Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Bis[(3,4-Dihydroxyphenyl)propanol]diamin-derivaten der Formel I. worin n für eine ganze Zahl von 2 bis 10 steht, und ihren Säureadditionssalzen.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin n obige Bedeutung besitzt und R1 Methyl, Äthyl oder Benzyl bedeutet, mit ätherspaltenden Mitteln behandelt, und die erhaltenen Verbindungen der Formel I in Form der freien Basen oder deren Säureadditionssalzen gewinnt.
In den Verbindungen der Formel I steht n vorzugsweise für eine ganze Zahl von 4 bis 8, insbesondere für 6.
Das erfindungsgemässe Verfahren kann nach an sich zur Ätherspaltung üblichen Methoden durchgeführt werden. So kann man beispielsweise die Verbindungen der Formel II mit Lewis-Säuren, z. B. mit Bortribromid oder Aluminiumchlorid, in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z. B. einem halogenierten Kohlenwasserstoff wie Methylenchlorid oder Tetrachlorkohlenstoff, oder einem aromatischen Kohlenwasserstoff wie Toluol oder Benzol, bei -80 bis +70 reagieren lassen, oder Verbindungen der Formel II kurzzeitig mit starken Mineralsäuren, wie z. B. mit Bromuasserstoff- oder Jodwasserstoffsäure, gegebenenfalls bei erhöhter Temperatur, z.
B. bei etwa 20 bis 100", behan deln. oder auf die Verbindungen der Formel II Hydrochloride, -Bromide oder -Jodide organischer Basen wie Anilin oder Pyridin bei erhöhter Temperatur einwirken lassen. Falls R1 in den Verbindungen der Formel II für Benzyl steht, kann die Ätherspaltung auch durch katalytische Hydrierung erfolgen.
Die hydrogenolytische Debenzylierung kann z. B. in Gegenwart eines Katalysators, vorzugsweise eines Palladiumkatalysators. in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in Essigester, einem niederen Alkanol wie Methanol oder Äthanol, bei Temperaturen zwischen 20 und 100" und bei 1 bis 200 Atmosphären Wasserstoffdruck durchgeführt werden.
Die Verbindungen der Formel I können auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden, die freien Basen lassen sich in üblicher Weise in ihre Säureadditionssalze überführen und umgekehrt.
Die Ausgangsverbindungen können beispielsweise wie folgt erhalten werden:
Verbindungen der Formel II können beispielsweise erhalten werden, indem man Verbindungen der Formel III, worin R2 für Wasserstoff oder, falls R1 Methyl oder Äthyl bedeutet, auch für Benzyl steht, R, und n obige Bedeutung besitzen und R3 für niederes Alkyl steht, mit gegebenenfalls komplexen Metallhydriden reduziert. Die Reduktion kann beispielsweise in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem Äther wie Diäthyläther, Tetrahydrofuran, Dioxan oder Dimethoxyäthan, mit Lithiumaluminiumhydrid oder Aluminiumhydrid erfolgen, wird vorzugsweise bei Temperatur zwischen Raumtemperatur und etwa 100" ausgeführt, und die Reaktionszeit kann 1/2 bis 5 Stunden betragen.
Bei der Reaktion erhaltene allfällige Verbindungen der Formel Ila, worin n obige Bedeutung besitzt und R11 Methyl oder Äthyl bedeutet, können anschliessend beispielsweise durch hydrogenolytische Debenzylierung z. B. bei den zur hydrogenolytischen Benzylätherspaltung angegebenen Bedingungen in Verbindungen der Formel II überführt werden.
Verbindungen der Formel III können z. B. erhalten werden. indem man Verbindungen der Formel IV, worin R2 und n obige Bedeutung besitzen, an Verbindungen der Formel V, worin R1 und R3 obige Bedeutung besitzen, addiert. Die Reaktion kann beispielsweise bei 20 bis 100" unter Verwendung äquimolarer Mengen der beiden Verbindungen und gegebenenfalls in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem niederen Alkohol, ausgeführt werden.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch inter essante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden. Sie besitzen insbesondere bronchospasmolytische Wirkungen, wie sich in Tierversuchen zeigt. So hemmen sie an der Katze in Dosen von etwa
0,0002 bis 0,01 mg/kg intravenös injiziert den durch Histamin induzierten Bronchospasmus.
Aufgrund ihrer bronchospasmolytischen Wirkungen können die Substanzen zur Behandlung von obstruktiven Atemwegserkrankungen wie Bronchospasmen, z. B. Asthma bronchiale, verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von 0.05 bis 0,15 mg/kg Körpergewicht p.o. appliziert erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 2 bis 10 mg. Für orale Applikationen enthalten die Teildosen etwa 0,5 bis 5 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Zur Inhalation können Sprays, welche etwa 1,0% der Verbindungen der Formel I neben geeigneten Trägersubstanzen enthalten, verwendet werden. Als besonders wirksam erwiesen sich beispielsweise das N,N'-Bis[3 -hydroxy-2-(3 ,4-dihydro- xyphenyl)-l-propyl]hexamethylendiamin und seine Säureadditionssalze.
Als Heilmittel können die Verbindungen der Formel I bzw.
ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1
N,N'-Bis[3 -hydroxy-2-(3,4-dihydroxyphenyl)- 1-propyl] hexamethylendiamin 13,0 g N,N'-Bis[3-hydroxy-2-(3,4-dimethoxyphenyl)-1- propyl]-hexamethylendiamin werden in 150 ml Methylenchlorid gelöst und auf -75" gekühlt. 30 g Bortribromid werden in Form einer einmolaren Lösung in Methylenchlorid so langsam unter Rühren hinzugetropft, dass die Temperatur nicht über -70" steigt. Man lässt das Gemisch sich langsam auf Raumtemperatur erwärmen, destilliert nach 5 Stunden das Lösungsmittel ab und erhitzt den Rückstand eine Stunde lang mit 100 ml Äthanol am Rückfluss. Nach dem Abdestillieren des Äthanols wird der Rückstand aus Äthanol/Äther umkristallisiert.
Smp. des Dihydrobromids der Titelverbindung: 86 bis 88".
Das Ausgangsmaterial wird wie folgt erhalten.
a) 12,8 g 1,6-Diaminohexan, 60,0 g 3,4-Dimethoxyatropasäureäthylester und 20 ml Äthanol werden 3 Stunden auf 60 erwärmt. Nach dem Abdestillieren des Lösungsmittels nimmt man in Äther auf, extrahiert mit 2n Salzsäure, stellt den salzsauren Extrakt mit 2n Natronlauge unter Kühlung alkalisch und extrahiert dann mit Äther. Die Ätherphase wird über Natriumsulfat getrocknet und eingedampft. Den Rück stand, das rohe N,N' -Bis [2-carbäthoxy-2-(3 ,4-dimethoxy- phenyl)-1-äthyl]hexamethylendiamin führt man in das Naphthalin-1,5-disulfonat über, das sich aus Äthanol umkristallisieren lässt. Smp. 176 bis 179".
b) 50 g des unter a) erhaltenen N,N'-Bis[2-carbäthoxy 2-(3 ,4-dimethoxyphenyl)-1-äthyl]hexamethylendiamin reduziert man in 500 ml Tetrahydrofuran mit 7,6 g Lithiumaluminiumhydrid 2 Stunden bei 70". Nach dem Zersetzen mit wenig Wasser wird abfiltriert, über Natriumsulfat getrocknet und eingedampft. Der Rückstand, das N,N'-Bis[3-hydroxy-2 (3 ,4-dimethoxyphenyl) -1 -propyl]hexamethylendiamin wird aus Essigester umkristallisiert, Smp. 92 bis 94 .
Beispiel 2
N,N' -Bis [3 -hydroxy-2-(3 ,4-dihydroxyphenyl)propyl] - tetramethylendiamin
N,N' -Bis [3-hydroxy-2-(3 ,4-dimethoxyphenyl)- 1-propyl] tetramethylendiamin wird nach dem in Beispiel 1 beschriebenen Verfahren umgesetzt. Smp. des Dihydrobromids der Titelverbindung 81 bis 85".
Ausgangsmaterial hergestellt analog Beispiel 1: a) Ausgehend von 3,4-Dimethoxyatropasäureäthylester und 1,4-Diaminobutan erhält man das ölige N,N'-Bis[2-carb äthoxy-2-(3,4-dimethoxyphenyl)- 1-äthyljtetramethylendi- amin und 4 b) daraus durch Reduktion das N,N'-Bis[3-hydroxy-2 (3,4-dimethoxyphenyl)-1-propyl]tetramethylendiamin. Smp.
des Dihydrochlorids 158 bis 1600.
Beispiel 3
N,N' -Bis [3 -hydroxy-2-(3,4-dihydroxyphenyl)- 1-propyl] äthylendiamin
11,0 g N,N'-Bis[3-hydroxy-2-(3,4-dimethoxyphenyl)-1- propyl]äthylendiamin wird nach dem in Beispiel 1 beschriebenen Verfahren umgesetzt.
Smp. des Dihydrobromids der Titelverbindung: 111" (Zersetzung).
Ausgangsmaterial, hergestellt analog Beispiel 1: a) Ausgehend von 3,4-Dimethoxyatropasäureäthylester und 1,2-Äthylendiamin erhält man das rohe N,N'-Bis[2-carb äthoxy-2-(3,4-dimethoxyphenyl)- 1-äthyl]äthylendiamin und führt dieses in das Dihydrochlorid über, das sich aus Äthanol umkristallisieren lässt.
Smp. 176-179 .
b) Durch Reduktion des N,N'-Bis[2-carbäthoxy-2-(3,4 dimethoxyphenyl)-l-äthyl]äthylendiamin erhält man das N,N'-Bis[3 -hydroxy-2-(3,4-dimethoxyphenyl) - 1 -propyl]- äthylendiamin, welches in das Dihydrochlorid übergeführt wird.
Smp. oberhalb 250 (Zersetzung).
Beispiel 4
N,N' Bis[3-hydroxy-2-(3 ,4-dihydroxyphenyl)propyl] - trimethylendiamin
N,N' -Bis [3-hydroxy-2-(3 ,4-dimethoxyphenyl) 1 -propyl]trimethylendiamin wird nach dem in Beispiel 1 beschriebenen Verfahren umgesetzt. Smp. des Dihydrobromids der Titelverbindung 78-81".
Ausgangsmaterial hergestellt analog Beispiel 1: a) Ausgehend von 3,4-Dimethoxyatropasäureäthylester und 1,3-Diaminopropan erhält man das ölige N,N'-Bis[2 carbäthoxy-2-(3,4-dimethoxyphenyl)-1-äthyl]trimethylendi- amin und b) daraus durch Reduktion das N,N'-Bis[3-hydroxy-2 (3 ,4-dimethoxyphenyl)-1 -propyl]trimethylendiamin.

Beispiel 5 N,N'-Bis[3 -hydroxy-2-(3 ,4-dihydroxyphenyl)-propyll- octamethylendiamin
N,N' -Bis[3 -hydroxy-2-(3 ,4-dimethoxyphenyl) - 1-propyl]- octamethylendiamin wird nach dem in Beispiel 1 beschriebenen Verfahren umgesetzt. Smp. des Dihydrobromids der Titelverbindung 66-69 .
Ausgangsmaterial hergestellt analog Beispiel 1: a) Ausgehend von 3,4-Dimethoxyatropasäureäthylester und Octamethylendiamin erhält man das ölige N,N' -Bis [2- carbäthoxy-2-(3,4-dimethoxyphenyl)- 1 -äthyl]octamethylendiamin und b) daraus durch Reduktion das N,N'-Bis[3-hydroxy-2- (3 ,4-dimethoxyphenyl) 1 1 -propyl]octamethylendiamin.
Smp. 80-82".
Beispiel 6
N,N' -Bist3 -hydroxy-2-(3,4-dihydroxyphenyl)-propyl]- decamethylendiamin N,N'-Bis [3 -hydroxy-2-(3,4-dimethoxyphenyl)- 1 -propyl]decamethylendiamin wird nach dem im Beispiel 1 beschriebenen Verfahren umgesetzt.
Smp. des Dihydrobromids der Titelverbindung 65-70".
Ausgangsmaterial hergestellt analog Beispiel 1: a) Ausgehend von 3,4-Dimethoxyatropasäureäthylester und Decamethylendiamin erhält man das ölige N,N'-Bis[2 carbäthoxy-2-(3,4 -dimethoxyphenyl) - -äthyl]decamethylen- diamin und b) daraus durch Reduktion das N,N'-Bis[3-hydroxy-2 (3,4-dimethoxyphenyl)- 1 -propyl]decamethylendiamin.
Smp. 70-74".
Beispiel 7
N,N' -Bis [3 -hydroxy-2-(3 ,4-dihydroxyphenyl)- 1-propyl] hexamethylendiamin
5,0 g N,N' -Bis[3 -hydroxy-2-(3,4-dibenzyloxyphenyl)- 1 - propyl]hexamethylendiamin löst man in 100 ml Methanol und hydriert nach Zusatz von 0,2 g Palladium/Kohle bei 5 atü Wasserstoffdruck und 40". Nach 4 Stunden wird der Katalysator abfiltriert und das Filtrat eingedampft. Die zurückbleibende Titelverbindung führt man mit äthanolischer Bromwasserstoffsäure in das Dihydrobromid über und kristallisiert dieses aus Äthanol/Äther um. Smp. des Dihydrobromids der Titelverbindung: 86 bis 88".
Das Ausgangsmaterial wird wie folgt hergestellt: a) 65,2 g a -(3 ,4-Dibenzyloxyphenyl)acrylsäureäthylester und 10,0 g 1,6-Diaminohexan werden in 30 ml Äthanol 4 Stunden am Rückfluss erhitzt. Man destilliert das Lösungsmittel im Vakuum ab und erhält als Rückstand das rohe N,N'-Bis-[2carbäthoxy-2-(3 ,4-dibenzyloxyphenyl)- 1-äthyl]hexamethylendiamin als dickes Ö1, welches ohne Reinigung weiterverarbeitet wird.
b) 25 g des vorstehend beschriebenen Produkts werden in 300 ml Tetrahydrofuran mit 4,5 g Lithiumaluminiumhydrid durch dreistündiges Erhitzen am Rückfluss reduziert. Man zersetzt dann unter Kühlung mit wenig Wasser, filtriert, trokknet die Tetrahydrofuranlösung über Natriumsulfat und destilliert das Lösungsmittel ab. Das rohe N,N'-Bis[3-hydroxy-2 (3,4-dibenzyloxyphenyl)- 1-propyl]hexamethylendiamin bleibt als harzartige Substanz zurück und wird ohne Reinigung weiterverarbeitet.

The compounds of the formula I and their pharmacologically acceptable acid addition salts have not yet been described in the literature. They are characterized by interesting pharmacodynamic properties and can therefore be used as remedies. In particular, they have bronchospasmolytic effects, as has been shown in animal experiments. Example 7
N, N '-Bis [3-hydroxy-2- (3, 4-dihydroxyphenyl) -1-propyl] hexamethylenediamine
5.0 g of N, N'-bis [3-hydroxy-2- (3,4-dibenzyloxyphenyl) -1-propyl] hexamethylenediamine are dissolved in 100 ml of methanol and, after addition of 0.2 g of palladium / carbon, hydrogenated at 5 atmospheric hydrogen pressure and 40 ". After 4 hours, the catalyst is filtered off and the filtrate is evaporated. The remaining title compound is converted into the dihydrobromide with ethanolic hydrobromic acid and this is recrystallized from ethanol / ether. Mp. of the dihydrobromide of the title compound: 86 to 88" .
The starting material is prepared as follows: a) 65.2 g of a - (3,4-Dibenzyloxyphenyl) acrylic acid ethyl ester and 10.0 g of 1,6-diaminohexane are refluxed in 30 ml of ethanol for 4 hours. The solvent is distilled off in vacuo and the crude N, N'-bis [2carbethoxy-2- (3, 4-dibenzyloxyphenyl) -1-ethyl] hexamethylenediamine is obtained as a thick oil, which is processed further without purification.
b) 25 g of the product described above are reduced in 300 ml of tetrahydrofuran with 4.5 g of lithium aluminum hydride by refluxing for three hours. It is then decomposed while cooling with a little water, filtered, the tetrahydrofuran solution is dried over sodium sulfate and the solvent is distilled off. The crude N, N'-bis [3-hydroxy-2 (3,4-dibenzyloxyphenyl) -1-propyl] hexamethylenediamine remains as a resinous substance and is processed further without purification.
