Gegenstand der Erfindung ist ein Verfahren zur Herstellung von neuen 3-Amino-tetrahydroindazo- len der allgemeinen Formel

(thiocarbomethylamino)-l-cyclohexanon-l-cyclohexen, 7,92 g (0,055 Mol) Phenylhydrazinhydrochlorid und 70 ml 95loges Äthanol gegeben.
Es wird 6 h lang unter Rückfluss erhitzt, das Lösungsmittel unter Vakuum verdampft und der Rückstand mit Äther digeriert, um das Morpholinhydrochlorid abzutrennen. Es wird filtriert, das Filtrat zur Trockne eingedampft, 20 ml1 : l-Sa1zsäure zum Rückstand zugegeben, 30 min lang mit Tierkohle gekocht und heiss filtriert. HCIBeispiel 4 : 3-Methylamino-4, 5,6, 7-tetrahydroindazol.
In einem 200 ml-Kolben werden 27 g (0, 1125 Mol) 1-N-Morpholino-2-thiocarbomethylamino- -l-cyclohexen, 11,7 g (0, 1125 Mol) Carbäthoxyhydrazin, 100 ml absolutes Äthanol und 30 ml Eisessig gegeben. Es wird 3 h lang am Rückfluss erhitzt, unter Vakuum zur Trockne eingedampft, der Rückstand wieder mit Äther aufgenommen, filtriert und die ätherische Lösung mit ätherischer Chlorwasserstoffsäure behandelt. Es wird ein sirupöser Niederschlag gebildet, der durch Dekantieren vom Äther abgetrennt und mit Aceton erhitzt wird, wobei Verfestigung eintritt.
Es wird filtriert und aus Aceton-Äthanol um- kristallisiert, wobei 12,8 g (75go) 3-Methylamino-4,5, 6, 7 - tetrahydroindazol-hydrochlorid mit dem Schmelzpunkt 192 bis 1940C erhalten werden.

Nach dem gleichen Verfahren wird durch Reaktion von äquimolaren Mengen 1-Morpholino-6-methyl-1-cyclohexen und Carbäthoxyhydrazin das 3-Methylamino-7-methyl-4,5, 6, 7-tetrahydroindazol- - hydrochlorid mit dem Schmelzpunkt 189 bis 1910C in einer Ausbeute von 56% erhalten.
Für C9H15N3 . HCl berechnet : N % 20, 89 ; gefunden : N% 20,96. Titer HC1 =100%.
Beispiel 5: 2-Methyl-3-methylamino-4, 5,6, 7-tetrahydroindazol.
Eine Lösung von 50,6 g (1, 1 Mol) Methylhydrazin in 100 ml absolutem Äthanol wird innerhalb 30 min in 240 g einer gerührten Mischung eingetropft, die aus 1 Mol rohem 1-N-Morpholino-2-thio- carbomethylamino-1-cyclohexen, 250 ml absolutem Äthanol und 120 g (2 Mol) Essigsäure besteht. Die Temperatur wird in der ersten Stufe unter 350C gehalten. Beim Ende der Zugabe, wenn die Gasentwicklung nachlässt, wird zum Sieden erhitzt und 3 h lang unter Rückfluss gekocht. Es wird dann unter Vakuum evaporiert, mit 1, 5 l Wasser verdünnt, etwa 80 g Weinsäure zugefügt und über Nacht stehengelassen.
Dann wird aktivierte Tierkohle zugegeben und von teerigen Produkten abdekantiert. Es wird im Vakuum auf 1/3 des Volumens konzentriert. Dieses wird mit festem Kaliumcarbonat übersättigt. Die organische Schicht wird in Benzol extrahiert, getrocknet über Natriumsulfat und im Vakuum eingedampft. Der Rückstand wird bei 115 bis 125 C/0, 05 bis 0, 1 Torr gesammelt, und das Destillat wird aus Isopropyl- äther umkristallisiert. Schmelzpunkt 92 bis 93 C.
Für C9H15N3 berechnet : N% 25, 45 ; gefunden : N% 25, 12. Titer 100, 3%.
Maleat Fp. 150 bis 153 C.
Beispiel 6 : 2-Methyl-3-äthylamino-4,5, 6, 7-tetrahydroindazol.
33,5 g (0, 2Mol) 1-N-Morpholino-1-cyclohexen und 17, 4 g (0, 2 Mol) Äthylisothiocyanat werden 7 h lang auf einem Wasserbad auf 1000C erhitzt. Nach Abkühlen werden 200 ml Ligroin zugegeben, und es wird lebhaft gerührt. Der unlösliche Anteil wird absetzen gelassen. Dann wird das Ligroin abdekantiert und der Rückstand (rohes 1-N-Morpholino-2-thiocarboäthylamino-1-cyclohexan)- wieder mit 80 ml Äthanol und 14 ml Essigsäure aufgenommen und mit 8 g (0, 175 Mol) Methylhydrazin behandelt. Die Mischung wird 4 h lang am Rückfluss erhitzt. Das Lösungsmittel wird im Vakuum entfernt. Der Rückstand wird wieder mit etwa 200 ml 10% iger wässeriger Weinsäurelösung aufgenommen, eine lange Zeit mit Tierkohle gerührt und filtriert. Nach Konzentration auf das halbe Volumen wird mit überschüssigem K CO alkalisch gemacht.
Das abgeschiedene Öl wird mit Benzol extrahiert, über NaSO. getrocknet und nach Entfernen des Lösungsmittels destilliert. Siedepunkt 125 bis 130 C/0, 6 Torr.
Zur Herstellung des Maleats wird das Destillat in wenig Äthanol gelöst. Dazu wird eine konzentrierte äthanolische Lösung von 17, 4g(0,15 Mol) Maleinsäure zugegeben. Dieerhaltene Lösung führt, wenn sie mit dem gleichen Volumen wasserfreiem Äther behandelt wird, zur Ausfällung eines kristallinen Produktes, welches. nach Umkristallisation aus wenig Äthanol-Äthyläther bei 137 bis 139 C schmilzt. Die Ausbeute beträgt 38%.
Für CH. C H berechnet : N% 14,26; gefunden : N% 13, 96.
Die Titration mit Perchlorsäure ergibt : 97, 9%.
Nach dem identischen Verfahren wurden die in der Tabelle 2 aufgeführten Verbindungen hergestellt.

11,6 g (0, 076 Mol) 3-Methylamino-4,5, 6, 7 -tetrahydroindazol werden auf einem Wasserbad in 10,5 g 98% tiger Ameisensäure gelöst. Dazu werden 6 ml 38, 250/oiges wässeriges Formaldehyd zugegeben, und es wird auf dem Wasserbad 4 h lang erhitzt. Dann wird unter Vakuum evaporiert, mit Kaliumcarbonat alkalisch gemacht und mit Chloroform extrahiert. Der Extrakt wird auf Natriumsulfat getrocknet, eingedampft, und der weisse feste Rückstand wird mit Äther extrahiert. Der unlösliche Anteil wird abfiltriert. Die Lösung wird getrocknet und mit ätherischem Chlorwasserstoff gefällt. Der Niederschlag wird filtriert, mit wenig Aceton erhitzt, abkühlen gelassen unter Bildung von Kristallen und filtriert. Fp. 220 bis 234 C.
Nach weiterer Kristallisation aus Aceton schmilzt die Verbindung bei 237 bis 239 C.
Für C9l\sNs. HCI Titer 99, 6%.
Nach dem gleichen Verfahren wird aus 2-Phenyl-3-methylamino-4,5, 6, 7-tetrahydroindazol das 2-Phenyl-3-dimethylamino-4,5, 6, 7-tetrahydroindazol-hydrochlorid in 55% tiger Ausbeute erhalten.
Fp. 1320C.

Nach analogem Verfahren werden aus äquimolaren Mengen 2, 6-Di- (thiocarbomethylamino)-1-cyclohexanon und substituierten Hydrazinen die in der Tabelle 3 angeführten Verbindungen erhalten.

Beispiel 9 : 2-Phenyl-3-methylamino-7-carboxy-4, 5,6, 7-tetrahydroindazol.
Eine Mischung von 3, 85 g (0,012 Mol) 2-Phenyl-3-methylamino-7-thiocarbomethylamino- - 4, 5, 6, 7 - tetrahydroindazol, 4 ml konzentrierter Schwefelsäure, 12,5 ml Essigsäure und 8 ml Wasser wird 12 h lang am Rückfluss erhitzt. Es wird dann mit 2 Volumina Wasser verdünnt. Die Schwefelsäure wird mit Kaliumcarbonat neutralisiert und es wird bald mit Äther extrahiert. Aus der ätherischen Lösung, die einige Stunden stehengelassen worden war, scheidet sich ein kristallines Pulver ab, welches nach Kristallisation aus Wasser-Äthanol. 2, 7 g (80%) 2-Phenyl-3-methylamino-7-carboxy-4, 5, 6, 7-tetra-hydroindazol, Fp. 156 bis 158 C, ergibt.

11.6 g (0.076 mol) of 3-methylamino-4,5,6,7-tetrahydroindazole are dissolved in 10.5 g of 98% formic acid on a water bath. 6 ml of 38.250% aqueous formaldehyde are added and the mixture is heated on the water bath for 4 hours.
