Préparation de nouveaux dérivés de l'acide salicylique
La présente invention concerne un procédé de préparation de nouveaux dérivés de l'acide salicylique de formule générale :

La réaction est exécutée de préférence en présence d'un solvant choisi du groupe comprenant les carbures aromatiques et les alcools à poids molécu- laires bas à une température comprise entre 600 C et 135 C et en présence d'un agent alcalin, accepteur d'hydracide, choisi du groupe comprenant le carbo- nate de potasse, la triéthylamine ou la pipérazine Nmono-substituée employée.
Les composés obtenus par le procédé selon l'in- vention sont, par suite : -dans le cas où Y = H : des dérivés de l'acide sali
cylique et ses esters ; -dans le cas où Y = CH3 : des dérivés des acides
ortho et méta crésotiniques, de l'acide méthyl-6
salicylique et leurs esters ; -dans le cas où Y = OH : des dérivés des acides
gentisiques (acide dihydroxy-2, 5 benzoïque);p
et y résorcyliques (acides dihydroxy-2, 4 et 2, 6
benzoïques) pyrocatéchique (acide dihydroxy-2, 3
benzoïques) et leurs esters.

L'invention sera mieux comprise grâce aux exem- ples qui suivent.
Dans ces exemples, la températures sont données en degrés Celsius. Les points de fusion ont été déterminés à la microplatine chauffante de Kofler.
Ces nouveaux dérivés peuvent être éventuellement purifiés par des méthodes physiques telles que : cristallisation, distillation, chromatographie, ou chimiques, telles que : formation de sels, cristallisation de ceux-ci et décomposition en milieu alcalin.
Les sels d'addition des composés qui sont des bases fortes peuvent être obtenus par action des nouveaux dérivés sur des acides dans des solvants appropriés ; comme solvants organiques, on utilise des alcools, éthers, cétones ; comme solvant minéral, on utilise avec avantage l'eau ; comme acides utilisés pour la formation de ces sels d'addition, on peut citer dans la série minérale : les acides chlorhydrique, bromhydrique, sulfurique, méthane-sulfonique, phosphorique ; dans la série organique : les acides acétique, maléique, fumarique, oxalique, tartrique, citrique, benzoïque, etc.
Enfin, certains de ces corps étant des esters et des acides, il est facile de passer de l'ester à l'acide correspondant par hydrolyse et, réciproquement, de l'acide à l'ester correspondant par estérification : par exemple, l'hydrolyse de 7, 8 g de (méthoxy-2'phényl)-1 (acétoxy-3'carbométhoxy-4'benzyl)-4 pipérazine, par ébullition une heure dans 37 cm3 d'acide chlorhydrique à 4"/o, donne, après cristallisation dans l'eau, 6, 8 g de dichlorhydrate de (méthoxy-2"phényl)-1 (hy droxy-3'carbométhoxy-4'benzyl)-4 pipérazine, fondant à 215-217 C. Rendement : 80 /o.
Exemple 1 :
(Hydroxy-4'carbométhoxy-3'benzyl)-l carbobenzoxy-4-pipérazine

A une solution de-60 g de carbobenzoxy pipérazine dans 300 cm de toluène anhydre contenant 27, 5 g de triéthylamine, on ajoute en 5 minutes, à 600C, 65, 7g de chlorure de carbométhoxy-3 hydroxy-4 benzyle fondant à 600 C. Puis on porte le mélange à reflux pendant 5 heures.
Au bout de ce temps, on refroidit et ajoute 700 cm3 d'eau pour dissoudre le chlorhydrate de triéthylamine qui a précipité ; on décante et extrait plusieurs fois les solutions toluéniques avec de l'acide méthane sulfonique à 10 /o. Les solutions acides sont réunies et lavées à l'éther. On alcalinise ensuite avec du carbonate de potassium et extrait la base huileuse avec du chloroforme.
Après séchage sur carbonate de potassium anhydre et évaporation du solvant, on-obtient 80 g-de-base titrant 99 /o de la théorie et dont l'oxalate acide préparé au sein de l'éthanol fond à 172 C.-
Le chlorure de carbométhoxy-3 hydroxy-4 benzyle de départ (Eb/0, 9 = 126 C, F = 63-65O C) a été préparé par chlorométhylation du salicylate de méthyle, au sein de l'acide chlorhydrique en présence de méthylal selon le brevet allemand Bayer No 113723 du 15 janvier 1899.
Selon le même procédé, ont été préparés : a) (Hydroxy-4'carboxy-3'benzyl)-l carbobenzoxy-4 pipérazine : e.'

Exemple 2 : Carbométhoxy-3'hydroxy-4'benzyl)-l pipérazine

A une suspension de 17 g d'hexahydrate de pipérazine dans 100 cm3 de méthanol, on ajoute 7, 4 cm3 d'acide chlorhydrique concentré (d = 1, 19) de façon à former le monochlorhydrate de pipérazine ; cette formation s'accompagne de dissolution.
A la solution obtenue, on ajoute, à 70O C, 9 g de chlorure de carbométhoxy-3 hydroxy-4 benzyle, en 5 minutes, puis on chauffe le mélange, pendant une heure, à 70O C, il se forme un abondant précipité de dichlorhydrate de pipérazine.
On refroidit ensuite, filtre le sel et chasse le solvant sous pression réduite ; le résidu huileux obtenu est acidifié à pH 2, avec de l'acide chlorhydrique à 20"/o ; extrait la solution plusieurs fois à l'éther et alcalinise fortement avec du carbonate de potassium. La solution alcaline saturée est extraite plusieurs fois au chloroforme, et le chloroforme séché sur CO3K2 anhydre.
Après évaporation du solvant sous pression réduite, on obtient 8, 75 g de base huileuse brute dont le dichlorhydrate préparé au sein de l'isopropanol cristallisé sous forme hydratée :

A une solution de 25 g de pipérazine anhydre dans 750 cm3 de benzène sec, on ajoute, en 10 minutes, à une température de 16 C, une solution de 23, 5 g de chlorure de (carbométhoxy-3 acétoxy-4) benzyle dans 100 cm3 de benzène sec. Il se forme un abondant précipité ; on laisse réagir 3 heures, à une température comprise entre 15 C et 250 C.
Au bout de ce temps, on filtre les cristaux de chlorhydrate de pipérazine formé, on lave la solution benzénique, à l'eau, puis extrait plusieurs fois, à température de 10 C, avec de l'acide méthane sulfonique à 10 /o. La solution acide, lavée à l'éther, est ensuite alcalinisée au CO3K2 et la solution alcaline extraite plusieurs fois à l'éther. Après séchage de l'éther sur CO3K2 et évaporation du solvant sous pression réduite, on recueille 11 g de base huileuse brute dont le dichlorhydrate préparé au sein de l'éthanol, fond à 212 C (déc.).
Le chlorure de départ (Eb/0, 140-146 C-K= 1, 5292) a été préparé par acétylation du chlorure de (carbométhoxy-3 hydroxy-4) benzyle au moyen de l'anhydride acétique, en présence d'acide sulfurique, à 700 C.
Les dérivés obtenus selon la présente invention sont intéressants en raison de leurs propriétés pharmacologiques et thérapeutiques remarquables. En particulier, leur activité anti-inflammatoire et analgésique est importante.
Expérimenté sur l'oedème de la patte de rat, provoqué par le kaolin, l'action inhibitrice de l'inflammation de certains de ces dérivés atteint 60 /o et est, à dose égale, plus importante que celle de la phénylbutazone.
L'action alnalgésique mesurée par la méthode de la plaque chauffante et exprimée en pourcentage d'augmentation du temps de léchage par rapport aux témoins, montre un accroissement atteignant 100 zozo pour certains dérivés.
En outre, la toxicité des produits obtenus par le procédé objet de l'invention étant faible : la DL 50 par voie intrapéritonéale chez la souris varie entre 218 et 789 mg/kg, la marge thérapeutique est assez large.

In these examples, temperatures are given in degrees Celsius. Melting points were determined using a Kofler microplatinum heater.
These new derivatives can optionally be purified by physical methods such as: crystallization, distillation, chromatography, or chemical methods, such as: formation of salts, crystallization of the latter and decomposition in an alkaline medium.
The addition salts of the compounds which are strong bases can be obtained by the action of the new derivatives on acids in suitable solvents; as organic solvents, alcohols, ethers, ketones are used; as inorganic solvent, water is advantageously used; as acids used for the formation of these addition salts, there may be mentioned in the mineral series: hydrochloric, hydrobromic, sulfuric, methanesulphonic and phosphoric acids; in the organic series: acetic, maleic, fumaric, oxalic, tartaric, citric, benzoic acids, etc.
