Procédé pour la préparation de 4-quinazolinones
La présente invention a pour objet la préparation de 1,2,3,4-tétrahydro-4-quinazolinones de formule générale:

atomes de carbone, 2) R1 et/ou 1L peuvent être également un radical
aralkyle. ayant jusqu'à 16 atomes de carbone, 3) et dans laquelle X représente un atome d'halo
gène, un radical alkyle ou un radical trihalo
méthyle.
Les quinazolinones préparées selon la présente invention sont des diurétiques et possèdent l'avantage d'opérer principalement comme agents ehloruré- tiques plutôt que comme excréteurs de bicarbonate.
Etant donné que l'acidose métabolique ne se développe pas dans ces conditions, on peut maintenir une diurèse beaucoup plus longue. En même temps les quinazolinones possèdent une faible toxicité et ne présentent pas les propriétés particulièrement indésirables des diurétiques mercuriels qui sont les agents chlorurétiques classiques de la technique antérieure.
Ces quinazolinones présentent un. avantage spécial étant donné qu'elles provoquent une faible potasse urésis lors de leur emploi.
Certains produits intermédiaires de la présente invention utilisés dans la préparation des 7-halogéno (ou 7-trihalogénométhyl- ou 7-allcyl)-6-sulfamyi- quinazolinones possèdent également une activité diurétique.
Lorsque les 1,2,3,4-tétrahydroquinazolinones de la présente invention possèdent des substituants différents sur l'atome de carbone en position 2 du noyau, le composé est produit sous forme de DL-racémate. Ces mélanges racématiques peuvent être dédoublés en leurs isomères D et L par les méthodes bien connues en utilisant des sels optiquement actifs. On peut prévoir que les isomères D et L ainsi isolés peuvent avoir différents ordres d'activité ou différents types d'aotivité. Les composés préférés sont les mélanges racémiques DL. Le coût élevé qu'entraîne la séparation rend l'isolement d'un iso- mère particulier D ou L indésirable.
On doit noter que les 1,2,3,4-tétrahydro-quinazolinones sont susceptibles d'exister sous une forme tautomère. Les membres typiques peuvent être représentés par la réaction d'équilibre suivante:

Pour plus de simplicité, dans les exemples et dans la description, les composés sont dormés sous leur forme cétonique plutôt que sous leur forme éno- lique.
Conformément à la présente invention, on fournit un procédé pour la préparation, des 4-quinazolinones répondant à la formule générale I donnée précédemment, qui est caractérisé en ce qu'on condense un dérivé de l'acide anthranilique de formule générale:

K représente NHR8, X, R, Rt, R2 et R3 dans lesdites formules ayant les significations susmentionnées.
Dans les exemples A à J sont indiqués les procédés typiques pour la préparation de certains composés intermédiaires utilisés dans le procédé de la prés ente invention.
Les données d'analyse à l'ultraviolet en unités m enregistrées dans les exemples ci-après sont mesurées sur des solutions du composé particulier dans des solutions de soude à 1/1 ON.
Dans les exemples suivants, les parties sont comptées en poids à moins d'indication contraire.
Préparation des produits intermédiaires
Exemple A
Préparation de la 5-chlor-4-sulfamyl-N-acétyltoluidine

On ajoute graduellement 17 parties de chlorure de sodium à 100 parties d'acide chlorosulfonique refroidi dans de la glace. La réaction s'amorce avec émission de HCl. On ajoute alors graduellement au mélange refroidi 58 parties de N-acéthyl-5-chlorotoluidine. A la fin de l'addition, on chauffe le mélange réactionnel jusqu'à 90-1000 C jusqu'à achèvement. Le dégagement de HCl se continue pendant une bonne partie de ce temps. Le mélange est ensuite refroidi et versé lentement sur 500 parties de glace, sous bonne agitation. Le produit brut obtenu est filtré et lavé à deux reprises par 50 parties d'eau glacée.
Au gâteau humide résultant, on ajoute sous agitation 500 parties d'une solution aqueuse d'am moniaque, la température étant portée à 90-1000 C.
A la fin de la réaction, on refroidit le mélange à la température ambiante et on le laisse au repos jusqu'à précipitation complète. Le précipité est ensuite séparé par filtration et le gâteau filtre est traité par 250 parties de NaOH(lN) puis on procède à la filtration et à la décoloration du filtrat par du charbon actif. Le filtrat est ensuite acidifié par de l'acide chlorhydrique concentré et le composé sulfonamide se sépare par précipitation.

On ajoute graduellement 40 parties du produit de l'exemple A à une solution, de 50 parties de permanganate de potassium dans 400 parties d'eau tiède. Le mélange est ensuite ohauffé à 90-1000 C jusqu'à la fin de la réaction.
Pendant la réaction, il se forme un précipité brun de MnO3 qui est séparé par filtration et lavé à 3 reprises par des portions de 50 parties de NaOH(lN). Les filtrats et les liqueurs de lavage combinés sont décolorés par du charbon actif et le filtrat résultant est acidifié à pH2.
Le produit solide précipité est l'acide 4whloro-5- sulfamyl-N-acétylan. thranilique.
Exemple C
Préparation de l'acide 4-chloro-5-sulfamyl
anthranilique

On dissout 10 parties du produit de l'exemple B dans un mélange tiède de 200 parties de HCL et 200 parties en volume d'éthanol. Le mélange est chauffé au reflux jusqu'à la fin de la réaction, puis refroidi et évaporé jusqu'à siccité sous pression réduite. Le produit solide obtenu est ensuite repris par 200 parties de NaOH(lN) décoloré par du charbon actif, et le filtrat est acidifié jusqu'à virage au rouge congo. Le produit qui se sépare est l'acide 4-chloro-5-sulfamylanthranilique qui peut être éventuellement purifié par recristallisation quoique sa purification ne soit pas nécessaire lorsqu'il est utilisé dans la production des quinazolones.
Exemple D
Deuxième méthode de préparation de l'acide 4-chloro-5-sulfamylanthranilique

On met en dispersion 10 parties du produit de l'exemple B dans 35 parties de NaOH aqueux (3N).
Le mélange est ensuite chauffé au reflux jusqu'à achèvement de la réaction. La solution est clarifiée par du charbon actif, filtrée et le filtrat est ensuite refroidi à 00 C. On obtient le sel sodique de l'acide 4-chloro-5-sulfamylanthranilique.
En vue de produire l'acide libre, on acidifie le sel de sodium par un acide minéral aqueux en utilisant du rouge congo comme indicateur. L1 se forme un précipité blanc sale qui est filtré et lavé à lJlusieurs reprises par de petites portions d'eau glacée. Le chlorure de sodium occlus est ainsi éliminé mais on ne doit effectuer le lavage qu'avec de très faibles quantités d'eau glacée, juste suffisantes pour dissoudre le dhlorure de sodium car l'acide luimême est légèrement soluble dans l'eau et l'utilisation de grandes quantités d'eau de lavage entraîne une perte de produit.
Exemple E
Préparation de l'acide 4-chloro-5-sulfamyl-N propionylanthranilique

On chauffe une partie de l'acide de l'exemple C avec 50 parties d'anhydride propionique à 90-100 C jusqu'à la fin de la réaction. Le mélange réactionnel est alors versé dans 200 parties d'eau glacée, sous agitation, l'eau glacée étant formée moitié de glace, moitié d'eau à peu près, il se forme deux couches qui se fusionnent ensuite et précipitent. On effectue la filtration et on lave le solide à l'eau froide pour donner l'acide 4-chloro-5-sulfamyl-N- propionylanthranilique.

Préparation de l'acide 4-bromo-5-sulfamyl
anthranilique

On répète le procédé de l'exemple A en remplaçant la N-acétyl-5-cllorotoluidine par un équivalent stoeshiométrique Ide N-acétyl-5¯bromotolu- idine préparée en faisant réagir la 5-hromo-o-tolu- idine avec de l'anhydride acétique. La Sbromo-4- sulfamyl-N-acétyltoluidine obtenue est utilisée dans le procédé de l'exemple B à la place du composé chloré correspondant. Toutefois, au lieu de 40 parties, on utilise 47 t/2 parties.
Le produit acétyle est ensuite hydrolysé de façon décrite à l'exemple C pour produire l'acide 4-bromo 5-sulfamyl anthranilique sous forme de cristaux blancs.

Préparation de l'acide 4-fluoro-5-sulfamyl-
anthranilique

On répète le procédé de l'exemple A en remplaçant la N-acétyl-5-cnlorotoluidine par un équivalent stoechiométrique de N-acétyl-5-fljuorotholuidine préparée en faisant réagir la 4-fluoro-o-toluidine avec de l'anhydride acétique. La 5-fluoro-4-sulfamyl
N-acétyltoluidine obtenue est utilisée dans le procédé d'oxydation de l'exemple B à la place du composé chloré correspondant. Le produit acétyle est ensuite hydrolysé de façon décrite à l'exemple C pour produire l'acide 4-41uoro-5-sulfamylanthranilique sous forme de cristaux blancs.

On dissout la 2-carbamyl-5-chloro-4-sulfamylaniline dans 60 parties d'éther dîméthylique du diéthylène-glycol, on ajoute un excès d'acétone, puis 4 parties d'acide chlorhydrique concentré. Après reflux pendant 3 heures, on porte la solution à siccité et on dissout le solide résultant dans une base et on le reprécipite par un acide minéral. Il se forme des cristaux blancs de 7-chloro-2,2-diméthyl-6-sulfamyl1,2,3,4-tétrahydro-4-quinazolinone.

Préparation de la 7-chloro-2-méthyl-1,2,3,4-
tétrahydro-sulfonamide-4-oxo-quinazoline

On chauffe au reflux, pendant une heure, une solution de 0,50 g (0,002 mole) de 4-chloro-5-sul- famyl-anthrainilamide dans 50 cm3 de diéthylène-gly- col-diméthyl-éther contenant 1 goutte de HCl concentré avec 0,32 cm3 (0,002 mole) d'acétaldéhydediéthylacétal. On évapore ensuite la solution et obtient 0,40 g d'un produit solide qu'on recristallise dans 50 % d'acétone aqueux. On obtient ainsi un rendement de 45 /o de produit fondant à 282-2830 C.

The present invention relates to the preparation of 1,2,3,4-tetrahydro-4-quinazolinones of general formula:

The quinazolinones prepared according to the present invention are diuretics and have the advantage of working primarily as hydrochloride agents rather than as bicarbonate excretors.
Since metabolic acidosis does not develop under these conditions, a much longer diuresis can be maintained. At the same time the quinazolinones have low toxicity and do not exhibit the particularly undesirable properties of the mercurial diuretics which are the conventional chloruretic agents of the prior art.
These quinazolinones exhibit a. special advantage given that they cause low potash uresis during their use.
Certain intermediates of the present invention used in the preparation of 7-halo (or 7-trihalogenomethyl- or 7-allcyl) -6-sulfamyinequinazolinones also possess diuretic activity.
When the 1,2,3,4-tetrahydroquinazolinones of the present invention have different substituents on the carbon atom at the 2-position of the ring, the compound is produced as a DL-racemate. These racematic mixtures can be resolved into their D and L isomers by well known methods using optically active salts. Provision may be made for the D and L isomers thus isolated to have different orders of activity or different types of activity. Preferred compounds are the racemic DL mixtures. The high cost of separation makes isolation of a particular D or L isomer undesirable.
It should be noted that 1,2,3,4-tetrahydro-quinazolinones are likely to exist in a tautomeric form. Typical members can be represented by the following equilibrium reaction:

In accordance with the present invention, there is provided a process for the preparation of 4-quinazolinones corresponding to the general formula I given above, which is characterized in that an anthranilic acid derivative of general formula is condensed:

Typical processes for the preparation of certain intermediates used in the process of the present invention are shown in Examples A to J.
The ultraviolet analysis data in m units recorded in the examples below are measured on solutions of the particular compound in 1: 1N sodium hydroxide solutions.
In the following examples, parts are counted by weight unless otherwise indicated.
Preparation of intermediate products
Example A
Preparation of 5-chlor-4-sulfamyl-N-acetyltoluidine

17 parts of sodium chloride are gradually added to 100 parts of ice-cold chlorosulfonic acid. The reaction begins with emission of HCl. 58 parts of N-acetyl-5-chlorotoluidine are then gradually added to the cooled mixture. At the end of the addition, the reaction mixture is heated to 90-1000 ° C. until complete. The evolution of HCl continues for a good part of this time. The mixture is then cooled and poured slowly onto 500 parts of ice, with good stirring. The crude product obtained is filtered and washed twice with 50 parts of ice-cold water.
To the resulting wet cake are added with stirring 500 parts of an aqueous solution of ammonia, the temperature being raised to 90-1000 C.

During the reaction, a brown precipitate of MnO3 forms which is filtered off and washed 3 times with 50 parts portions of NaOH (1N). The combined filtrates and washings are decolorized with activated charcoal and the resulting filtrate is acidified to pH2.
The solid product precipitated is 4whloro-5-sulfamyl-N-acetylan acid. thranilic.
Example C
Preparation of 4-chloro-5-sulfamyl acid
anthranilic

10 parts of the product of Example B are dissolved in a warm mixture of 200 parts of HCL and 200 parts by volume of ethanol. The mixture is heated at reflux until the end of the reaction, then cooled and evaporated to dryness under reduced pressure. The solid product obtained is then taken up in 200 parts of NaOH (1N) decolorized with activated charcoal, and the filtrate is acidified until turning to congo red. The product which separates is 4-chloro-5-sulfamylanthranilic acid which can optionally be purified by recrystallization although its purification is not necessary when used in the production of quinazolones.
Example D
Second method of preparing 4-chloro-5-sulfamylanthranilic acid

10 parts of the product of Example B are dispersed in 35 parts of aqueous NaOH (3N).
The mixture is then heated to reflux until the reaction is complete. The solution is clarified with activated carbon, filtered and the filtrate is then cooled to 00 ° C. The sodium salt of 4-chloro-5-sulfamylanthranilic acid is obtained.
In order to produce the free acid, the sodium salt is acidified with an aqueous mineral acid using congo red as an indicator. L1 forms a dirty white precipitate which is filtered and washed several times with small portions of ice water. The occluded sodium chloride is thus removed but the washing should only be carried out with very small quantities of ice water, just sufficient to dissolve the sodium chloride since the acid itself is slightly soluble in water and using large amounts of wash water results in product loss.

The 2-carbamyl-5-chloro-4-sulfamylaniline is dissolved in 60 parts of diethyl ether of diethylene glycol, an excess of acetone is added, then 4 parts of concentrated hydrochloric acid. After refluxing for 3 hours, the solution is brought to dryness and the resulting solid is dissolved in a base and reprecipitated with mineral acid. White crystals of 7-chloro-2,2-dimethyl-6-sulfamyl1,2,3,4-tetrahydro-4-quinazolinone form.

A mixture of 5 parts of 4-chloro-5-sulfamyl-anthranil-amide, 2 parts of propionacetal and 50 parts of diethylene glycol dimethyl ether containing one part of diethylene glycol is heated in a water bath with stirring for 1-2 hours. 'HCl gas, dissolved. The reaction mixture is then concentrated to dryness in vacuo and the solid residue is recrystallized from aqueous acetone to give 2-ethyl-7-chloro-6-sulfamyl1,2,3,4-tetrahydro-4-quinazolinone, mp. 248-250.

tetrahydro-sulfonamide-4-oxo-quinazoline

A solution of 0.50 g (0.002 mol) of 4-chloro-5-sul-famyl-anthrainilamide in 50 cm3 of diethylene-glycol-dimethyl-ether containing 1 drop of HCl is refluxed for one hour. concentrated with 0.32 cm3 (0.002 mol) of acetaldehyde diethyl acetal. The solution is then evaporated to obtain 0.40 g of a solid product which is recrystallized from 50% aqueous acetone. This gives a yield of 45 / o of product melting at 282-2830 C.

By substituting 1.0 g of paraformaldehyde instead of the acetal used according to Example 4, a product is obtained in a yield of 50%. This product melts at 254-2560 C.
