Die Erfindung betrifft ein Verfahren zur Herstellung von gegebenenfalls weiter substituierten 9-Aminoalkoxy-9,10-dihydro - 9,10 - äthano anthrazenen mit dem Kern der Formel

wobei das Stickstoffatom mit dem Alkylenrest zu einem Ring verbunden sein kann und in der Aminealkoxy- gruppe das Sauerstoffatom vom Stickstoffatom durch mindestens zwei Kohlenstoffatome getrennt ist, und ihren Salzen.
In den neuen Verbindungen kann die Aminogruppe des Aminoalkylrestes unsubstituiert sein. Vorzugsweise ist sie jedoch mono- oder disubstituiert. Dabei kommen als Substituenten vor allem niedere aliphatische, cycloaliphatische, araliphatische oder aromatische Kohlen wasserstoffreste in Frage, wobei die taliphatischen bzw.
cycloaliphatischen Reste auch durch Heteroatome, wie Sauerstoff, Schwefel oder Stickstoff, unterbrochen und/ oder mit dem Alkylenrest verbunden sein können und wobei aliphatische Reste durch freie Hydroxy-, Aminooder Mercaptogruppen oder Halogenatome, wie Fluor, Chlor, Brom oder Jod, und aromatische Gruppen durch niedere Alkyl-, Alkoxy- oder Alkenyloxygruppen, Halogenatome, wie Fluor, Chlor, Brom, Jod oder das Pseudohalogen Trifluormethyl, wie z. B. die weiter unten genannten, substituiert sein können.
Als niedere Kohlenwasserstoffreste sind vor allem zu nennen: niedere AlkylL oder Alkenylreste, wie Methyl-, Athyl-, Propyl-, Isopropyl-, gerade oder verzweigte, in beliebiger Stelle verbundene Butyl-, Pentyl-, Hexyl- oder Heptylreste, Allyl- oder Methallylreste, unsubstituierte oder alkyl-substituierte Cycloalkyl- oder Cycloalkenylreste, wie Cyclopentyl-, Cyclohexyl-, Cyclheptyl-, Cyclopentenyl-, Cyclohexenylreste, unsubstituierte oder alkylsubstituierte Cycloalkyl- oder -alkenylalkylreste, wie Cyclopentyl- oder Cyclohexenylmethyl-, -äthyl- oder -propylreste, Aralkyl- oder Araikenyl-, wie Phenylmethyl-, -äthyl-, vinyl- oder -propyireste, oder Aryl-,
insbesondere Phenylreste, oder Alkylen- oder Alkenylenreste, wie z. B. Butylen-(1,4), Pentylen-(1,5), 1,5-Dimethyl-pentylen-(1,5), Hexylen-(1,6), Hexylen-(1,5).
Durch Heteroatome unterbrochene Reste dieser Art sind z. B. Alkoxyalkyl- oder Oxa-cycloalkylallkylreste, wie Methoxyäthyl, Äthoxyäthyl, Propoxyäthyl, Butoxy äthyl, Methoxypropyl, Methoxyäthoxyäthyl, Tetrahydrofurylmethyl, Methylmercaptoäthyl, Oxa-, Aza- oder Thiaalkylen- oder -alkenylenreste, wie 3-Aza-, Oxaoder Thiapentylen (1,5), 3 -Azahexylen-( 1,6), 1,5-Di- methyl-3-aza-pentylen-(1,5), 3-Methyl-3-aza-pentylen (1,5) oder 3-Hydroxyäthyl-3-aza-pentylen-(1,5).
Die Aminogruppe ist vor allem eine Diniederalkylaminogruppe, wie die Dimethylamino-, Diäthylamino-, Dipropylamino-, N-Methyl-N-äthylaminogruppe, eine N Niedelralkyll N-cycloalkylaminogruppe, wie die N-Me- thyl-N-cyclopentyl- oder -cyolohexylgruppe, eine N-Dicycloalkylaminogruppe, eine gegebenenfalls C-nieder alkylierte Pyrrolidino-, Piperidine, Morpholino-, Thiomorpholino-, Piperazino- oder N-Niederalkyl oder Hydroxy-niederalkyl-piperazinogruppe, wie die P yrroll- dino-, Piperidino-, Morpholino-, Piperazino-, N-Methyl-N-Äthyl- oder N-ss-Hydroxyäthyl-piperazinogruppe.
Die Aminogruppe kann aber auch eine Mono-nieder- alkylaminogruppe, wie die Methylamino-, Athylamino-, Propylamino- oder N-Cycloal'kylaminogruppe sein.
Aminoalkoxyreste, bei denen ein Substituent der Aminogruppe mit dem Alkylenrest verbunden ist, sind beispielsweise N-Alkyl-pyrrolidinyl-3-alkoxyreste oder N-Alkyl-piperidyl-3-oder-4-alkoxyreste.
Der die endständige Aminogruppe mit dem am Kern sitzenden Sauerstoffatom verbindende Alkylenrest ist vor allem ein niederer, gerader oder verzweigter Alkylenrest, vorzugsweise mit 2-4 Kohlenstoffatomen, wobei die endständige Aminogruppe vom am Kern sitzenden Sauerstoffatom durch mindestens zwei Kohlenstoffatome getrennt ist, wie z.B. der Propylen-(1,2)-, Propylen-(1,3)-, Butylen-(1,2)-, Butylen-(1,3)-, Butylen-(2,3)- oder Butylen-(1,4)-rest, oder ganz besonders ein solcher mit zwei Kohlenstoffatomen, wie ein Äthylen-(1,2)-rest. Die Alkylengruppe kann auch mit einem Substituenten der Aminogruppe verbunden sein.
Die neuen Verbindungen können noch weiter substituiert sein. So können sie z. B. in den Stellungen 11 und 12 durch niedere Alkylreste, wie z. B. die genannten, substituiert sein.
Ferner können sie z. B. in den Stellungen 1-8 des Anthrazenringes niedere Alkyl-, Alkoxy- oder Alkenyloxy- oder Alkylmercaptogruppen, Halogenatome, wie Fluor, Chlor, Brom oder Jod, oder das Pseudohalogen Trifluormethyl, Alkylsulfonyl-, Alkanoyl-, Nitro- oder Aminogruppen tragen, wobei z. B. als Alkylreste der Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, i-Butyi-, oder tert. Butylreste, als Alkoxy- oder Alkenyloxygrup- pen, die Methoxy-, Äthoxy-, Allyloxy- oder Methylendioxygruppe, als Alkylmereaptogruppen die MethylL oder Athylmercaptogruppe, und als Alkanoylreste vor allem der Acetyl-, Propionyl- oder Butyrylrest zu nennen smd.
In 10-Stellung können die neuen Verbindungen vor allem einen aliphatischen Kohlenwasserstoffrest, wie einen der obengenannten niederen Alkyl- oder Alkenyl; reste, oder auch ein Halogenatom aufweisen.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So weisen sie neben einer antiallergischen Wirkung insbesondere eine Antihist- amin-Wirkung auf, wie sich im Tierversuch, z. B. am Meerschweinchendarm, und beim Histaminschock, zeigt. Ferner besitzen sie, z. B. im Tierversuch, wie z. B. an Mäusen, eine Anti-Mescalin-Wirkung. Die neuen Verbindungen können daher als Antihistaminika und Antiallergica und psychotrope Pharmaka Verwendung finden. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere von pharmakologisch wirksamen Verbindungen.
Besonders wertvoll sind 9-Aminoalkoxy-9,10-di- hydro-9,10-äthano-anthrazene der Formel

worin A eine Alkylengruppe mit 24 Kohlenstoffatomen bedeutet, die das Sauerstoffatom vom Rest R durch 2-3 Kohlenstoffatome trennt, vor allem den Äthylen-(1,2)- oder den Propylen-(1,3)-rest, R ein Pyrroli- dino-, Piperidino-, Morpholino- oder Piperazinorest, z.
B. ein N-Niederalkylpiperazinorest oder ein N-(2 Hydroxyäthyl)-piperazinorest, vor allem aber eine Diniederalkylamino- oder Mono-niederalkylaminogruppe, R1 ein Halogenatom, eine Trifluormethyl-, Niederalkyl-, Niederalkoxy- oder Nitrogruppe oder besonders ein Wasserstoffatom ist, und die Buchstaben n und m für die Zahlen 1 oder 2 stehen, wobei die Reste R1 gleichartig oder verschieden sein können.
Speziell zu erwähnen sind folgende Verbindungen: Das 9-(2-Methylamino-äthoxy)-9, 1 0-dihydro-9, 10- äthano-anthrazen, das 9-(2-Methylamino-propoxy)-9, 1 0dihydro-9, 10- äthano-anthrazen, das 9-(2-Dimethylamino-1-methyl-äthoxy)-9,10 dihydro-9,10-äthano-anthrazen, das 9-(2Diäthylamino-äthoxy)-9, 1 0-dihydro-9, 10- äthSano-anthrazen und msbeson dere das 9-(2-Dimethyl mino-äthoxy) -9,1 0-dihydro-9, 10- äthano-anthrazen, das beispielsweise in Form seines Hydrochlorids an Meerschweinchen bei peroraler Gabe in Dosen von 0,1 bis 1,0 mg/kg eine ausgesprochene Schutzwirkung gegenüber Tod durch i. v. Injektion von Histamin, aufweist.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man in einem entsprechenden 9-Z-9,10-dihydro9,1 0-äthano-anthrazen, worin Z einen durch Reduktion einer in Nachbarstellung zum Stickstoff stehenden Carbonylgruppe in eine Aminoalkoxygruppe überfühf- baren Rest bedeutet, den Rest Z zu einer Aminoalkoxygruppe reduziert.
Der Rest Z ist insbesondere ein Carbamylalkoxyrest, ein enlsprechenlder Acylaminoalkoxy- oder N Acylcarbamylalkoxyrest. Die Reduktion kann beispielsweise durch Einwirkung eines zur Amidreduktion geeigneten Hydridionen abgebenden Mittels, wie eines entsprechenden Dileichtmetallhydrides, speziell eines Alkalimetall-aluminium-hydrides, wie Lithium- oder Natriumaluminiumhydrid, erfolgen. Falls notwendig, können die Reduktionsmittel auch gemeinsam mit Aktivatoren, z. B. Aluminiumchlorid, angewendet werden.
Die Reduktion kann auch zum Beispiel elektrolytisch an Kathoden mit hoher Überspannung, wie Quecksil; ber-, Bleiamalgam- oder Bleikathoden, erfolgen.
In erhaltenen Verbindungen kann man im Rahmen der Definition der Endstoffe in üblicher Weise Substituenten abwandeln, einführen oder abspalten.
So kann man in erhaltene primäre oder sekundäre Aminogruppen in üblicher Weise noch weitere Sub stituenten einführen.
Beispielsweise kann man die Aminogruppe alkylie- ren, z. B. durch Umsetzung mit reaktionsfähigen Estern von Alkanolen. Als reaktionsfähige Ester kommen dabei insbesondere die Obengenannten in Betracht. Die Alkylierung kann auch reduktiv erfolgen, d. h. durch Umsetzung mit einer entsprechenden Carbonylverbindung und anschliessende oder Igleichzeitige Reduktion.
Ebenso lassen sich andere Substituenten im Molekül in üblicher Weise abwandein, z. B. Nitrogruppen an den aromatischen Resten zu Aminogruppen reduzieren.
Die Erfindung betrifft auch diejenigen AusfüXhrungs- formen des Verfahrens, bei denen man von einer auf irgendeiner Stufe als Zwischenprodukt erhältlichen Verbindung ausgeht und die fehlenden Schritte durchführt, oder das Verfahren auf irgendeiner Stufe abbricht oder bei denen ein Ausgangsstoff unter den Relaktionsbedin- gingen gebildet oder in Form eines Salzes verwendet wird.
Die genannten Reaktionen können in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensations- undloder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt werden.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Verfahren gewonnen werden.
Je nach den Verfahrensbedingungen und Ausgangs- stoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Salze, So können beispielsweise basische, neutrale, saure oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesqui- oder Polyhydrate davon erhalten werden. Die Salze der Endstoffe können in an sich bekannter Weise, z. B. mit Alkalien oder Ionenaustauschern in die freien Basen übergeführt werden. Von den letzteren lassen sich durch Umsetzung mit organischen oder anorganischen Säuren, insbesondere solchen, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen.
Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffs äuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure; aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Asoorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxy-benzoe-, Salicyl- oder p-Amino- salicylsäure, Embonsäure, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenben zolsulfon-, Toluolsulfon-, Naphthalinsulfons äuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können Rauch zur Reinigung der erhaltenen Blasen dienen, indem man die Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freisetzt. Infolge der engen Beziehung zwischen den Basen in freier Form und in Form ihrer Salze sind im vorausgegangenen und nachfolgend unter den freien Basen sinn- und zweckgemäss, gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die neuen Verbindungen können als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden, welche diese Verbindungen zusammen mit pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägerstoffen, die für enterale, z. B.
orale, oder parenterale Gabe geeignet sind, enthalten.
Die neuen Verbindungen können auch in der Tiermedizin, z. B. in einer der obengenannten Formen, oder bei der Aufzucht und Ernährung von Tieren in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden.
In den folgenden Beispielen sind die Temperaturen in Celsiusgradeu angegeben.
Beispiel 1
Zu einer Suspension von 5,0 g Lithiumaluminium- hydrid in 150 ml Tetrahydrofuran werden 15,0 g N-Carbäthoxy-9-(2-methylaminoäthoxy)-9,10 dihydro-9, 1 0-äthano-'anthrazen in 50 ml Tetrahydrofuran getropft. Anschliessend kocht man 4 Stunden unter Rühren. Nach dem Abkühlen lässt man nacheinander 5 ml Wasser, 5 ml 15 % ige wässerige Natronlauge und nochmals 15 ml Wasser zufliessen.
Die unlöslichen anorganischen Bestandteile werden abgenutscht und mit Tetrahydrofuran ausgewaschen. Das Filtrat wird eingedampft, der Rückstand in 200 ml 2n Salzsäure aufgenommen und zweimal mit je 100 ml Äther extrahiert. Die wässerig-salzsaure Phase wird mit 5n wässeriger Natronlauge alkalisch gemacht. Die ausfallende Base extrahiert man auf übliche Weise mit Methylenchlorid und erhält so nach dem Trocknen und Eindampfen des Lösungsinittels das rohe 9-(2-Dimethylaminoäthoxy)-9, 1 dihydro-9, 10 äthano-anthrazen der Formel

In analoger Weise wie im Beispiel 1 beschrieben, kann man ausgehend von den entsprechenden 9-Acyl amlinoalkoxy-Verbindungen oder den entsprechenden 9-Carbamylalkoxy-Verbindungen, d. h. z.
B. von entsprechen den Verbindungen, die an einem dem Stickstoffatom benachbarten Kohlenstoffatom eine Oxogruppe tragen, durch Reduktion mit Lithiumaluminiumhydrid die folgenden Verbindungen herstellen: a) 9-(2-Diäthylamino-äthoxy)-9,10-dihydro-9,10 äthano-anthrazen-hydrochlorid, F. nach Umkristal lisation aus Äthanol-Äther 152-153 ; b) 9-(2-Dimethylamino-1-methyl-äthoxy)-9,10 dihydro-9,10-äthano-anthrazen-hydrochlorid,
