Verfahren zur Herstellung von 2, 6-Pyridindimethanol-bis-carbamat-Derivaten
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 2, 6-Pyridindimethanol-bis carbamat-Derivaten, der allgemeinen Formel :

worin Ri ein Wasserstoff-oder Halogenatom oder eine Alkylgruppe, Rg ein Wasserstoffatom oder eine Alkylgruppe und R3 ein Wasserstoffatom oder eine Alkyl-, Fluoralkyl-, Hydroxyalkyl-, Alkenyl-, Aralkyl-oder Arylgruppe ist, wobei die Alkylgruppen 1-10 C-Atome aufweisen oder R9 und R3 bilden zusammen mit dem N-Atom, an welches sie gebunden sind,
eine gesättigte heterocyclische Gruppe mit insgesamt 1 oder 2 Hetero- atomen.
2, 6-Pyridindimethanol-bis-carbamat-Derivate sind nützlich bei der Behandlung entzündlicher Erkrankungen, elinschliesslich rheumatischer Affektionen, wie z. B.
Gelenkrheumatismus und rheumatische Arthritis. Es wurden n bereits mehrere Verfahren zur Herstellung der 2, 6-Pyridindimethanol-bis-carbamat-Derivate, vorgeschlagen. Diese Verfahren beruhen auf folgenden Reaktionen : a) Reaktion von Pyridindimethanol mit einem Isocyanat, b) Reaktion von Pyridindimethanol mit einem
Carbamoylchlorid und c) Reaktion eines durch die Umsetzung von Pyridin dimethanol mit Phosgen erzeugten Pyridindimethanol bis-chloroformiats mit einem Amin.
Diese Verfahren sind mit verschiedenen Nachteilen verbunden : z. B. sind einifge der Isocyanate nicht leicht erhältlich, besonders wenn eine industrielle Herstellung erwünscht ist, und die Verfahren b) und c) sind, was die Ausbeute betrifft, nicht befriedigend, insbesondere im Fall der nicht substituierten bis-Carbamate.
Nun ist ein neues und einfaches Verfahren zur Herstellung von 2, 6-Pyridindimethanol-bis-carbamat- Derivaten gefunden worden, das die oben erwähnten Nachteile ausschaltet.
Nach dem erfindungsgemässen Verfahren werden die 2, 6-Pyridinedimethanol-bis-carbamat-Derivate obiger Formel so hergestellt, indem man eine Verbindung der Formel :

Die angeführten Alkylgruppen mit 1-10 C-Atomen können gerad-und verzweigtkettige gesättigte einwertige aliphatische Kohlenwasserstoffradikale sein, wie z. B.
Methyl, Äthyl, Propyl, Isopropyl, Butyl, sekundäres Butyl, tertiäres Butyl, Amyl, Decyl usw. Die bevor- zugten Alkylgruppen sind diejenigen, die 1-6 Kohlen stoffatome aufweisen.
In einer zweckmässigen Ausführungsform der vorliegenden Erfindung wird die Verbindung der Formel II mit überschüssigem Ammoniak oder Amin der Formel III bei einer Temperatur von 0 bis 150 C, insbesondere von Raumtemperatur bis 50 C, oder, wenn erwünscht, bei einer Temperatur von unter 0 C, z. B. dem Siedepunkt des wässrigen Ammoniaks, in Gegenwart oder in Abwesenheit von Lösungsmitteln oder Verdünnungs- mitteln umgesetzt.
Das Amin der Formel III kann ein primäres oder sekundäres Amin sein. Es kann ein aliphatisches, Aralkyl-, aromatisches oder cycloaliphatisches Amin verwendet werden. Als Beispiel für das Amin kann erwähnt werden : Methyl-, Äthyl-, Propyl-, Butyl-, Dimethyl-, Diäthyl-, Dipropyl-, Benzylamin. Ein Anilin, Chloranilin, Anisidin, Pyrrolidin, Naphthylamin und Piperidin kann ebenfalls verwendet werden. Es können auch Amine verwendet werden, die 1 oder 2 Heteroatomc neben dem Stickstoffatom, woran mindestens ein reaktions fähiges Wasserstoffatom gebunden ist, aufweisen.
Als Beispiele seien hier erwähnt : 2-Aminoäthanol, 3-Amino- propanol, Morpholin, N-(2-Aminoäthyl)-morpholin, N Alkylpiperazin, unsym-Dimethyläthylendiamin, unsym- Diäthyläthylendiamin und unsym-Dimethyläthyltetra- methylendiamin. Gewöhnlich werden mindestens 2 Mol Ammoniak oder Amin pro Mol der Verbindung II verwendet. Wenn erwünscht, kann die Reaktion mit weniger als 2 Mol des Amins ausgeführt werden, doch ist dies mit keinem Vorteil verbunden. Eine gröBere Menge Ammoniak oder Amin kann verwendet werden, ohne dass die Ausbeute vermindert wird.
Die Reaktion wird vorzugsweise bei ungefähr Raumtemperatur ausgeführt ; wenn nötig, kann auch eine höhere Temperatur von bis zu 150 C oder sogar darüber verwendet werden.
Gewöhnlich wird die Reaktion bei atmosphärischem Druck ausgeführt. Wenn erwünscht, kann die Reaktion bei erhöhtem oder vermindertem Druck ausgeführt werden.
Die erforderliche Zeit, um die Reaktion zu Ende zu führen, hängt von der Natur der Reaktionsteilnehmer und den Reaktionsbedingungen, wie z. B. der Reaktions- temperatur, ab. Im allgemeinen genügt eine Reaktions- zeit von 30 Minuten bis 24 Stunden, um die Reaktion bei Raumtemperatur zu Ende zuführen.
Die Reaktion kann entweder in Abwesenheit oder in Gegenwart eines Lösungs- oder Verdünnungsmittels ausgeführt werden. Es können anorganische oder orga- nische Lösungsmittel verwendet werden, die fiir orga- nische Synthesen im allgemeinen benutzt ! werden und dem Reaktionssystem gegenüber inert sind. Beispielshalber seien hier erwähnt : Wasser, Alkohole, wie z. B.
Methanol, Äthanol und Propanol, Aceton, Äthylacetat, Dioxan und Kohlenwasserstoffe wie z. B. Benzol, Toluol und Xylol-sowie Gemische davon.
Eine Verbindung der Formel II, die als Ausgangs- stoff dient, kann nach dem nachstehend als Beispiel angegebenen Verfahren hergestellt werden.
Eine Verbindung der Formel II, worin Z eine Aryloxygruppe ist, das heisst 2, 6-Pyridindimethanol-bis aryloxyformiat oder dessen in 4-Stellung substituiertes Produkt, kann z. B. durch Reaktion von 2, 6-Pyridindimethanol oder dessen 4-substituierten-2, 6-Pyridin- dimethanol, das in 4-Stellung durch Wasserstoff, Halogen oder Alkyl substituiert ist, mit Arylchlorformiat hergestellt werden. Es ist von Vorteil, wenn die Reaktion in Gegenwart eines Dehydrochlorierungsmittels ausgeführt wird. Als Dehydrochlorierungsmittel kom- men tertiäre Amine oder anorganische Alkali in Frage.
Bevorzugte Dehydrochlorierungsmittel sind z. B. Tri äthylamin, Dimethylanilin, Pyridin und Chinolin. Die Reaktion kann entweder in Gegenwart oder in Ab wesenheit eines Lösungsmittels ausgeführt werden. Es kann jedes Lösungsmittel, das den Ausgangstoff gut auflöst und Arylchlorformiat, einem Acylierungsmittcl gegenübe. r inert ist, verwendet werden, doch ist die Verwendung von Pyridin am vorteilhaftesten, da es gleichzeitig auch als Dehydrochlorierungsmittel wirkt.
Obwohl die Reaktion bei Raumtemperatur gut vor sich geht und zu Ende geführt wird, ist es zur Vermeidung von Nebenreaktionen erwünscht, die Reaktion in einem frühen Stadium bei einer Temperatur von 5 bis 10 C auszuführen. Die Arylgruppe des Arylchlorformiats ist vorzugsweise eine Phenylgruppe. Diese Phenylgruppe kann durch ein Halogenatom oder eine Alkyl-oder Alkoxygruppe substituiert sein. Statt der Arylgruppe kann auch eine Naphthylgruppe verwendet) werden.
Anstelle des Arylchlorformiats kann Arychlorthioformiat verwendet werden, doch ist dies vom wirtschaftlichen Standpunkt kaum von Vorteil.

ist, das heil3t 2, 6-Pyridindimethanol-bis-azidformiat oder dessen 4-substituiertes-2, 6-Pyridinmethanol-bis-azidformiat, das in 4-Stellung durch Wasserstoff, Halogen oder Afkyl substituiert ist, kann z. B. hergestelltl werden, indem das oben erwähnte 2, 6-Pyridindimethanol-bisaryloxyformiat oder dessen in 4-Stellung substituiertes Produkt mit Hydrazin umgesetzt wird und das dabei erzielte 2, 6-Pyridin-dimethyl-bis-carbazat oder dessen in 4-Stellung substituiertes Produkt dann mit Salpetersäure umgesetzt wird. Vorzugsweise wird die Umsettzung bei niedriger Temperatur, wie z.
B. -10 bis 10 C, und gewöhnlich in der Nähe von 0 C ausgeführt. Das erwähnte Carbazat ist wasserlöslich, doch bildet es bei Zusatz von Essig-oder Salzsäure ein Salz, da es ein basisches Säurehydrazidradikal aufweist, und wird wasserlöslicher. Wird eine wässrige Lösung eines S, alzes von Salpetersäure in eine wässrige Lösung des erwähnten Carbazatsalzes gegeben, so wirkt die gebildete Salpeter- säure sofort auf das erwähnte Carbazatsalz, was zur Bildung des erwähnten Azidformiats führt. Da es sich in Wasser schlecht auflöst, kristallisiert es aus der wässrigen Reaktionsflüssigkeit aus.
Es wird dann filtriert und mit Wasser gewaschen, worauf es entweder bei Raumtemperatur getrocknet oder mit einem organischen Lösungsmittel, wie z. B. Äther oder Benzol, extrahiert wird. Der Extrakt wird getrocknet und konzentriert, wobei das gewünschte Produkt in Form von Kristallen in hoher Ausbeute erzielt wird.
Die entzündungshemmende Wirkung wurde mittels eines Versuchs an Ratten bestimmt, bei denen ein Hinterpfotenödem durch Carragecn (isländisches Moos) als Reizmittel hervorgerufen wurde. Einige der Ergeb- nisse sind in der nachstehenden Tabelle als ED3o ausgedrückt, das heisst die minimale orale Dosis der betreffenden Verbindung, die eine 30 %ige Hemmung der Ödembildung bewirkt, zusammen mit der akuten Toxi zität bei oraler Verabreichung an Mäusen, als LD50 ausgedrückt, aufgeführt.

Eine orale Verabreichung von bis zu 400 mg/kg 2, 6-Pyridindimethanol-bis-(N-methylcarbamat) im Tag an Mäuse während 6 Monaten bewirkte keine chronische Toxizität, was aus einer gründlichen patho histologischen Untersuchung hervorging. Es wurde weder eine teratogene Wirkung noch eine zusätzliche Neigung zu Affektionen entdeckt.
Beim erfindungsgemässen Verfahren können die Produkte nach herkömmlichen Methoden, wie sie nachstehend beschrie, ben werden, getrennt und gereinigt werden. Bei Verwendung von bis-Aryloxyformiaten als Verbindungen der Formel II wird es oft vorgezogen, das bei der Reaktion entstandene Phenol bei vermindertem Druck abzudestillieren oder mit einer wässrigen Alkalilösung zu extrahieren.
Die vorliegende Erfindung wird durch folgende Bei- spiele veranschaulicht : wenn nicht anders angegeben, sind die Teile Gewichtsteile.
Beispiel I a) Herstellung von 2, 6-Pyridindimethanol-bis phenoxyformiat
In eine gerührte Lösung von 69, 5 Teilen 2, 6 Pyridindimethanol in 300 Volumteilen Pyridin werden 164 Teile Phenylchlorformiat bei einer Temperatur von 5-10 C tropfenweise gegeben. Nach 3stündigem Rüh ren bei Raumtiemperatur wird das Reaktionsgemisch in Wasser gegossen.
Das erzielte ölige Produkt, das bald hart wird, wird abfiltriert und aus Methanol umkristalli- siert ; man erhält 165 Teile 2, 6-Pyridindimethanol-bis phenoxyformiat, F = 50-51 C. b) Herstellung von 2, 6-Pyridindimethanol-bis carbamat
37, 9 Teile wie in Beispiel 1, Abschnitt a), herge stellites 2, 6-Pyridindimethanol-bis-phenoxyformiat wird mit 33 Teilen 30%iger methanolischer Methylaminlösung unter Rühren und Abkühlen behandelt. Nachdem die anfängliche exotherme Reaktion aufgehört hat, wird das Reaktionsgemisch mehrere Stunden bei Raumtemperatur stehengelassen. Das Gemisch wird mit Methanol verdünnt und dann auf einem Wasserbad erhitzt ; man erhält eine klare Lösung, die man sich auf Raumtemperatur abkühlen lässt.
Die ausgefällten Kristalle, nämlich 21 Teile, werden abfiltriert Nachdem das als Nebenprodukt der Reaktion gebildete Methanol und Phenol im Vakuum abdestilliert werden, wird eine weitere Menge Kristalle aus der Mutterlauge zurückgewonnen. Die kombinierten Kristalle werden aus Methanol umkristallisiert ; man erhält 24, 4 Teile 2, 6-Pyridindimethanol-bis-N-methylcarbamat, F = 136 bis 137 C. Die Ausbeute beträgt 96%.

Beispiel 2
In eine Lösung von 19 Teilen 2, 6, Pyridindimethanol- b, is- (phenoxyformiat) in 30 Volumteilen Benzol werden 9 Teile Allylamin bei 25-30 C gegeben. Das Reak tionsgemisch wird über Nacht bei Raumtemperatur stehen gelasse, mit Benzol verdünnt und nacheinander mit einer 10% igen wässrigen Lösung von 4, 5 Teilen Natriumhydroxyd und mit Wasser gewaschen. Die Benzollösung wird isoliert und bei vermindertem Druck konzentriert. Man erhält 14, 1 Teile 2,6-Pyridindimethanol-bis-(N-allylcarbamat), F = 83-84 C.

Beispiel 3 a) Herstellung von 2, 6-Pyridindimethanol-bis (p-chlorphenoxyformiat)
In eine Lösung von 13, 9 Teilen 2, 6-Pyridindi- methanol in 140 Volumteilen Pyridin werden 40, 1 Tei len pChlorphenylchlorformiat unter Rühren und Kühlen auf eine Temperatur von unter 0 C tropfenweise gege- ben. Nach erfolgter Beigabe wird das Reaktionsgemisch mit Eiswasser behandelt, worauf die ausgefällten Kri- stalle abfiltriert, mit Wasser und dann mit Methanol gewaschen und getrocknet werden. Man erhält 42 Teile 2,6-Pyridindimethanol-bis-(p-chlorphenoxyformiat), was einer 94%igen Ausbeute entspricht.
Nach Umkristalli sieren aus Isopropanol-Benzol schmilzt das Produkt bei 121 C. b) Herstellung von N-substituiertem bis-Carbamat von 2, 6-Pyridindimethanol
In ein gerührtes Gemisch von 15 Teilen wie in Beispiel 3, Abschnitt a), hergestelltem 2, 6Pyridindi- methanol-bis-(p-chlorphenoxformiat) und 15 Volumteilen Methanol werden 12 Teile Morpholin gegeben.
Nachdem die exotherme Reaktion aufgehört hat, wird das gesamte Volumen des Reaktionsgemisches durch Zusatz von Methanol auf 50 Volumteile erhöht, worauf das Gemisch auf einem Wasserbad erhitzt wird, um eine klare Lösung zu erhalten. Man lässt die Lösung sich auf Raumtemperatur abkühlen, und die ausgefällten Kristalle werden abfiltriert. Man erhält 12, 5 Teile 2, 6 Pyridindimethanol-bis-N-morpholincarbamat, F = 157 bis 158 C.

In ähnlicher Weise wie in den obigen Beispielen werden folgende bis-Aryloxyformiate von 2, 6-Pyridin- dimethanol umgesetzt, wobei die entsprechenden 2, 6 Pyridindimethanol-bis-carbamat-Derivate in ähnlicher Ausbeute erzeugt werden. Die Ergebnisse werden in der nachstehenden Tabelle angegeben.

Beispiel 16
Ammoniakgas wird bei Raumtemperatur in eine Suspension von 19 Teilen von 2, 6-Pyridindimethanolbis-phenoxyformiat in 20 Volumteilen Methanol einge- leitet.
Währensd die Temperatur steigt, wird das Gemisch eine Lösung, worauf Kristalle ausfallen,. Nach dem Abkühlen werden Kristalle abfiltriert und getrocknet. Man erhält 10 Teile 2, 6-Pyridindimethanol-bis-carbamat, F = 165-166 C. Die Verwendung von Ammoniakflüssigkeit ohne Verdünnungsmittel führt zu einer leich- ten Erhöhung der Ausbeute an bis-Carbamat.

Beispiel 17 a) Herstellung von 2, 6-Pyridindimethanol-bis-carbazat
In ein gerührtes Gemisch von 189, 7 Teilen wie in Beispiel 1, Abschnitt a) hergestelltem 2, 6-Pyridindi methanol-bis-phenoxyformiat und 380 Volumteilen Methanol werden 55, 1 Teile Hydrazinhydrat bei Ab- kühlung auf 20-30 C tropfenweise gegeben. Das Reak tionsgemisch wird zu einer klaren Lösung, während eine exotherme Reaktion vor sich geht. Nach erfolgter Beigabe des Hydrazins fallen weisse Kristalle aus der Lösung. Nach Abkühlen werden die Kristalle abfiltriert, mit Methanol gewaschen und getrocknet.
Man erhält 2, 6-Pyridindimethanol-bis-carbazat, F = 140-142 C (118, 7 Teile), was einer 93% igen Ausbeute entspricht. b) Herstellung der 2, 6wPyridindimethanol- bis-azidoformiat
127, 6 Teile wie in diesem Beispiel unter a) herge stelltem 2, 6-Pyridindimethanol-bis-carbsazat werden in einem Gemisch von 200 Volumteilen Essigsäure und 220 Volumteilen Wasser gelöst. Die erzielte Lösung wird 30 Minuten unter Rühren und Abkühlen auf -5 bis 0 C mit einer wä#rigen Lösung von 7, 6 Teilen Natriumnitrit behandelt. Die Lösung wind weitere 30 Minuten bei etwa 0 C gerührt, worauf sie mit Wasser verdünnt wird.
Der erzielte Niederschlag wird abfiltriert, mit Wasser gewaschen und bei vermindertem Druck getrocknet. Man erhält 131, 8 Teile 2, 6-Pyridin- dimethanol-bis-azidoformiat, F = 42-43 C, was einer 95% igen Ausbeute entspricht. c) Herstellung von bis-Carbamat von
2, 6-Pyridinmethanol
14 Teile wie in diesem Beispiel unter b) erzieltes 2, 6-tPyridinmethanol-bis-azidoformiat werden in 16 Vo lumteilen Benzol gelöst, wobei allfällig unlösliche Substanz abfiltriert wird. Die Lösung wird tropfenweise in eine gekühlte Lösung von 10 Teilen Diäthylamin in 20 Volumteilen Benzol gegeben.
Das Reaktionsgemisch wird mehrere Stunden bei 20-25 C stehengelassen, wobei das pH des Gemisches durch Zusatz von Di äthylamin alkalisch gehalten wird. Nach Beendigung dieser Reaktion wird das Reaktionsgemisch mit Benzol verdünnt und nacheinander mit Wasser, einer wässrigen Natriumbicarbonatlösung und wiederum Wasser gewaschen. Die organische Schicht wird abgetrennt und bei vermindertem Druck konzentriert ; man erhält 16, 5 Teile eines öligen Rückstands. Durch Behandlung dieser öligen Substanz mit Salzsäure werden 15 Teile
2, 6aPyridindimethanol-bis-(N, N-diäthylcarbamat)- hydrochlorid, F = 131-132 C, erzielt ; nach Umkristallisieren aus Aceton-Ather schmilzt das Produkt bei 132-133 C.

Der Rückstand wird aus Methanol umkristallisiert ; man erhält 2, 6-Pyridindimethanol-bis (N-phenylcarbamat), F = 153-154 C.

2,6-Pyridinedimethanol-bis-carbamate derivatives are useful in the treatment of inflammatory diseases, including rheumatic affections, such as e.g. B.
Rheumatoid arthritis and rheumatoid arthritis. Several processes for the preparation of the 2,6-pyridinedimethanol-bis-carbamate derivatives have already been proposed. These processes are based on the following reactions: a) reaction of pyridinedimethanol with an isocyanate, b) reaction of pyridinedimethanol with an
Carbamoyl chloride and c) reaction of a pyridine dimethanol bis-chloroformate produced by the reaction of pyridine dimethanol with phosgene with an amine.
These methods have several disadvantages: e.g. B. some of the isocyanates are not readily available, especially when industrial production is desired, and processes b) and c) are unsatisfactory in terms of yield, especially in the case of the unsubstituted bis-carbamates.
A new and simple process for the preparation of 2,6-pyridinedimethanol-bis-carbamate derivatives has now been found which eliminates the disadvantages mentioned above.
In the process according to the invention, the 2,6-pyridinedimethanol-bis-carbamate derivatives of the above formula are prepared by adding a compound of the formula:

Examples include: 2-aminoethanol, 3-aminopropanol, morpholine, N- (2-aminoethyl) morpholine, N-alkylpiperazine, unsym-dimethylethylenediamine, unsym- diethylethylenediamine and unsym-dimethylethyltetra- methylenediamine. Usually at least 2 moles of ammonia or amine are used per mole of compound II. If desired, the reaction can be carried out with less than 2 moles of the amine, but there is no benefit in doing so. A larger amount of ammonia or amine can be used without reducing the yield.
The reaction is preferably carried out at about room temperature; if necessary, a higher temperature of up to 150 C or even above can be used.
Usually the reaction is carried out at atmospheric pressure. If desired, the reaction can be carried out under increased or reduced pressure.
The time required for the reaction to complete depends on the nature of the reactants and the reaction conditions, e.g. B. the reaction temperature. In general, a reaction time of 30 minutes to 24 hours is sufficient to complete the reaction at room temperature.
The reaction can be carried out either in the absence or in the presence of a solvent or diluent. Inorganic or organic solvents, which are generally used for organic syntheses, can be used! and are inert to the reaction system. It is advantageous if the reaction is carried out in the presence of a dehydrochlorinating agent. Tertiary amines or inorganic alkali can be used as dehydrochlorination agents.
Preferred dehydrochlorinating agents are e.g. B. triethylamine, dimethylaniline, pyridine and quinoline. The reaction can be carried out either in the presence or in the absence of a solvent. Any solvent that dissolves the starting material well and aryl chloroformate, an acylating agent, can be used. r is inert, but the use of pyridine is most advantageous because it also acts as a dehydrochlorinating agent.
Although the reaction goes well and is completed at room temperature, in order to avoid side reactions, it is desirable to carry out the reaction at an early stage at a temperature of 5 to 10 ° C. Oral administration of up to 400 mg / kg of 2,6-pyridinedimethanol-bis- (N-methylcarbamate) per day to mice for 6 months did not cause any chronic toxicity, which was evident from a thorough patho-histological examination. Neither a teratogenic effect nor an additional tendency to affections was discovered.
In the process according to the invention, the products can be separated and cleaned by conventional methods as described below. When using bis-aryloxyformates as compounds of the formula II, it is often preferred to distill off the phenol formed in the reaction under reduced pressure or to extract it with an aqueous alkali solution.

Example I a) Preparation of 2,6-pyridinedimethanol bis phenoxyformate
164 parts of phenyl chloroformate are added dropwise at a temperature of 5-10 ° C. to a stirred solution of 69.5 parts of 2.6 pyridine dimethanol in 300 parts by volume of pyridine. After 3 hours of stirring at room temperature, the reaction mixture is poured into water.

The combined crystals are recrystallized from methanol; 24.4 parts of 2,6-pyridinedimethanol-bis-N-methylcarbamate are obtained, F = 136 to 137 C. The yield is 96%.

Example 2
9 parts of allylamine are added at 25-30 ° C. to a solution of 19 parts 2, 6, pyridinedimethanol- b, is- (phenoxyformate) in 30 parts by volume of benzene. The reaction mixture is left to stand overnight at room temperature, diluted with benzene and washed successively with a 10% strength aqueous solution of 4.5 parts of sodium hydroxide and with water. Example 3 a) Preparation of 2,6-pyridinedimethanol-bis (p-chlorophenoxyformate)
40.1 parts of p-chlorophenyl chloroformate are added dropwise to a solution of 13.9 parts of 2,6-pyridinedi-methanol in 140 parts by volume of pyridine, with stirring and cooling to a temperature below 0 C. After the addition, the reaction mixture is treated with ice water, whereupon the precipitated crystals are filtered off, washed with water and then with methanol and dried. 42 parts of 2,6-pyridinedimethanol-bis (p-chlorophenoxyformate) are obtained, which corresponds to a 94% yield.
After recrystallization from isopropanol-benzene, the product melts at 121 C. b) Production of N-substituted bis-carbamate of 2,6-pyridinedimethanol
12 parts of morpholine are added to a stirred mixture of 15 parts as in Example 3, section a), 2,6-pyridinedi-methanol-bis (p-chlorophenoxformate) and 15 parts by volume of methanol.
After the exothermic reaction has ceased, the total volume of the reaction mixture is increased to 50 parts by volume by adding methanol, and the mixture is then heated on a water bath to obtain a clear solution. The solution is allowed to cool to room temperature and the precipitated crystals are filtered off. 12.5 parts of 2.6 pyridine dimethanol-bis-N-morpholine carbamate are obtained, F = 157 to 158 C.

Ammonia gas is introduced at room temperature into a suspension of 19 parts of 2,6-pyridinedimethanol bis-phenoxyformate in 20 parts by volume of methanol.

Example 17 a) Preparation of 2,6-pyridinedimethanol-bis-carbazate
In a stirred mixture of 189.7 parts of 2,6-pyridinedi methanol-bis-phenoxyformate prepared as in Example 1, section a) and 380 parts by volume of methanol, 55.1 parts of hydrazine hydrate are added dropwise while cooling to 20-30.degree. The reaction mixture becomes a clear solution while an exothermic reaction occurs. After the hydrazine has been added, white crystals fall out of the solution. After cooling, the crystals are filtered off, washed with methanol and dried.
2,6-Pyridinedimethanol-bis-carbazate, F = 140-142 C (118.7 parts), which corresponds to a 93% yield, is obtained. b) Preparation of the 2,6w pyridine dimethanol bis-azidoformate
127.6 parts as in this example under a) prepared 2,6-pyridinedimethanol-bis-carbsazate are dissolved in a mixture of 200 parts by volume of acetic acid and 220 parts by volume of water. The solution obtained is treated with an aqueous solution of 7.6 parts of sodium nitrite for 30 minutes while stirring and cooling to -5 to 0 C. The solution is stirred for a further 30 minutes at about 0 C, after which it is diluted with water.
The precipitate obtained is filtered off, washed with water and dried under reduced pressure. 131.8 parts of 2,6-pyridine-dimethanol-bis-azidoformate, F = 42-43 ° C., are obtained, which corresponds to a 95% yield. c) Production of bis-carbamate from

The procedure of this example is repeated, but without triethylamine and using tert-butylamine instead of aniline in benzene. A spontaneous reaction occurs at room temperature, whereby
