Verfahren zur Herstellung von neuen Penicillinderivaten
Die vorliegende Erfindung bezieht sich auf neue Penicillinderivate bzw. auf neue Derivate von 6-Aminopenicillansäure.

Es wurde nun gemäss der vorliegenden Erfindung gefunden, dass neue PenicilUnderivate mit wertvollen antibiotischen Eigenschaften erhalten werden können, wenn man Aminoacylsubstituentengruppen in die Aminogruppe von 6-Aminopenicillansäure einführt.
Die neuen Penicillinderivate, welche gemäss der vorliegenden Erfindung erhalten werden, besitzen die folgende allgemeine Formel :

endet. In allen andern Fällen aber, d. h., wenn das dem Kohlenstoffatom, welches die zu reduzierende Carbobenzoxyaminogruppe trägt, benachbarte Kohlenstoffatom aliphatisch ist, kann es notwendig sein, um eine vollständige Reduktion zu erzielen, mit zwei oder mehr hintereinander zugesetzten Anteilen des Katalysators zu hydrieren.
Wenn die aminosubstituierte Carbonsäure mehr als eine Carboxylgruppe enthält, beispielsweise a-Aminoadipinsäure, ist es notwendig, um ein einwandfreies Acylderivat herzustellen, entweder eine der Carboxylgruppen vor der Reaktion mit 6-Aminopenicillansäure zu schützen oder ein sicheres reaktionsfähiges Derivat von nur einer Carboxylgruppe herzustellen.
Da einige der nach dem erfindungsgemässen Verfahren erhaltenen antibiotischen Substanzen relativ instabile Verbindungen sind, welche leicht chemischen Veränderungen unterliegen, woraus sich ein Verlust an antibiotischer Aktivität ergibt, ist es zweckmässig, hinreichend milde Reaktions- und Isolationsbedingungen auszuwählen, um deren Zersetzung zu vermeiden.
Die folgenden Beispiele sollen den Gegenstand der vorliegenden Erfindung erläutern, ohne dass diese jedoch hierauf beschränkt werden soll.
Beispiel l : Herstellung von Aminomethyl-Penicillin.
Eine Suspension von Palladium auf Bariumcarbonat (7, 3 g, 30%ig) in 200 ml Wasser wurde in einer Wasserstoffatmosphäre bei Raumtemperatur und atmosphärischem Druck 2 h lang geschüttelt. Nach dieser Behandlung wurde der Katalysator abgetrennt und gut mit Wasser gewaschen, wobei man darauf achtete, dass er niemals trocken wurde. Hierauf wurden 4, 5 g Natriumsalz von Carbobenzoxyaminomethyl-peni-

schrieben reduziert, wurde zugesetzt und die Hydrierung weitere 45 min lang fortgesetzt. Hierauf wurde der Katalysator abfiltriert, mit Wasser gewaschen und das Filtrat wurde, nachdem das PH mit n-Salzsäure auf 7, 0 eingestellt worden war, unter vermindertem Druck bei einer Temperatur unter 20 C zur Trockne eingedampft.
DasAminomethylpenicillin (IV) wurde als gelbes Pulver in einer Menge von 3, 0 g und einer Reinheit von 33% erhalten. Die Papierchromatographie zeigte, dass dieses Material nur ein Antibiotikum enthielt, welches einen vom Ausgangsmaterial beträchtlich verschiedenen Rf-Wert aufwies. Es zeigte sich, dass das Produkt in saurer Lösung stabil ist und Staphylococcus aureus in einer Konzentration von 1. 25 Jl g/ml hemmt.

Nach 5 min wurde die resultierende Suspension, welche das gemischte Anhydrid enthielt, langsam mit einer eiskalten Lösung von 4, 3 g 6 -A minopenicillansäure und 170 ml 3%iger wässeriger Natriumbikarbonatlösung behandelt. Während des Zusatzes wurde die Mischung auf 00C gehalten, aber hernach wurde sie während einstündigem weiterem Rühren auf Raumtemperatur steigen gelassen. Die Mischung wurde dreimal mit je 250 ml Äther gewaschen und die Waschlösungen wurden verworfen.
Die wässerige Phase wurde mit Salzsäure auf ein PH von 2 gebracht und die freie Penicillansäure wurde zunächst mit 40 ml, dann zweimal mit je 10 ml Butanol extiahiert. Das Natriumsalz von Carbobenzoxyaminomethyl-Penicillin wurde durch

neuerliches Extrahieren der gewaschenen Butanollosung mit dreimal je 5 ml Wasser, zu welchem jeweils genügend 3% igue wässerige Natriumbikarbonatlösung zugesetzt wurde, um die wässerige Phase auf ein PH von 7 zu bringen, gewonnen. Die kombinierten wässerigen Extrakte wurden mit Äther gewaschen und dann im Vakuum bei einer Temperatur unter 200C eingedampft. Es hinterblieben 4, 7 g des festen Natriumsal- zes.
Beispiel 2 : Herstellung von ss-Aminoäthyl-Penicillin,
Das Natriumsalz von ss-Carbobenzoxyaminoäthyl-Penicillin (50% zig, rein), wurde in Gegenwart von drei hintereinander zugesetzten Anteilen an vorbehandeltem Palladium/Bariumcarbonatkatalysator, wie in Beispiel 1 beschrieben, zu ss -Aminoäthylpenicíl1in mit roziger Reinheit mit 40%figer Ausbeute hy- driert. Die Papierchromatographie zeigte nur ein Antibiotikum, dessen Rf-Wert von dem des Ausgangs- materials beträchtlich verschieden war. Es war in saurer Lösung stabil und zeigte Hemmung von Staphylo- coccus aureus in einer Konzentration von 2, 5 ug/ml.
Das in diesem Beispiel verwendete ss-Carbobenzoxyaminoäthyl-Penicillin wurde wie folgt herge- stellt : 3, 75 ml Chlorameisensäureäthylester wurden tropfenweise zu einer eiskalten gerührten Lösung von
9, 9g N-Carbobenzoxy-ss-alanin und 6,7 ml Triäthylamin in 330 ml trockenem Azeton zugesetzt. Nach
5 min wurde die resultierende Suspension, welche das gemischte Anhydrid enthielt, langsam mit einer

tur während weiterem einstündigem Rühren auf Raumtemperatur steigen gelassen. Die Mischung wurde dreimal mit je 300 mlÄther gewaschen und die Waschflüssigkeiten wurden verworfen. Die wässerige Pha- se wurde mit Salzsäure auf ein pH von 2gebracht und die freie Penicillansäure wurde zunächst mit 50 ml, dann zweimal mit je 20 ml Butanol extrahiert.
Das Natriumsalz von ss-Carbobenzoxyaminoäthyl-penicillin wurde durch Extrahieren der gewaschenen Butanollösung mit dreimal je 10 ml Wasser wiedergewonnen, wobei jeder Wasseranteil mit genügend 3% figer wässeriger Natriumbikarbonatlösung versetzt wurde, um die wässerige Phase auf einen pH-Wert von 7 zu bringen. Die kombinierten wässerigen Extrakte wurden mit Äther gewaschen und dann im Vakuum unter 200C eingedampft. Es hinterblieben 6, 68 g des festen Natriumsalzes.
Beispiel 3 : Herstellung von 5-Aminopentyl-Penicillin.
Das Natriumsalz von 5-Carbobenzoxyaminopentyl-Penicillin (690/0 rein) wurde in Gegenwart von vier hintereinander zugesetzten Anteilen an vorbehandeltem Palladium/Bariumcarbonat-Katalysator, wie in Beispiel 1 beschrieben, zu 5-Aminopentylpenicillin mit 52% figer Reinheit in 55% tiger Ausbeute hydriert.
Die Papierchromatographie zeigte nur ein Antibiotikum, dessen Rf-Wert von dem des Ausgangsmaterials beträchtlich verschieden war.
Das in diesem Beispiel verwendete 5-Carbobenzoxyaminopentyl-penicillin wurde in piger Ausbeute aus 6-Aminopenicillansäure und einem gemischten Anhydrid, hergestellt aus 6-Carbobenzoxy- -aminohexansäure und Chlorameisensäure-äthyleseter, gemäss dem in Beispiel 1 angegebenen allgemeinen Verfahren hergestellt.
Die benötigte 6-Carbobenzoxy-aminohexansäure, Fp 64 C, wurde in 81% tiger Ausbeute aus 6-Aminohexansäure und Chlorameisensäure-benzylester in kalter wässeriger Natriumhydroxydlösung hergestellt.
Beispiel 4 : Herstellung von α-Aminopentyl-Penicillin,
Das Natriumsalz von (x-Carbobenzoxyaminopentyl-penicillin (56'% rein) wurde in Gegenwart von drei hintereinander zugesetzten Anteilen an vorbehandeltem Palladium/Bariumcarbonat-Katalysator, wie in Beispiel 1 beschrieben, zu a-Aminobenzylpenicíl1in mit 34% figer Reinheit in zuiger Ausbeute hydriert.
Die Papierchromatographie zeigte nur ein Antibiotikum, dessen Rf-Wert von dem des Ausgangsmaterials beträchtlich verschieden war. Es war in saurer Lösung stabil und zeigte Hemmung von Staphylococcus aureus in einer Konzentration von 0, 12 ug/ml.
Das in diesem Beispiel verwendete α-Carbobenzoxyaminopentyl-penicillin wurde in 64% tiger Ausbeute aus 6-Aminopenicillansäure und einem gemischten Anhydrid, hergestellt aus N-Carbobenzoxy-DL- - norleucin und Chlorameisensäure-äthylester, gemäss dem in Beispiel 1 angegebenen allgemeinen Verfahren hergestellt.
N-Carbobenzoxy-DL-norleucin, Fp 106-107 C. wurde in 70%figer Ausbeute aus DL-Norleucin und Chlorameisensäurebenzylester in kalter wässeriger Natriumhydroxydlösung hergestellt.

Es war in saurer Lösung stabil und zeigte Hemmung von Staphylococcus aureus in einer Konzentration von 0,12 g/ml.
Das in diesem Beispiel verwendete α-Carbobenzoxyaminocyclohexylmethyl - Penicillin wurde in 51%iger Ausbeute aus 6-Aminopenicillansäure und einem gemischten Anhydrid, hergesteut aus Dl-α-Carbo- benzoxyaminocyclohexylessigsäure und Chlorameisensäure-äthylester, gemäss dem in Beispiel 1 angegebenen allgemeinen Verfahren hergestellt.
Die DL-α-Carbobenzoxyaminocyclohexyl-essigsäure, Fp 136-137 C, wurde in 80% figer Ausbeute aus

ZurHerstellung von α-Carbobenzoxyamino-ss-phenyläthyl-penicillin wurde N-carbobenzoxy-ss-phe- nyl-DL-alanin mit Chlorameisensäureäthylester zum gemischten Anhydrid umgesetzt ; dieses wurde dann unter den in Beispiel 1 angegebenen Bedingungen mit 6-Aminopenicillansäure reagieren gelassen.
Beispiel 8 : Herstellung von α-Amino-ss-3-indolyläthyl-Penicillin.
Diese Verbindung wurde erhalten durch Hydrieren des Natriumsalzes von ct-Carbobenzoxyamino- -ss-3-indolyläthyl-Penicillin (77% rein) gemäss Beispiel 2 mit 44% figer Reinheit in 47% tiger Ausbeute. Die Papierchromatographie zeigte nur ein Antibiotikum, dessen Rf-Wert von dem des Ausgangsmaterials beträchtlich verschieden war. Es war in saurer Lösung stabil und zeigte Hemmung von Staphylococcus aureus in einer Konzentration von 0, 5 g/ml.
Das in diesem Beispiel verwendete a-Carbobenzoxy-amino-ss-3-indolyläthyl-penicillin wurde in 83%iger Ausbeute aus 6-Aminopenicillansäure und einem gemischten Anhydrid, hergestellt aus N-Carbobenzoxy-DL-tryptophan in Chlorameisensäureäthylester, gemäss dem in Beispiel l angegebenen allgemeinen Verfahren, hergestellt ; Fp = 167 C.

Das in diesem Beispiel verwendere α-Carbobenzoxyamino-ss-hydroxyäthyl-penicillin wurde in 67%iger Ausbeute aus 6-Amino-penicillansäure und einem gemischten Anhydrid aus N-Carbobenzoxy-DL-serin und Chlorameisensäure-äthylester, gemäss dem in Beispiel 1 beschriebenen allgemeinen Verfahren hergestellt.
Beispiel 10: Herstellung von α-Amino-ss-hydroxy-ss-phenyläthyl-Penicillin.
Dieses wurde durch Hydrieren des Natriumsalzes von a-Carbobenzoxyamino-ss -hydroxy- ss-phenyl- äthyl-penicillin(60% rein), wie in Beispiel 2 beschrieben, in 53%iger Ausbeute und mit 39% figer Reinheit hergestellt.Die Papierchromatographie zeigte nur ein Antibiotikum mit einem vom Ausgangsmaterial beträchtlich verschiedenen Rf-Wert. Es war in saurer Lösung stabil und hemmte Staphylococcus aureus.
Das in diesem Beispiel verwendete α-Carbobenzoxyamino-ss-hyroxy-ss-phenyläthyl-penicillin wurde in 46% figer Ausbeute aus 6-Amino-penicillansäure und einem gemischten Anhydrid aus N-Carbobenzoxy- -Dl-threo-ss-phenylserin und Chlorameisensäure-äthylester, gemäss dem in Beispiel 1 beschriebenen allgemeinen Verfahren hergestellt.
N-Carbobenzoxy-DL-threo-ss-phenylserin, Fp 63 - 64 C, wurde in 30% figer Ausbeute aus DL-Threo- - ss-phenylserin und Chlorameisensäure-benzylester in wässeriger Natriumhydroxydlösung hergestellt.
Beispiel 11 : Herstellung von α-Aminobenzyl-Penicillin.
EineSuspension vonPalladium-Bariumcarbonat (3,7 g mit 30%) in 20 ml Wasser wurde in einer Wasserstoffatmosphäre bei Raumtemperatur und Atmosphärendruck 11/2 h geschüttelt. Der Katalysator wurde dann abfiltriert und gut mit Wasser gewaschen, wobei man darauf achtete, dass er nicht trocken wurde. Eine Lösung von 4g des Natriumsalzes von α-Carbobenzoxyaminobenzyl-Penicillin in 20 ml Wasser wurde zu dem vorbehandelten Katalysator zugesetzt und die Suspension wurde in einer Wasserstoffatmosphäre und bei Raumtemperatur und-druck l h lang geschüttelt. Der Katalysator wurde dann abfiltriert, gut mit Wasser gewaschen und das mit den Waschflüssigkeiten vereinigte Filtrat wurde mit n-Salzsäure auf ein PH von 7, 0 eingestellt.
Die resultierende Lösung wurde dann im Vakuum bei einer Temperatur unter 200C eingedampft. Es wurden 2, 4 g α-Aminobenzyl-Penicillin(ineres Salz) erhalten, mit 48%iger Reinheit und 74% Ausbeute. Die Papierchromatographie zeigte, dass dieses Material nui ein Antibiotikum enthielt, welches einen Rf-Wert aufwies, der von dem des Ausgangsmaterials beträchtlich verschieden war. Es war in saurer Lösung stabil und hemmte Staphylococcus aureus in einer Konzentration von 0,12 g/ml.
Das Natriumsalz von a-Carbobenzoxyaminobenzyl-Penicillin, welches für die obige Darstellung benötigt wurde, wurde erhalten, indem zunächst 1 Äquivalent DL-α-Carbobenzoxyaminophenylessigsäure

Das Natriumsalz von α-Carbobenzoxyamino-2-furylmethyl-Penicillin, welches für das obige Verfahren benötigt wurde, wurde erhalten, indem zunächst ein Äquivalent DL-a-Carbobenzoxyamino-2-furyl- essigsäure mit einem Äquivalent Chlorameisensäureäthylester zum reaktiven gemischten Anhydrid umgesetzt wurde. Dieses wurde dann mit 6-Aminopenicillansäure unter den in Beispiel 1 beschriebenen Bedingungen umgesetzt. Es wurde ein Feststoff mit 40% figer Reinheit in 32% figer Ausbeute erhalten.
DL-α-Carbobenzoxyamino-2-furylessigsäure, Fp 121-1220C, wurde in 40% figer Ausbeute durch Reaktion von α-Amino-2-furylessigsäure mit Chlor a meisensäurebenzylesterf in verdünnter Natriumhydroxyd- lösung erhalten.
Analyse : Berechnet C14H13O5N: C 61,1% H 4, 80/0 N 5, 1% Gefunden : 61, 31o H 4, 7% N4, 9%.
