Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer therapeutisch wirksamer Derivate der 7-Aminocephalosporansäure (ACA) der Formel I

worin Rl einen S-Aminotetrazolylrest bedeutet und R2 Wasserstoff oder eine freie oder durch eine Carbonsäure oder Thiocarbonsäure veresterte Hydroxylgruppe ist, u.
ihrer Salze.
Eine wie erwähnt veresterte Hydroxylgruppe R2, die sich von einer Carbonsäure oder Thiocarbonsäure ableitet. ist beispielsweise eine gegebenenfalls z.B. durch Ha logenatom e, besonders Chlor, substituierte Niederalkan- oyloxygruppe wie Propionyloxy, Butyryloxy, Pivaloyloxy, Chloracetoxy, insbesondere Acetoxy, oder eine gegebenenfalls z.B. durch Niederalkyl-, Niederalkoxy- oder Nie deralkylmereaptoreste, Halogenatome oder die Nitrogruppe substituierte mono- oder dicyclische Arylcarbonyloxy- oder Arylthiocarbonyloxygruppe, insbesondere die Thiobenzoyloxygruppe.
R2 kann auch eine substituierte Carbamoyloxygruppe, z.B. eine Gruppe der Formel -O-CO-NiH-R, sein, worin R3 ein aliphatischer, aromatischer, araliphatischer oder heterocyclischer Rest, besonders ein unsubstituierter oder substituierter, vorzugsweise durch eine oder mehrere Niederalkoxygruppen oder Halogenatome substituierter, gerader oder verzweigter Niederalkylrest. wie der Methyl-, Äthyl-, vor allem aber der ss-Chloräthylrest, ist.
Die Salze der neuen Verbindungen können Metallsalze, vor allem solLte von therapeutisch anwendbaren Alkali- oder Erdalkalimetallen, wie Natrium, Kalium, Ammonium, Calcium, oder Salze mit organischen Basen. z.B. Triäthylamin, N-Äthylpiperidin, Dibenzylamin, N-Benzyl-y-phenetylamin, N,N'-Dibenzyläthylendiamin, Procain, Ephenamin sein. Ist :R2 basisch, so können sich innere Salze bilden.
Die neuen Verbindungen weisen eine besonders gute antibakterielle Wirkung auf. Sie sind sowohl gegenüber grampositiven wie vor allem auch gegenüber gramnegativen Bakterien wirksam, z. B. gegen Staphylococcus aureus penicillin-resistent, Escherichia coli, Kiebsiella pneumoniae, Salmonelsla typhosa und Bacterium proteus, wie sich auch im Tierversuch, z.B. an Mäusen, zeigt. An diesen sind bei subeutaner Anwendung, je nach Art der bakteriellen Infektion, 0,1-100 mg/kg chemotherapeutisch wirksam.
Die Verbindungen können daher zur Bekämpfung von Infektionen, die durch solche Mikroorganismen hervorgerufen werden, verwendet werden, ferner als Fut termittelzusätze, zur Konservierung von Nahrungsmitteln oder als Desinfektionsmittel. Besonders wertvoll sind Verbindungen, in denen R die Acetoxygruppe oder eine unsubstituierte Pyin.diniumgruppe ist.
Die Verbindungen der vorliegenden Erfindung können nach an sich bekannten Methoden hergestellt werden. Erfindungsgemäss werden sie erhalten, wenn man eine Verbindung der Formel II

worin Z für Wasserstoff steht und R2 die angegebene Bedeutung hat, oder ein Salz davon durch die Gruppe R1-CH2-CO-, worin R1 die angegebene Bedeutung hat, acyliert. Wenn erwünscht, können erhaltene Verbindungen, in denen R2 für die Acetoxygruppe steht, in Verbindungen mit freier Hydroxylgruppe R2 und Verbindungen mit freier Hydroxylgruppe R in Verbindungen, welche eine andere Estergruppe als die Acetoxygruppe enthalten, übergeführt werden.
Man kann auch erhaltene Verbindungen, in denen R für eine durch eine Carbonsäure oder Thiocarbonsäure veresterte Hydroxylgruppe steht, in Verbindungen, worin R2 eine quaternäre Aminogruppe, z. B. eine gegebenenfalls substituierte Pyridiniumgruppe, bedeutet, umwandeln. Wenn erwünscht, können die erhaltenen Verbindungen in ihre therapeutisch verwendbaren Metall-, wie Alkali- oder Brdalkali- metallsalze, oder Salze mit Ammomak oder organischen Basen übergeführt oder aus erhaltenen Salzen die freien Carbonsäuren oder gegebenenfalls inneren Salze gebildet werden.
Die Acylierung der Verbindung II, worin Z für Wasserstoff steht wird in der für die Acylierung von Aminosäuren bekannten Weise, z.B. mittels eines Säurehalogenids, besonders Säurechlorids, oder Säureazids oder eines Säureanhydrids, insbesondere eines gemischten Anhydrids, z.B. eines mit monoveresterter Kohlensäure, Pivalinsäure oder Trichloressigsäure gebildeten gemischten Anhydrids, oder mit der freien Säure selbst in Gegenwart eines Kondensationsmittels wie eines Carbodiimids, z.B. des Dicyclohexylcarbodiimids, vorgenommen. Man kann die Acylierung der Verbindung II auch in der Weise vornehmen, dass man die Verbindung II, worin Z für Wasserstoff steht, zunächst silyliert oder stannyliert, das Silylierungs- bzw.
Stannylierungsprodukt mit der Säure oder einem reaktionsfähigen Säurederivat, welches die Gruppe Rl-CH2-CO- enthält, acyliert und gegebenenfalls vorhandene Silyl- oder Stanylgruppen durch Alkohol oder Wasser abspaltet, vgl. z.B. die britischen Patente 1073 530 und 1 211 694.
Die als Ausgangsstoffe verwendeten 7 -Am inocepha - losporansäurederivate sind bekannt oder können nach an sich bekannten Verfahren hergestellt werden. Verbindungen der Formel II, worin Z für Wasserstoff steht und R2 eine andere Estergruppe als die Acetoxygruppe ist, werden vorteilhaft nach dem im Schweizer Patent Nr. 507 983 beschriebenen Verfahren hergestellt.
Die Verbindung der Formel I, worin R2 für die Acetoxygruppe steht, kann in an sich bekannter Weise durch Desacetyllerung mittels Acetylesterase in eine Verbindung mit freier Hydroxylgeuppe R2 umgewandelt werden. Verbindungen der Formel 1, worin R2 für eine durch eine Carbonsäure oder Thiocarbonsäure veresterte gHy- droxylgruppe steht, können in an sich bekannter Weise durch Umsetzung mit einem tertiären Amin in Verbin zungen, worin R2 eine quaternäre A,minogruppe ist, überführt werden.
Die neuen Verbindungen können als Heilmittel, z.B.
in Form pharmazeutischer Präparate, Verwendung finden. Diese enthalten die Verbindungen in Mischung mit einem für die enterale, topicale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial.
Die Erfindung wird in den folgenden Beispielen beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
In der Dünnschichtchromatographie auf Silicagel platten werden die folgenden Systeme verwendet: System 52 = n-Butanol Eisessig-Wasser (75 : 7,5 : 21), System 101 A = n-Butanol-Pyridin-Eisessig-Wasser (42: 24:4:30).
Beispiel I
In einem mit Rührer versehenen 1 50-ml-Sulfierkol- ben wird eine Lösung von 1,58 g 5-Amino-l-tetrazolylessigsäure und 1,5 ml Triäthylamin in 30 ml Tetrahydrofuran unter Stickstoffatmosphäre auf - 500C abgekühlt und mit 1,2 mi Trichloractylchlorid versetzt. Man lässt unter Rühren während 15 Minuten bei der gleichen Temperatur reagieren. Dann wird rasch unter intensivem Rühren und Kühlen eine kalte Lösung (ca. - 500C) von
1,13 g 7-Aminocephalosporansäure und 2,5 ml Tri äthylamin in 75 ml Methylenchlorid zugegeben und noch während 45 Minuten weitergerührt.
Hierauf wird das Reaktionsgemisch auf 30 ml 5%ige wässerige Kaliumhydrogenphosphat-Lösung gegossen, geschüttelt und auf pH 6,5 eingestellt. Man trennt die Schichten und extrahiert die wässerige Phase zuerst mit 50 ml Methylenchlorid und dann mit 50 ml Essigester nach. tDie organischen Phasen werden zweimal mit je 10 ml 0,1 m.
Phosphatpuffer pH 6,7 zurückgewaschen und dann verworfen. Die vereinigten wässerigen Phasen werden mit 200 ml Essigester überschichtet, mit 4-n. Salzsäure unter Schütteln auf pH 2,2 eingestellt und die Phasen getrennt.
Nach Sättigung mit Kochsalz wird die wässerige Phase noch zweimal mit je 100 ml Essigester extrahiert, die organischen Phasen sukzessive zweimal mit je 20 ml gesättigter Kochsalzlösung gewaschen, mit Natriumsulfat getrocknet und anschliessend durch eine Säule von 20 g Silicagel (0 27 mm, Höhe 70 mm) filtriert. Man wäscht die Säule mit 100 ml Essigester nach und dampft das filtrat im Vakuum zur Trockne ein. Der amorphe, schaumige Rückstand wird in Methanol aufgenommen und mittels Natn.um-oc-äthylhexanoat in ein Rohkristallisat des Natriumsalzes übergeführt.
Daraus erhält man, wie im schweizer Patent Nr. 507 985 beschrieben, die kristallisierte Säure und das reine kristallisierte Natriumsalz der 7-[5-Aminotetrazolyl(1)-acetylamino]-cephalo- sporansäure.

ausser den für die 7-ACA charakteristischen Signalen tritt ein Singlett bei N = 4,92 ppm auf, welches der -CH2-Gruppe des Aminotetrazolyl- acetylrestes zugeordnet werden kann, und ein Singlett mit breiter Basis bei 8 = 6,7 ppm für die am Kohlenstoff des Tetrazolringes stehende NH2EGruppe.
In analoger Weise kann man die 3-(Desacetoxyme thyl) - 3 - thiobenzoylmethyl-7-[5-aminotetrazolyl-acetyl- amino]-cephalosporansäure, herstellen.

Beispiel 2
2,51 g 3-(Desacetoxymethyl)-3-thiobenzoylmethyl-7- - [5 - aminotetrazolyl - acetylaminol - cephalosporansäure werden in 41,5 ml eines Gemisches aus gleichen Teilen Dioxan und Pyridin gelöst, mit 12,2 ml 40%iger wässeriger Quecksilberperchloratlösung versetzt und während 45 Minuten bei 450 und 45 KHz Ultraschall unter Stickstoff reagieren gelassen. Man kühlt ab, versetzt mit 6,5 ml Thiobenzoesäure und schüttelt 5 Minuten. Die Lösungsmittel werden im Vakuum abdestilliert und eine Lösung des Rückstandes in 65 ml Wasser wird durch Celite abfiltriert.
Das Filtrat wird nacheinander mit 50 ml Toluol, zweimal mit je 33 ml Amberlite LA-2 in 66 ml Toluol und zweimal mit je 50 ml Toluol gewaschen. Anschliessend filtriert man die wässerige Phase durch eine Säule, die von unten nach oben 4,1 ml Sephadex CM C-25 (H+-form), 16,5 ml Alox , 4,1 ml Zeo-Karb 226 (H+-form), 16,5 ml Alox , 4,1 ml Dowex 1 (Acetatform) und 4,1 ml Sephadex CM C-25 (11+-form) enthält. Celite)y, organische Phasen und die Säule werden zweimal mit je 15 ml Wasser nachextrahiert, die Säule zudem mit weiteren 150 ml Wasser eluiert.
Man engt die vereinigten Eluate im Vakuum ein, entfernt eine kleine Menge von Präcipitat durch Filtration und dampft zur Trockne ein. Der Rückstand wird mit ca. 50 ml abs.
Äthanol digeriert und ergibt die reine 3-(Desacetoxyme thyl) -3 -pyridiniomethyl-7 - [5-Aminotetrazolyl-acetylami no] -cephalosporansäure.

Compounds of formula 1 in which R 2 is a hydroxyl group esterified by a carboxylic acid or thiocarboxylic acid can be converted in a manner known per se by reaction with a tertiary amine in compounds in which R 2 is a quaternary A, amino group.
The new compounds can be used as medicaments, e.g.
in the form of pharmaceutical preparations, use. These contain the compounds in a mixture with a pharmaceutical, organic or inorganic, solid or liquid carrier material suitable for enteral, topical or parenteral administration.
The invention is described in the following examples. The temperatures are given in degrees Celsius.

Example I.
In a 150 ml sulfonation flask equipped with a stirrer, a solution of 1.58 g of 5-amino-1-tetrazolylacetic acid and 1.5 ml of triethylamine in 30 ml of tetrahydrofuran is cooled to -50 ° C. under a nitrogen atmosphere and with 1.2 ml Trichloractyl chloride added. Phosphate buffer pH 6.7 was washed back and then discarded. The combined aqueous phases are covered with 200 ml of ethyl acetate, with 4-n. Hydrochloric acid adjusted to pH 2.2 with shaking and the phases separated.
