das Filtrat ein und erhält das N,N' Bis[2-hydroxy-2-(3-hydroxymethyl-4-methoxyphenyl)äthyl]- 2,2'-(o-phenylen)-bis(äthylamin) als farbloses Harz, das direkt weiterumgesetzt werden kann.

The compounds of the formula I can be isolated and purified from the reaction mixture in a manner known per se and, if desired, can be converted into their acid addition salts.
The compounds of formula II can, for. B. be obtained by reducing compounds of the formula III in which R and n have the above meanings and R2 is lower alkyl.
The reduction can be carried out, for example, in a solvent which is inert under the reaction conditions, e.g. B. in an ether such as diethyl ether, tetrahydrofuran, dioxane, dimethoxyethane with lithium aluminum hydride or aluminum hydride.
Compounds of formula III can, for. B. can be obtained by reacting compounds of the formula IV in which R1 has the above meaning and X is chlorine or bromine with compounds of the formula V in which n has the above meaning.
