Verfahren zur Herstellung neuer Derivate von Dihydro-D-lysergsäure- und 1-Methyl-dihydro-D-lysergsäure-a-hydroxyäthylamid Die vorliegende Erfindung bezieht sich auf ein Ver fahren zur Herstellung neuer Derivate von Dihydro-D lysergsäure-a-hydroxyäthylamid der Formel:

worin R' den ss-Carbohydrazid-ss-carbobenzoxyamino äthylrest; einen S- oder 6gliedrigen Zykloalkylrest oder einen Alkylrest mit 1-6 Kohlenstoffatomen, der durch eine Alkoxy-, Halogen-, Aryl-, Amino-, Acylamino- gruppe substituiert ist, wobei die Acylgruppe den Rest einer aliphatischen organischen Säure mit 1-9 Kohlen stoffatomen, den Rest der Benzyloxycarbonsäure, oder den Rest einer substituierten oder unsubstituierten Ben zoesäure, Nikotin- und Isonikotinsäure darstellt, bedeuten.
Im Hauptpatent sind neue Amidderivate der Di- hydro-D-lysergsäure der obenangeführten Formel be schrieben, worin R' Wasserstoff oder einen Alkylrest mit 1 bis 6 Kohlenstoffatomen bedeutet, die auf das Zentralnervensystem wirken und eine hypotensive, ge- fässerweiternde, antienteraminische Wirkung aufweisen.
Es wurde nun gefunden, dass auch die erfindungs- gemäss hergestellten Derivate auf das Zentralnerven system wirken und hypotensive, gefässerweiternde, anti- enteraminische Wirkung aufweisen und nur wenig toxisch sind.
Das erfindungsgemässe Verfahren, das im folgenden Reaktionsschema dargestellt wird, ist nun da durch gekennzeichnet, dass D-Lysergsäure-a-hydroxy- äthylamid (I) in Gegenwart eines Hydrierungskatalysa- tors hydriert wird und das so erhaltene Dihydro-D- lysergsäure-a-hydroxyäthylamid (II) mit einer Verbin dung der Formel R'OH in Anwesenheit einer organi schen Säure veräthert wird, worauf die so erhaltenen Stereoisomeren des O-substituierten Dihydro-D-lyserg säure-a-hydroxyäthylamids (IV) durch fraktionierte Kristallisation getrennt werden.

Das Verfahren kann beispielsweise wie folgt durch geführt werden: Die katalytische Hydrierung des D- Lysergsäure-a-hydroxyäthylamids I erfolgt in saurer wässeriger Lösung in Gegenwart von Platinoxyd oder Palladium auf Kohle.
Man führt die Verätherung in Gegenwart einer Säure, vorzugsweise einer organischen Säure, wie Ma leinsäure, Oxalsäure oder Weinsäure, bei einer Tempe ratur zwischen 50 und 120 C während eines Zeitraumes von 30 Minuten bis 4 Stunden, gegebenenfalls in An wesenheit von Lösungsmitteln, aus.
Die Lösung wird dann im Vakuum zur Trockene eingedampft, der Rückstand in einer leicht alkalischen wässerigen Lösung aufgeschlämmt und mit einem orga nischen Lösungsmittel, wie Chloroform, Methylenchlo- rid oder Butanol, extrahiert. Der so erhaltene Auszug enthält zwei Stereoisomere des O-substituierten Derivats des Dihydro-D-lysergsäure-a-hydroxyäthylamids (III), die Stereoisomere A und B bezeichnet werden, wo bei das Stereoisomer A weniger löslich und das Ste- reoisomer B besser löslich in organischen Lösungs mitteln ist.
Die zwei Stereoisomere können durch Kon zentrieren voneinander abgetrennt werden; durch Kon zentrieren der Auszüge bis auf 1/10 des ursprünglichen Volumens fällt das Stereoisomer A und nach weite rem Konzentrieren das Stereoisomer B aus.
Durch Methylieren der erhaltenen O-substituierten Derivate von Dihydro-D-lysergsäure-a-hydroxyäthyl amid (III) in denen R Wasserstoff ist, erhält man die entsprechenden 1-methyl-substituierten Verbindungen.
Diese Methylierung wird vorzugsweise durch Um setzung der Verbindung III mit Methyljodid in flüssi gem Ammoniak und in Gegenwart von metallischem Kalium durchgeführt.
Das so erhaltene 1-Methylderivat kann hierauf mit einem organischen Lösungsmittel, wie Chloroform oder Methylchlorid, extrahiert, durch Eindampfen des Aus zuges isoliert und durch Umkristallisieren des Rück standes gereinigt werden.
Nach dem Verfahren herstellbare O-substituierte Derivate von Dihydro-D-lysergsäure- und 1-Methyl-di- hydro-D-lysergsäure-a-hydroxyäthylamid sind z. B. die O-Hydroxypropyl-, O-Hydroxybutyl-, O-Methoxyäthyl-, O-Methoxypropyl-, O-Methoxybutyl-, O-Chloräthyl-, O-Chlorpropyl-, O-Chlorbutyl-, O-Bromoäthyl-, O- Aminoäthyl-, O-Phenyläthyl-, O-Phenylbutyl-, O-Zyklo- pentyl-, O-Zyklohexyl-, O-Formylaminoäthyl-, O-For- mylaminopropyl-, O-Acetamidoäthyl-, O-Acetamido- butyl-, O-Propionamidoäthyl-, O-Benzamidoäthyl-,
O- Benzamidopropyl-, O-Carbobenzoxyaminoäthyl-, O-Ni- kotinoylaminoäthyl-, O-Benzyl- und O-(N-Carbobenz- oxy-L-alanyl-hydrazid)-Derivate. Die Produkte können in Form von Kristallpulver erhalten werden und sind in den üblichen organischen Lösungsmitteln löslich. Sie weisen eine gute antienteraminische, hypotensive und gefässerweiternde Wirksamkeit auf, wobei sie auf das Zentralnervensystem einwirken, und besitzen eine nied rige Toxizität.
Beim Menschen können sie entweder oral, intra muskulär, subkutan oder intravenös verabreicht werden. Die Produkte sind besonders bei allen Formen von Hypertension, Herzkrankheiten, Blutgefässstörungen, Tachykardie, Migräne, Kopfschmerzen nützlich.
Sie können auch in der Psychiatrie als antidepressi ves, beruhigendes und für das psychische Gleichgewicht nützliches Mittel Anwendung finden. Vorzugsweise kön nen sie bei endogenen Depressionen in der depressiven Phase bei Geisteskrankheiten, bei psychasthenischen und neurotischen Depressionen angewendet werden.
<I>Beispiel 1</I> Dihydro-D-lysergsäure-a-(acetamido-äthoxy)-äthylamid Eine Mischung von 10 g durch Hydrierung von D- lysergsäure-a-hydroxyäthylamid in Gegenwart eines Hy- drierungskatalysatorserhaltenem Dihydro-D-lysergsäure- a-hydroxyäthylamid, 55g N-Acetylamino-äthanol und 4,5 g Maleinsäure wird eine Stunde bei 75 C gerührt. Nach Abkühlen verdünnt man mit 250 ml Wasser und macht mit Natriumbikarbonat alkalisch. Dann wird mit Methylenchlorid extrahiert; der Auszug wird im Va kuum eingedampft. Der Rückstand wird aus Aceton um kristallisiert. Man erhält 1,6 g des Isomers A , das bei 227-230 C schmilzt.
Bei weiterem Konzentrieren des Filtrats erhält man 1,7 g des Isomers B , das bei 205 bis 210 C schmilzt.
<I>Beispiel 2</I> Dihydro-D-lysergsäure-a-(2'-phenyläthoxy)-äthylamid Eine Mischung von 1 g durch Hydrierung von D- lysergsäure-a-hydrbxyäthylamid in Gegenwart eines Hy- drierungskatalysators erhaltenem Dihydro-D-lysergsäure- a-hydroxyäthylamid, 10 ml 2-Phenyläthylalkohol und 0,6 g Maleinsäure wird anderthalb Stunden bei 70 C gerührt. Nach Abkühlen wird sie dreimal mit etwas Äthyläther geschüttelt, wobei man jedesmal das Lö sungsmittel durch Dekantieren abtrennt.
Das hinterblei bende Öl wird in 20 ml 5o/oiger wässeriger Natriumbi- karbonatlösung gelöst und mit Methylenchlorid ausge- zogen. Die organische Lösung wird im Vakuum einge dampft. Der Rückstand wird aus Äthyläther umkristal lisiert. Man erhält 350 mg Dihydro-D-lysergsäure-a-(2'- phenyläthoxy)-äthylamid (Mischung der Isomere A und B), das bei 167-170 C schmilzt;

<I>Beispiel 3</I> 1-Methyl-dihydro-D-lysergsäure-a-(2'-acetamido- äthoxy)-äthylamid Man geht gemäss Beispiel 1 vor und rührt eine Mi schung von 2,6 g durch Hydrierung von D-lysergsäure- a-hydroxyäthylamid in Gegenwart eines Hydrierungs- katalysators erhaltenem Dihydro-D-lysergsäure-a-(2'- acetamidoäthoxy)-äthylamid (Isomer A) in 150 ml flüs sigem Ammoniak, enthaltend 0,51g metallisches Ka lium, wird 30 Minuten bei -40 C, worauf 1,64 g Me- thyljodid in 5 ml wasserfreiem Äther zugefügt werden.
Das Reaktionsgemisch wird weitere 30 Minuten bei -40 C gerührt und dann lässt man das Ammoniak bei Raumtemperatur verdampfen. Der Rückstand wird mit Wasser aufgenommen und die wässerige Lösung mit Methylenchlorid extrahiert, der Auszug im Vakuum ein gedampft, der Rückstand mit Aceton aufgenommen und filtriert. Nach Umkristallisieren aus Aceton erhält man 1,3 g Produkt; Fp = 219-220 C, (c=1 in 96%igem Äthanol) (Isomer A).
