Die Erfindung betrifft ein Verfahren zur Herstellung neuer Bis[(3 ,4-dihydroxyphenyl)propanol]diamin-derivate der Formel I, worin n eine ganze Zahl von 2 bis 4 bedeutet, sowie ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Bis[(3,4-dihydroxyphenyl)propanol]diamin-derivaten der Formel I, und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin n obige Bedeutung besitzt und R1 je Methyl, Äthyl oder Benzyl bedeutet, mit ätherspaltenden Mitteln behandelt und die erhaltenen Verbindungen der Formel I in Form der freien Basen oder ihrer Säureadditionssalze gewinnt.
In den Verbindungen der Formel I bedeutet n vorzugsweise 2 oder 3.
Das erfindungsgemässe Verfahren kann nach an sich zur Ätherspaltung üblichen Methoden durchgeführt werden. So kann man beispielsweise die Verbindungen der Formel II mit Lewis-Säuren, z. B. mit Bortribromid oder Aluminiumchlorid, in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z. B. einem halogenierten Kohlenwasserstoff, wie Methylenchlorid oder Tetrachlorkohlenstoff, oder einem aromatischen Kohlenwasserstoff, wie Toluol oder Benzol, bei80 bis +70 reagieren lassen, oder Verbindungen der Formel II kurzzeitig mit starken Mineralsäuren, wie z. B.
mit Bromwasserstoff- oder Jodwasserstoffsäure, gegebenenfalls bei erhöhter Temperatur, z. B. bei etwa 20 bis 100 , behandeln, oder auf die Verbindungen der Formel II Hydrochloride, -bromide oder -jodide organischer Basen, wie Anilin oder Pyridin, bei erhöhter Temperatur einwirken lassen.
Falls Rl in den Verbindungen der Formel II für Benzyl steht, kann die Ätherspaltung auch durch hydrogenolytische Debenzylierung erfolgen, z. B. indem man die Verbindungen in Gegenwart eines Katalysators, vorzugsweise eines Palladiumkatalysators, in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z. B. in Essigester oder einem niederen Alkohol bei Temperaturen zwischen 20 und 100" mit 1 bis 200 Atmosphären Wasserstoffdruck hydriert.
Die Verbindungen der Formel I können auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden und können gewünschtenfalls in ihre Säureadditionssalze überführt werden.
Die Ausgangsverbindungen können beispielsweise wie folgt erhalten werden:
Verbindungen der Formel II können beispielsweise erhalten werden, indem man Verbindungen der Formel III, worin R2 für Wasserstoff oder, falls R1 Methyl oder Äthyl bedeutet, auch für Benzyl steht, R1 und n obige Bedeutung besitzen und R3 für niederes Alkyl steht, mit gegebenenfalls komplexen Metallhydriden reduziert. Die Reduktion kann beispielsweise in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem Äther wie Diäthyl äther, Tetrahydrofuran, Dioxan oder Dimethoxyäthan, mit Lithiumaluminiumhydrid oder Aluminiumhydrid erfolgen, wird vorzugsweise bei Temperatur zwischen Raumtemperatur und etwa 100" ausgeführt, und die Reaktionszeit kann l/2 bis 5 Stunden betragen.
Bei der Reaktion erhaltene allfällige Verbindungen der Formel IIa, worin n obige Bedeutung besitzt, R11 Methyl oder Äthyl bedeutet und R21 für Benzyl steht, können anschliessend beispielsweise durch hydrogenolytische Debenzylierung z. B. bei den zur hydrogenolytischen Benzylätherspaltung angegebenen Bedingungen in Verbindungen der Formel II überführt werden.
Verbindungen der Formel III können z. B. erhalten werden, indem man Verbindungen der Formel IV, worin R2 und n obige Bedeutung besitzen, an Verbindungen der Formel V, worin R1 und R3 obige Bedeutung besitzen, addiert.
Die Reaktion kann beispielsweise bei 20 bis 100" unter Verwendung äquimolarer Mengen der beiden Verbindungen und gegebenenfalls in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem niederen Alkohol, ausgeführt werden.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden. Sie besitzen insbesondere bronchospasmolytische Wirkungen, wie sich in Tierversuchen zeigt. So hemmen sie an der Katze in Dosen von etwa 0,0002 bis 0,01 mg/kg intravenös injiziert den durch Histamin induzierten Bronchospasmus.
Aufgrund ihrer bronchospasmolytischen Wirkungen können die Substanzen zur Behandlung von obstruktiven Atemwegserkrankungen wie Bronchospasmen, z. B. Asthma bronchiale verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von 0,05 bis 0,15 mg/kg Körpergewicht p.o.
appliziert erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden.
Für grössere Säugetiere liegt die Tagesdosis bei etwa 2 bis 10 mg. Für orale Applikationen enthalten die Teildosen etwa 0,5 bis 5 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Zur Inhalation können Sprays, welche etwa 1,0% der Verbindungen der Formel I neben geeigneten Trägersubstanzen enthalten, verwendet werden. Als besonders wirksam erwiesen sich beispielsweise das 1,4-Bis(2-E3 -hydroxy-2-(3,4-di- hydroxyphenyl)-1-propanolamino]äthyl}benzol und dessen Säureadditionssalze.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In dem nachfolgenden Beispiel, das die Erfindung näher erläutert, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel
Bis- {N-[2-(3,4-dihydroxyphenyl)-3-hydroxy-1-propyl]- 2-amino- 1-äthylj-p-phenylen
10,2 g Bis-{N-[2-(3,4-dimethoxyphenyl)-3-hydroxy 1-propyl]-2-amino-1-äthyl}-p-phenylen werden in 250 ml Methylenchlorid gelöst und bei -70 bis -75" tropfenweise unter Rühren mit 25,0 g Bortribromid in Form einer lmola- ren Lösung in Methylenchlorid versetzt. Man lässt auf Raumtemperatur kommen und destilliert dann das Lösungsmittel im Vakuum ab. Der Rückstand wird mit 100 ml Äthanol am Rückfluss erhitzt, beim Abkühlen kristallisiert die Titelverbindung als Dihydrobromid aus. Smp. 78".
Das Ausgangsmaterial kann wie folgt hergestellt werden: a) 65,0 g 3,4-Dimethoxyatropasäureäthylester und 19,5 q Bis(2-Amino-1-äthyl)-p-phenylen werden mit 40 ml Äthanol 3 Stunden bei 70" gerührt. Anschliessend destilliert man das Lösungsmittel im Vakuum ab und löst den Rückstand in 500 ml Äther. Man extrahiert mit 200 ml 2n Salzsäure, trennt ab und stellt den sauren Extrakt mit 2n Natronlauge alkalisch, extrahiert wiederum mit 500 ml Äther, trocknet die Ätherphase, dampft ein und führt das zurückbleibende ölige Bis-(N-[2-(3,4 -dimethoxyphenyl) -2-carbäthoxy- 1-äthyl]-2-amino-1-äthyl}-p-phenylen ins Dihydrochlorid über. Nach der Umkristallisation aus Äthanol/Äther zeigt das Salz einen Schmelzpunkt von 106-108".
b) 38,1 g des vorstehend beschriebenen Produktes in Form der freien Base werden in 400 ml absolutem Tetrahydrofuran mit 5,7 g Lithiumaluminiumhydrid durch 2stündiges Erhitzen am Rückfluss reduziert. Man zersetzt unter Kühlung mit wenig Wasser, filtriert, trocknet die Tetrahydrofuran-Lösung und destilliert das Lösungsmittel ab. Der ölige Rückstand kristallisiert aus Äther/Petroläther, Smp.
des Bis-(N-[2-(3,4 -dimethoxyphenyl) -3-hydroxy-l -propyl]- 2-amino-l-äthyl)-p-phenylen 112".
