Verfahren zur Herstellung neuer Pyrimidine
Gegenstand der Erfindung ist die Herstellung von Pyrimidinverbindungen der Formel
Py-NH-Ph-X-Ph'-NH-Py', worin Py und Py' gegebenenfalls substituierte Pyrimidyl-(2)-reste bedeuten, die in 4-Stellung eine Aminogruppe (Y) enthalten, die durch einen, eine gegebenenfalls substituierte Aminogruppe tragenden, aliphatischen Rest mit mehr als 1 C-Atom zwischen den N-Atomen substituiert ist, Ph und Ph'gegebenenfalls substituierte Phenylenreste darstellen und X eine direkte Bindung, Sauerstoff oder einen niederen divalenten aliphatischen Kohlenwasserstoffrest darstellt.
Die mindestens 2 Kohlenstoffatome aufweisenden aliphatischen Reste, die eine Aminogruppe tragen, sind vor allem niedere Alkylenreste, die auch durch Heteroatome, wie Sauerstoff, Stickstoff oder Schwefel, in der Kohlenstoffkette unterbrochen und/oder durch freie Oxy- oder Aminogruppen substituiert sein können.
Diese Reste weisen zweckmässig höchstens 8 Kohlenstoffatome auf. Zu nennen sind vor allem Äthylen-, gerade oder verzweigte, an beliebigen, verschiedenen Kohlenstoffatomen mit den Aminogruppen verbundenen Propylen-, Butylen-, Pentylen-, Hexylen- oder Heptylenreste, insbesondere Äthylen, Propylen-(1,3), Propylen (2,3), Butylen-(2,4), Butylen-(1,4), Pentylen-(1,5), Pentylen-(2,5), Hexylen-(1, 6) oder Hexylen-(2,6).
Die den genannten aliphatischen Rest substituierende Aminogruppe ist unsubstituiert oder substituiert. Als Substituenten sind z. B. zu nennen: niedere Kohlenwasserstoffreste aliphatischen Charakters, die auch durch Heteroatome, wie Sauerstoff, Schwefel oder Stickstoff, in der Kohlenstoffkette unterbrochen und/oder durch freie Hydroxylgruppen substituiert sein können. Niedere Kohlenwassertoffreste aliphatischen Charakters sind vor allem gesättigte oder einfach ungesättigte, höchstens 8 Kohlenstoffatome aufweisende Alkyl-, Cycloalkyl- oder Cycloalkyl-alkylreste. Durch Heteroatome unterbrochene Reste dieser Art sind vor allem Oxa- oder Azaalkyl-, Oxaalkylen-, Azaalkylen-oder Oxacycloalkylalkylreste.
Zu nennen sind insbesondere Methyl-, Athyl-, Allyl-, Propyl-, Isopropyl-, gerade oder verzweigte, in beliebiger Stelle verbundene Butyl, Pentyl-, Hexyt- oder Heptylreste, 3-Oxabutyl-, 3-Oxapentyl-, 3-Oxaheptyl-, 2-Hydroxyäthyl-, 3-Hydroxypropyl-, Butylen-(1,4)-, Butylen-(1,5)-, Hexylen-(1,5)-, Hexylen (1,6)-, Hexylen-(2,5)-, Heptylen-(1,7)-, Heptylen-(2,7)-, Heptylen-(2, 6)-, 3-Oxapentylen-( 1,5)-, 3-Aza-penltylen (1,5)-, 3-Niederalkyl-3-aza-pentylen-(1,5)-, wie 3-Methyl-3-aza-pentylen-(1,5)-, 3-Hydroxy-niederalkyl-3-azapentylen-(1,5)-, wie 3-Hydroxy-äthyl-3-aza-pentylen (1,5)-, 3-Oxa- oder -Azahexylen-(1,6)-,
Cyclopentyloder Cyclohexylreste oder Cyciopentyl- oder Cyclo hexylmethyl oder -üthyfreste. Die Aminogruppe ist vorteilhaft eine sekundäre Aminogruppe, insbesondere eine Niederalkylaminogruppe, z. B. eine Methyl-, Äthyl-, und vor allem eine Propyl-, Butyl- oder Pentylaminogruppe, worin die Alkylreste, vorzugsweise nicht primärer Natur sind, z. B. eine Isopropyl- oder sek.-Butylaminogruppe. Ganz besonders günstig sind jedoch ter tiäre Aminogruppen, insbesondere Di-niederalkylamino gruppen, wie Dimethyl-, Diäthyl-, N-Methyl-N-äthyl-, Dipropyl-, Di-isopropyl-, Dibutyl-, Di-sek,-butyl- oder Di-amylaminogruppen.
Die beiden Phenylenreste Ph und Ph' sind vorzugsweise p-Phenylenreste; sie können aber auch oder o-Phenylenreste und in dieser Beziehung voneinander verschieden sein.
Ein bivalenter aliphatischer Kohlenwasserstoffrest X ist z. B. ein niederer Alkylen oder Alkylidenrest, z. B.
Methylen, Athylen, Propylen oder ethyliden, oder ein niederer Alkenylenrest, z. B. Äthenylen.
Die neuen Verbindungen können in beliebiger Weise weitere Substituenten tragen. So können die Pyrimidylreste in 6-Stellung z. B. durch Halogen, Niederalkyl, Niederalkoxy, Niederalkoxyalkyl oder Niederalkoxyniederalkoxy und/oder in 5-Stellung z. B. durch Niederalkyl, Niederalkoxy oder Halogen substituiert sein.
Dabei können Niederalkylreste in 5- und 6-Stellung mit einander zu einem Alkylenrest, z. B. einem Butylen (1, 4)-rest, verbunden sein.
Die Phenylenreste können ebenfalls substituiert sein, vorzugsweise durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl, Oxy- oder Aminosulfonyl, Nitro oder Amino.
Dabei sind niedere Alkylreste, insbesondere solche mit 1-5 Kohlenstoffatomen, wie Methyl-, Äthyl-, n-Propyl- oder Isopropylreste, oder gerade oder verzweigte, in beliebiger Stellung verbundene Butyl- oder Pentylreste. Niederalkoxyreste sind beispielsweise solche, die die oben genannten Alkylreste enthalten, insbesondere Methoxy-, Äthoxy- oder Propoxyreste.
Die in den Niederalkoxyniederalkylresten enthaltenen Niederalkoxyreste sind beispielsweise die vorstehend genannten. Die das Sauerstoffatom mit dem Pyrimidinkern verbindenden Alkylenreste stehen vorzugsweise für Alkylenreste mit 1-5, insbesondere 1-3 Kohlenstoffatomen, wie Methylenreste, in beliebiger Stellung verbundene Athylen-, Propylen-, Butylen oder Pentylenreste. Niederalkoxyniederalkylreste sind insbesondere Reste der Formel RO-(CH2)n-, worin R einen Alkylrest mit 1-3 Kohlenstoffatomen und n eine ganze Zahl von 1-3 bedeutet.
Die in den Niederalkoxyniederalkoxyresten enthaltenen Alkoxygruppen sind beispielsweise die oben genannten. Der diese Alkoxygruppen mit dem am Pyrimidinkern befindlichen Sauerstoffatom verbindende Alkylenrest besitzt zwischen der Alkoxygruppe und dem genannten Sauerstoffatom mindestens zwei Kohlenstoffatome und erhält vorzugsweise 2-5 Kohlenstoffatome und steht speziell für 1,2-Äthylen, ferner für 1,2-, 2,3- oder 1,3-Propylen, gerades oder verzweigtes, in beliebiger Stellung verbundenes, jedoch die benachbarten Sauerstoffatome durch mindestens zwei Kohlenstoffatome trennendes Butylen oder Pentylen.
Als Halogenatome kommen insbesondere Brom-, Chlor-oder Fluoratome in Frage.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. Insbesondere wirken sie gegen Bakterien, wie Tuberkelbazillen, vor allem aber gegen Protozoen, speziell Plasmodien, z. B. in der Maus, und gegen Piroplasmen, wie Babesien, Babesiellen und Theilerien. Dabei sind sie auch gegen solche Plasmodien wirksam, die gegenüber bekannten Antimalariamittein resistent sind. Ferner wirken die neuen Verbindungen entzündungshemmend. Die neuen Verbindungen können daher pharmakologisch am Tier oder als Medikamente, z. B. bei Malaria, Babesienie und andern Infektionen, sowie bei diversen Entzündungszuständen, Verwendung finden. Sie können auch als tierfuttermittel bzw. als Zusatz zu Tierfutter verwendet werden.
Sie sind auch wertvolle Zwischenprodukte für die Herstellung anderer als Heilmittel wertvoller Verbindungen.
Besonders wertvoll, vor allem in bezug auf ihre Wirksamkeit gegenüber Plasmodien und Babesien, sowie als entzündungshemmende Stoffe, sind die Verbindungen der Formel

Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man in Verbindungen der Formel Pyl-NH-Ph-X-Ph¯NH-Pyi worin Ph, Ph' und X die eingangs gegebene Bedeutung haben und Pyl und Py1, gegebenenfalls weiter substituierte Pyrimidyl-(2)-reste darstellen, von denen einer in 4-Stellung die Gruppe X' und der andere ebenfalls die Gruppe X' oder die oben definierte Gruppe Y enthält, wobei X' eine Aminogruppe bedeutet, die durch einen aliphatischen Rest substituiert ist, der einen direkt gegen eine gegebenenfalls substituierte Aminogruppe austauschbaren Rest Z enthält, wobei Z vom Aminostickstoffatom durch mindestens 2 Kohlenstoffatome getrennt ist, den oder die Reste Z direkt gegen eine gegebenenfalls substituierte Aminogruppe austauscht.
Ein direkt gegen eine Aminogruppe austauschbarer Rest Z ist vor allem eine reaktionsfähig veresterte Hydroxylgruppe, wie eine mit einer starken anorganischen Säure oder einer starken organischen Sulfonsäure veresterte Hydroxylgruppe, besonders ein Halogenatom, wie Chlor, Brom oder Jod, oder eine Sulfonyloxygruppe, wie eine Benzolsulfonyloxygruppe. Die reaktionsfähig veresterte Hydroxylgruppe wird dann in üblicher Weise durch Umsetzung mit Ammoniak oder einem primären oder sekundären Amin in die Aminogruppe umgewandelt
Je nach der Natur der Reste Z, die gleich oder verschieden sein können, kann man die Reste Z gleichzeitig oder nacheinander gegen die Aminogruppe austauschen.
Die genannten Reaktionen können in üblicher Weise, in An- oder Abwesenheit von Verdünnungsund/oder Kondensationsmitteln und/oder Katalysatoren, bei tiefer, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss und/oder unter einer Inertgasatmosphäre durchgeführt werden.
Je nach der Arbeitsweise erhält man die Verbindungen in freier Form oder in Form ihrer Salze. Aus den Basen können therapeutisch verwendbare Salze mit Säuren gebildet werden, z. B. von therapeutisch verwendbaren Säuren, wie der Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Per chlorsäure; aliphatischer, alicyclischer, aromatischer oder heterocyclischer Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milch-, Apfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-, Dioxymalein- oder Brenztraubensäure Phenylessig-, Benzoe, pAminobenzoe-, Anthranil-, p-Oxybenzoe-, Salicyl-, Embon- oder p-Aminosalicylsäure;
Methansulfon-, Athansulfon-, Oxyäthansulfon-, Athy- lensulfonsäure; Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin. Erhaltene Salze lassen sich in die freien Verbindungen umwandeln.
Die Salze lassen sich auch zur Reinigung der freien Verbindungen verwenden. Infolge der engen Beziehungen zwischen den Verbindungen in freier Form und in Form ihrer Salze werden im vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckgemäss gegebenenfalls auch die entsprechenden Salze verstanden.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden. So kann man eine Verbindung der Formel
H2N-Ph-X-Ph'-Q", worin Ph, Ph' und X die oben gegebene Bedeutung haben, und Q" eine freie Aminogruppe oder eine Nitrooder Acetylaminogruppe ist, mit einem 4-Oxy-2-Z'- pyrimidin umsetzen, worin Z' einen gegen eine Aminogruppe austauschbaren Rest, wie vor allem ein Halogenatom oder eine freie oder verätherte Mercaptogruppe, darstellt, hierauf die Oxygruppe durch Halogenierung, z. B. mit Phosphoroxyhalogeniden, gegen ein Halogenatom austauschen oder durch Schwefelung, z. B. mit Phosphorpentasulfid, gegen die Mercaptogruppe austauschen und diese gegebenenfalls in üblicher Weise veräthern.
Der Substituent in 4-Stellung kann dann in üblicher Weise gegen eine Aminogruppe, die durch einen, den Rest Z enthaltenden aliphatischen Rest substituiert ist, ausgetauscht werden. Dabei hat Z die oben genannte Bedeutung. Falls Q" keine Aminogruppe war, kann man den Substituenten in 4-Stellung gegen die Aminogruppe Y austauschen, die Nitro-oder Acylaminogruppe in die freie Aminogruppe umwandeln und diese, wenn erwünscht, der gleichen Reaktionsfolge, wie die erste Aminogruppe, unterwerfen.
Die erhaltenen Endstoffe und ihre Salze können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Im folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
10 g 4,4'-Bis-[4-(ss-chloräthylamino)-6-methyl-pyrimidyl-2- amino]-diphenyl-dihydrochlorid werden mit 100 ml ammoniakalischem Äthanol 12 Stunden bei 1500 erwärmt. Nach Erkalten wird das Äthanol im Vakuum abdestilliert, der Rückstand in Wasser aufgenommen, die Lösung mit Aktivkohle behandelt und filtriert. Die Lösung wird mit 50 ml 10% iger äthanoli- scher Salzsäure versetzt und das ausfallende Tetrahydrochlorid des 4,4'-Bis-[4-(ss-aminoäthylamino)-6-methyl pyrimidyl-2-amino] 4iphenyis der Formel

abgenutscht und aus 70 % igem Äthanol umkristallisiert, F. > 310.
Das Ausgangsmaterial wird wie folgt erhalten:
30 g 2-Methylthio-6-methyl-4-oxy-pyrimidin werden mit 17,4 g 4,4'-Diamino-diphenyl innig gemischt und im Ölbad langsam auf 1700 erhitzt, wobei die anfänglich geschmolzene Masse langsam erstarrt. Nach 301 Minuten bei dieser Temperatur wird pulverisiert und dreimal mit Athanol ausgekocht. Der Rückstand wird in 2n Natronlauge gelöst, die Lösung über Tierkohle filtriert und hierauf mit Eisessig angesäuert. Der Niederschlag wird abfiltriert, mit Wasser gewaschen und vorsichtig getrocknet. Man erhält so das 4,4'-Bis-(6-methyl4-oxy-pyrimidyl-2-amino)-diphenyl (F. 3000). 38 g dieses Produktes werden mit 350 ml Phosphoroxychlorid 3 Stunden am Rückfluss zum Sieden erhitzt.
Hierauf wird auf Eiswasser ausgegossen und nach 30 Minuten mit konzentriertem wässrigem Ammoniak schwach alkalisch gestellt. Nun wird der Niederschlag abgenutscht, mit verdünntem wässrigem Ammoniak und Wasser gewaschen und vorsichtig getrocknet. Man erhält so das rohe 4,4'-Bis-(6-methyl-4-chlor-pyrimidyl-2-amino)-diphenyl, das in dieser Form verarbeitet wird.
Eine Mischung von 23 g 4, 4'-Bis-(6-methyl-4-chlor- pyrimidyl-2-amino)-diphenyl und 50 ml Äthanolamin wird 4 Stunden unter Rückfluss erhitzt. Der Überschuss an Amin wird durch Destillation entfernt und der Rückstand unter starkem Rühren in Eiswasser gegossen.
Das ausfallende 4,4'-Bis-[4-(ss-hydroxyäthylamino)-6methyl-pyrimidyl-2-amino]-diphenyl wird abgenutscht und getrocknet. Davon werden 10 g mit 50 mi Thionylchlorid 4 Stunden unter Rückfluss erhitzt, das über- schüssige Thionylchlorid im Vakuum gut abdestilliert und das verbleibende 4,4'-Bis-[4-(ss-chloräthylamino)-6- methyl-pyrimidyl-2-amino]-diphenyl-dihydrochlorid direkt weiter umgesetzt.
In ähnlicher Weise, ausgehend von den entsprechenden 4,4'-Bis-(4chloralkylamino-pyrimidyl-2-amino)diphenyl-verbindungen und durch Umsetzung mit dem gewünschten Amin erhält man: das Tetrahydrochlorid des 4,4'-Bis-[4(ss-diäthytamino- äthylamino)-6-methoxymethyl-pyrimidyl-2-amino] 3,3'-dimethoxydiphenyls, F. 255-257 aus Äthanol; das 4,4'-Bis-[4-(5-diäthylamino-pentyl-2-amino)
6-methoxymethyl-pyrimidyl-2amino]-3,3'-di methoxy-diphenyl-tetrahydrochlorid,
