Die Erfindung betrifft ein Verfahren zur Herstellung von 2- (Phenylamino) -imidazolin- (2) -deri- vaten der allgemeinen Formel

Verbindungen mit dieser Struktur finden Anwendung als antihypertensive Arzneimittel.
Zu diesem Zweck wird die Verbindung 2- (2'. 6'-Dichlorphenylamino)-imidazolin- (2). die in der Therapie unter den Namen Clonidin (Chlophasolin) bekannt ist. weitgehend verwendet.

Basische N-Phenyl-S-alkylisothiocarbamide und Äthylendiamin- - mono-p-toluolsulfonat können auch gemäss der DL-PS Nr. 68511 benutzt werden.
2- (Phenylamino)-imidazolin- (2)-derivate können noch hergestellt werden, ausgehend von : Phenylthiocarbamid und Äthylendiamin gemäss der NL-PS Nr. 6411516. der GB-PS Nr. l, 034. 938. der DE-PS Nr. 2163306 ; N-Phenyl-NI-nitroguanidinen mit Äthylendiamin gemäss den BG-PS Nr. 12390 und Nr. 12393. Phenylguanidin mit Äthylendiamin in Form einer Base oder als Mono-p-toluolsulfonat gemäss der DL-PS Nr. 68510. FR-PS Nr. 1. 577. 129. GB-PS Nr. 1. 230. 583 ; Phenylisocyanid-dihalogeniden mit Äthylendiamin gemäss der DL-PS Nr. 71554. FR-PS Nr. 1. 566. 036 und Nr. 1. 577. 128. GB-PS Nr. 1, 229. 993, SA-PS Nr. 6706503 ; Phenylcyanamiden mit Monosalzen des Äthylendiamin. wie es in der DL-PS Nr. 68509 und in der GB-PS Nr. 1. 230. 482 beschrieben ist.
Es werden verschiedene 2- (Phenylamino) -imidazolin- (2) -derivate durch Einführung von Substituenten in den aromatischen Kern gemäss der FR-PS Nr. 1. 566. 035 und Nr. 1. 566. 036 hergestellt ; durch Reaktion der Aniline mit Bis- (2-oxo-l-imidazolinyl)-phosphinchlorid gemäss der GB-PS Nr. 12. 391. Nr. 12. 394 oder mit l-Acylimidazolin-2-on-Derivaten gemäss einem noch im unveröffentlichten Vorschlag. Es sind auch andere Varianten der angeführten bekannten Verfahren vorhanden.
Ein Nachteil von manchen bekannten Verfahren sind die unzureichenden Ausbeuten. In andern Fällen sind die nicht stabilen und einer Reinigung schwer zu unterziehenden Zwischenprodukte ein Nachteil, wie z. B. die Cyanamide. Eine schwache Seite der bekannten Verfahren sind ebenso die Schwierigkeiten mit denen die Isolierung und Reinigung der 2- (Phenylamino)- - imidazolin- (2)-derivate verbunden sind.
Der Erfindung liegt die Aufgabe zugrunde, eine neue einfachere Methode für Synthese von 2- (Phenylamino)-imidazolin- (2)-derivaten aus leicht zugänglichen Ausgangsprodukten, die leicht einer Reinigung unterzogen werden können, zu erarbeiten, wodurch die Herstellung der gewünschten Stoffe mit einer guten Ausbeute und einem hohen Reinheitsgrad gesichert wird.
Erfindungsgemäss werden die 2- (Phenylamino)-imidazolin- (2)-derivate und deren Salze der allgemeinen Formel (I), in der R, und Ru die Stellungen 2 bis 4 im Phenylkern einnehmen und unabhängig voneinander Wasserstoff, Halogen oder eine Nitrogruppe bedeuten, synthetisiert, indem Verbindungen der allgemeinen Formel (II), in der R, und R die oben angeführten Bedeutungen aufweisen, R 3 einen niedrigen Alkyl-oder Phenylrest bedeutet, und R. ein Alkylrest mit 1 bis 6 Kohlenstoffatomen sein kann, mit Äthylendiamin-mono-p-toluolsulfonat in Anwesenheit eines Lösungsmittels bei Temperaturen von 100 bis 200 C umgesetzt werden.

Ausgezeichnete Ergebnisse können gemäss der Erfindung erreicht werden, wenn Äthylendiamin- - mono-p-toluolsulfonat mit N-Phenyl-N'-acyl-S-alkylisothiocarbamid in Molarverhältnissen grösser als 2 : 1 kondensiert wird.
Eine andere vorgezogene Ausführung der Erfindung erfordert, dass der Ringschluss in einem Medium von Alkohol oder Alkoholgemischen, deren Gerüst 5 bis 8 Kohlenstoffatome enthält und die in einem Temperaturbereich von 130 bis 180 C sieden, unter Erhitzung von einigen Stunden erfolgt.
Gemäss der Erfindung wird die Synthese der 2- (Phenylamino)-imidazolin- (2)-derivate auf eine neue und viel einfachere Weise verwirklicht, dank der neu entdeckten Reaktion. Auf Grund der bekannten chemischen Vorgänge, die die Bildung der Imidazolin-derivate durch Ringschluss von N-Phenyl-S-alkylisothioronium Salze mit Äthylendiamin oder Äthylendiamin-mono-p-toluolsulfonat und durch Eliminierung des Alkylmercaptans und Ammoniaks beschreiben, war es keinesfalls zu erwarten, dass auch eine Ringbildung der N-Phenyl-N'-acyl-S-alkylisothiocarbamide unter der Wirkung des Äthylendiamin-mono-p-toluolsulfonats möglich ist. weil der Verlauf der letzten Umsetzung unter Eliminierung von grösseren Molekülen, wie z. B. der Amide. Acetamid.
Benzamid u. a.. wenig möglich erscheint, obwohl er in Wirklichkeit so verläuft.

(Phenylamino) -imidazolin- ( 2) -derivateamide-können gemäss den folgenden Beispielen 9 bis 24 hergestellt werden.
Dieser Vorteil ist insbesondere beim Vergleich des erfindungsgemässen Verfahrens mit der Synthese des 2- (2', 6'-Dichlorphenylamino)-imidazolins auf bekannte Weise aus Äthylendiamin oder Äthylendiamin-mono-p-toluolsulfonat und 2, 6-Dichlorphenyl-S-methyl-isothiuroniumsalz oder 2. 6-Dichlorphenylcyanamid ersichtlich. Beide Zwischenprodukte werden in drei Stufen aus 2. 6-Dichloranilin hergestellt, während letzteres in das gewünschte N- (2. 6-Dichlorphenylamino) -NI-acyl- - S-alkylisothiocarbamid in nur zwei Stufen leicht umgesetzt werden kann.
Ausserdem erlaubt es das erfindungsgemässe Verfahren, eine Ausbeute an reine 2- (2', 6'-Di- chlorphenylamino)-imidazolin- (2)-Base von 74, 4% der Theorie zu realisieren. Diese angeführte Ausbeute steht den besten Resultaten von andern Verfahren nicht nach : z. B. wird 2- (2', 6'-Dichlor- phenylamino)-imidazolin- (2)-Base gemäss DL-PS Nr. 68511, Beispiel 1 mit einer Ausbeute von 69% erhalten. Gemäss DL-PS Nr. 68509 wird oben genannte Base mit einer Ausbeute von 71% der Theorie erhalten.
An Hand der nachstehend angeführten Beispiele soll die Erfindung näher erläutert werden.

: 2- (2'. 6'-Dichlorphenylamino)-imidazolin- (2)-hydrochloridÄthylendiamin-mono-p-toluolsulfonat werden in 280 ml Isoamylalkohol 20 h am Rückfluss erwärmt und dann eine Nacht bei Zimmertemperatur stehengelassen. Der ausgefallene Niederschlag von Äthylendiamin-mono- ul1d -di-p-toluolsulfonat wird abfiltriert. Das Filtrat wird im Vakuum bis zu einem ölartigen Rückstand konzentriert. Eine Lösung aus 12. 7 g Natriumcarbonat in 140 ml Wasser wird zu dem Rückstand zugegeben. Das Gemisch wird 1 h gerührt und dann eine Nacht stehengelassen. Der ausgefallene Niederschlag wird abfiltriert und zweimal mit einer Natriumcarbonatlösung und danach mit Wasser gewaschen.
Das Produkt wird getrocknet, und man erhält 17, 1 g 2- (2', 6'-Dichlorphenylamino)-imidazolin- (2) mit einem Schmelzpunkt von 134 bis 139 C.
Die Ausbeute beträgt 74, 4% der Theorie.
Zu 17. 1 g von dem erhaltenen Produkt werden 75 ml Wasser. 7. 5 ml konzentrierte Salzsäure und 2 g aktive Kohle zugegeben. Das Reaktionsgemisch wird auf einem Wasserbad erwärmt und danach filtriert. Aus dem wässerigen Filtrat fallen 12, 4 g (Ausbeute : 62. 6% gegenüber der Base)

Beispiel 2 : 2- (2', 6'-Dichlorphenylamino)-imidazolin- (2)-hydrochlorid

Äthylendiamin-mono-p-toluolsulfonat werden am Rückfluss in 20 ml Benzyl-Alkohol erwärmt. Das Reaktionsgemisch wird mit 20 ml gesättigter wässeriger Natriumcarbonatlösung gewaschen. Beide Schichten werden in einem Scheidetrichter abgetrennt und die Alkoholschicht wird noch zweimal mit je 20 ml Wasser gewaschen. wonach sie mit Natriumsulfat (entwässertes) getrocknet wird.
Aus der getrockneten Lösung werden mit Chlorwasserstofflösung in Äthylacetat 1, 55 g 2- (2'. 6'-Di- chlorphenylamino)-imidazolin- (2)-hydrochlorid mit einem Schmelzpunkt von 303 bis 3060C ausge- scheden. Die Ausbeute beträgt 58. 3% der Theorie.
Beispiel 3 : 2- (2', 6'-Dichlorphenylamino)-imidazolin- (2)
2, 8 g (0. 01 Mol) N- (2. 6-Dichlorphenyl) -N'-acetyl-S-methylisothiocarbamid. 7. 0 g (0, 03 Mol) Äthylendiamin-mono-p-toluolsulfonat und 25 ml Cyclohexanol werden 5 h am Rückfluss erwärmt.
Das Lösungsmittel wird im Vakuum abdestilliert und der Rückstand wird mit einer wässerigen Natriumcarbonatlösung verrührt. Das erhaltene ölartige Produkt wird in Äthylacetat aufgelöst und eine Lösung von trockenem Chlorwasserstoff in Äthylacetat wird zugegeben. Der erhaltene Niederschlag wird abfiltriert und mit Aceton gewaschen. Man erhält 1. 3 g 2- (2'. 6'-Dichlorphenyl- amino)-imidazolin- (2)-hydrochlorid, welches in Wasser aufgelöst wird und die wässerige Lösung wird mit gesättigter wässeriger Natriumcarbonatlösung alkalisiert. Das ausgeschiedene Produkt wird abfiltriert.
Nach dem Trocknen erhält man 0, 8 g 2- (2. 6-Dichlorphenylamino)-imidazolin- (2)

Nachdem das Lösungsmittel abdestilliert wird, wird der Rückstand mit einer wässerigen Natriumcarbonatlösung vermischt. Der ausgeschiedene Niederschlag wird filtriert und mit Wasser gewaschen. Nach dem Trocknen erhält man 1. 4 g 2- (2', 6'-Dichlorphenylamino)-imidazolin- (2) vom Schmp. 138 bis 141 C. Die Ausbeute beträgt 61% der Theorie.
Beispiel 5 : 2- (2 1, 6' -Dichlorphenylamino) -imidazolin- (2) -hydrochlorid 3, 4 g (0, 01 Mol) N- (2, 6-Dichlorphenyl)-NI-benzoyl-S-methylisothiocarbamid, 7, 0 g (0, 03 Mol) Äthylendiamin-mono-p-toluolsulfonat und 40 ml n-Amylalkohol werden 15 h am Rückfluss erwärmt.
Die Reaktionslösung wird zweimal mit je 50 ml Wasser gewaschen. Das Lösemittel wird abdestilliert. Der Rückstand wird zweimal mit je 20 ml gesättigter wässeriger Natriumcarbonatlösung gewaschen. Zu dem auf diese Weise gewaschenen Rückstand wird Salzsäure (1 : 1) zugegeben und unter leichtem Erwärmen aufgelöst. Die erhaltene Lösung wird im Vakuum bis zur Trockensubstanz konzentriert. Das erhaltene Produkt wird mit Aceton gewaschen und filtriert. Nach dem Trocknen erhält man 0, 9 g 2- (21, 61-Dichlorphenylamino) -imidazolin- (2) -hydrochlorid.
Beispiel 6 : 2- (21, 6'-Dichlorphenylamino) -imidazolin- (. 2) -hydrochlorid 2, 9 g (0, 01 Mol) N- (2, 6-Dichlorphenyl)-NI-propionyl-S-methylisothiocarbamid, 7, 0 g (0, 03 Mol) Äthylendiamin-mono-p-toluolsulfonat und 40 ml n-Amylalkohol werden 75 h am Rückfluss erwärmt.
Mit dem Reaktionsgemisch wird wie im Beispiel 5 angegeben. verfahren. Es werden 1. 2 g 2- (2', 6'-Dichlorphenylamino)-imidazolin- (2)-hydrochlorid isoliert.
Beispiel 7 : 2- (2', 5'-Dichlorphenylamino)-imidazolin- (2) 2, 8 g (0, 01 Mol) N- (2. 5-Dichlorphenyl) -N'-acetyl-S-methylisothiocarbamid, 7, 0 g (0, 03 Mol) Äthylendiamin-mono-p-toluolsulfonat und 30 ml n-Amylalkohol werden 5 h am Rückfluss erwärmt.
Die Reaktionslösung wird mit 50 ml gesättigter Natriumcarbonatlösung und mit 50 ml Wasser gewaschen. Der Amylalkohol wird im Vakuum bis zu einem trockenen Rückstand abdestilliert. welcher nach Umkristallisieren aus Äthlacetat 1,5 g 2-(2',5'-Dichlorphenylamino)-imidazolin-(2) mit einem Schmelzpunkt von 179 bis 182 C ergibt. Die Ausbeute beträgt 65% der Theorie. Das Jodwasserstoff-Salz der oben angeführten Verbindung weist einen Schmelzpunkt von 275 bis 2770C auf. Gemäss der US-PS Nr. 2. 899, 426 ist der Schmelzpunkt vom 2- (2', 5'-Dichlorphenylamino)- - imidazolin- (2)-hydrochlorid 275. 4 bis 277 C.
Beispiel 8 : 2- (0-Nitrophenylamino)-imidazolin- (2) 2. 5 g (0,01 Mol) N-(O-Nitrophenyl)-N'-acetyl-S-methylisothiocarbamid, 7,0 g (0, 03 Mol) Äthylendiamin-mono-p-toluolsulfonat und 30 ml n-Amylalkohol werden 6 h am Rückfluss erwärmt.
Die Lösung wird mit 50 ml gesättigter wässeriger Natriumcarbonatlösung und 50 ml Wasser gewaschen. Der Amylalkohol wird im Vakuum abdestilliert. Der Rückstand wird aus Äthanol umkristallisiert. Man erhält 0. 8 g 2- (0-Nitrophenylamino)-imidazolin mit einem Schmelzpunkt von 162 bis 166 C. Die Ausbeute beträgt 38, 8% der Theorie.
Die folgenden Beispiele veranschaulichen die Herstellung von neuen Ausgangsprodukten :
Beispiel 9 : N-(2,6-Dichlorphenyl)-N'-acetyl-S-methylisothiocarbamid
26, 3 g (0. 1 Mol) N-(2,6-Dichlorphenyl)-N'-acetylthiocarbamid, 9,9 g (0.06 Mol) gemahlenes 2 K. COs. 3 Haro, 8,3 ml (0. 13 Mol) Methyljodid und 200 ml Aceton werden unter Rückfluss 5 h erwärmt und energisch gerührt. Das Aceton wird abdestilliert und der Rückstand mit Wasser gewaschen und filtriert. Der Niederschlag auf dem Filter wird noch ein-bis zweimal mit Wasser gewaschen. Nach dem Trocknen wird das Produkt aus Isopropanol 1 : 6 (Masse : Volumen) umkristallisiert. Man erhält 22, 0 g N- (2, 6-Dichlorphenyl)-N'-acetyl-S-methylisothiocarbamid isoliert.
Ausbeute 79, 4% der Theorie, Fp. 137 bis 140'C.

Das Aceton wird abdestilliert. Der Rückstand wird mit Wasser gewaschen und danach filtriert. Es werden 6, 8 g (80. 5% der Theorie) N- (2. 6-Dichlorphenyl) - - N'-acetyl-S-äthylisothiocarbamid mit einem Fp. 97 bis 1000C erhalten. Nach Umkristallisieren aus Äthylalkohol wird ein Produkt vom Fp. 99 bis 1020C erhalten. Die Ausbeute beträgt 56% der Theorie.

Beispiel 16: N-(2,6-Dichlorphenyl)-N'-propionyl-S-methylisothiocarbamid
33, 2 g (0,12 Mol) N-(2,6-Dichlorphenyl)-N'-propionylthiocarbamid, 10,5 g (0.08 Mol) gemahlenes Kaliumkarbonat, 9. 6 ml (0, 15 Mol) Methyljodid und 300 ml Aceton werden 4 h unter Rückfluss erwärmt und energisch gerührt. Nach Ablauf der oben erwähnten Reaktionszeit wird das Aceton abdestilliert. Zum Rückstand werden 300 ml Wasser zugesetzt und verrührt. Danach wird der Niederschlag filtriert und getrocknet.
Es werden 34, 4 g (98, 5% der Theorie) N- (2, 6-Dichlor- phenyl)-NI-propionyl-S-methylisothiocarbamid vom Fp. 165 bis 170 C erhalten. Nach Umkristallisieren aus Äthylalkohol werden 23. 5 g Produkt (68,2% des Rohproduktes) vom Fp. 173 bis 1760C gewonnen.
