Verfahren zur Herstellung von Azetidino [3 ,2-djthiazolidinverbindungen
Gegenstand der vorliegenden Erfindung ist ein Methodikverfahren zur Herstellung von Azetidino[3,2- d]thiazolidinverbindungen, das zur Darstellung von wertvollen Zwischenprodukten und insbesondere bei der erstmaligen synthetischen Herstellung der 7-Amino-cephalosporansäure und ihrer Derivate Anwendung fand und zu dieser eigenartigen Synthese besonders geeignet ist.

Derivate sind in erster Linie N-Acylverbindungen, worin Acylreste insbesondere diejenigen von wirksamen N-Acylderivaten der 7-Arnino-cephalosporan- säure, wie der Thienylacetyl-, z. B. 2-Thienylacetyl-, Cyanacetyl-, Chloräthylcarbamyl- oder Phenylacetylrest. oder leicht abspaltbare Acylreste, wie der Rest eines Halbesters der Kohlensäure, z. B. der tert.-Butyl oxycarbonylrest, bedeuten.
Die Synthese dieser für die Herstellung wertvoller Arzneimittel wichtigen Verbindung und ihrer Derivate beruht auf der Idee, von einer 3,5-unsubstituierten 2,2disubstituierten Thiazolidin-4-carbonsäure, z. B. einer Verbindung der Formel I

worin R1 eine freie oder veresterte Carboxylgruppe darstellt, mit einem zur ss-Lactambildung geeigneten Mittel behandelt. Wenn erwünscht, kann in einer erhaltenen Verbindung ein Substituent in einen anderen übergeführt werden.
Je nachdem man eine freie oder veresterte fi-Aii nocarbonsäure als Ausgangsstoff verwendet, wird die obige Reaktion unterschiedlich durchgeführt. So gelangt man durch Behandeln der freien ss-Aminocarbonsäure z. B. mit wasserabspaltenden Mitteln, wie Carbodiimiden z. B. Dicyclohexylcarhodiimid, zu den er wünschen Azetidino [3 ,2-d] thiazolidin-2-on-Verbin- dungen.
Ester der als Ausgangsprodukte verwendeten ,-Aminocarbonsäuren sind insbesondere aliphatische oder araliphatische Ester, wie Niederalkyl- oder substituierte, z. B. Halogen-eizthaltende Niederalkyl-, sowie Phenyl-niederalkylester, sowie aromatische Ester, z. B.
Phenylester, können durch Behandeln mit organischen Metallverbindungen, wie z. B. organischen Magnesium Grignardverbindungen, insbesondere gehinderten organischen Magnesiumhalogeniden, wie -chloriden oder -bromiden, z. B. tert.-Butyl-magnesium-halogeniden oder Mesityl-magnesium-halogeniden in die erwünschten Azetidino [3,2-d]thiazolidin-2-on-Verbindungen übergeführt werden. Als besonders geeignet zur Bildung des Azetidinrings haben sich organische Aluminiumverbindungen herausgestellt. Es handelt sich dabei in erster Linie um Aluminiumverbindungen, welche 1-3 organische Reste aliphatischen oder cycloaliphatischen. aromatischen oder araliphatischen Charakters, insbesondere Niederalkyl, z. B.
Methyl-, Athyl-, n-Propyl-, Isopropyl-, n-B utyl-, Isobutyl-, sec. Butyl-, n-Pentyl- oder n-Hexylreste, sowie Cycloalkyl-, wie Cyclohexylreste, oder aliphatisch oder cycloaliphatisch verätherte Hydroxylgruppen, wie Niederalkoxy-, z. B.
Isopropyloxy-, n-Butyloxy- oder Isobutyloxyreste, enthalten.
Aluminiumverbindungen dieser Art sind z. B.
Tri-niederalkylaluminium, wie Trimethylaluminium, Tri-isobutyl-aluminium, Di-niederalkylaluminium-hydride oder -halogenide, wie Di-isobutylaluminiumhydrid, Dimethylaluminiumchlorid, Diäthylaluminiumhydrid, Di-isobutylaluminium-chlorid oder Diäthy! aluminiumchlorid, oder Tri-cycloalkylalunninium, wie Tri-cyclohexylaluminium, oder Dicycloalkylaluminium-hydride oder -halogenide, wie Dicyclohexylaluminium-chlorid, (wobei die Aluminium-halogenid-Verbindungen vorteil hafterweise in Gegenwart einer Base verwendet werden), sowie Tri-niederalkoxyaluminium, wie Aluminium-isopropylat.
Die verfahrensgemässe Lactambildung wird vorzugsweise in Anwesenheit eines Verdünnungsmittels, unter Kühlen, bei Zimmertemperatur oder unter Er wäntn en, wenn notwendig, in einer Inertgasatmosphäre und/oder in einem geschlossenen Gefäss vorgenommen.
Lösungsmittel, welche sich insbesondere in Gegenwart der bevorzugten organischen Aluminiumverbindungen anwenden lassen, sind aliphatische oder aromatische Kohlenwasserstoffe, wie Pentan, Hexan, Benzol, Toluol oder Xylol, oder geeignete AXther- oder Thio ätherverbindungen, wie Tetrahydrofuran, ARthylengly- coldimethyAäther, Diäthylenglycoldimethyläther, Dioxan oder Tetrahydrothiopen.
In erhaltenen Verbindungen können Substituenten nach an sich bekannten Verfahren abgespalten und/ oder in andere übergeführt werden. So lassen sich z. B.
tert.-Butylester unter sauren Bedingungen spalten, und in eilialtenen Verbindungen mit einer freien Aminogruppe kann diese, z. B. durch Behandeln mit geeigneten Acylierungsmitteln, substituiert werden.
In den erfindungsgemäss erhaltenen Verbindungen sind Acylreste Ac in erster Linie solche, welche in pharmaliologisch wirksamen N-Acylderivaten der 7-Amino-cephalosporansäure vorkommen, wie der Thienylacetyl-, z. B. der 2-Thienylacetyl-, Chloräthylcarbamyl- oder Phenylacetylrest, oder leicht abspaltbare Acylreste, wie der Rest eines Halbesters der Kohlensäure, z. B. der tert.-Butyloxycarbonylrest.
Der Rest -X- steht insbesondere für die Gruppe der Formel

Äthyl-, n-Propyl-, Isopropyl- oder vorzugsweise Methylgruppen, sowie aromatische, insbesondere Phenylgruppen, oder araliphatische Kohlenwasserstoffreste, insbesondere Phenylalkyl-, z. B. B. durch Niederalkyl-, wie Methyl- oder Athylgruppen, Niederalkoxy-, wie Methoxy- oder Athoxygruppen, Halogen-, wie Fluor-, Chlor- oder Bromatome, Halogenalkyl-, wie Trifluormethylgruppen oder andere geeignete Gruppen, substituiert sein.
Das Verfahren umfasst auch diejenigen Ausführungsformen, wonach als Zwischenprodukte anfallende Verbindungen als Ausgangsstoffe verwendet und die restlichen Verfahrensschritte mit diesen durchgeführt werden, oder das Verfahren auf irgendeiner Stufe abgebrochen wird; ferner können Ausgangsstoffe in Form von Derivaten, z. B. von Salzen, verwendet oder während der Reaktion gebildet werden.
Vorzugsweise werden solche Ausgangsstoffe verwendet und die Reaklionsbedingungen so gewählt, dass man zu den eingangs als besonders bevorzugt aufgeführten Verbindungen gelangt.
Die im obigen Verfahren verwendeten Ausgangsstoffe können z. B. nach dem in der Anmeldung G.Nr.
497 451 (Wo 7) erhalten werden. Bevorzugte Ausgangsstoffe sind in erster Linie Verbindungen der Formel

wobei Ac und X die oben gegebene Bedeutung haben und Ra für den Rest eines Alkohols, wie eines aliphatischen oder araliphatischen Alkohols, insbesondere eines Niederalkanols oder eines substituierten Niederalkanols, wie eines Halogen-niederalkanols, sowie eines Phenylniederalkanols steht.
Erfindungsgemäss erhaltene Verbindungen können, wie im Formelschema gezeigt wird, in die 7-Aminocephaiosporansäure und deren Derivate umgewandelt wrden; die Umwandlung kann z. B. nach dem in den Anmeldungen G.Nr. 497 371 (wo 9), G.Nr. 497 369 (Wo 10), G.Nr. 497 457 (Wo 11), G.Nr. 497 460 (Wo 12), G.Nr. 497 379 (Wo 13) und G.Nr. 497 461 (Wo 14) beschriebenen Verfahren erfolgen.
Die Erfindung wird in den folgenden Beispielen näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiell
Ein C;emisch von 3,832 g L-2, 2-Dimethyl-3-tert.-butyloxy-carbonyl- 5-amino-thiazolidin-4-carbonsäuremethylester in 160 ml absolutem Toluol wird mit Eis gekühlt und unter Rühren und in einer Stickstoffatmosphäre mit 29 ml einer 0,96-m. Lösung von Triisobutyl-aluminium in Toluol langsam versetzt. Nach 64-stündigem Rühren bei 70 wird das Gemisch mit Eis unter Eisbadkühlung während 2 Stunden gerührt und durch ein Filterhilfsmittel filtriert. Letzteres wird mit Toluol und Chloroform gut nachgewaschen; das Filtrat wird über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck eingedampft.
Der Rückstand wird in 75 ml Cyclohexan und 10 ml Hexan gelöst; unter Kühlen bei -18 entsteht ein kristalliner Niederschlag, der abfiltriert, mit Hexan gewaschen und aus Cyclohexan umkristallisiert wird; das so in kleiner Menge erhaltene Produkt stellt den N-(/3-Amino-a-formyl-ethenyl)- carbaminsäure-tert.-butylester der Formel

dar, welches nach einer weiteren Kristallisation aus Benzol bei 145,5-147 schmilzt.
Das Filtrat wird eingedampft und in einem 1 42,5:7,5-Gemisch Benzol und Essigsäureäthylester an 320 g Silicagel chromatographiert, wobei jeweils Fraktionen von 150 ml entnommen werden. Mit 127:23bis 125:25-Gemisch von Benzol und Essigsäureäthylester (450 ml) erhält man eines der Isomeren des 2,9-Dimethyl-3-tert.-butyloxycarbonyl- 4-aminomethylen-thiazolidin-5-ons der Formel in 2 ml absolutem Toluol wird unter einer Stickstoffat- mosphäre und bei Zimmertemperatur 0,4 ml einer 20 %igen Lösung von Diisobutyl-aluminiumhydrid in Toluol zugegeben: das Reaküonsgemisch erwärmt sich auf etwa 350 und verfärbt sich gelb.
Nach 20 Minuten wird Eis zugegeben und geschüttelt, dann mit Cyclohexan verdünnt und die Aluminiumrückstände abzentrifugiert, und 2 mal mit Cyclohexan aufgerührt und jeweils centrifugiert. Die abdekantierten organischen Lösungen werden über Natriumsulfat getrocknet und verdampft.
Der Rückstand wird an 8 g Silicagel chro matographiert; mit drei 25 ml-Fraktionen eines 22:3 Gemisches von Benzol und Essigsäureäthylester wird ein Produkt eluiert, das nochmals an 5 g Silicagel chro matographiert, durch Eluieren mit einem 23,25:1,75 Gemisch von Benzol und Essigsäureäthylester erhält man das gewünschte 3 -tert. -Butyloxycarbonyl-4,4-dimethyl azetidino[3,2-djthiazolidin-2-on, welches im Infrarotspektrum bei 5,64 eine starke Bande zeigt.
Beispiel 3
Ein Gemisch von 0,145 g AL-2,2-DimeíAlyl-3-tert.-butyloxycarboilyl- Sa-amino-thiazolidin-4-carbonsäure- methylester in 4 ml absolutem Toluol und 2 ml einer 0,975-m.
Lösung von Aluminium-isopropylat in absolutem Toluol (hergestellt aus frisch destilliertem Aluminium- isopropylat) wird während 15 Minuten bei 1000 erhitzt.
Nach weiterem Erhitzen bei 115 (15 Minuten) kann im Infrarot-Absorptionsspektrum (in Methylenchlorid) das Auftreten einer starken ss-Lactambande bei 5,63 u und einer nur schwachen Esterbande bei 5,73 festgestellt werden. Das erwünschte -tert. -Butyloxycarbonyl-4,4-dimethyl- azetidino [3 ,2-d] thiazolidin-2-on kann nach dem in den vorhergehenden Beispielen beschriebenen Verfahren erhalten werden.
Beispiel 4
Ein Gemisch von 1,162 g L-2,2-Dimethyl-3 -tert. -butyloxycarbonyl Sa-amAino-thiazolidin-4-carbonsäure- methylester und 1,4 ml Äthyl-diisopropylamin in 75 ml absolutem Toluol wird unter Rühren im Eisbad gekühlt, mit Stickstoff entgast und darauf mit 5,76 ml einer 8 mMol Diäthylaluminiumchlorid enthaltenden Toluollösung versetzt. Nach 32-stündigem Rühren bei 70 wird das Reaktionsgemisch mit Chloroform verdünnt und mit Eis und einer gesättigten Natriumhydrogencarbonatlösung während 10 Minuten gerührt und filtriert. Die wässrige Phase wird einige Male mit Chloroform extrahiert. Die vereinigten organischen Extrakte werden mit Hilfe eines Filtrierhilfsmittels filtriert, letzteres mit Chloroform gewaschen und die vereinigten organischen Lösungen werden über Natriumsulfat getrocknet und verdampft.
Der Rückstand wird an 115 g Silicagel chromatographiert; die Kolonne wird mit einem 95:5 Gemisch von Benzol und Essigsäureäthylester hergestellt und jeweils Fraktionen von 100 ml entnommen.
Mit 200 ml von 87,5:12,5- bis 87:13-Gemischen Benzol und Essigsäureäthylester wird eine kleine Menge

welches bei 1050 schmilzt. Mit 300 ml 124:26- bis 123:27-Gemischen Benzol und Essigsäureäthylester wird das Gemisch der beiden Isomeren und mit 300 ml 122:28- bis 121:29-Gemischen derselben Lösungsmittel das andere Isomere eluiert; letzteres schmilzt nach Umkristallisieren aus Hexan bei 109,5-1100; in einer anderen Modiikation schmilzt das Produkt bei S9.S-C0".
Durch Eluierung mit 750 ml von 120:30- bis 116:34-Gemischen von Benzol und Essigsäureäthylester erhält man als Hauptprodukt das 3 -tert.-Butyloxycarb onyl-4,Sdimethyl- azetidino[3,2-d]thiazoliAdin-2-on der Formel

Nach Umkristallisieren aus Hexan schmilzt das Produkt bei 120,50; [a]D = +2740 (in Chloroform); Infrarot-Absorptionsbanden (in Methylenchlorid) bei 2,95 , 5,62 , 5,90 , 7,25 , 7,35 , 7,75 lu, 8,65 9,36 , 10,60 , 11,65 und 12,30 ; es hat die Konfiguration des L-2,2-Dimethyl-3-tert.-butyloxyzarbonyl- 5α-amino-thiazolidin-4-carbonsäureesters beibehalten.
Aus dem Chromatogramm kann durch Eluieren mit 115:35- bis 100:50-Gemischen Benzol und Essigsäureäthylester (600 ml) unreagiertes Ausgangsmaterial erhalten werden: eine weitere Menge der erwünschten Tactam-Verbindung kann durch erneutes Chromatographieren der Kristallisationsmutterlauge und der Nebenfraktionen an Silicagel erhalten werden.
Beispiel 2
Zu 0,059 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5t-amino-thiazolidin4-carbonsäure- methylester eines Isomeren des 2,2-Dimethyl-3-tert.-butyloxycarbonyl4-aminomethylen-thiazolidin-5-ons, F. 105"; Infrarot-Absorptionsbanden (in Methylenchlorid) bei 2,85,u, 3,02 , 5,97 , 6,05 lt 6,22,u, 6,60 lt 7,23 lt 7,46,u, 8,63 lt, 9,25 und 9,92 lt mit 200 ml 86,5:13,5- bis 86:14-Gemischen (200 ml) ein Gemisch der beiden Isomeren, und mit 200 ml von 85,5:145- bis 85:15-Gemischen das andere Isomere eluiert;
F. 109-110 nach Umkristallisieren mit Hexan; Infrarot-Absorptionsbanden (in Methylenchlorid) bei 2,88,, 3,00 lt 5,98,m, 6,15,u, 7,23 , 7,48,a, 8,65 , 9,23 , 9,90 lt und 11,51 , wobei letzteres schon das 3-tert.-Butyloxvycarbonyl-4,4-dimethyl- azetidino [3,2-d]thiazolidin-2-on als Verunreinigung enthält.
Die Hauptmenge des er erwünschten Produkts wird mit 400ml von 84,5:15,5bis 83:17-Gemischen von Benzol und Essigsäureäthylester eluiert; das 3-tert.-Butyloxycarbonyl-4,4-dimethyl- azetidino [3 ,2-d] thiazolidin-2-on schmilzt nach Umkristallisieren aus Hexan bei 120-121 . Die Mutterlauge wird mit dem mittels 200 ml von 82,5:17,5- bis 82:1 8-Gemischen eluierten Produkt in Hexan bei -18" stehen gelassen und man erhält eine weitere Menge des erwünschten Produktes.
Beispiel 5
Zu einer Lösung von 0,132 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5α-amino-thiazolidin-4-carbonsäuremthylester in 05 ml Äther wird unter einer Stickstoffatmosphäre bei 700 mti 0,4 ml einer 1,35-n. Lösung von Mesitylmagnesiumbromid in Äther tropfenweise versetzt. Nach 5 Minuten wird das Kühlbad entfernt, das Gemisch während 20 Minuten gerührt und dann auf 2 ml einer 10 %igen wässrigen Ammoniumchloridlösung gegossen.
Das Gemisch wird mit Methylenchlorid extrahiert, der organische Extrakt getrocknet und verdampft und der viskose Rückstand an 10 g gereinigtem Silicagel mit einem 9:1-Gemisch Benzol und Essigsäureäthylester chromatographiert, wobei Fraktionen zu 5 ml abgetrennt werden. Fraktionen 8 und 9 enthalten laut Dünnschichtchromatogramm und Infrarot-Absorptionsspektrum (in Methylenchlorid, charakteristische Bande bei 5,60 lt) das erwünschte 3 -tert.-Butyloxycarbonyl-4,4-dimethylazetidino[3,2-d]thiazolidin-2-on, das nach den in obigen Beispielen beschriebenen Verfahren isoliert werden kann.
Beispiel 6
Ein Gemisch von 0,005 g 4,4-Dimethyl-3-tert.-butyloxycarbonyl- azetidinor3,2-d]thiazolidin-2-on und 1 ml Trifluoressigsäure wird während 2 Stunden bei Zimmertemperatur stehen gelassen. Das Lösungsmittel wird im Wasserstrahlvakuum abgedampft und der Rückstand in Methylenchlorid aufgenommen; die organische Lösung wird mit einer Natriumhydrogencarbonatlösung neutral gewaschen und eingedampft.
Man erhält so das 4,4-Dimethyl-azetidino [3 ,2-d] thiazolidin-2-on der Formel

welches im Infrarotspektrum (Methylenchlorid) bei 2,92 sl und 5,66 ,t! charakteristische Banden zeigt.
Beispiel 7
Ein Gemisch von 0,103 g 3-tert.-Butyloxycarbonyl-4,4-dimethylazetidino[3,2-d]thiazolidin-2-on und 7,5 ml Trifluoressigsäure wird 20 Minuten bei Raumtemperatur stehen gelassen und dann unter Wasserstrahlvakuum eingedampft. Nach dem Neutralisieren mit wässriger Natriumhydrogencarbonatlösung wird das Reaktionsgemisch mit Methylenchlorid extrahiert; der organische Extrakt wird mit Wasser gewaschen, getrocknet und eingedampft und man erhält das kristalline 4,4-Dimethyl-azetidino[3 ,2-d] thiazolidin-2-on, welches bei 114-117 schmilzt. Im Gegensatz zum Ausgangsmaterial erhält man eine positive Nitroprussidnatrium-Reaktion, was darauf hindeutet, dass das Produkt zum Teil in der Form des 3-Isopropylidenamino4-in ercapto-azetidin-2-ons der Formel

Im Infrarot-Absorptionsspektrum (in Methylenchlorid) beobachtet man Banden bei 2,93,u, 5,67,u, 9,20 lt und 10,58 .
Beispiel 8
Eine Suspension von 1,652 g B-Amino-p-phenyl- propionsäure in 50 ml absolutem Toluol wird mit 1,72 ml einer Benzollösung enthaltend 20 mMol Triisobutylaluminium bei-18 und unter einer Stickstoffatmosphäre versetzt. Nach 16-stündigem Rühren bei 25 werden weitere 9 ml einer Benzollösung enthaltend 1û,5 mMol Triisobutylaluminium bei 25 zugegeben, worauf das bis anhin ungelöste Material schnell in Lösung geht und diese sich gelb verfärbt. Nach 6-stündigem Rühren wird das Reaktionsgemisch während 17 Stunden und unter einer Stickstoffatmosphäre erhitzt, dann mit Eis und Chloroform versetzt; die organische Phase wird durch ein Filterhilfsmittel filtriert, mit Natriumsulfat getrocknet und verdampft.
Der Rückstand wird an 100 g Silicagel chromatographiert, wobei die Kolonne in einem 95,5:0,5-Gemisch Benzol und Essigsäureäthylester hergestellt wird und 100 ml Fraktionen entnommen werden. Mit 200 ml von 92:8- bis 91:9-Gemischen von Benzol und Essigsäureäthylester und 200 ml von 87:13 bis 86:14-Gemischen des gleichen Lösungsmittelgemisches werden Nebenprodukte ausgewaschen, während man mit 600 ml 77:23- bis 70:30-Gemischen von Benzol und Essigsäureäthylester das erwünschte 4-phenyl-azetidin-2-on eluiert, welches nach Umkristalllsieren aus Hexan bei 105-105,5 schmilzt.
Beispiel 9
Ein Gemisch. von 1,162 g L-2,2-Dimethyl-3-tert.-butylcarbonyl- 5ss-amino-thiazolidin-4-carbonsäure- methylerter und 1,4 ml Äthyldiisopropyiamin in 75 ml absolutem Toluol wird unter Rühren, mit Eis gekühlt, mit Stickstoff entgast und mit 5,76 ml Diätliylalmniniumchiorid in Toluol mittels Zutropfen aus einer hypodermischen Nadel versetzt. Nach 32-stündigem Stehenlassen bei t-75 wird das Reaktionsgemisch mit Chloroform verdünnt und mit einem Gemisch von Eis und einer gesättigten Natriumhydrogencarbonatlösung während 10 taIinuten geschüttelt, durch ein Filterhilfsmittel filtriert und der Rückstand mit Chloroform nachgewaschen.
Die organische Lösung wird über Natriumsulfat ge trnckner, filtriert und eingedampft; der Rückstand wird an 115 g Silicagel chromatographiert, wobei die Kolonne mit einem 95:5-Gemisch von Benzol und Essigsäureäthylester hergestellt wird und Fraktionen von 100 ml entnommen werden.
Mit 200 ml 87,5:12,5- bis 87:13-Gemischen von Benzol und Essigsäureäthylester wird eines der Isomeren des 2,2-Dimethyl-3-tert.-butyloxycarbonyl4-aminomethylen-thiazolidin-5-ons, F. 105", mit 200 ml von 86,5:13,5- bis 86:14-Gemischen von Benzol und Essigsäureäthylester ein Gemisch der beiden Isomeren des 2,2-Dimethyl-3-tert.-butyloxycarbonyl4-aminomethylen-thiazolidin-5-ons und mit 200 ml von 85,5:14,5- bis 85:15-Gemischen von Benzol und Essigsäureäthylesters das beinahe reine zweite Isomere erhalten;
es enthält schon eine kleine Menge des 3-tert.-Butyloxycarbonyl-4,4 dimethyl-azetidino [3,2-d]thiazolidin-4-ons und schmilzt nach Umkristallisieren aus, Hexan bei 109-110". Mit 400 ml von 84,5:15,5- bis 83:17-Gemischen von Benzol und Essigsäureäthylester wird das 3-tert.-Butyloxycarbonyl-4,4-dimethyl azetidino [3 ,2-d] thiazolidin-2-on eluiert, das nach dem Kristallisieren aus Hexan bei 120-i210 schmilzt. Die Mutterlauge wird mit dem durch 82,5:17,5- bis 82:18-Gemische (200 ml) von Benzol-Essigsäureäthylester eluierten Produkt vereinigt und aus Hexan umkristallisiert; man erhält so eine weitere Menge des erwünschten Produkts. Unreagiertes Ausgangsmaterial kann mit weiteren Benzol-EssigsäureäthylesteryGemischen eluiert werden.
Beispiel 10
Eine Lösung von 5,805 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-amino-thiazolidin-4-carbonsäuremethylester in 400 ml absolutem Toluol und 7,5 ml athyldiisopro- pylamin wird nach Entgasen mit Stickstoff gekühlt und bei -15" kräftig gerührt.
Darauf werden etwa 20 ml einer ca. 1,75-molaren Lösung von Dimethyl-aluminiumchlorid in Toluol tropfenweise innert 45 Minuten zugegeben; die Lösung verfärbt sich gelb und die Temperatur steigt auf -100. Nach 45-minütigem Rühren bei -10" bis -150 werden wiederum 15 ml des Reagens während 10 Minuten zugegeben, nach 60 Minugen weitere 3-4 ml. Nach 30-minütigem Rühren wird das Gemisch bei -5 auf 200 ml Toluol, 400 ml Eis und 50 ml einer gesättigten Natriumhydrogencarbonatlösung ausgegossen, während 10 Minuten kräftig gerührt und durch ein Filterhilfsmittel filtriert, welches mit mehreren Portionen Methylenchlorid ausgewaschen wird.
Die wässrige Phase wird 3 mal mit Methylenchlorid extrahiert; die vereinigten organischen Lösungen werden über Natriumsulfat getrocknet und eingedampft und der Rückstand an 370 g Silicagel chromatographiert; die Kolonne wird in einem 90:10-Gemisch von Benzol-Essigsäureäthylester hergestellt. Mit je 500 ml von 87,5:12,5-, 85:15-, 82,5:17,5- und 80:20 Gemischen von Benzol-Essigsäureäthylester erhält man die 2,2-Dimethyl-3-tert.-butyloxycarbonyl 4-annomethylen-thiazolidin-5-one, deren Gemisch man mit 200 ml eines 79:21-Gemisches von Benzol-Essigsäureäthylesters eluiert.
Mit 300 ml eines 79:21-, 500 ml eines 78:22-, 500 ml eines 77:23und 300ml eines 76:24-Gemisches von Benzol-Essigsäureäthylesters wird das 3 -iert.-Butyloxycarbonyl-4,4-dimethyl-aze ,2- d] thiazolidin-2-on eluiert, welches spontan kristallisiert und aus Hexan umkristallisiert wird; durch erneutes CCh,romatographie- ren der Mutterlauge erhält man eine weitere Menge des erwünschten Produktes.
Aus dem Chromatogramm wird mit 200 ml eines 76:24-, 50 ml eines 65:35- und 500 mi eines 50:50: Gemisches von Benzol-Essigsäureäthylester unreagiertes Ausgangsmaterial eluiert; der L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-aminoqhiazolidin-4-carbonsäure- methylester kristallisiert spontan und schmilzt bei 55-58 .
Beispiel 11
Eine Lösung von 0,587 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-amino-thiazolidin-4-carbonsäure- methylester in 40 ml absolutem Dioxan wird unter Rühren und in einer Stickstoffatmosphäre auf 800 erhitzt und dann mit 3 ml einer 1,6-molaren Lösung von Triäthylaluminium in absolutem Toluol rasch versetzt. Man rührt während 15 Minuten bei 800 und unter der Stickstoffatmosphäre weiter und versetzt das Reaktionsgemisch dann mit 5 ml tert.-Butanol und giesst dann auf ein Gemisch von 20 ml 20 /oiger wässriger Zitronensäure und 100 g Eis aus und extrahiert 2 mal mit je 150 ml Methylenchlorid. Der organische Extrakt wird mit 50 ml Wasser gewaschen, getrocknet und eingedampft.
Das als Rückstand erhaltene klare Öl weist im Infrarot Absorptionsspektrnm (in Methylenchlorid) die für das 3 -tert.-Butyloxycarbonyl-4,4 dimethyl-azetidino[3,34]thiazolidin-2-on charakteristischen Banden bei 2,85,u, 5,60 lot und 9,35,u, sowie die für die beiden Isomeren 2,2-Dimethyl-3-tert.-butyloxycarbonyl4-Aminomethylen-thiazolidin-5-one charakteristischen Banden bei 2,75 lot 6,0 lt 6,15 lt und 9,25 lt auf.
Die quantitative Analyse des Kernre sonanzspektrums ergibt folgende Zusammensetzung des Produktes: 10 O/o L-2,2-Dimethyl-3-tert. -butyloxycarbonyl- Sa-amino-thiazolidin-4-carbonsäure methylester, 60 Oio 3 -tert.-Butyloxycarbdnyl-4,4,-dimethyl- azetidino [3 ,2-d] thiazolidin-2-on und 30 O/o der isomeren 2,2-Dimethyl-3-tert.-butyloxycarbonyl- 4-aminomethylen-thiazolidin-5-one; das Gemisch kann z. B. wie im Beispiel 1 beschrieben, getrennt werden.
Beispiel 12
Eine Lösung von 0,587 g L-2,2-Dimethyl-3 -tert.-butyloxycarbonyl- 5ayamino-thiazolidin-4-carbon,säure- methylester in 40 ml Toluol wird auf 80" erwärmt und unter Rühren mit Stickstoff gespült. Nach einer Stunde werden 0,093 g Anilin und kurz darauf 4,8 mMol Triäthylaluminium in Toluol zugegeben. Nach 15-minütigem Rühren bei 800 wird das Reaktionsgemisch mit 5 ml tert.-Butanol versetzt und das Reaktionsgemisch wie im Beispiel 14 beschrieben aufgearbeitet.
Man erhält ein öliges Produkt, welches im Infrarot-Absorptionsspektrum (in Methylenchlorid) die für das 3-tert.-Butyloxycarbonyl-4,4 dimethyl-azetidino [3,2-d]thiazolidin-2-on charakteristischen Banden bei 2,85etc, 5,60u und 9,35,u, sowie die Banden der beiden isomeren 2,2-Dimethyl-3 -tert.-butyloxycarbonyl4-Aminomethylen-thiazolidin-5-one bei 2,75,a, 6,0 6,15, und 9,25lt zeigt.Diequan- titative Analyse des Kernresonanzspektrums zeigt folgende Zusammensetzung:
