La présente invention a pour objet un procédé pour préparer des dérivés des 1,1-bioxydes de 7-sulfamyl- 1,2,4-benzothiadiazine substituée en 3 ayant la formule générale

dans laquelle A et Z ont la signification ci-dessus et X représente de l'halogène, avec un mercaptan ; si l'on désire obtenir le sulfoxyde ou le sulfone, on oxyde le produit de réaction.
La condensation avec le mercaptan s'effectue, de préférence, en présence d'une base. Il est :préférable de l'effectuer à une température de 20 à l20 C, envi ron, pendant 1 à 12 heures, et de préférence, en pré sence d'au moins un équivalent molaire d'un hydroxyde d'un métal alcalin. ou alcalino-ferreux, d'une amine ter tiaire dont l'action est stériquement retardée, ou d'un carbonate ou bicarbonate :d'un métal alcalin, ou alca- lino-ferreux.
Des amines tertiaires, dont l'action est stériquement retardée, sont, par exemple, les pyridines mono-, di- et tri-substituées comme la 2,4,6-collidine, la lutidine, la 2-chloropyridine, la 2,4-dichloropyridine et analogues, de même que certaines trialcoylamines. Un hydroxyde métallique est utilisé dans les meilleures conditions sous forme d'une solution aqueuse contenant généralement entre environ 5 et environ 20% en poids d'hydroxyde, mais on préfère -de 5 à 15 0/0, car on obtient de meil leurs rendements.
Bien que la réaction progresse d'une manière satisfaisante en solution :aqueuse, l'addition d'un solvant organique facilite notablement la produc tion :des composés désirés, en établissant un contact plus intime entre les réactifs qui ont une solubilité limi tée dans l'eau.
Les solvants organiques qui conviennent à cet effet sont les cétones. tels que L'acétone et l'éthyl- méthyl-cétone, les alcanols inférieurs tels que le métha nol, l'éthanol et les propanols, mais de préférence, le diméthylformamide et :des formamides faiblement alcoy- lés analogues.
Bien qu'on ait constaté qu'une proportion équi moléculaire des réactifs fournisse des rendements appré ciables en produit, on préfère généralement utiliser un excès de thiol pour obtenir des .rendements meilleurs. Un excès atteignant jusqu'à environ 40 moles en % est particulièrement utile, alors que des excès plus grands, bien qu'ils puissent être adoptés, ne procurent pas un avantage appréciable.
La réaction peut avantageusement avoir lieu à des températures comprises entre 20 et 1200 :pendant envi ron 1 à environ 12 heures. Le chauffage a des tempé ratures plus élevées et pendant des durées plus longues donne lieu à un rendement réduit en produit désiré.
Quand la réaction est terminée, le produit peut être isolé par les méthodes usuelles. Par exemple, le produit se sépare du mélange de réaction par refroidissement et il est séparé et purifié par recristallisation dans des solvants appropriés tels que l'acétone, les alcanols infé rieurs, les mélanges acétone-éther, les mélanges acétone- alcanol et analogues.
La conversion des agents thérapeutiques en question dans lesquels, Y est S, en produits dans lesquels Y est SO ou S02 :

en peroxyde :d'hydrogène peuvent être adoptés. Des permanganates de métaux alcalins peuvent égale ment être utilisés comme agents oxydants, mais leur usage peut donner lieu à :des rendements inférieurs en produit désiré.
La réaction peut avantageusement se faire :dans un solvant tel qu'un acide alcanoïque infé rieur, par exemple l'acide acétique. D'autres solvants miscibles dans l'eau peuvent avantageusement être uti- lisés pour ce procédé d'oxydation, par exemple des alcanols inférieurs tels que l'éthanol, le méthanol, le propanol,
ou des cétones tels que l'acétone et l'éthyl- méthyl-cétone. Ces composés sont préparés en utilisant au moins des quantités stochiométriques de peroxyde d'hydrogène. Comme celui-ci a tendance à se décom poser, il est bon d'utiliser un excès de peroxyde d'hy drogène.
Evidemment et comme bien connu par les spécialistes. dans ce domaine, on doit éviter de se servir de l'agent oxydant en excès :extrêmement grands pour préparer les composés en question dans lesquels Y est SO pour réduire nu minimum la coproduction des com posés. dans. .lesquels Y est S02. Une méthode particu lièrement avantageuse pour préparer ces premiers pro duits consiste à faire réagir :
la substance avec un excès d'environ 100% en peroxyde d'hydrogène à la tempé rature ambiante (20 a 300), mais des températures infé rieures peuvent être utilisées, par exemple des tempé ratures aussi basses que 00.
L'usage de températures plus élevées peut réduire, d'une manière appréciable, le rendement en produit désiré et est donc de préférence évité. La préparation des composés dans lesquels Y est S02 peut se faire avantageusement en utilisant un grand excès :de peroxyde, par exemple un excès de 200 à 400% ou plus. Le chauffage du mélange de réaction est particulièrement utile car il diminue nota blement la durée de la réaction et permet d'obtenir une conversion complète.
Il est évident que ces derniers composés. peuvent également être préparés à partir de ceux dans lesquels Y est SO et qui forment un étage d'oxydation intermédiaire. Quand la réaction est ter minée,. le produit est obtenu par les méthodes usuelles, par exemple par concentration et précipitation.
Les agents thérapeutiques, préparés par le procédé selon l'invention, possèdent plusieurs des propriétés exi gées d'un diurétique idéal de sorte qu'ils sont des com posés -de valeur pour leur application thérapeutique.
Des examens sur des rats ont démontré que des composés représentatifs, préparés selon le nouveau pro cédé sont des diurétiques plus efficaces que le chloro- thiazide. Ces composés se sont montrés supérieurs au chloro- thiazide sur la base d'une courbe à 3 points .respective à la dose.
L'efficacité relative des composés est indi quée par la valeur RP dans le tableau I, laquelle est de 0,13 pour le chlorothiazide. (Les composés restants du tableau II se sont montrés supérieurs au chloro- thiazide sur la base :

Ces agents, peuvent se présenter sous forme de sels d'acides .et de bases pharmacologiquement acceptables. Des exemples d'anions pharmacologiquement accepta bles sont .l'iodure, le chlorure, le bromure, le sulfate, le méthylsulfate, l'acétate, le propionate, le tartrate, le citrate, le gluconate, etc.
Les benzothiadiazines de départ peuvent être obte nues à partir de 2,4-disulfamylanilines substituées en 5, qui peuvent être préparées par des méthodes décrites dans. la littérature technique, par exemple par la méthode générale, décrite dans les Monatsch. Chem.
Vol. 48, page 87 (1927), qui consiste à traiter une aniline métasubstituée avec 10 à 20 .parties en poids d'acide chlorosulfonique, après quoi on ajoute graduel lement environ 90 à environ 170 .parties en poids de chlorure de sodium. Le mélange obtenu est chauffé à environ 1500 pendant environ 2 heures, après quoi le mélange de réaction est versé dans l'eau et le chlorure de 2,4-disulfonylaniline substituée en 5 est filtré et traité ensuite avec de l'hydroxyde d'ammonium concentré par les méthodes usuelles pour obtenir le disulfonamide correspondant.
L'haloacylanilide substituée obtenue est ensuite transformée en bioxyde de benzothiadiazine 3-(halo- alcoyle)-substituée.
Les exemples ci-dessous illustrent l'invention. Exemple 1 Préparation du 1,1-bioxyde de 3-benzylthiométhyl- 6-chloro-7-sulfamyl-1,2,4-benzothiadiazine Un mélange .de 7,24 g (0,02 mole) de 5-chloro-2,4- disulfamyl (chloro)acétanilide et de 2,98 g (0,024 mole) de benzylmercaptan dans 20 ml d'une solution à 10 d'hydroxyde de sodium, à laquelle on ajoute 20 ml de diméthylformamide,

Le précipité formé est filtré et recristallisé dans l'acétone. Le produit fond à 231- 232 . L'analyse élémentaire du C15H14N3O4S3Cl donne, par calcul, C = 41,71 ; H = 3,3 ; N = 9,7.
On trouve C=41,2 et H=3,3.
On prépare le chlorhydrate du 1,1 bioxyde de 3 - benzyléthiométhyl- 6 -chloro-7- sulfamyl-1,2,4-benzo- thiadiazine en dissolvant ce composé dans une solution aqueuse contenant une quantité molaire équivalente d'acide chlorhydrique. Ensuite,
on concentre la solution sous une pression réduite pour obtenir le chlorhydrate correspondant. On recourt à ce procédé pour préparer d'autres sels acides en utilisant les acides bromhydrique, iodhydrique et sulfurique. On obtient des résultats com parables, en répétant le procédé avec une solution de chlorure d'hydrogène dans de l'éthanol.
Le sel de potassium de 1,1-bioxyde de 3-benzyléthio- méthyl-6-chloro-7-sulfamyl-1,2,4-benzo@thiadiazine est préparé en dissolvant ce composé dans une solution aqueuse contenant une quantité molaire équivalente de carbonate de potassium. La solution qui en résulte est concentrée sous, une pression réduite pour obtenir le sel de potassium.
On répète ce procédé en utilisant du bicarbonate de sodium aqueux, @de l'hydroxyde de baryum et de l'hydroxyde @de sodium alcoolique pour obtenir les sels métalliques correspondants.

5g du produit de l'exemple I dans 25 ml d'acétone et on traits le mélange avec 2,5 mi d'uns solution aqueuse à 30% de peroxyde d'hydro gène dans 10 ml d'acide acétique glacial. Le mélange est agité pendant 7 heures, après quoi on le laisse au repos pendant 8 heures. Le précipité cristallisé qui se forme est filtré, lavé avec -de l'acétone et ensuite avec de l'eau, après quoi il est séché. La recristallisation dans un mélange méthanol-acétone donne un produit qui fond à 2250.
L'analyse élémentaire du C15H14N305S3Cl donne, par calcul, C = 40,22 ; H = 3,15 ; N = 9,38. On trouve C = 40,1 ; H = 3,1 ; N = 9,1.
Exemple III Préparation du 1,1-bioxyde de 3-benzylsulfonyl- méthyl - 6- chloro -7-sulfamyl -1,2,4-benzothiadiazine On dissout 3,5 g du produit de l'exemple I dans 25 ml d'acétone et on traite le mélange avec 5,0 ml d'une solution aqueuse à 30% de peroxyde d'hydro gène dans 20 ml d'acide acétique glacial. Le mélange obtenu est chauffé sur un bain de vapeur pendant 4 heures,
après quoi on obtient le produit par concen- tration. La recristallisation dans un mélange méthanol- acétone -donne un produit fondant à 299-300 (avec dé composition). L'analyse élémentaire du C15H14N306S3C1 donne, par calcul, C = 38,83 ; H = 3,04 ; N = 9,06. On trouve C =<B>38,36;</B> H = 3,03 et N = 8,80.
Exemple 1V Le chlorhydrate de 1,1-bioxyde de 3-p-amino- phénylthiométhyl- 6 - chloro -7- sulfamyl-1,2,4-benzothia- diazine est dissous dans l'eau à laquelle on ajoute ensuite une quantité molaire équivalente d'acétate d'ar gent. Le précipité de chlorure d'argent, qui se forme, est filtré et le filtrat est concentré sous pression réduite pour obtenir le sel d'addition de l'acide acétique.
D'une manière analogue, on prépare les sels d'ad dition de l'acide tartrique, citrique, gluconique et pro- pionique.
Exemple V A - On chauffe 3 g du 5-chloro-2,4-disulfamyl(α- chloro)acétanilide jusqu'à la température de fusion (env. 2360. En à peu près 5 minutes, le composé initias liqué fié se solidifie et forme une matière solide beige clair (2,7 g). Le 1,1-bioxyde de 3-(chlorométhyl)-6-chloro-6- sulfamylbenzothiadiazine obtenu .est recristallisé par pré cipitation dans une solution de carbonate de sodium avec du HCl dilué.
B - Un mélange de 1,1-bioxyde de 3-chlorométhyl-6- chloro-7-sulfamylbenzothiadiazine (0,02 mole) et de benzylmercaptan (0,024 mole) est agité dans 20.m1 d'une solution à 10 % d'hydroxyde de sodium et 20 ml de diméthylformamide à la température ambiante pendant six heures. Après avoir été chauffé pendant 10 minutes au bain de vapeur, le mélange est refroidi et acidifié avec du HCl 6N. Le précipité est du. 1,1-bioxyde de 3-benzylthiométhyl-6-chloro-7-sulfamyl-1,2,4-benzo- thiadiazine.

The condensation with the mercaptan is preferably carried out in the presence of a base. It is preferable to carry it out at a temperature of about 20 to 120 ° C for 1 to 12 hours, and preferably in the presence of at least one molar equivalent of an alkali metal hydroxide. or alkaline-ferrous, of a tertiary amine whose action is sterically delayed, or of a carbonate or bicarbonate: of an alkali metal, or alkaline-ferrous.
Tertiary amines, the action of which is sterically delayed, are, for example, the mono-, di- and tri-substituted pyridines such as 2,4,6-collidine, lutidine, 2-chloropyridine, 2,4 -dichloropyridine and the like, as well as some trialkylamines. A metal hydroxide is best used in the form of an aqueous solution generally containing between about 5 and about 20% by weight of hydroxide, but 5 to 15% by weight is preferred because the better yields are obtained. .
Although the reaction progresses satisfactorily in aqueous solution, the addition of an organic solvent significantly facilitates the production of the desired compounds, by establishing more intimate contact between the reactants which have limited solubility in water. 'water.
Organic solvents which are suitable for this purpose are ketones. such as acetone and ethyl methyl ketone, lower alkanols such as methanol, ethanol and propanols, but preferably dimethylformamide and analogous weakly alkylated formamides.
Although it has been found that an equi molecular proportion of the reactants will provide appreciable yields of product, it is generally preferred to use an excess of thiol to obtain better yields. An excess of up to about 40 mole% is particularly useful, while larger excesses, although they can be adopted, do not provide an appreciable advantage.
The reaction can advantageously take place at temperatures between 20 and 1200: for about 1 to about 12 hours. Heating at higher temperatures and for longer times results in a reduced yield of desired product.
When the reaction is complete, the product can be isolated by the usual methods. For example, the product separates from the reaction mixture on cooling and is separated and purified by recrystallization from suitable solvents such as acetone, lower alkanols, acetone-ether mixtures, acetone-alkanol mixtures and the like.

The reaction can advantageously be carried out: in a solvent such as a lower alkanoic acid, for example acetic acid. Other solvents miscible in water can advantageously be used for this oxidation process, for example lower alkanols such as ethanol, methanol, propanol, etc.
or ketones such as acetone and ethyl methyl ketone. These compounds are prepared using at least stochiometric amounts of hydrogen peroxide. As this tends to decompose, it is good to use an excess of hydrogen peroxide.
Obviously and as well known by specialists. in this field, one should avoid using the oxidizing agent in excess: extremely large to prepare the compounds in question in which Y is SO to minimize the co-production of the compounds. in. .which Y is S02. A particularly advantageous method for preparing these first products consists in reacting:

The use of higher temperatures can appreciably reduce the yield of the desired product and is therefore preferably avoided. The preparation of the compounds in which Y is SO 2 can advantageously be carried out using a large excess of peroxide, for example an excess of 200 to 400% or more. Heating the reaction mixture is particularly useful because it significantly shortens the reaction time and allows complete conversion to be achieved.
It is obvious that these latter compounds. can also be prepared from those in which Y is SO and which form an intermediate oxidation stage. When the reaction is complete ,. the product is obtained by the usual methods, for example by concentration and precipitation.
The therapeutic agents, prepared by the process according to the invention, possess several of the properties required of an ideal diuretic so that they are valuable compounds for their therapeutic application.
Tests on rats have shown that representative compounds prepared according to the new process are more effective diuretics than chlorothiazide. These compounds were shown to be superior to chlorothiazide based on a dose-sensitive 3 point curve.
The relative efficacy of the compounds is indicated by the RP value in Table I, which is 0.13 for chlorothiazide. (The remaining compounds in Table II were shown to be superior to chlorothiazide based on:

These agents can be provided in the form of salts of pharmacologically acceptable acids and bases. Examples of pharmacologically acceptable anions are iodide, chloride, bromide, sulfate, methylsulfate, acetate, propionate, tartrate, citrate, gluconate, etc.
The starting benzothiadiazines can be obtained from 5-substituted 2,4-disulfamylanilines, which can be prepared by methods described in. technical literature, for example by the general method, described in Monatsch. Chem.
Flight. 48, page 87 (1927), which involves treating a metasubstituted aniline with 10 to 20 parts by weight of chlorosulphonic acid, after which about 90 to about 170 parts by weight of sodium chloride are gradually added. The resulting mixture is heated at about 1500 for about 2 hours, after which the reaction mixture is poured into water and the 5-substituted 2,4-disulfonylaniline chloride is filtered and then treated with ammonium hydroxide. concentrated by the usual methods to obtain the corresponding disulfonamide.
The resulting substituted haloacylanilide is then converted to 3- (haloalkyl) -substituted benzothiadiazine dioxide.

3-Benzylethiomethyl-6 -chloro-7-sulfamyl-1,2,4-benzothiadiazine 1,1 hydrochloride is prepared by dissolving this compound in an aqueous solution containing an equivalent molar amount of hydrochloric acid. Then,
the solution is concentrated under reduced pressure to obtain the corresponding hydrochloride. This process is used to prepare other acidic salts using hydrobromic, hydroiodic and sulfuric acids. Comparable results are obtained by repeating the process with a solution of hydrogen chloride in ethanol.
3-Benzylethio-methyl-6-chloro-7-sulfamyl-1,2,4-benzo @ thiadiazine 1,1-dioxide potassium salt is prepared by dissolving this compound in an aqueous solution containing an equivalent molar amount of potassium carbonate. The resulting solution is concentrated under reduced pressure to obtain the potassium salt.
This process is repeated using aqueous sodium bicarbonate, barium hydroxide and alcoholic sodium hydroxide to obtain the corresponding metal salts.

5 g of the product of Example I in 25 ml of acetone and the mixture is treated with 2.5 ml of a 30% aqueous solution of hydrogen peroxide in 10 ml of glacial acetic acid. The mixture is stirred for 7 hours, after which it is allowed to stand for 8 hours. The crystalline precipitate which forms is filtered off, washed with acetone and then with water, after which it is dried. Recrystallization from a methanol-acetone mixture gives a product which melts at 2250.
Elemental analysis of C15H14N305S3Cl gives, by calculation, C = 40.22; H = 3.15; N, 9.38. We find C = 40.1; H = 3.1; N, 9.1.
Example III Preparation of 3-benzylsulfonyl-methyl-6-chloro -7-sulfamyl -1,2,4-benzothiadiazine 1,1-dioxide 3.5 g of the product of Example I are dissolved in 25 ml of acetone and the mixture is treated with 5.0 ml of a 30% aqueous solution of hydrogen peroxide in 20 ml of glacial acetic acid. The mixture obtained is heated on a steam bath for 4 hours,
after which the product is obtained by concentration. Recrystallization from a methanol-acetone mixture gives a product melting at 299-300 (with decomposition). Elemental analysis of C15H14N306S3C1 gives, by calculation, C = 38.83; H = 3.04; N, 9.06. We find C = <B> 38.36; </B> H = 3.03 and N = 8.80.
Example 1V 3-p-Amino-phenylthiomethyl-6-chloro -7-sulfamyl-1,2,4-benzothia-diazine 1,1-dioxide hydrochloride is dissolved in water to which a molar amount is then added. equivalent of silver acetate. The precipitate of silver chloride, which forms, is filtered and the filtrate is concentrated under reduced pressure to obtain the addition salt of acetic acid.
In a similar manner, the addition salts of tartaric, citric, gluconic and propionic acid are prepared.
Example VA - 3 g of 5-chloro-2,4-disulfamyl (α - chloro) acetanilide are heated to melting temperature (approx. 2360. In about 5 minutes, the liquefied initias compound solidifies. and formed a light beige solid (2.7 g). The obtained 3- (chloromethyl) -6-chloro-6-sulfamylbenzothiadiazine 1,1-dioxide was recrystallized by precipitation from sodium carbonate solution with sodium carbonate. Dilute HCl.
B - A mixture of 3-chloromethyl-6-chloro-7-sulfamylbenzothiadiazine 1,1-dioxide (0.02 mol) and benzylmercaptan (0.024 mol) is stirred in 20.m1 of a 10% solution of sodium hydroxide and 20 ml of dimethylformamide at room temperature for six hours. After being heated for 10 minutes on a steam bath, the mixture is cooled and acidified with 6N HCl. The precipitate is due. 3-Benzylthiomethyl-6-chloro-7-sulfamyl-1,2,4-benzothiadiazine 1,1-dioxide.
