Gegenstand der Erfindung ist die Herstellung neuer therapeutisch wirksamer Derivate der 7-Aminocephalosporansäure (ACA) der Formel I

worin R1 einen unsubstituierten oder niederalkalysubstituierten Amino-pyridiniumrest bedeutet une R2 Wasserstoff oder eine freie oder durch eine Carbonsäure veresterte Hydroxylgruppe, in der Estersauerstoffatome durch Schwefelatome ersetzt sein können, oder eine gegebenenfalls N-substituierte Carbamoyloxy- oder Thiocarbamoylmercaptogruppe ist. Pyridinrest enthält die Aminogruppe in ortho- oder vorzugsweise in para- oder meta-Stellung. Er kann als niedere Alkylgruppen z. B. Methyl- oder Äthylreste aufweisen.
Eine veresterte Hydroxylgruppe R2, in der Sauerstoffatome durch Schwefel ersetzt sein k6nnen, leitet sich von einer Car bonsäure ab und ist beispielsweise eine gegebenenfalls z. B.
durch Halogenatome, besonders Chlor, substituierte Niederalkanoyloxygruppe wie Formyloxy, Propionyloxy, Butyryloxy, Pivaloyloxy-Chloracetoxy, insbesondere Acetoxy, oder eine gegebenenfalls z. B. durch Niederalkyl-, Niederalkoxy- oder Niederalkylmercaptoreste, Halogenatome oder die Nitrogruppe substituierte mono- oder dicyclische Arylcarbonyloxyoder -thiocarbonyloxy-, Arylcarbonylmercapto- oder thiocarbonylmercaptogruppe, insbesondere die Benzoylmercaptogruppe.
R2 kann auch eine gegebenenfalls substituierte Carbamoyloxygruppe, z. B. eine Gruppe der Formel -O-CO-NH-R3 sein, worin R3 für Wasserstoff steht oder ein aliphatischer, @romatischer, araliphatischer oder herteroxyclischer Rest, besonders ein unsubstituierter oder substituierter, vorzugsweise durch eine oder mehrere Niederalkoxygruppen oder Halogenatome substituierter, gerader oder verzweigter Niederalkylrest, wie der Methyl-, Athyl-, vor allem aber der ss-Chlor- äthylrest, ist.

Die neuen Verbindungen weisen eine besonders gute antibakterielle Wirkung auf. Sie sind sowohl gegeniiber grampositiven wie vor allem auch gegeniiber gramnegativen Bakterien wirksam, z. B. gegen Staphylococcus aureus penicillin-resistent, Escherichia coli, Klebsielle pneumoniae und Salmonella typhosa, wie sich auch im Tierversuch, z. B. an Mäusen, zeigt.
an diesen sind bei subcutaner Anwendung, je nach Art der bakteriellen Infektion, 1-1000 mg/kg chemotherapeutisch wirksam. Die Verbindungen können daher zur Bekimpfung von Infektionen, die durch solche Mikroorganismen hervorgerufen werden, verwendet werden, ferner als Futterzusätze, zur Konservierung von Nahrungsmitteln oder als Desinfektionsmittel. Besonders wertvoll sind Verbindungen, in denen der Aminopyridiniumrest der para-Aminopyridiniumrest und R2 die Acetoxygruppe oder die Methyl-, Athyl- oder ss-Chlor äthylcarbamoyloxygruppe ist.
Die Verbindungen der vorliegenden Erfindung können nach an sich bekannten Methoden hergestellt werden. So werden sie erhalten, wenn man eine Verbindung der Formel II

Wenn erwünscht, können erhaltene Verbindungen, in denen R2 für die Acetoxygruppe steht, in Verbindungen mit freier Hydroxylgruppe R2 oder in Verbindungen, welche statt der Acetoxygruppe eine gegebenenfalls N-substituierte Thiocarbamoylmercaptogruppe enthalten, und Verbindungen mit freier Hydroxylgruppe R2 in Verbindungen, welche eine andere Estergruppe als die Acetoxygruppe enthalten (in der gegebenenfalls Sauerstoffatome durch Schwefelatome ersetzt sein k6nnen) oder in eine gegebenenfalls N-substituierte Carbamoyloxygruppe,
in der Sauerstoffatome durch Schwefelatome ersetzt sein kdnnen, iibergefiihrt werden.
Die Acylierung von Verbindung II wird in an sich bekannter, insbesondere in für die Acylierung von schwach basischen Aminogruppen durch Aminosäuren bekannter Weise durchgeführt, z. B. indem man die Verbindung II mit einem Säureadditionssalz, insbesondere dem Salz einer Halogenwasserstoffsäure wie oben erwähnt, z. B. Chlorwasserstoffsäure, des Säurehalogenids wie -fluorids, -bromids, -jodids, in erster Linie -chlorids, einer Säure der Formel RrCH2COOH umsetzt. Weiter kann die Acylierung durch Behandeln der Verbindung II mit einem N-Carboxyanhydrid (Leuch'ssches Anhydrid) einer Säure der Formel
R1-CH2-COOH erfolgen.
Die als Ausgangsstoffe verwendeten Cephalosporinderivate sind bekannt oder können nach an sich bekannten Verfahren hergestellt werden. Verbindungen der Formel II, worin R2 eine andere Estergruppe als die Acetoxygruppe ist, werden vorteilhaft nach dem im französischen Patent 1 588 507 beschriebenen Verfahren hergestellt.
Die Überführung der Verbindung der Formel I, worin R2 für die Acetoxygruppe oder für eine freie Hydroxylgruppe steht, in Verbindungen, worin R2 für eine andere Gruppe oder für Wasserstoff steht, erfolgt nach bekannten Methoden So kann man eine Verbindung der Formel I, worin R2 für die Acetoxygruppe steht, durch Reduktion in eine Verbindung, die statt der Acetoxygruppe ein Wasserstoffatom enthält, überführen. Die Umwandlung einer Acetoxygruppe R2 in eine Hydroxylgruppe erfolgt durch Behandlung mit desacetylierenden Mitteln, z. B. durch enzymatische Desacetylierung mit Citrus-Acetylesterase oder mit Acetylesterase aus Mikroorganismen wie Bacillus subtilis.
Durch Umsetzung mit einem Alkalimetall- oder Ammoniumsalz eines N-substituierten Dithiokohlensäureamids kann man die Acetoxygruppe in eine N-substituierte Thiocarbamoylmercaptogruppe überführen.
Eine freie Hydroxylgruppe R2 wird durch Umsetzung mit einem Isocyansäureester in eine N-substituierte Carbamoyloxygruppe übergeführt. Die Veresterung einer freien Hydroxylgruppe R2 mit einer anderen Carbonsäure als Essigsäure erfolgt z. B. nach dem im oben genannten französischen Patent angegebenen Verfahren.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, nach denen man von einer auf irgendeiner Stufe des Verfahrens als Zwischenprodukt erhältlichen Verbindung ausgeht und die fehlenden Verfahrensschritte durchführt, oder das Verfahren auf irgendeiner Stufe abbricht, oder bei denen man die Ausgangsstoffe unter den Reaktionsbedingun- gen bildet, oder bei denen die Reaktionskomponenten gegebenenfalls in Form ihrer Salze vorliegen.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden.
Diese enthalten die Verbindungen in Mischung mit einem für die enterale, topicale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial.
Die Erfindung wird in dem folgenden Beispiel beschrieben.
Die Temperaturen sind in Celsiusgraden angegeben.
In der Dünnschichtchromatographie auf Silicagelplatten werden die folgenden Systeme verwendet: System 52A = n-Butanol-Eisessig-Wasser (67:10:23) System 101 = n-Butanol-Pyridin-Eisessig-Wasser (38:24:8:30) System 101D = n-Butanol-Pyridin-Eisessig-Wasser (34:24:12:30) System 110 = Essigester-n-Butanol-Pyridin-Eisessig-Wasser (42:21:21:6:

Beispiel
Man stellt ein gemischtes Anhydrid aus p-Aminopyridini umessigsäure und Trichloressigsäure her, indem man eine Mischung von 0,2 ml Trichloracetylchlorid und 0,5 ml absolutem Dimethylformamid unter Rühren und leichtem Kühlen auf 240 ml p-Aminopyridiniumessigsäure tropft und die gelbe Lösung noch 15 Minuten bei 200 rührt. Zu dieser Lösung gibt man innerhalb von 15 Minuten tropfenweise unter leichter Kühlung und unter Rühren eine auf 0 abgekühlte Lösung von 365 mg 7-Aminocephalosporansäure und 0,23 ml Triäthyl- amin in 2 ml Dimethylformamid. Die Mischung wird noch 1 Stunde bei 200 gerührt. Dann fügt man 0,05 ml Triäthylamin zu und rührt weiter während einer Stunde.
Der Niederschlag wird abfiltriert und das Filtrat im Vakuum und dann im Hochvakuum eingedampft. Den Rückstand nimmt man in 2 ml Methanol auf, filtriert den Niederschlag ab und versetzt das Filtrat unter Rühren mit 50 ml Essigester. Man rührt eine Stunde bei 0 und nutscht dann den Niederschlag ab. Er wird weiter gereinigt, indem man ihn in 3 ml Methanol suspendiert, mit 2-n. Salzsäure auf pH 2 ansäuert, mit Aktivkohle behandelt und dann mit Triäthalamin auf pH 4 einstellt. Die Lösung wird hierauf langsam und unter Rühren in das 10-fache Volumen Essigester, der mit Impfkristallen versehen ist, einfliessen gelassen. Dabei scheidet sich die 7-(p-Aminopyridinium)acetylamino-cephalosporansäure in kristallisierter Form ab.
Im Ultraviolett-Spektrum (in Wasser) ist fmaX = 271 m (# = 30 900).
Auf Silicagel dünnschichtchromatographiert werden nach Anfärben mit Joddampf folgende Rf-Werte erhalten: Rf52 = 0,08 (für Bromacetyl-7-ACA ist Rfs2A = 0,39); Rf101 = 0,37 (für Cephaloridin ist Rf101 =0,39); Rf,("D = 0,44 für Cephaloridin ist RflolD = 0,44).
Die p-Aminopyridiniumessigsäure kann wie folgt hergestellt werden: Eine Lösung von 5,9 g Bromessigsäure und 5,5 g N,N-Di-isopropyläthylamin in 8 ml absolutem Methanol wird unter Rühren in eine filtrierte Lösung von 4,0 g Aminopyridin in 10 ml absolutem Methanol eingetropft. Wenige Minuten nach beendeter Zugabe beginnt die p-Aminopyridiniumessig- säure auszukristallisieren. Man filtriert, wäscht mit kaltem Methanol und erhält die Säure nahezu quantitativ in praktisch reiner Form. F. 303O (Zers.)
In gleicher Weise kann die 7-[(meta-Aminopyridinium)- acetylamino]-cephalosporansäure hergestellt werden.
Im UV-Spektrum (in Wasser) sind #max = 256 nm (# = 15 500) und #max = 328 nm (# = 3450). Die optische Dre hung,[α]20D = # 150 # 1 (c = 0,79 in Wasser).
Im Dünnschichtchromatogramm auf Silicagel (mit Jodspray) erhält man folgende Rf-Werte: Rfs2A = 0,07 (für Bromacetyl-7-ACA ist Rfs2A = 0,39); Rflol = 0,35 (für Cephaloridin ist Rflol = 0,39); RflolD = 0,43 (für Cephaloridin ist Rfloo = 0,44).

The new compounds have a particularly good antibacterial effect. They are effective against both gram-positive and especially against gram-negative bacteria, e.g. B. against Staphylococcus aureus penicillin-resistant, Escherichia coli, Klebsielle pneumoniae and Salmonella typhosa, as also found in animal experiments, e.g. B. on mice shows.

The acylation of compound II is carried out in a manner known per se, in particular in a manner known for the acylation of weakly basic amino groups by amino acids, e.g. B. by the compound II with an acid addition salt, in particular the salt of a hydrohalic acid as mentioned above, e.g. B. hydrochloric acid, the acid halide such as fluoride, bromide, iodide, primarily -chloride, an acid of the formula RrCH2COOH. The cephalosporin derivatives used as starting materials are known or can be prepared by processes known per se. Compounds of the formula II in which R2 is an ester group other than the acetoxy group are advantageously prepared by the process described in French patent 1,588,507.
The conversion of the compound of the formula I in which R2 stands for the acetoxy group or for a free hydroxyl group into compounds in which R2 stands for another group or for hydrogen is carried out by known methods. For example, a compound of the formula I in which R2 is the acetoxy group is, converted by reduction into a compound which contains a hydrogen atom instead of the acetoxy group. The conversion of an acetoxy group R2 into a hydroxyl group is carried out by treatment with deacetylating agents, e.g. B. by enzymatic deacetylation with citrus acetyl esterase or with acetyl esterase from microorganisms such as Bacillus subtilis.
The acetoxy group can be converted into an N-substituted thiocarbamoyl mercapto group by reaction with an alkali metal or ammonium salt of an N-substituted dithiocarbonamide.
A free hydroxyl group R2 is converted into an N-substituted carbamoyloxy group by reaction with an isocyanic acid ester. The esterification of a free hydroxyl group R2 with a carboxylic acid other than acetic acid takes place, for. B. according to the method indicated in the above French patent.
The invention also relates to those embodiments of the process in which one starts with a compound obtainable as an intermediate at any stage of the process and carries out the missing process steps, or the process is terminated at any stage, or in which the starting materials are formed under the reaction conditions , or in which the reaction components are optionally in the form of their salts.
The new compounds can be used as remedies, e.g. B. in the form of pharmaceutical preparations, use.
These contain the compounds in a mixture with a pharmaceutical, organic or inorganic, solid or liquid carrier material suitable for enteral, topical or parenteral administration.

A mixed anhydride is prepared from p-aminopyridinium acetic acid and trichloroacetic acid by adding a mixture of 0.2 ml of trichloroacetyl chloride and 0.5 ml of absolute dimethylformamide with stirring and slight cooling to 240 ml of p-aminopyridinium acetic acid and the yellow solution for a further 15 minutes at 200 stirs. A solution, cooled to 0, of 365 mg of 7-aminocephalosporanic acid and 0.23 ml of triethylamine in 2 ml of dimethylformamide is added dropwise to this solution over the course of 15 minutes, with slight cooling and with stirring. The mixture is stirred at 200 for a further hour. Then 0.05 ml of triethylamine is added and stirring is continued for one hour.
The precipitate is filtered off and the filtrate is evaporated in vacuo and then in a high vacuum. The residue is taken up in 2 ml of methanol, the precipitate is filtered off and the filtrate is treated with 50 ml of ethyl acetate while stirring. The mixture is stirred for one hour at 0 and then the precipitate is filtered off with suction. It is further purified by suspending it in 3 ml of methanol with 2-n. Acidified hydrochloric acid to pH 2, treated with activated charcoal and then adjusted to pH 4 with triethalamine. The solution is then allowed to flow slowly and with stirring into 10 times the volume of ethyl acetate, which is provided with seed crystals. The 7- (p-aminopyridinium) acetylamino-cephalosporanic acid separates out in crystallized form.
