Verfahren zur Herstellung von Dibenzocycloheptenen
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen 5H-Dibenzo-[a, d]-cycloheptenen und 10,1 i-Dihydro-5H-dibenzo[a,d] cycloheptenen und deren Salze, welche am Kohlenstoff der 5-Stellung durch einen Aminopropylrest substituiert sind.
Diese Verbindungen können zur Behandlung von Geisteskrankheiten verwendet werden, da sie als Mittel gegen Depressionszustände bzw. zur Aufheiterung des Gemüts und als psychische Energiespender wirksam sind. Für solche Fälle kann die tägliche Dosis 5 bis 250 mg betragen, welche vorzugsweise in Teildosen eingenommen wird. Diese Verbindungen werden vorzugsweise in Form ihrer mit Säuren erhältlichen Salze verabreicht.
Die nach dem erfindungsgemässen Verfahren herstellbaren Dibenzocycloheptene haben folgende Formel

Die punktierte Linie zwischen den Stellungen 10 und 11 bedeutet, dass die Verbindungen an dieser Stelle gesättigt oder ungesättigt sein können, wobei die gesättigte Verbindung als die 10,11-Dihydroverbindung bezeichnet wird.

Die Bildung eines primären Amins erfolgt demnach durch Umsetzung mit einer Alkoxypropyl-Grignardverbindung am Kohlenstoffatom in 5-Stellung, gegebenenfalls bei gleichzeitiger Reduktion der Oxygruppe; hierauf Ersatz der Alkoxygruppe durch Jod und Bildung des Amins durch Umsetzung mit Kaliumphthalamid und anschliessende Einwirkung von Hydrazinhydrat in absolutem Alkohol oder Abspaltung des Phthalrestes durch Hydrolyse. Das gebildete Grignardaddukt wird vorzugsweise unter neutralen Bedingungen hydrolysiert, z. B. unter Verwendung einer Ammoniumchloridlösung. Hierauf verfährt man zweckmässig wie folgt: Man isoliert das Produkt durch Verdampfung des Lösungsmittels und behandelt es in Gegenwart eines geeigneten anderen Lösungsmittels, z. B. Essigsäure oder Essigsäureanhydrid, mit Jodwasserstoffsäure.
Dann extrahiert man die Jodverbindung, z. B. mit Benzol, und isoliert sie durch Verdampfung des letzteren.
Hierauf setzt man die Jodverbindung mit Kaliumphthalimid in Gegenwart eines Lösungsmittels um, z. B.
Dimethylformamid, und erhitzt schliesslich gelinde um die Umsetzung zu beschleunigen und zu Ende zu füh- ren. Das gebildete Phthalimidaddukt isoliert man aus dem Reaktionsgemisch durch Extraktion.
Die anschliessende Behandlung mit Hydrazinhydrat in absolutem Methanol oder Äthanol kann gewünschtenfalls unter Erhitzung bis zum Rückfluss erfolgen.
Dann wird das gebildete Phthalhydrazid abgetrennt und das primäre Amin aus der Lösung isoliert. Die Phthalimidverbindung kann wie gesagt auch durch Hydrolyse in das Amin übergeführt werden.
Nun wird das erhaltene primäre Amin z. B. mit Essigsäureanhydrid bei Raumtemperatur oder unter Erwärmen und Rühren umgesetzt und das erhaltene Amid mit einem Metallhydrid, z. B. Lithiumaluminiumhydrid, reduziert.
Wenn von einem 5-Halogen (vorzugsweise Chlor oder Brom)-Derivat des 5H-Dibenzo(a,d) cyclohepten ausgegangen wird, so kann die Umsetzung mit dem Grignardreagens in Gegenwart eines üblichen Lösungsmittels, z. B. von trockenem Äther oder Tetrahydrofuran, durchgeführt werden. Dabei findet eine langsame Additionsreaktion statt, so dass dadurch eine wesentliche Temperaturerhöhung über Raumtemperatur verhindert wird.
Beispiel 1
Herstellung von 3-Chlor-5-(3-äthylaminopropyl)- 5H-dibenzo(a,d)cyclohepten
A. 3-Chlor-5-(3-Äthoxypropyl)-5H-dibenzo(a,d)cy- clohepten:
Eine Lösung von 3,5-Dichlor-5H-dibenzo(a,d)cy- clohepten in abs. Äther wird unter Rühren tropfenweise einer Äther-Lösung zugefügt, welche ein Äquivalent 3-Äthoxypropyl-magnesiumbromid enthält. Hierauf rührt man die Mischung während 1 Stunde bei Raumtemperatur, dann während 2 Stunden bei Rückflusstemperatur. Nach Abkühlung wird die Mischung mit verdünnter Salzsäure versetzt, die Ätherschicht abgetrennt, mit Wasser gewaschen und getrocknet.
Durch Verdampfen des Athers erscheint das Produkt als Rückstand.
B. 3-Chlor-5-(3-jodopropyl)-5H-dibenzo[a,d] cyclohepten:
Man löst 0,03 Mol 2-Chlor-5-(3-äthoxypropyl)-5Hdibenzo[a,d]cyclohepten in 45 ml Essigsäureanhydrid, erwärmt die Lösung auf 650 C, lässt Stickstoff durchströmen und fügt allmählich 30 ml Jodwasserstoffsäure (Sp. Gew. = 1,5) zu, ohne dass die Temperatur auf 900 C steigt. Dann erwärmt man auf 800 C während 30 Minuten, kühlt ab und giesst in Eiswasser. Das Produkt wird mit Äther extrahiert, die Ätherlösung mit verdünnter Natriumthiosulfatlösung, dann mit verdünnter Natriumbicarbonatlösung und schliesslich mit Wasser gewaschen. Durch Verdampfen des Äthers bleibt das Produkt zurück.
C. 3-Chlor-5-(3-phthalimidopropyl)-5H-diben zo[a,d] cyclohepten:
Dieses Produkt erhält man aus 3-Chlor-5-(3-jod- propyl)-5H-dibenzo [a, d] cyclohepten nach dem Verfahren gemäss Beispiel 2 C.
D. 3-Chlor-5-(3-aminopropy -5H-dibenzo [a,d] cyclohepten:
Dieses Produkt gewinnt man aus 3-Chlor-5-(phthal imidopropyi)-5H-dibenzo[a,d] cydohepten nach dem Verfahren gemäss Beispiel 2 D.
E. 3-Chlor-5-(3-acetamidopropyl)-5H-diben zo[a,d] cyclohepten:
Man löst 3 -Chlor-5-(3-aminopropyl)-5H-diben- zo[a,d] cyclohepten in Essigsäureanhydrid und erhitzt die Lösung während 20 Minuten zum Rückfluss. Nach Abkühlung giesst man in ein grosses Quantum Wasser und rührt die Mischung bis der Überschuss an Reagens hydrolysiert ist. Hierauf extrahiert man mit Äther, wäscht mit Natriumbicarbonatlösung, dann mit Wasser und trocknet über Natriumsulfat. Durch Verdampfung des Athers erhält man das Produkt.
F. 3 -Clüor-5-(3 -äthylaminopropyl)-H-dibenzo[a,d] cyclohepten:
Man löst 3-Chlor-5-(3-acetamidopropyl)-5H-diben zo[a,d]cyclohepten in abs. Äther und fügt diese Lösung unter Rühren tropfenweise einer 1,25 molaren Lithiumaluminiumhydridlösung in Äther zu. Die Mischung wird während 2 Stunden bei Raumtemperatur gerührt und dann während 15 Stunden auf Rückflusstemperatur. Nach Abkühlung setzt man unter Rühren behutsam Wasser und dann Natriumhydroxyd zu. Die Ätherschicht wird abgetrennt, mit Wasser gewaschen und über Natriumsulfat getrocknet. Nach Verdampfung des ethers wird das Produkt durch Destillation gereinigt.
Verwendet man als Ausgangsstoff das entsprechende 10, 11-Dihydro-SH-dibenzo (a, d) cyclohepten, so erhält man das 3-Chlor-1 0,1 1-Dihydro-5-(3-Äthyl- aminopropyl)-5H-dibenzo(a,d)cyclohepten.
Beispiel 2
Herstellung von 5-(3-Äthylaminopropyl)
SH-dibenzo [a,d] cyclohepten
A. 5-(3-Athoxypropyl)-5-oxy-5H-dibenzo [a, d]- cyclohepten:
Das Grignardreagens bereitet man aus 12, 55 g (0,075 Mol) 3-Äthoxypropylbromid und 1,82 g (0,075 Mol) Magnesium in l2ither. Nach Abkühlung der Lösung in einem Eisbad fügt man ihr unter Rühren eine Lösung von 5, 15 g (0,025 Mol) 5H-Dibenzo[a,d]cyclohepten-5-on in trockenem Äther tropfenweise zu. Dann rührt man die Mischung während 2 Stunden bei Raumtemperatur und anschliessend während 30 Minuten bei Rückflusstemperatur.
Hierauf zersetzt man das Grignardaddukt durch Abgiessen der Reaktionsmischung in eine Mischung von Eis-Ammo niumchloridlösung, trennt von der Ätherschicht und extrahiert die wässrige Schicht mit weiteren Mengen Äther. Nach Verdampfung des Äthers erhält man das Produkt als eine ölige kristalline Substanz. Nach Umkristallisieren aus Alkohol ergab sich ein Smp. des Produktes von 129-130 C.
Analyse für C20H22O2 berechnet: 81,600/0 C und 7,540/0 H; gefunden: 81,28 O/c C und 7, 50 /o H.
B. 5-(3-Jodpropyl)-5H-dibenzo [a,d] cyclohepten:
Eine Lösung von 9,25 g (0,0314 Mol) 5-(3-Äthoxy propyl)-5-oxydibenzo[a,d]cyclohepten, das wie oben angegeben hergestellt wurde, in 45 ml Essigsäureanhydrid, wird auf einem Dampfbad während 90 Minuten erhitzt und hierauf auf 650 C abgekühlt. Man leitet einen Stickstoffstrom durch die Lösung während 30 ml Jodwasserstoffsäure (1,5 spez. Gew.) portionenweise zugefügt werden, so dass die Temperatur unterhalb 90n C bleibt. Während dieser Operation entsteht in der Lösung eine dunkelbraune Jodfärbung. Die Lösung wird während 30 Minuten auf 80" C erhitzt, dann abgekühlt und in Eiswasser gegossen.
Das ausgeschiedene braune Öl wird in Benzol aufgenommen und die Lösung mit Wasser, mit verdünnter Natriumthiosulfatlösung und schliesslich mit Wasser gewaschen. Nach Verdampfung des Benzols erhält man 5-(3-Jodpropyl)-5H-dibenzo[a,d]cyclohepten als gelbes Öl, welches mit alkoholischem Silbernitrat eine Fällung ergibt.
C. 5-(3-Phthalimidpropyl)-5H-dibenzo [a,d] cyclohepten:
Man löst 1,8 g (0,005 Mol) 5-(3-Jodpropyl)-5Hdibenzo[a,d]cyclohepten in 10 ml Dimethylformamid und fügt unter Rühren 930 mg (0,0051 Mol) Kaliumphthalimid zu. Hierauf wird die Mischung während 40 Minuten auf 85-90 C erhitzt, dann abgekühlt, mit Chloroform verdünnt und in Wasser gegossen. Nach Abtrennung der Chloroformschicht extrahiert man die wässrige Schicht mit weiteren Mengen Chloroform. Nach Verdampfung des Chloroforms erhält man das Produkt als eine ölige kistalline Substanz.
Nach mehrmaliger Umkristallisation aus Isopropylalkohol schmilzt das Produkt bei 125, 5-131, 5 C.
Analyse für C26H21O2N berechnet: 82, 30 /o C; 5,58 0/o H; 3,69 0/0 N; gefunden: 82,46 0/0 C; 5, 60 /o H; 3, 70 /o N.
D. 5-(3-Aminopropyl)-5H-dibenzo[a,d] cyclohepten:
Man löst 2,15 g (0,0054 Mol) 5-(3-Phthalimidpro pyl)-5H-dibenzo[a,d]cyclohepten in 50 ml heissem 95 0/obigem Äthanol, setzt 0,6 ml Hydrazinhydrat zu und erhitzt die Lösung während 2 Stunden zum Rückfluss.
Nach Abkühlung wird angesäuert und man filtriert vom ausgefallenen Phthalhydrazid ab. Dann wird das Äthanol des Filtrats unter vermindertem Druck abgedampft und der Rückstand in 40 ml Wasser gelöst.
Dann verdampft man die filtrierte wässrige Lösung unter reduziertem Druck bis zu einem Volumen von ca. 20 ml. Durch Abkühlung fällt das salzsaure Salz von 5-(3-Aminopropyl)-5H-dibenzo [a,d] cyclehepten in einer Ausbeute von 1,35 g (87,5 O/o) aus. Nach Umkristallisation aus einer Äthanol-Äthermischung und aus Isopropylalkohol schmilzt das Analysenmuster bei 263-265 C unter Zersetzung.
Analyse für Ct8H19N. HC1 berechnet: 75,640/0 C; 7,05 0/ H; 4,900/0 N; gefunden: 75,54 O/o C; 6,770/U H; 4, 78 /o N.
Hierauf verfährt man gemäss Beispiel 1E und F.
Beispiel 3
Verwendet man an Stelle des unsubstitutierten Ketons gemäss Beispiel 2, ein kemsubstitutiertes Keton als Ausgangsstoff, so entsteht ein entsprechendes Endprodukt, wobei die Radikale X und X' die oben angegebene Bedeutung haben können.
Beispiel 4
Wenn als Ausgangsstoff 10,1 1-Dihydro-5H-diben- zo[a,d]cyclohepten-5-on verwendet und nach dem Verfahren gemäss Beispiel 2 gearbeitet wird, so entsteht als Endprodukt 5-(3-Aminopropyl)-10,11-dihy dro-5H-dibenzo[a,d] cyclohepten.
Beispiel 5
Herstellung von 5-(3-Äthylaminopropyl)-5H-diben- zo[a,d] cyclohepten:
A. 5-(3-Äthoxypropyl)-5H-dibenzo [a,d] cyclohepten:
Das Grignardreagens erhält man aus 6,44 g (0,0386 Mol) 3-Sithoxypropylbromid und 940 mg (0,0386 g Atom) Magnesium in Äther. Dieser Lösung fügt man unter Rühren tropfenweise eine Lösung von 5,67 g (0,025 Mol) 5-Chlor-5H-dibenzo[a,d]cyclohep- ten in einer Mischung von trockenem Äther und Tetrahydrofuran und rührt während einer weiteren Stunde bei Raumtemperatur. Hierauf wird das Grignardaddukt durch Abgiessen der Reaktionsmischung in eine Mischung von Eis und einer Ammoniumchloridlösung zersetzt. Die Ätherschicht wird abgetrennt und dei wässrige Schicht mit Äther extrahiert.
Nach Abdampfen des Äthers und Destillation des Rückstandes unter reduziertem Druck erhält man das Produkt als gelbes, Öl, das bei 145-148 (0,1 Torr) siedet.
Analyse für C20H22O berechnet: 86,28 /o C und 7,97 /o H; gefunden: 86,84 /o C und 7,97 O/o H.
B. 5-(3-Iodopropyl)-5H-dibenzo [a,d] cyclohepten:
Dieses Produkt erhält man nach dem Verfahren gemäss Beispiel 2B, jedoch unter Verwendung von 5-(3 -Äthoxypropyl)-H-dibenzo [a,d] cyclohepten als Ausgangsstoff. Daraus erhält man das gewünschte Endprodukt durch Anwendung des restlichen Verfahrens gemäss Beispiel 2.

A primary amine is accordingly formed by reaction with an alkoxypropyl Grignard compound on the carbon atom in the 5-position, optionally with simultaneous reduction of the oxy group; then replacement of the alkoxy group by iodine and formation of the amine by reaction with potassium phthalamide and subsequent action of hydrazine hydrate in absolute alcohol or elimination of the phthalic radical by hydrolysis. Preparation of 3-chloro-5- (3-ethylaminopropyl) -5H-dibenzo (a, d) cycloheptene
A. 3-chloro-5- (3-ethoxypropyl) -5H-dibenzo (a, d) cycloheptene:
A solution of 3,5-dichloro-5H-dibenzo (a, d) cyclohepten in abs. Ether is added dropwise with stirring to an ether solution which contains one equivalent of 3-ethoxypropyl magnesium bromide. The mixture is then stirred for 1 hour at room temperature, then for 2 hours at reflux temperature. After cooling, the mixture is treated with dilute hydrochloric acid, the ether layer is separated off, washed with water and dried.
Evaporation of the ether causes the product to appear as a residue.
B. 3-chloro-5- (3-iodopropyl) -5H-dibenzo [a, d] cycloheptene:
0.03 mol of 2-chloro-5- (3-ethoxypropyl) -5Hdibenzo [a, d] cycloheptene is dissolved in 45 ml of acetic anhydride, the solution is heated to 650 ° C., nitrogen is allowed to flow through and gradually 30 ml of hydriodic acid (Sp . = 1.5) without the temperature rising to 900 C. The mixture is then heated to 800 ° C. for 30 minutes, cooled and poured into ice water. The product is extracted with ether, the ether solution is washed with dilute sodium thiosulphate solution, then with dilute sodium bicarbonate solution and finally with water. C. 3-chloro-5- (3-phthalimidopropyl) -5H-diben zo [a, d] cycloheptene:
This product is obtained from 3-chloro-5- (3-iodopropyl) -5H-dibenzo [a, d] cycloheptene by the method according to Example 2 C.
D. 3-chloro-5- (3-aminopropy -5H-dibenzo [a, d] cycloheptene:
This product is obtained from 3-chloro-5- (phthalimidopropyi) -5H-dibenzo [a, d] cydoheptene by the method according to Example 2 D.
E. 3-chloro-5- (3-acetamidopropyl) -5H-diben zo [a, d] cycloheptene:
3-chloro-5- (3-aminopropyl) -5H-dibenzo [a, d] cycloheptene is dissolved in acetic anhydride and the solution is refluxed for 20 minutes. After cooling, it is poured into a large amount of water and the mixture is stirred until the excess of reagent is hydrolyzed. It is then extracted with ether, washed with sodium bicarbonate solution, then with water and dried over sodium sulfate. The product is obtained by evaporation of the ether.
F. 3-chloro-5- (3-ethylaminopropyl) -H-dibenzo [a, d] cycloheptene:
3-Chloro-5- (3-acetamidopropyl) -5H-diben zo [a, d] cycloheptene is dissolved in abs. Ether and this solution is added dropwise to a 1.25 molar lithium aluminum hydride solution in ether while stirring. If the corresponding 10, 11-dihydro-SH-dibenzo (a, d) cycloheptene is used as the starting material, the 3-chloro-1 0.1 1-dihydro-5- (3-ethyl aminopropyl) -5H- is obtained dibenzo (a, d) cycloheptene.
Example 2
Production of 5- (3-ethylaminopropyl)
SH-dibenzo [a, d] cycloheptene
A. 5- (3-Athoxypropyl) -5-oxy-5H-dibenzo [a, d] - cycloheptene:
The Grignard reagent is prepared from 12.55 g (0.075 mol) of 3-ethoxypropyl bromide and 1.82 g (0.075 mol) of magnesium in l2ither. After the solution has been cooled in an ice bath, a solution of 5.15 g (0.025 mol) of 5H-dibenzo [a, d] cyclohepten-5-one in dry ether is added dropwise with stirring. The mixture is then stirred for 2 hours at room temperature and then for 30 minutes at reflux temperature.
The Grignard adduct is then decomposed by pouring the reaction mixture into a mixture of ice-ammonium chloride solution, separating from the ether layer and extracting the aqueous layer with further amounts of ether. After evaporation of the ether, the product is obtained as an oily crystalline substance. After recrystallization from alcohol, the product had a melting point of 129-130 C.

B. 5- (3-iodopropyl) -5H-dibenzo [a, d] cycloheptene:
A solution of 9.25 g (0.0314 mol) of 5- (3-ethoxy propyl) -5-oxydibenzo [a, d] cycloheptene, which was prepared as indicated above, in 45 ml of acetic anhydride, is on a steam bath for 90 Heated for minutes and then cooled to 650.degree. A stream of nitrogen is passed through the solution while 30 ml of hydriodic acid (1.5 specific weight) are added in portions so that the temperature remains below 90NC. C. 5- (3-phthalimidopropyl) -5H-dibenzo [a, d] cycloheptene:
1.8 g (0.005 mol) of 5- (3-iodopropyl) -5Hdibenzo [a, d] cycloheptene are dissolved in 10 ml of dimethylformamide, and 930 mg (0.0051 mol) of potassium phthalimide are added with stirring. The mixture is then heated to 85-90 ° C. for 40 minutes, then cooled, diluted with chloroform and poured into water. After the chloroform layer was separated, the aqueous layer was extracted with additional amounts of chloroform. After evaporation of the chloroform, the product is obtained as an oily, crystalline substance.
After repeated recrystallization from isopropyl alcohol, the product melts at 125, 5-131, 5 C.

D. 5- (3-aminopropyl) -5H-dibenzo [a, d] cycloheptene:
2.15 g (0.0054 mol) of 5- (3-phthalimidopropyl) -5H-dibenzo [a, d] cycloheptene are dissolved in 50 ml of hot 95 0 / above ethanol, 0.6 ml of hydrazine hydrate is added and the mixture is heated Solution to reflux for 2 hours.
After cooling, it is acidified and the precipitated phthalhydrazide is filtered off. The ethanol from the filtrate is then evaporated off under reduced pressure and the residue is dissolved in 40 ml of water.

A solution of 5.67 g (0.025 mol) of 5-chloro-5H-dibenzo [a, d] cycloheptene in a mixture of dry ether and tetrahydrofuran is added dropwise to this solution while stirring, and the mixture is stirred for a further hour at room temperature. This product is obtained by the process according to Example 2B, but using 5- (3-ethoxypropyl) -H-dibenzo [a, d] cycloheptene as the starting material. The desired end product is obtained therefrom by applying the rest of the process according to Example 2.
