Verfahren zur Herstellung von neuen N'-substituierten N-Arylsulfonylharnstoffen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer N-Arylsulfonylharnstoffe.
Verbindungen der allgemeinen Formel I,

in welcher R1 Wasserstoff, Halogen bis Atomnummer 35, eine niedere Alkyl-, Alkoxy-, Alkylthio- oder Alkanoylgruppe oder die Aminogruppe, R2 Wasserstoff oder R1R2 die Trimethylen- oder die Tetramethylengruppe bedeutet, sowie ihre Salze mit anorganischen oder organischen Basen sind bisher nicht bekannt geworden.
Wie nun gefunden wurde, besitzen die neuen Verbindungen sowie ihre pharmazeutisch annehmbaren Salze wertvolle pharmakologische Eigenschaften. Sie zeigen überraschenderweise bei peroraler oder parenteraler Verabreichung hypoglykämische Wirkung. Die neuen Verbindurngen eignen sich zu Behandlung der Zuckerkrankheit.
In den Verbindungen der allgemeinen Formel I kann Rl die o-, oder p-Stellung einnehmen und beispielsweise folgende Bedeutungen haben: als niedere Alkylgruppe die Methyl-, itthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, sek.Butyl-, tert.Butyl-, Pentyl-, Isopentyl- oder 2,2-Dimethyl-propylgruppe;
als niedere Alkoxygruppe die Methoxy-, Äthoxy-, Propoxy-, Isopropoxy-, Butoxy-, Isobutoxy-, sek-Butoxy-, tert.Butoxy-, Pentoxy-, Isopentoxy- sowie die 2,2-Dimethyl- propoxygruppe; als niedere Alkylthiogruppe die Methylthio-, iithylthio-, Propylthio-, Isopropylthio-, die Butylthio-, Isobutylthio-, sek.Butylthio-, tert.Butylthio-, Pentylthio-, Isopentylthio- sowie die 2,2-Dimethyl-propylthiogruppe und als niedere Alkanoylgruppe die Acetyl-, Propionyl-, 2-Methyl-propionyl-, Butyryl-, 2-Methyl-butyryl- sowie die 3-Methyl-butyrylgruppe.
Nach dem erfindungsgemässen Verfahren stellt man Verbindungen der allgemeinen Formel I her, indem man eine Verbindung der allgemeinen Formel II,

X bedeutet als Halogenatom vorzugsweise das Chloratom, als niedere Alkoxy oder Alkylthiogruppe z. B. die Methoxy-, Athoxy- bzw. die Methylthio- oder A ; thylthiogruppe.
Falls X ein Halogenatom bedeutet, wird die Hydrolyse zweckmässig mit Hilfe einer anorganischen Base, z. B. mit Natriumhydroxid vorgenommen und falls X eine niedere Alkoxy- oder Alkylthiogruppe bedeutet, verwendet man anstelle der anorganischen Base eine Halogenwasserstoffsäure, z. B. Salzsäure. Die Reaktion wird zweckmässig in einem organischen Lösungsmittel durchgeführt, das sich mit Wasser mischt. Solche Lösungsmittel sind z. B. Ketone, wie Aceton oder Methyläthylketon, ätherartige Flüssigkeiten, wie Dioxan oder Tetrahydrofuran und ferner Carbonsäureamide, wie N,N-Dimethylfermamid.
Die anschliessende Umwandlung einer Gruppe R1, des Reaktionsproduktes in die freie Aminogruppe, welche dieses in eine Verbindung der allgemeinen Formel I überführt, wird je nach der Art der Gruppe R1, durch eine Hydrolyse, Reduktion oder reduktive Spaltung vorgenommen.
Ist der Substituent R,' durch Hydrolyse in die Aminogruppe überführbar, so kann diese Hydrolyse auch gleichzeitig, wie die erfindungsgemässe Umsetzung erfolgen.
Durch Hydrolyse in die freie Amingruppe überführbare Reste R1, sind beispielsweise Acylaminoreste, wie z. B. die Acetamidogruppe, oder Alkoxy- oder Phenoxycarbonylaminoreste, wie z. B. die Äthoxycarbonylamino- oder Phenoxycarbonylaminogruppe. Weitere Beispiele sind substituierte Methylenaminoreste, wie z. B. die Benzylidenamino- oder die p-Dimethylamino benzylidenammogruppe. Die Hydrolyse zur Freisetzung der Aminogruppe kann z. B. in saurem Medium, wie durch Erhitzen in verdünnter methanolischer Salzsäure, oder, falls R1, einen Alkoxy- oder Phenoxycarbonylaminorest bedeutet, auch unter milden alkalischen Bedingungen, z. B. mittels l-n. bis 2-n. Natronlauge, bei Raumtemperatur erfolgen.
Ein Beispiel für einen durch Reduktion in die Aminogruppe überführbaren Rest R1, ist die Nitrogruppe und Beispiele für solche Reste, die durch reduktive Spaltung zur Aminogruppe führen, sind die Phenylazooder p-Dimethylamino-phenylazogruppen. Die Reduktion dieser Reste kann allgemein katalytisch, z. B. mittels Wasserstoff in Gegenwart von Raney-Nickel, Palladium- oder Platin-Kohle, in einem inerten Lösungsmittel, wie z. B. Äthanol, erfolgen. Neben diesen kommen auch andere übliche Reduktionsverfahren in Betracht, beispielsweise die Reduktion von Nitrogruppen oder die reduktive Spaltung von Azogruppen mit Hilfe von Eisen in Essigsäure oder Salzsäure.
Ausgangsstoffe der allgemeinen Formel II sind z. B. solche Verbindungen, deren Substituenten R1, und R2 mit den Gruppen übereinstimmen, die anschliessend an Formel I oder II für die Reste R1, R2 oder RXR2 bzw. R1,,R2 oder R1,R2 aufgezählt sind.
Eine solche Gruppe von Ausgangsstoffen sind z. B. durch R1, und R2 im Benzolring substituierte N-Phenylsulfonyl-decahydro1H-3-benzazepin-3-carboximidoylchloride.
Diese werden z. B. aus Phosgen und aus entsprechend substituierten N-Phenylsulfonyldecahydro-1H3-benzazepin-3-thiocarboxamiden erhalten, die Ausgangsstoffe des zweiten Verfahrens sind. Die Umsetzung kann z. B. in Tetrahydrofuran vorgenommen werden. Die erhaltenen Reaktionsprodukte können in eine weitere Gruppe von Ausgangsstoffen der allgemeinen Formel II, deren Rest X eine niedere Alkylthiogruppe bedeutet, übergeführt werden, wenn man sie z. B. mit dem Natriumsalz eines niederen Alkanthiols, wie z. B. des Methanthiols umsetzt.
Diese zweite Gruppe von Ausgangsstoffen sind N-Phenylsulfonyl-decahydro
1H-3-benzazepin-3-thiocarboximid säure-niederalkylester z. B. N-Phenylsulfonyl-decahydro-1H
3-benzazepin-3-thiocarboximidsäure methylester, die in den Benzolkernen durch die Reste R1, und R2 substituiert sind. Entsprechende N-Phenylsulfonyl-decahydro-lH- 3-benzazepin-3 -carboximidsäurealkylester-Verbindungen der allgemeinen Formel II, in welcher X eine niedere Alkoxygruppe bedeutet, werden z. B. erhalten, wenn man anstelle eines Natriumsalzes eines niederen Alkanthiols ein Natriumsalz eines niederen Alkanols als Reaktionskomponente in die Reaktion einsetzt.
Dieselben Ausgangsstoffe können ferner auch ausgehend von Decahydro-lH-3-benzazepin hergestellt werden. Dieses wird z. B. mit Bromcyan in das Deca hydro-1H-3 -benzazepin-3-carbonitril überführt, welches in einer Lösung von Chlorwasserstoff in einem niederen Alkanol in einen 2-Niederalkyl-3-(decahydro-1H3-benzazepin-3-yl)-pseudoharnstoff umgewandelt wird. Dieser Pseudoharnstoff kann z. B. mit einem Benzolsulfochlorid, das durch die Reste R1, und R2 substituiert ist, unter Chlorwasserstoffabspaltung kondensiert werden.
Die neuen Wirkstoffe oder die pharmazeutisch annehmbaren Salze derselben werden vorzugsweise peroral verabreicht. Zur Salzbildung können anorganische oder organische Basen, wie beispielsweise Alkali- oder Erdalkalihydroxide, Carbonate oder Bicarbonate, Triäthanolamin, Cholin, N1-Dimethyl- oder N1--Phe- nyläthyl)-biguanid, verwendet werden. Die täglichen Dosen bewegen sich zwischen 50 und 1000 mg für er wachsen Patienten. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, enthalten vorzugsweise 25-500mg eines erfindungsgemässen Wirkstoffes, und zwar 20 bis 80 O/o einer Verbindung der allgemeinen Formel I. Zu ihrer Herstellung kombiniert man den Wirkstoff z.
B. mit festen pulverförmigen Trägerstoffen, wie Lactose, Saccharose, Sorbit, Mannit; Stärken, wie Kartoffelstärke, Maisstärke oder Amylopektin, ferner Laminariapulver oder Citruspulpenpulver; Cellulosederivaten oder Gelatine, gegebenenfalls unter Zusatz von Gleitmitteln, wie Magnesium-oder Calciumstearat oder Polyäthylenglykolen zu Tabletten oder zu Dragee Kernen. Letztere überzieht man beispielsweise mit konzentrierten Zuckerlösungen, welche z. B. noch arabischen Gummi, Talk und/oder Titandioxid enthalten, oder mit einem in leichtflüchtigen organischen Lösungsmitteln oder Lösungsmittelgemischen gelösten Lack. Diesen Überzügen können Farbstoffe zugefügt werden, z. B. zur Kennzeichnung verschiedener Wirk stoffdosen.
Die folgenden Vorschriften sollen die Herstellung von Tabletten und Dragees näher erläutern: a)1000g N-(p-Tolylsulfonyl)-decahydro
1H-3-benzazepin-3-carboxamid werden mit 550 g Lactose und 292 g Kartoffelstärke vermischt, die Mischung mit einer wässerigen Lösung von 8,0 g Gelatine befeuchtet und durch ein Sieb gra nuliert.
Nach dem Trocknen mischt man 60,0 g Kartoffelstärke, 60,0 g Talk, 10,0 g Magnesiumstearat und 20,0 g kolloidales Siliciumdioxid zu und presst die
Mischung zu 10 000 Tabletten von je 200 mg Gewicht und 100 mg Wirkstoffgehalt, die gewünschtenfalls mit
Teilkerben zur feineren Anpassung der Dosierung ver sehen sein können. b) Aus 1000 g N-(p-Chlor-phenylsulfonyl)-decahydro1H-3-benzazepin-3-carboxamid, 379 g Lactose und der wässerigen Lösung von 6,0 g Gelatine stellt man ein Granulat her, das man nach dem Trocknen mit 10,0 g kolloidalem Siliciumdioxid, 40,0 g Talk, 60,0 g Kartoffelstärke und 5,0 g Magnesiumstearat mischt und zu 10 000 Dragee-Kernen presst.
Diese werden anschliessend mit einem konzentrierten Sirup aus 533,5 g krist. Saccharose 20,0g Schellack, 75,0 g arabischem Gummi, 250 g Talk, 20 g kolloidalem Siliciumdioxid und 1,5 g Farbstoff überzogen und getrocknet. Die erhaltenen Dragees wiegen je 240 mg und enthalten je 100 mg Wirkstoff.
Das nachfolgende Beispiel erläutert die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellt jedoch keineswegs die einzige Ausführungsform derselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1 a) 36,9 g N-(p-Tolylsulf onyl)-decahydro lH-3-benzazepin- 3-carboximidoylchlorid werden in 200 ml Dioxan gelöst und mit 200 ml 2-n.
Natronlauge eine Stunde auf dem Wasserbad erwärmt.
Nach dem Abkühlen wird das Reaktionsgemisch mit 2-n. Salzsäure angesäuert. Die ausgefallenen Kristalle werden aus Essigsäureäthylester umkristallisiert. Das reine N-(p-Tolylsulfonyl)decahydro-1H-3-benzazepin3-carboxamid schmilzt bei 167,5-1690.
Der Ausgangsstoff, das N-(p-Tolylsulfonyl)-decahydro-1H-3benzazepin-3-carboximidoylchlorid, wird wie folgt hergestellt: b) In eine Lösung von 36,7 g N-(p-Tolylsulfonyl)-decahydro1H-3-benzazepin-3-thiocarboxamid in 500 ml abs. Tetrahydrofuran wird während 5 Stunden Phosgen eingeleitet. Die Reaktion ist schwach exotherm. Hierauf wird überschüssiges Phosgen mit Stickstoff verdrängt und die Lösung unter Vakuum eingedampft. Man kristallisiert den Rückstand aus Essigsäureäthylester um, wonach man das reine N-(p-Tolylsulfonyl)-decahydro lH-3-benzazepin-3- carboximidoylchlorid vom Smp. 122-124 erhält.

Radicals R1 which can be converted into the free amine group by hydrolysis are, for example, acylamino radicals, such as. B. the acetamido group, or alkoxy or phenoxycarbonylamino radicals, such as. Starting materials of the general formula II are, for. B. those compounds whose substituents R1 and R2 match the groups which are listed after formula I or II for the radicals R1, R2 or RXR2 or R1, R2 or R1, R2.
Such a group of starting materials are z. B. N-phenylsulfonyl-decahydro1H-3-benzazepine-3-carboximidoylchloride substituted by R1 and R2 in the benzene ring.
These are z. B. obtained from phosgene and from appropriately substituted N-phenylsulfonyldecahydro-1H3-benzazepine-3-thiocarboxamides, which are starting materials for the second process. The implementation can e.g. B. be made in tetrahydrofuran. The reaction products obtained can be converted into a further group of starting materials of the general formula II, the radical X of which is a lower alkylthio group, if they are, for. B. with the sodium salt of a lower alkanethiol, such as. B. the methanethiol implemented.
This second group of starting materials is N-phenylsulfonyl-decahydro

The same starting materials can also be produced starting from decahydro-1H-3-benzazepine. This is z. B. with cyanogen bromide in the decahydro-1H-3-benzazepine-3-carbonitrile, which in a solution of hydrogen chloride in a lower alkanol into a 2-lower alkyl-3- (decahydro-1H3-benzazepin-3-yl) - pseudourea is converted. This pseudourea can e.g. B. with a benzenesulfonyl chloride, which is substituted by the radicals R1 and R2, be condensed with elimination of hydrogen chloride.
The new active ingredients or the pharmaceutically acceptable salts thereof are preferably administered orally. The following example explains in more detail the preparation of the new compounds of general formula I and of intermediates not previously described, but is by no means the only embodiment of the same. The temperatures are given in degrees Celsius.
Example 1 a) 36.9 g of N- (p-tolylsulfonyl) -decahydro lH-3-benzazepine-3-carboximidoyl chloride are dissolved in 200 ml of dioxane and mixed with 200 ml of 2-n.
