Verfahren zur Herstellung neuer Derivate der 7-Aminocephalosporansäure
Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer therapeutisch wirksamer Derivate der 7-Aminocephalosporansäure (ACA) der Formel I

worin Rt einen 5-Aminotetrazolylrest bedeutet und R2 Wasserstoff oder eine freie oder durch eine Carbonsäure oder Thiocarbonsäure veresterte Hydroxylgruppe, eine N-substituierte Carbamoyloxygruppe, in der Sauerstoffatome durch Schwefel ersetzt sein können, oder eine quaternäre Aminogruppe ist, und ihrer Salze.
Eine veresterte Hydroxylgruppe R2, in der Sauerstoffatome durch Schwefel ersetzt sein können, leitet sich von einer Carbonsäure ab und ist beispielsweise eine gegebenenfalls z. B. durch Halogenatome, besonders Chlor, substituierte Niederalkanoyloxygruppe wie Formyloxy, Propionyloxy, Butyryloxy, Pivaloyloxy, Chloracetoxy, insbesondere Acetoxy, oder eine gegebenenfalls z. B. durch Niederalkyl-, Niederalkoxy-, oder Niederalkylmercaptoreste, Halogenatome oder die Nitrogruppe substituierte mono- oder dicyclische Arylcarbonyloxy- oder -thiocarbonyloxy-, Arylcarbonylmercapto- oder -thiocarbonylmercaptogruppe, insbesondere die Benzoylmercaptogruppe.
R2 kann auch eine substituierte Carbamoyloxygruppe, z. B. eine Gruppe der Formel -O-CO-NH-Rs sein, worin R ein aliphatischer, aromatischer, araliphatischer oder heterocyclischer Rest, besonders ein unsubstituierter oder substituierter, vorzugsweise durch eine oder mehrere Niederalkoxygruppen oder Halogenatome substituierter, gerader oder verzweigter Niederalkylrest, wie der Methyl-, Äthyl-, vor allem aber der p-Chloräthylrest, ist.

worin R5 für Wasserstoff oder eine oder mehrere Niederalkyl-, Niederalkoxycarbonyl-, Carbamoyl- oder Carboxylgruppen oder ein oder mehrere Halogenatome steht.
Die Salze der neuen Verbindungen sind Metallsalze, vor allem solche von therapeutisch anwendbaren Alkali- oder Erdalkalimetallen, wie Natrium, Kalium, Ammonium, Calcium, oder Salze mit organischen Basen, z. B. Triäthylamin, N-Athylpiperidin, Dibenzylamin, N-Benzyl-ss-phenetylamin, N,N'-Dibenzyläthylendiamin, Procain, Ephenamin. Ist R2 basisch, so können sich innere Salze bilden.
Die neuen Verbindungen weisen eine besonders gute antibakterielle Wirkung auf. Sie sind sowohl gegenüber gramposiüven wie vor allem auch gegenüber gram-negativen Bakterien wirksam, z. B. gegen Staphylococcus aureus penicillin-resistent, Escherichia coli, Klebsiella pneumoniae, Salmonella typhosa und Bacterium proteus, wie sich auch im Tierversuch, z. B. an Mäusen, zeigt. An diesen sind bei subcutaner Anwendung, je nach Art der bakteriellen Infektion, lOlOOmg/kg chemotherapeutisch wirksam. Die Verbindungen können daher zur Bekämpfung von Infektionen, die durch solche Mikroorganismen hervorgerufen werden, verwendet werden, ferner als Futtermittelzusätze, zur Konservierung von Nahrungsmitteln oder als Desinfektionsmittel.
Besonders wertvoll sind Verbindungen, in denen R2 die Acetoxygruppe, die -Chlor äthylcarbamoyl- oder eine unsubstituierte oder, wie oben angegeben, substituierte Pyridinogruppe ist.
Die Verbindungen der vorliegenden Erfindung können nach an sich bekannten Methoden hergestellt werden. So werden sie erhalten, wenn man eine Verbindung der Formel II

worin Z für einen Halogenacetylrest wie den Fluor-, Chlor-, Jod- oder vor allem Bromacetylrest steht und R2 Wasserstoff oder eine freie oder durch eine Carbonsäure oder Thiocarbonsäure veresterte Hydroxylgruppe ist mit 5-Aminotetrazol umsetzt. Wenn erwünscht, können erhaltene Verbindungen, in denen R2 für die Acetoxygruppe steht, in Verbindungen mit freier Hydroxylgruppe R2 und Verbindungen, mit freier Hydroxylgruppe R2 in Verbindungen, welche eine andere Estergruppe als die Acetoxygruppe enthalten, übergeführt werden.
Man kann auch erhaltene Verbindungen, in denen R2 für eine durch eine Carbonsäure oder Thiocarbonsäure veresterte Hydroxylgruppe steht, in Verbindungen, worin R2 eine quaternäre Aminogruppe bedeutet, umwandeln. Wenn erwünscht, können die erhaltenen Verbindungen in ihre therapeutisch verwendbaren Metall-, wie Alkali- oder Erdalkalimetallsalze, oder Salze mit Ammoniak oder organischen Basen übergeführt oder aus erhaltenen Salzen die freien Carbonsäuren oder gegebenenfalls inneren Salze gebildet werden.
Die Umsetzung der Verbindung II, worin Z für eine Halogenacetylgruppe steht, mit 5-Aminotetrazol findet bei Zimmertemperatur oder bei leicht erhöhter oder erniedrigter Temperatur, vorzugsweise bei 20-40 C statt. Sie wird vorzugsweise in einem inerten organischen Lösungsmittel wie Methylenchlorid, Chloroform, Tetrachlorkohlenstoff, Tetrahydrofuran, Dioxan, Dimethylformamid oder Acetonitril in Gegenwart eines halogenwasserstoffbindenden Mittels, z. B. einer schwachen anorganischen Base wie eines Alkalicarbo nahes -bicarbonates oder -acetates oder eines tertiären Amins, besonders eines Triniederalkylamins, vorzugsweise Diisopropyläthylamin (Hünigbase) vorgenommen.
Die als Ausgangsstoffe verwendeten Cephalosporinderivate sind bekannt oder können nach an sich bekannten Verfahren hergestellt werden.
Die tjberführung der Verbindung der Formel I, worin R für die Acetoxygruppe steht, in eine Verbindung mit freier Hydroxylgruppe und deren Veresterung mit anderen Säuren als Essigsäure oder deren tZberfüh- rung in Carbamoylderivate oder Verbindungen, worin R eine quaternäre Aminogruppe ist, erfolgt in an sich bekannter Weise.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, nach denen man von einer auf irgendeiner Stufe des Verfahrens als Zwischenprodukt erhältlichen Verbindung ausgeht und die fehlenden Verfahrensschritte durchführt, oder das Verfahren auf irgendeiner Stufe abbricht, oder bei denen man die Ausgangsstoffe unter den Reaktionsbedingungen bildet, oder bei denen die Reaktionskomponenten gegebenenfalls in Form ihrer Salze vorliegen.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwen dung finden. Diese enthalten die Verbindungen in Mischung mit einem für die enterale, topicale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssgen Trägermaterial.
Die Erfindung wird in den folgenden Beispielen beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
In der Dünnschichtchromatographie auf Silicagelplatten werden die folgenden Systeme verwendet:
System 52 = n-Butanol-Eisessig-Wasser (75:7,5:21)
System 101 A = n-Butanol-Pyrildin-Eisess,ig-Wasser (42:24:4:30)
Beispiel 1
7,86 g Bromacetylaminocephalosporansäure werden unter Zugabe von 6,9 ml N,N-Diisopropyläthylamin in 40 ml Methylenchlorid gelöst. Dazu gibt man eine Lösung, die durch Auflösen von 2,46 g 5-Aminotetrazol-Monohydrat in 20 ml Dimethylformamid und anschliessendes Verdünnen mit 30 ml Methylenchlorid hergestellt worden ist. Mit 10ml Methylenchlorid wird nachgespült.
Nach 45 Stunden wird das Methylenchlorid im Wasserstrahlvakuum und das Dimethylformamid bei 0,3 mm Vakuum und einer Temperatur von 300 möglichst vollständig abdestilliert. Das zurückbleibende dicke Öl wird in 100 ml Wasser gelöst, mit 200 ml Essigester überschichter und das pH der wässrigen Phase unter wiederholtem Schütteln mittels 2n-Natriumcarbonatlösung auf 5,4 eingestellt. Nach der Abtrennung wird die wässrige Phase ein zweites Mal mit 200 ml Essigester extrahiert. Die organischen Phasen werden zweimal mit je 20 ml Puffer von pH 6 gewaschen und dann verworfen. Die vereinigten wässerigen Lösungen überschichtet man mit 800 ml Essigester und stellt das pH unter Schütteln mittels 2n-Salzsäure auf 1,8 ein.
Nach der Phasentrennung wird die wässerige Phase mit 400 ml Essigester überschichtet, mit Koch- salz gesättigt und später nochmals mit 200 ml Essigester nachextrahiert. Die Essigesterlösungen werden mit 40 ml gesättigter Kochsalzlösung gewaschen, mit Natriumsulfat getrocknet und im Vakuum zur Trokkene eingedampft. 6,3 g dieses Rohproduktes werden in 36 ml Methanol suspendiert und durch Zugabe von 7,7 ml 3m.-methanolischer Natrium-a-äthylhexanoatlö sung gelöst. Unter gutem Rühren werden 4,5 ml abs.
Äthanol eingetropft. Das unreine Präcipitat wird abfil triert und verworfen. Das Filtrat wird in gleicher Weise mit 300 ml abs. Äffianol versetzt, abgekühlt und 30
Minuten bei -i00 stehen gelassen. Man nutscht ab, wäscht mit 40 ml Äthanol und erhält das Natriumsalz der 7-[5-Aminotetrazolyl-(5)-acetylamino]-cephalospo- ransäure. Die dazu gehörige Säureform lässt sich durch Rückextraktioil in Essigester bei pH 2 gewinnen.

Natriumsalz) U.-V.-Spektrum in Wasser als Natriumsalz: Amax: 227 m, (± = 8350) und 260 mm (e = 8200); NMR-Spektrum in Deutero-Dimethylsulfoxyd (100 Mc); ausser den für die 7-ACA charakteristischen Signalen tritt ein Singlett bei a = 4,92 ppm auf, welches der -CH2-Gruppe des Aminotetrazolylacetylrestes zugeordnet werden kann, und ein Singlett mit breiter Basis bei a = 6,7 ppm für die am Kohlenstoff des Tetrazolringes stehende NH2-Gruppe.
Beispiel 2
9,42 g 3 -(Des acetoxymethyl)-3-benzoylthio- methyl-7-bromacetylamino-cephalosporansäure werden unter Zugabe von 7,0 ml N,N-Diisopropyl äthylamin in 60 ml Dimethylformamid gelöst. Dazu gibt man eine Lösung von 2,46 g 5-Aminotetrazol Monohydrat in 20 ml Dimethylformamid. Mit 13 mol Dimethylformamid wird nachgespült.
Nach 36-stündigem Stehen bei Raumtemperatur wird das Lösungsmittel bei 0,3 mm Vakuum gegen einen Kühler von -600 abdestilliert. Das zurückbleibende Öl wird in ein stark gerührtes Zweiphasensystem aus 500 ml Essigester und 250 ml Phosphatpuffer pH 6,5 eingetragen. Durch Zugabe von wenig 2n-Natriumcarbonat-Lösung wird die wässerige Phase anschliessend auf pH 6.5 eingestellt. Man trennt sofort die Phasen und extrahiert die wässerige Phase nochmals mit 20 ml Essigester. Die organischen Phasen werden nacheinander mit 50 ml Phosphatpuffer vom pH 6,5 gewaschen und verworfen.
Nun werden die vereinigten wässerigen Lösungen unverzüglich mit 500 ml Essigester überschichtet und das pH wird unter starkem Rühren durch Zugabe von 2n-Salzsäure auf 2 eingestellt. Ein dabei gebildetes Präcipitat lässt sich durch Filtration entfernen. Nach der Phasentrennung wird die wässerige Lösung mit Kochsalz gesättigt und mit 300 ml und 200 ml Essigester nachextrahiert. Die organischen Phasen werden nacheinander zweimal mit je 50ml gesättigter Kochsalzlösung gewaschen, mit Natriumsulfat getrocknet und durch eine Säule (Durchmesser 4,5 cm) von 60 g Silicagel filtriert. Das gesammelte Filtrat dampft man im Vakuum ein. Der Rückstand wird in 30 ml Tetra hydrofuran aufgelöst und mit soviel Methanol versetzt (ca. 100 ml), dass eine leichte Färbung entsteht.
Durch leichtes Erwärmen im Luftbad und Abblasen im Stickstoffstrom wird die Lösung dann langsam konzentriert, wobei man die kristalline 3-(Desacetoxymethyl)3-benzoylthiomethyl-7-[5-aminotetrazolyl-(5) acetplaminol-cephalosporansäure erhält. F. 190-1930 (Zers.).

U-V.Spektrum (in O,1 mol Nakiumbicarbonat): Ämax bei 240 m,u (e = 16500) und bei 276 m,u (e = 19900)
Das Ausgangsmaterial kann wie folgt hergestellt werden: Eine Lösung von 17,5 g 3-(Desacetoxymethyl) -3 -benzoylthiomethyl)- 7-amino-cephalosporansäure (vgl. belg. Patent 650 444) und 12,5 ml Triäthylamin in 1 1 Dimethylformamid wird während einer Stunde in eine gut gerührte und bei -13" bis -15" gehaltene Lösung von 9,2 ml Bromacetylbromid in 100 ml Methylenchlorid eingetropft (Stickstoffatmosphäre).
Man lässt die Temperatur während 1 1/2 Stunden langsam auf 100 steigen und hält sie dort noch während einer halben Stunde. Dann wird der grösste Teil der Lösungsmittel im Vakuum bei 0,5-1 mm Hg gegen einen Kühler von Trockeneis-Aceton abdestilliert. Das ölige Produkt wird auf einen Phosphatpuffer von pH 6 gegossen und mit 1 1 Essigester geschüttelt. An der Grenze der beiden Phasen entsteht ein Präcipitat, das durch Filtration oder Zentrifugieren abgetrennt wird.
Dann stellt man das pH der wässrigen Phase auf 2 ein, sättigt diese mit Kochsalz und trennt die organische Phase ab. Die wässerige Phase wird mit 600 und 400 ml Essigester nachextrahiert. Nach dem Waschen mit gesättigter Kochsalzlösung werden die organischen Phasen über Natriumsulfat getrocknet und nacheinander durch eine Säule von 100 mg Silicagel filtriert. Die Filtrate werden im Vakuum zur Tockne eingeengt, der Rückstand mit 30 ml Äthanol versetzt und bei -20" auskristallisiert. Man erhält 7,8 g 3-(Desacetoxymethyl)-3-benzoylthiomethyl- 7-bromacetyl-amino-cephalosporansäure vom Schmelzpunkt 137-138 .
Rf52 = 0,55. Das Natriumsalz zeigt im U-V.
Spektrum in Wasser: Amax 243 my (± = 16800) und
275 m,u (e = 20600).
[a]D20 = -47 + 10 (c = 1; in 0,1 mol. Natrium bicarbonat-Aceton (1:1).
Beispiel 3
2,51 g 3-(Desacetoxymethyl)-3-benzoylthio methyl-7- [5-aminotetrazolyl-(5)- acetylamino] -cephalosporansäure werden in 415 ml eines Gemisches aus gleichen Teilen Dioxan und Pyridin gelöst, mit 12,2 ml 40 0/obiger wässeriger Quecksilberperchloratlösung versetzt und während 45 Minuten bei 45" und 45 KHz Ultraschall unter Stickstoff reagieren gelassen. Man kühlt ab, versetzt mit 6,5 ml Thiobenzoesäure und schüttelt 5 Minuten. Die Lösungsmittel werden im Vakuum abdestilliert und eine Lösung des Rückstandes in 65 ml Wasser wird durch Celite abfiltriert.
Das Filtrat wird nacheinander mit 50 ml Toluol, zweimal mit je 33 ml Amberlite LA-2 in 66 ml Toluol und zweimal mit je 50 ml Toluol gewaschen Anschliessend filtriert man die wässerige Phase durch eine Säule, die von unten nach oben 4,1 ml Sephadex CM C-25 (H+-form), 16,5 ml Alox , 4,1 ml Zeo-Karb 226 (H+-form), 16,5 ml Alox , 4,1 ml Dowex 1 (Acetatform) und 4,1 ml Sephadex CM C-25 (H+-form) enthält.
Celite , organische Phasen und die Säule werden zweimal mit je 15 ml Wasser nachextrahiert, die Säule zudem mit weiteren 150 ml Wasser eluiert. Man engt die vereinigten Eluate im Vakuum ein, entfernt eine kleine Menge von Präcipitat durch Filtration und dampft zur Trockne ein. Der Rückstand wird mit ca. 50 ml abs.
Äthanol digeriert und ergibt die reine 3-(Desacetoxymethyl)3-pyridiniomethyl-7-[5-Aminotetrazolyl-(5) acetylaminoj-cephalosporansäure.

NMR-Spektrum in Deutero-Dimethylsulfoxyd (100 Mc): Ausser den für den Grundkörper charakteristischen Signalen tritt ein Singlett bei a = 4,96 ppm auf, welches der -CH2-Gruppe der Aminotetrazolylacetylrestes zugeordnet werden kann, und ein Singlett mit breiter Basis bei d = 6,76 ppm für die am Kohlenstoff des Tetrazolringes stehende -NH2-Gruppe.

UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man von einer Verbindung der Formel II ausgeht, worin Z für den Bromacetylrest steht.
2. Verfahren nach Patentanspruch I oder Unteranspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel I herstellt, worin R1 die angegebene Bedeutung hat und R2 für die Acetoxygruppe steht.
3. Verfahren nach Patentanspruch I oder Unteranspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel I herstellt, worin R1 die angegebene Bedeutung hat und R2 für eine gegebenenfalls substituierte Pyridiniogruppe steht.
4. Verfahren nach Patentanspruch I oder Unteranspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel I herstellt, worin Rt die angegebene Bedeutung hat und R2 für die Pyridiniogruppe steht.
5. Verfahren nach Patentanspruch I oder Unteranspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel I herstellt, worin Rt die angegebene Bedeutung hat und R2 für eine Niederalkylcarbamoyloxygruppe steht.
6. Verfahren nach Patentanspruch I oder Unteranspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel I herstellt, worin Rl die angegebene Bedeutung hat und R2 eine Carbamoyloxygruppe der Formel O-C O-NH-R3 ist, worin R3 ein durch ein oder mehrere Niederalkoxygruppen oder Halogenatome substituierter Niederalkylrest ist.
7. Verfahren nach Patentanspruch I oder Unteranspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel I herstellt, worin R1 die angegebene Bedeutung hat und R2 eine Carbamoyloxygruppe der Formel -O-CO-NH-R3 ist, worin R5 ein durch ein oder mehrere Chloratome substituierter Niederalkylrest ist.
8. Verfahren nach Patentanspruch I oder Unteranspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel I herstellt, worin Rt die angegebene Bedeutung hat und R2 die ss-Chloräthylcarbamoyloxygruppe ist.
9. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der
Formel I, worin R1 die angegebene Bedeutung hat und
R2 für die Acetoxygruppe steht, mit desacetylierenden
Mitteln behandelt.

They are effective against both gram-positive bacteria and especially against gram-negative bacteria, e.g. B. against Staphylococcus aureus penicillin-resistant, Escherichia coli, Klebsiella pneumoniae, Salmonella typhosa and Bacterium proteus, as has also been found in animal experiments, e.g. B. on mice shows. With subcutaneous application, depending on the type of bacterial infection, 100 mg / kg are chemotherapeutically effective on these. The compounds can therefore be used to combat infections caused by such microorganisms, and also as feed additives, for the preservation of food or as disinfectants.
Particularly valuable are compounds in which R2 is the acetoxy group, the -chloroethylcarbamoyl- or an unsubstituted or, as indicated above, substituted pyridino group.
The compounds of the present invention can be prepared by methods known per se. This is how they are obtained when a compound of the formula II

where Z is a haloacetyl radical such as fluorine, chlorine, iodine or, above all, bromoacetyl radical and R2 is hydrogen or a free hydroxyl group or a hydroxyl group esterified by a carboxylic acid or thiocarboxylic acid and reacts with 5-aminotetrazole. If desired, compounds obtained in which R2 stands for the acetoxy group can be converted into compounds having a free hydroxyl group R2 and compounds having a free hydroxyl group R2 into compounds which contain an ester group other than the acetoxy group.
Compounds obtained in which R2 is a hydroxyl group esterified by a carboxylic acid or thiocarboxylic acid can also be converted into compounds in which R2 is a quaternary amino group. If desired, the compounds obtained can be converted into their therapeutically useful metal salts, such as alkali or alkaline earth metal salts, or salts with ammonia or organic bases, or the free carboxylic acids or, if appropriate, internal salts can be formed from the salts obtained.
The reaction of the compound II, in which Z is a haloacetyl group, with 5-aminotetrazole takes place at room temperature or at a slightly elevated or reduced temperature, preferably at 20-40.degree. It is preferably in an inert organic solvent such as methylene chloride, chloroform, carbon tetrachloride, tetrahydrofuran, dioxane, dimethylformamide or acetonitrile in the presence of a hydrogen halide binding agent, e.g. B. a weak inorganic base such as an alkali carbonate close -bicarbonates or acetate or a tertiary amine, especially a tri-lower alkylamine, preferably diisopropylethylamine (Hünig base).
The cephalosporin derivatives used as starting materials are known or can be prepared by processes known per se.
The conversion of the compound of the formula I in which R represents the acetoxy group into a compound with a free hydroxyl group and its esterification with acids other than acetic acid or its conversion into carbamoyl derivatives or compounds in which R is a quaternary amino group takes place in itself known way.
The invention also relates to those embodiments of the process in which one starts from a compound obtainable as an intermediate product at any stage of the process and carries out the missing process steps, or the process is terminated at any stage, or in which the starting materials are formed under the reaction conditions, or in which the reaction components are optionally in the form of their salts.
The new compounds can be used as remedies, e.g. B. in the form of pharmaceutical preparations, use find. These contain the compounds mixed with a pharmaceutical organic or inorganic, solid or liquid carrier material suitable for enteral, topical or parenteral administration.
The invention is described in the following examples. The temperatures are given in degrees Celsius.
The following systems are used in thin layer chromatography on silica gel plates:
System 52 = n-butanol-glacial acetic acid-water (75: 7.5: 21)
System 101 A = n-butanol-pyrildine-glacial, ig-water (42: 24: 4: 30)
example 1
7.86 g of bromoacetylaminocephalosporanic acid are dissolved in 40 ml of methylene chloride with the addition of 6.9 ml of N, N-diisopropylethylamine. A solution is added which has been prepared by dissolving 2.46 g of 5-aminotetrazole monohydrate in 20 ml of dimethylformamide and then diluting with 30 ml of methylene chloride. Rinse with 10ml methylene chloride.
After 45 hours, the methylene chloride is distilled off as completely as possible in a water jet vacuum and the dimethylformamide is distilled off at 0.3 mm vacuum and a temperature of 300. The thick oil that remains is dissolved in 100 ml of water, covered with a layer of 200 ml of ethyl acetate and the pH of the aqueous phase is adjusted to 5.4 with repeated shaking using 2N sodium carbonate solution. After the separation, the aqueous phase is extracted a second time with 200 ml of ethyl acetate. The organic phases are washed twice with 20 ml of pH 6 buffer each time and then discarded. The combined aqueous solutions are covered with a layer of 800 ml of ethyl acetate and the pH is adjusted to 1.8 with 2N hydrochloric acid while shaking.
After the phases have separated, the aqueous phase is covered with a layer of 400 ml of ethyl acetate, saturated with sodium chloride and later extracted again with 200 ml of ethyl acetate. The ethyl acetate solutions are washed with 40 ml of saturated sodium chloride solution, dried with sodium sulfate and evaporated to dryness in vacuo. 6.3 g of this crude product are suspended in 36 ml of methanol and dissolved by adding 7.7 ml of 3m-methanolic sodium a-ethylhexanoate solution. With thorough stirring, 4.5 ml of abs.
Dripped in ethanol. The impure Precipitat is filtered off and discarded. The filtrate is in the same way with 300 ml of abs. Affianol added, cooled and 30

9.42 g of 3 - (De acetoxymethyl) -3-benzoylthio-methyl-7-bromoacetylamino-cephalosporanic acid are dissolved in 60 ml of dimethylformamide with the addition of 7.0 ml of N, N-diisopropylamine. A solution of 2.46 g of 5-aminotetrazole monohydrate in 20 ml of dimethylformamide is added. Rinsing is carried out with 13 mol of dimethylformamide.
After standing for 36 hours at room temperature, the solvent is distilled off under 0.3 mm vacuum against a condenser of -600. The remaining oil is introduced into a vigorously stirred two-phase system of 500 ml of ethyl acetate and 250 ml of phosphate buffer pH 6.5. The aqueous phase is then adjusted to pH 6.5 by adding a little 2N sodium carbonate solution. The phases are separated immediately and the aqueous phase is extracted again with 20 ml of ethyl acetate. The organic phases are washed successively with 50 ml of phosphate buffer pH 6.5 and discarded.
The combined aqueous solutions are then immediately covered with a layer of 500 ml of ethyl acetate and the pH is adjusted to 2 by adding 2N hydrochloric acid while stirring vigorously. A precipitate formed in the process can be removed by filtration. After the phases have separated, the aqueous solution is saturated with sodium chloride and re-extracted with 300 ml and 200 ml of ethyl acetate. The organic phases are washed twice in succession with 50 ml of saturated sodium chloride solution each time, dried with sodium sulfate and filtered through a column (diameter 4.5 cm) of 60 g of silica gel. The collected filtrate is evaporated in vacuo. The residue is dissolved in 30 ml of tetrahydrofuran and mixed with enough methanol (approx. 100 ml) that a light color develops.
The solution is then slowly concentrated by gentle warming in an air bath and blowing off in a stream of nitrogen, giving the crystalline 3- (deacetoxymethyl) 3-benzoylthiomethyl-7- [5-aminotetrazolyl- (5) acetaminol-cephalosporanic acid. F. 190-1930 (decomp.).
Rf 52 = 0.40, Rf ,,, A = 0.55.

The starting material can be prepared as follows: A solution of 17.5 g of 3- (desacetoxymethyl) -3-benzoylthiomethyl) -7-aminocephalosporanic acid (cf. Belgian Patent 650 444) and 12.5 ml of triethylamine in 1 l of dimethylformamide is added dropwise to a well-stirred solution of 9.2 ml of bromoacetyl bromide in 100 ml of methylene chloride kept at -13 "to -15" (nitrogen atmosphere) over one hour.
The temperature is allowed to rise slowly to 100 over 1 1/2 hours and is held there for a further half an hour. Most of the solvents are then distilled off from dry ice-acetone in vacuo at 0.5-1 mm Hg against a condenser. The oily product is poured onto a phosphate buffer of pH 6 and shaken with 1 liter of ethyl acetate. A precipitate is formed at the boundary between the two phases and is separated off by filtration or centrifugation.
The pH of the aqueous phase is then adjusted to 2, this is saturated with sodium chloride and the organic phase is separated off. The aqueous phase is extracted with 600 and 400 ml of ethyl acetate. After washing with saturated sodium chloride solution, the organic phases are dried over sodium sulfate and successively filtered through a column of 100 mg of silica gel. The filtrates are concentrated to dryness in vacuo, the residue is treated with 30 ml of ethanol and crystallized at -20 ". 7.8 g of 3- (deacetoxymethyl) -3-benzoylthiomethyl-7-bromoacetylamino-cephalosporanic acid with a melting point of 137 are obtained. 138.
Rf52 = 0.55. The sodium salt shows in the U-V.

Example 3
2.51 g of 3- (Desacetoxymethyl) -3-benzoylthio methyl-7- [5-aminotetrazolyl- (5) - acetylamino] -cephalosporanic acid are dissolved in 415 ml of a mixture of equal parts of dioxane and pyridine, with 12.2 ml of 40 0 / above aqueous mercury perchlorate solution are added and the mixture is allowed to react for 45 minutes at 45 "and 45 KHz ultrasound under nitrogen. It is cooled, mixed with 6.5 ml of thiobenzoic acid and shaken for 5 minutes. The solvents are distilled off in vacuo and a solution of the residue in 65 ml of water is filtered off through Celite.
The filtrate is washed successively with 50 ml of toluene, twice with 33 ml each time of Amberlite LA-2 in 66 ml of toluene and twice with 50 ml each time of toluene. The aqueous phase is then filtered through a column containing 4.1 ml of Sephadex from bottom to top CM C-25 (H + form), 16.5 ml Alox, 4.1 ml Zeo-Karb 226 (H + form), 16.5 ml Alox, 4.1 ml Dowex 1 (acetate form) and 4.1 ml Contains Sephadex CM C-25 (H + form).
Celite, organic phases and the column are re-extracted twice with 15 ml of water each time, and the column is also eluted with a further 150 ml of water. The combined eluates are concentrated in vacuo, a small amount of precipitate is removed by filtration and evaporated to dryness. The residue is with about 50 ml of abs.
Ethanol digests and gives the pure 3- (deacetoxymethyl) 3-pyridiniomethyl-7- [5-aminotetrazolyl- (5) acetylaminoj-cephalosporanic acid.
