5-(3-Aminopropyl)-5H-dibenzo (a,d)cyclohepten
A. 5-(3-thoxypropyl- 5-oxy - 5H - dibenzo-(a,d)- cyclohepten: Das Grignardreagens bereitet man aus

These compounds can be used to treat mental illnesses because they are effective as an antidepressant agent or to cheer up the mind and as a psychic energizer. In such cases, the daily dose can be 5 to 250 mg, which is preferably taken in divided doses. These compounds are preferably administered in the form of their salts obtainable with acids.

The process according to the invention for the preparation of dibenzocycloheptenes of the formula indicated is now characterized in that a compound of the formula

making a compound of formula
EMI2.4
arises, which with a metal hydride or by catalytic hydrogenation to form a compound of the formula

is reduced.
The primary amine is accordingly formed by reaction with an alkoxypropyl Grignard compound on the carbon atom in the 5-position, optionally with simultaneous reduction of the oxy group; then replacement of the alkoxy group by iodine and formation of the amine by reaction with potassium phthalamide and subsequent action of hydrazine hydrate in absolute alcohol or elimination of the phthalic radical by hydrolysis. The Grignard adduct formed is preferably hydrolyzed under neutral conditions, e.g. B. using an ammonium chloride solution. The appropriate procedure is as follows: The product is isolated by evaporation of the solvent and treated in the presence of another suitable solvent, e.g. Acetic acid or acetic anhydride, with hydriodic acid.
Then the iodine compound is extracted, e.g. With benzene, and isolates them by evaporation of the latter.
The iodine compound is then reacted with potassium phthalimide in the presence of a solvent, e.g. B.
Dimethylformamide, and finally heated gently to accelerate the reaction and to complete it. The phthalimide adduct formed is isolated from the reaction mixture by extraction.
The subsequent treatment with hydrazine hydrate in absolute methanol or ethanol can, if desired, be carried out with heating to reflux.
The phthalhydrazide formed is then separated off and the primary amine is isolated from the solution. As mentioned, the phthalimide compound can also be converted into the amine by hydrolysis.
Now you bring the primary amine with a ketone or an aldehyde in a solvent, e.g. B. benzene or toluene, by heating at reflux to the reaction and thereupon, or after evaporation of the solvent, the remaining alkylideneaminopropyl compound with a metal hydride or by catalytic hydrogenation.
Instead of separating the alkylideneamino compound, the primary amine and the carbonyl compound can be dissolved in a suitable solvent, e.g. B. a lower alcohol, dissolved and then catalytically hydrogenated.
These methods are particularly suitable in cases where there are larger alkyl groups than methyl groups.
If a 5-halogen (preferably chlorine or bromine) derivative of 5H-dibenzo- (a, d) -cyclohepten is assumed, the reaction with the Grignard reagent in the presence of a conventional solvent, for. B. of dry ether or tetrahydrofuran. A slow addition reaction takes place, so that a substantial increase in temperature above room temperature is easily prevented.

Production of
5- (3-aminopropyl) -5H-dibenzo (a, d) cycloheptene
A. 5- (3-thoxypropyl-5-oxy - 5H - dibenzo- (a, d) - cycloheptene: Prepare the Grignard reagent
12.55 g (0.075 mol) 3-ethoxypropyl bromide and 1.82 g (0.075 mol) magnesium in ether. After the solution has been cooled in an ice bath, a solution of 5.15 g (0.025 mol) of 5H-dibenzo (a, d) cyclohepten-5-one in dry ether is added dropwise with stirring. The mixture is then stirred for 2 hours at room temperature and then for 30 minutes at reflux temperature.
The Grignard adduct is then decomposed by pouring the reaction mixture into a mixture of ice-ammonium chloride solution, separated from the ether layer, and the aqueous layer is extracted with further amounts of ether. After evaporation of the ether, the product is obtained as an oily crystalline substance. After recrystallization from alcohol, the product had a melting point of 129-130 C.

The mixture is then heated to 85-90 ° C. for 40 minutes, then cooled, diluted with chloroform and poured into water. After the chloroform layer was separated, the aqueous layer was extracted with additional amounts of chloroform. After evaporation of the chloroform, the product is obtained as an oily crystalline substance. After repeated recrystallization from isopropyl alcohol, the product melts at 125.5-131.5 "C.
Analysis for C26H2, O2N calculated: 82.30 0 / o C; 5.58 0 / o H; 3.69% N; found: 82.46 0 / o C; 5,600 / 0H; 3,700 / 0 N.

After cooling, it is acidified and the precipitated phthalhydrazide is filtered off. The ethanol from the filtrate is then evaporated off under reduced pressure and the residue is dissolved in 40 ml of water. The filtered aqueous solution is then evaporated under reduced pressure to a volume of approx. 20 ml. The hydrochloric acid salt of 5- (3-aminopropyl) -5H-dibenzo (a, d) cyclohepten precipitates on cooling in a yield of 1.35 g (87.5 o / o).
After recrystallization from an ethanol / ether mixture and from isopropyl alcohol, the analytical sample melts at 263-265 C with decomposition.
Analysis for Ct8Ht9N. HCl calculated: 75.64 / o C; 7.05 0 / o H; 4.90 0/0 N; found: 75.54 54 / o C; 6.77 o / o H; 4.78 0 in N.
