Verfahren zur Herstellung eines neuen Antibiotikums Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung eines neuen Antibiotikums, welches im folgenden als Rifamycin bezeichnet werden soll, sowie der Komponenten A, B, C, D und E des Rifamycins.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man einen Stamm eines Rifamycin produzierenden Mikroorganismus, z. B. Streptomyces mediterranei oder eines natürlich vor kommenden oder künstlich erzeugten Mutanten oder Varianten davon, in einem Medium züchtet, welches assimilierbare Kohlehydrate, Stickstoffverbindungen und organische Salze enthält. Aus dem Kulturmedium kann man hierauf das Rifamycin bzw. seine Kom ponenten gewinnen.
Der Stamm Streptomyces mediterranei wurde aus einer bei St. Raphael (Frankreich) erhobenen Boden probe isoliert. Er hat die im folgenden beschriebenen Eigenschaften, welche auf schrägen Kulturen bei 28 während einer Versuchsdauer von bis zu 20 Tagen auf verschiedenen Kulturmedien beobachtet worden sind. Die Farbbestimmungen wurden nach Maerz and Pauls Dictionary of Colour; Mc Graw Hill <B>1950</B> gemacht.
Hafermehl Agar: Gutes Wachstum mit glatter Oberfläche. Vegeta tives Mycel glasig bis gelblich, Rückseite rosa. Weissliches oder irdisches Mycel mit Rosafär- bung. Spuren eines gelblichen löslichen Pigmentes. Hefeextrakt-Glucose-Agar: Reichliches Wachstum, gelblich bis rosa mit rauher Oberfläche. Dürftiges oberirdisches Mycel. Keine Pigmentation der Medien. Emerson-Glucoseagar: Reichliches Wachstum, gelblich bis hellorange mit rauher Oberfläche.
Oberirdisches Mycel wird rötlich (2/B/8). Bernsteinfarbiges lösliches Pigment.
Hafermehl-Mischagar: (enthält 20g Filtrat von gekochten Haferflocken, 5 g enzymatisches Caseinhydrolysat, 3 g Fleisch extrakt, 10 g Saccharose, 40 g Agar in 1 Liter Wasser. pH auf 6,9 korrigiert.) Gutes Wachstum, glatt, gelblich mit leichter oranger Tönung. Bernsteinfarbiges Pigment. Benett-Agar: Gutes Wachstum, gelblich bis orangegelb, Ober irdisches Mycel wird rosa. Helles bernstein farbiges Pigment.
Penassay-Agar ( Difcoy>): Schlechtes Wachstum. Hefeextrakt-Melasseagar: (enthält 7,5g Glycerin, 7,5g Melasse, 5 g Hefe extrakt, 4 g NaC1, 30 g Agaragar, 1 Liter dest. Wasser. pH korrigiert auf 6,0.) Reichliches rauhes Wachstum, farblos bis gelb lich.
Weissliches oberirdisches Mycel. Dunkles bernsteinfarbiges lösliches Pigment (12/H/10). Csapek-Dox Saccharoseagar: Schlechtes Wachstum, dünn und farblos bis schwach melonenfarbig (11/A/8). Spuren eines rötlichweissen oberirdischen Mycels. Kein lös liches Pigment. Kartoffelagar: Schlechtes Wachstum, dünn und farblos. Spuren eines weisslichen oberirdischen Mycels. Kein lösliches Pigment.
Kartoffelbrei: Schlechte Entwicklung, glasig, Farbe des Breis nicht verändert.
Glucose-Asparagin-Agar: Gutes Wachstum mit glatter Oberfläche. Dünnes vegetatives Mycel, orange bis rosa (10/B/12), gelbliche Rückseite. Kein oberirdisches Mycel. Wenig hellgelbes lösliches Piment. Glycerin-Asparagin-Agar: Wie Glucose-Asparagin-Agar. Nähragar: Mässiges Wachstum mit glatter Oberfläche, hell rosa bis orange (2/B/10) mit gelblich oranger Rückseite. Oberirdisches Mycel rötlichweiss. Kein lösliches Pigment.
Pridham-Agar: Mässiges Wachstum, glatt, farblos mit hummer roten (2/G/11) Flecken. Oberirdisches Mycel rosa. Keine Pigmentation des Mediums. Stärke-Agar: Schlechtes Wachstum, farblos bis hellorange rosa. Stärkehydrolyse zweifelhaft. Wenig weisses oberirdisches Mycel.
Dextrose-Trypton-Agar: Reichliches Wachstum, orangerosa mit gold gelber bis orangerosa Rückseite. Oberirdisches Mycel rosa. Hellgoldgelbes (9/J/6) lösliches Pigment.
Hickey und Tresners Cobalt-Agar: Mässiges Wachstum, glasig bis hellrosaorange. Wenig oberirdisches Mycel. Wenig gelbliches lösliches Pigment.
Tyrosin Agar: Schlechtes Wachstum. Calciummaleat Agar: Gutes Wachstum, farblos. Oberirdisches Mycel weisslich mit Rosatönung. Kein lösliches Pigment. Teilweise Verdauung von Calciummaleat. Dorsets Albuminagar: Gutes Wachstum, rosa. Kein oberirdisches Mycel und keine Pigmentation des Mediums.
Gelatine: Keine Pigmentation. Langsame und unvollstän dige Verflüssigung.
Nitrate: Oberflächenwachstum mit rosafarbigem ober- irdischem Mycel. Keine Reduktion zu Nitrit. Extrakt wird gelblich.
Lackmusmilch: Keine Peptisation oder Koagulation. Leicht alkalische Reaktion. Cellulose: Kein Wachstum auf Filterpapierscheiben, die auf kohlehydratfreies mineralisches Agar gebracht werden.
Temperatur: Optimum 28 ; bei 37 findet noch Wachstum statt, doch sind die Eigenschaften weniger aus geprägt.
Morphologie der Kolonien: Auf Benett-Agar bildet dieser Stamm kleine Kolonien (Durchmesser gewöhnlich nicht mehr als 2 mm) mit gut abgegrenztem Umfang und dichter, aber rauher Oberfläche, bedeckt mit weiss lichem oberirdischem Mycel. Eine grosse Zahl von auf Agar gezogenen Kolonien bilden kein oberirdisches sondern nur ein orangerotes vege tatives Mycel aus, welches bis zu erheblicher Dicke wächst, bis die Kolonien ein felsartiges Aussehen haben.
Sporenbildung: ME/83 bildet auf den obengenannten Kultur medien gewöhnlich keine Sporen. Falls Sporu- lation erfolgt, erscheinen die Sporen in langen, etwas verdrehten Ketten; Spiralen sind selten. Die Sporen sind ellipsoidartig bis länglich (0,8-1,0 X 3,0-5,0, u).
Test für die Verwendung von Kohlenstoffquellen: Kulturen auf mineralischem Agar mit verschie denen Quellen gemäss Pridham und Gottlieb er gaben kein Wachstum auf: Raffinose, Acetat, Citrat, Glycin.
Alle andern ausprobierten Kohlenstoffquellen erga ben mässiges bis gutes Wachstum: Xylose, Arabinose, Glucose, Rhamnose, Fructose, Mannose, Galactose, Lactose, Saccharose, Maltose, Glycerin, Mannit, Inosit, Succinat und Salicin.
Streptomyces mediterranei kann wie folgt charak terisiert werden: 1. Er gehört nicht zur chromogenen Gruppe der Streptomycetes.
2. Auf den meisten Medien ergibt das vegetative Wachstum eine gelbliche bis leicht rosaorange, jedoch nie eine intensive orange bis rote Farbe.
3. Das oberirdische Mycel ist rötlich weiss, hat jedoch nie eine orange oder lavendelartige Fär bung. Auf den meisten Medien ist die Sporulation schwach. 4. Acetat, Citrat, Raffinose und Glycin werden nicht als Kohlenstoffquellen verwendet.
Ein Stamm von Streptomyces mediterranei ist bei der American Type Culture Collection unter Num mer 13 685 hinterlegt worden. Varianten oder Mu tanten eines Rifamycin produzierenden Mikroorga nismus können durch Behandlung von Sporensuspen- sionen mit mutagenen Substanzen, wie z. B. UV- Strahlen und Senfgas, erzeugt werden.
Zur Herstellung von Rifamycin kann so vorge gangen werden, dass man Streptomyces mediterranei oder, wie gesagt, irgendeinen anderen Rifamycin pro duzierenden Mikroorganismus, bei Temperaturen von 25-37 , vorzugsweise bei 28 , in belüfteten Kulturen in einem wässrigen Kulturmedium züchtet.
Als assimilierbare, gut brauchbare Stickstoffquellen kommen beispielsweise Aminosäuren einzeln oder gemischt, Peptide und Proteine und ihre Hydrolysate wie etwa Peptone, Fleischextrakt, Hefeextrakt, Soja bohnenmehl, Getreidebreiextrakt, Fischmehl und -extrakt, wässrige Auszüge von Samenkörnern und Destillationsrückstände in Betracht.
Als Kohlenstoff quelle können beispielsweise die folgenden Substan zen verwendet werden: Glucose, Xylose, Lactose, Saccharose, Maltose, Glycerin, Mannit, Inosit, Succinatstärke und Dextrine. Die Gärung kann wäh rend 24-120 Stunden durchgeführt werden. Das ge wöhnlich auf 7-8 eingestellte Anfangs-pH sinkt im Verlauf der Gärung auf 5-7. Normalerweise werden die besten Resultate bei einer Gärdauer von 48 bis 72 Stunden erhalten. Nach dieser Zeit wird eine aus gezeichnete Ausbeute an Antibiotikum erhalten.
Nach Beendigung der Gärung kann Rifamycin wie folgt isoliert werden: Das Gärmedium wird beim End-pH von 5-7 oder nachdem man das pH auf 7,5-8,5 eingestellt hat, bei 26-28 oder einer tieferen Temperatur fil triert. Das Filtrat wird sofort angesäuert, um die beste Stabilisierung der antibiotischen Substanzen zu erreichen. Die aktiven Substanzen werden mit in Wasser unlösbaren Lösungsmitteln wie Butanol, Äthyl-, Propyl-, Butyl- oder Amylacetat oder Chloro form extrahiert.
Das Verhältnis zwischen dem Volu men des Gärmediums und dem Volumen des Lö sungsmittels wird je nach dem Lösungsmittel ver schieden gewählt, es kann 2: 1 bis 10: 1 betragen.
Ein Teil des antibiotischen Substanzgemisches ist in saurem Medium unlöslich und kann daraus direkt, etwa durch Filtration gewonnen werden. Auf alle Fälle muss hierbei aber das Filtrat mit orga nischen Lösungsmitteln extrahiert werden, um soviel wie möglich der antibiotisch aktiven Substanzen zu gewinnen. Der in Wasser unlösliche Teil kann im gleichen Lösungsmittel gelöst werden und mit dem organischen Extrakt aus der Extraktion des Filtrates vereinigt werden. Nach der Extraktion enthält das wässrige Medium immer noch antibiotisch aktive Substanzen, welche jedoch mit üblichen organischen Lösungsmitteln nicht extrahiert werden können.
Diese Substanzen können jedoch in leicht saurem Milieu durch Tierkohle adsorbiert und daraus durch wasser lösliche Alkohole, wie Methanol oder Äthanol, eluiert werden.
Das Mycel hält einige mikrobiologisch aktive Substanzen zurück. Diese können daraus durch die genannten Lösungsmittel oder durch mehrmaliges Waschen mit Wasser extrahiert werden. Sobald die meisten der antibiotisch aktiven Sub stanzen im organischen Lösungsmittel gelöst sind, wird dieses hierbei unterhalb 30 im Stickstoffstrom im Vakuum auf ein kleines Volumen eingedampft.
Nach Zugabe von Petroläther oder einem andern Kohlenwasserstoffgemisch fällt eine braune Substanz aus, welche eine hohe Aktivität gegen Mycobakterien und grampositive Bakterien aufweist. Dieses Rohpro dukt ist ein hell bis dunkelbraunes amorphes Pulver, welches manchmal eine grünliche Nüance besitzt. Es ist ein Gemisch von Substanzen mit antibiotischer Wirkung.
Eine Chromatographie auf Whatman-Nr.1- Papier. (Lösungsmittel Wasser mit 3 % Ammonium chlorid und 1 % Ascorbinsäure, Entwicklung auf einer mit Sarcina lutea geimpften Agarplatte) ergibt mindestens fünf Flecken, welche mit den Buchstaben A-E bezeichnet worden sind, und zwar gemäss der Zunahme des Rf-Wertes vom Startpunkt aus. Die Flecken C und D sind normalerweise grösser als die übrigen. Das Vorhandensein weiterer Fraktionen ist nicht ausgeschlossen.
Anstatt die antibiotische Mischung aus dem orga nischen Extrakt auszufällen, kann man diesen auch mit Pufferlösungen zunächst bei pH 7,0-7,5 und dann bei pH 10,0-10,5 extrahieren. Die Wahl geeig neter pH-Bereiche erlaubt es, eine angenäherte Tren nung der verschiedenen Fraktionen des rohen Pulvers zu erreichen. Bei pH 7,0-7,5 wird sodann das Rifamycin B, welches stark sauren Charakter hat, selektiv extrahiert. Die andern Fraktionen werden bei höheren pH-Werten extrahiert.
Da diese Komponen ten in neutraler und alkalischer Lösung wenig stabil sind, ist es nötig, Pufferlösungen mit 0,5 bis 3 % Ascorbinsäure oder andern in ähnlicher Weise stabi lisierend wirkenden Substanzen zu verwenden. Das Volumenverhältnis zwischen organischer Lösung und alkalischer Pufferlösung schwankt in breiten Gren zen, je nach den verwendeten organischen Lösungs mitteln und dem pH-Wert der Pufferlösung.
Aus einer Äthylacetatlösung, welche 1 % aktive Substanz enthält, werden bei pH 7,0-7,5 in drei Portionen mit je 1/1o des Volumens der Pufferlösung die Frak tion B und bei pH 10,0-10,5 mit drei Portionen mit je 1/5 des Volumens der Pufferlösung, welche 1 Ascorbinsäure enthält, die restlichen Fraktionen in die wässrige Phase gebracht.
Aus der wässrigen Phase können die Antibiotica in halbreiner Form gewonnen werden, indem man die wässrige Phase ansäuert und die Substanzen mit einem Lösungsmittel, wie z. B. Äthylacetat oder Chloroform, extrahiert, die erhaltene Lösung ein dampft und die Substanzen mit Petroläther ausfällt.
Man kann die Antibiotica auch aus der wässrigen Lö sung durch Ansäuern ausfällen und abfiltrieren.
Die Reinigung der rohen Mischung kann durch Chromatographie an verschiedenen Adsorbentien er folgen, z. B. an Florisils>, welches mit methanolischer Salzsäure vorbehandelt worden ist. Die rohe Mi schung wird hierzu in einem Lösungsmittel, z. B. Äthylacetat, gelöst und auf eine Kolonne von Floro- sil gegeben. Die Elution erfolgt mit dem gleichen Lösungsmittel oder mit Lösungsmitteln oder -ge- mischen mit zunehmender Polarität.
Die Eluate mit den höchsten mikrobiologischen Werten sind die jenigen, welche Rifamycin C und D enthalten.
Nachfolgend wird ein zweckmässiges Vorgehen zur Isolierung der fünf Hauptkomponenten der rohen Antibioticamischung beschrieben, und es werden einige ihrer chemischen und physikalischen Daten angeführt, die ihre Identifikation erlauben.
Rifamycin <I>A.</I> Es besitzt einen Rf-Wert von 0,00 bis 0,10 bei der oben beschriebenen Papierchromato- graphie. Rifamycin A kann aus der rohen Mischung durch Gegenstromverteilung im Zweiphasensystem Methanol + 0,01n Salzsäure - Benzol + Petroläther (10:5:15:5) gewonnen werden. Nach 160 Über tragungen kann Rifamycin A direkt aus den leichten Phasen der Gefässe 150-160 gewonnen werden.
Nach dem Eindampfen und Ausfällen in Petroläther erhält man Rifamycin A in Form eines dunkelbraunen amorphen Pulvers. Rifarnycin A ist unlöslich in Was ser, löslich in wässrigen Alkalien. In Äthylacetat ge löst zeigt es im sichtbaren Gebiet ein Absorptions maximum bei 460 mg.
Nach Schütteln der Athyl- acetatlösung mit einer sauren wässrigen Lösung von <I>1</I> 2ö Stannochlorid ist das Maximum nach<I>422</I> my verschoben. Derselbe Effekt kann auch durch Schüt teln mit einer einprozentigen wässrigen Ascorbin- säurelösung erreicht werden. Das Maximum von 460 my kann wieder erreicht werden, wenn man die ÄthyIacetatlösung mit einer 0,1 ö igen Lösung von Kaliumferricyanid schüttelt.
Vermutlich hat Rifamycin A eine Chinonstruktur, welche reversibel reduzierbar und oxydierbar ist.
Rifamycin B. Es besitzt einen Rf-Wert von 0,20 bis 0,30 bei der oben beschriebenen Papierchromato- graphie. Rifamycin B ist stark sauer. Es kann isoliert werden, indem man die rohe Mischung in einem Lösungsmittel wie z. B. Äthylacetat oder Chloroform auflöst, und die Substanz bei pH 6,5-7,5 mit wäss- rigem Phosphatpuffer extrahiert.
Aus der wässrigen Phase kann Rifamycin B entweder durch Ansäuern oder durch erneutes Extrahieren der angesäuerten wässrigen Phase mit Äthylacetat gewonnen werden. Beim Eindampfen der Äthylacetatlösung fällt die Fraktion B in halbreinem Zustand aus. Durch Um kristallisieren aus Äthylacetat oder Benzol erhält man das reine Produkt.
Rifamycin B bildet gelbe glänzende Kristalle. Es wird dunkel bei 165 und schmilzt unterhalb 300 noch nicht. Seine gesättigte wässrige Lösung hat ein pH von 3,6. Es ist sehr wenig löslich in Wasser und verdünnten Mineralsäuren, jedoch löslich in Bicar- bonatlösungen unter Entwicklung von Kohlensäure.
In konzentrierter Schwefelsäure löst es sich unter Rotfärbung. Es ist schlecht löslich in Methanol, wenig in Äthanol, Chloroform, Äthylacetat und Aceton, sehr schlecht in Benzol und praktisch un löslich in Petroläther, Tetrachlorkohlenstoff, Athyl- äther und Kohlenwasserstoffen. Es entfärbt Kalium permanganat, reduziert Fehlingsche Lösung und Tollensreagenz, absorbiert Brom und wird mit Was serstoff katalytisch reduziert.
Bei der Behandlung mit Nitrit in alkalischem Milieu erfolgt eine Rot färbung. Es reagiert nicht mit Ninhydrin, jedoch er folgt die Reaktion nach einer kräftigen sauren Hydro lyse. Es ist nicht diazotierbar und reagiert nicht mit aromatischen Diazokörpern. Es ergibt keine Molish- reaktion und reduziert Triphenyltetrazolium nicht. Es reagiert jedoch mit Ferrichlorid unter Rotfärbung.
Die Elementaranalyse ergab die folgenden Haupt werte: C 61,75 %; H 6,72 %; N 1,88 ö; O 29,22 7o. Daraus kann die allgemeine Formel C."H47_,53NO14 abgeleitet werden. Rifamycin B ist optisch aktiv: [a] 589= - 11 , c = 1, Methanol, photoelektrisches Polarimeter.
Eine potentiometrische Titration in Wasser zeigt zwei saure Gruppen: pK, etwa 2,8 (Aquivalentge- wicht 780), pK2 etwa 6,7 (Äquivalentgewicht 765).
Das Molekulargewicht wurde in Cyclohexanol nach Rast mit etwa 750 ermittelt. Das UV- und sichtbare Spektrum in Wasser zeigt eine Schulter bei 220 bis 230 my (Ei @m = 513), und zwei Maxima bei 305 mci (E i % = 246) und 430 m (E i 5m = 202).
In Äthyl- acetat liegt das Maximum im Sichtbaren bei 415 mu. Das UV- und das sichtbare Spektrum von Rifamycin B in Phosphatpuffer vom pH 7,3 zeigt Maxima bei 223 my (E i @m 555), 304 my (E i %R# 275) und bei 425 my (E i %l 220).
Das IR-Spektrum von Rifamycin B weist Ab sorptionsmaxima bei den folgenden Werten (in cm-') auf: 3465, 3330, 1742, 1730 (Schulter), 1705 (Schul ter), 1673, 1602, 1575, 1550, 1445, 1342, 1300, 1267, 1240 (Schulter), 1205, 1180, 1165, 1102, 1073, 1048, 1020, 975, 948, 915, 905, 850, 795, 760, 730, 680.
Da Rifamycin B sauer reagiert, bildet es sowohl mit anorganischen wie mit organischen Basen Salze. Es wurden das Mono- und Dinatriumsalz dargestellt, von denen das erste wenig und das zweite gut in Wasser löslich ist. Die Lösungen zeigen ein pH von 5,0 bzw. etwa 7,0. Diese Lösungen sind sehr stabil.
Mit den folgenden organischen Basen sind Salze gebildet worden: Dibenzylamin, Chinin, 1-(p-Chlor benzyl)-2-pyrrolidylmethyl- benzoimidazol, Dibenzyläthylendiamin, 2-Methyl-6-amino-heptan und andern. Alle diese Salze sind in Wasser wenig löslich und versprechen, für therapeutische Zwecke nützlich zu sein.
Rifamycin <I>C und D.</I> Diese beiden Fraktionen zeigen beinahe gleiche Rf-Werte bei der oben be schriebenen Papierchromatographie (Rf = 0,45 bis 0,55). Die Flecke sind auf dem Chromatogramm miteinander durch einen breiten Hals verbunden. Rifamycin C und D können auf der rohen Mischung durch Gegenstromverteilung in einem Zweiphasen system bestehend aus Methanol + O,Oln Salzsäure Benzol + Petroläther (10: 5 :<B><I>15:</I></B> 5) gewonnen wer den.
Bei 85 Übertragungen wird die Hauptmenge an Rifamycin C zwischen den Gefässen 55-65 und die Hauptmenge an Rifamycin D zwischen den Gefässen 35-45 angereichert. Durch Extrahieren dieser Ge fässe mit Benzol, Äthylacetat, Chloroform oder ähn lichen Lösungsmitteln, Konzentrieren des Extraktes und Ausfällen mit Petroläther oder andern Kohlen wasserstoffmischungen kann man die beiden Rifamy- cine C und D als amorphe Pulver von hoher Rein heit erhalten.
Rifamycin C und D erscheinen als dunkelbraune Pulver mit grüner Nuance. Sie sind in den üblichen organischen Lösungsmitteln löslich und in Wasser praktisch unlöslich. Eine wässrige Lösung in einem Puffer vom pH 8,0-10,0 kann man herstellen, wenn man Ascorbinsäure als Stabilisator zugibt. Die UV- und sichtbaren Spektren der beiden Rifamycine C und D sind ähnlich. Die Maxima bei 385-390 my und bei 460 mlc (in Methanol und Äthylacetat) sind sehr charakteristisch.
Das Maximum bei 460 mu wird nach 422 my verschoben, wenn man die Äthylacetat- lösung von Rifamycin C oder D mit einer 0,1 %igen wässrigen Stannochloridlösung oder mit einer 1 % igen wässrigen Ascorbinsäurelösung schüttelt.
Nach weite rem Schütteln mit einer 0,1 % igen wässrigen Lösung von Kaliumferricyanid erscheint das Maximum von 460 my wieder. Dies zeigt, dass Rifamycin C und D reversibel oxydier- und reduzierbare Substanzen, wie z. B. die Chinone, sind.
Eigenschaften von Rifamycin C: Elementar analyse: C 61,51 %; H 6,73 %; N 4,21 %; O 27,55 (Differenz). Rifamycin C gibt eine positive Reaktion mit Fehling- und Tollensreagens, mit Ferrichlorid, Jodoform und Permanganat. Die Ninhydrinreaktion ist negativ, wird jedoch nach kräftig saurer Hydro lyse positiv.
Das sichtbare Spektrum in Äthylacetat zeigt Maxima bei 388 mg (E I %m78) und bei 460 mu

Eigenschaften von Rifamycin D: Elementar analyse: C 62,17 %; H 6,58 %; N 3,53 %; O 27,72 (Differenz). Rifamycin D gibt eine positive Reak tion mit Tollens und Fehlingreagens, Ferrichlorid, Jodoform und Permanganat. Die Ninhydrinreaktion ist vor und nach Hydrolyse negativ. Das sichtbare Spektrum in Äthylacetat zeigt Maxima bei 388 mu (E i @m 79) und bei 460 my (E i %m 115).
Es ist un löslich in Wasser, Mineralsäuren, Bicarbonaten, Äthyläther und Petroläther, löslich in Soda, Methanol, Äthylacetat, Chloroform und Aceton.
Rifamycin <I>E.</I> Diese Fraktion besitzt einen Rf- Wert von etwa 0,7 bis 0,8 bei der oben beschrie benen Papierchromatographie. Es kann aus der rohen Mischung durch Gegenstromverteilung in einem Zweiphasensystem bestehend aus Methanol + 0,01n Salzsäure - Benzol + Petroläther <B>(10:5:15:5)</B> gewonnen werden. Nach 160 Über tragungen hat sich die Hauptmenge des Rifamycins E in den ersten 10 Gefässen angereichert.
Durch Extraktion der sauren wässrigen Phase mit Äthyl- acetat, Eindampfen des Extrakts und Fällen mit Petroläther kann man das Rifamycin E gewinnen. Man erhält ein hellbraunes amorphes Pulver.
Das sichtphase Spektrum in Äthylacetat weist ein Maxi mum bei 400 my auf, welches nach Schütteln der Äthylacetatlösung mit verdünnten wässrigen Lösun gen von Kaliumferricyanid oder Stannochlorid nicht verschoben wird.
Wie oben erwähnt, zeigen Rifamycin und seine Komponenten einen hohen Wirkungsgrad gegen gram-positive Bakterien und Mycobakterien. Biolo gische Versuche wurden getrennt mit Rifamycin B und mit einer Mischung der Rifamycine A, C, D und E, welche nachfolgend mit Rifamycinkomplex bezeichnet wird, ausgeführt.
Die folgende Tabelle gibt Aufschluss über die antibakterielle Wirkung in vitro von Rifamycin B und dem Rifamycinkomplex auf eine grosse Zahl von gram-positiven und gram-negativen Mikroorga nismen.
Die angegebenen Werte sind die minimalen Inhibitionskonzentrationen in gamma/cmg.

dass der Rifamycinkomplex bei Mäusen eine sehr starke Wirkung gegen Streptococcus- und Staphylococcus- infektionen und allgemein gegen gram-positive Bak terien ausübt. Eine 100%ige therapeutische Dosis von 50 mg/kg erscheint als sehr niedrig für ein Anti- Die Wirkung von Rifamycin B und Rifamycin- komplex wurde in vivo mit sehr befriedigenden Resultaten an Mäusen untersucht, welchen stark pathogene gram-positive Microorganismen injiziert worden waren.
Alle Injektionen waren intraperitoneale Injek tionen von 10-100 Letaldosen von 15-20 Stunden alten Kulturen der pathogenen Mikroorganismen. Während die Kontrolltiere in jedem Fall 2-3 Tage nach der Injektion starben, wurde die Zahl der schliesslich überlebenden Tiere jeweils 12 Tage nach Beginn der Experimente ermittelt.
Einige der erhaltenen Resultate sind in der fol genden Tabelle zusammengestellt. bioticum, welches eine intravenöse LD.o von 250 bis 300 mg/kg aufweist.
Rifamycin B, obwohl etwas weniger aktiv, weist einen noch bessern therapeutischen Koeffizienten auf infolge seiner extrem niedrigen Toxizität. Bei Men schen werden gute Antibiotica-Blutspiegel mit täg- lichen intramuskulären Verabreichungen von 500 bis 1000 mg erreicht.
Die Rifamycine haben eine sehr gute Wirkung bei Patienten gezeigt, welche unter verschiedenen Abszessen, Mastitis, eitriger Mittelohrentzündung und dergleichen litten. Die verabreichte Tagesdosis betrug im allgemeinen 2 g, durch zwei intramuskuläre Injektionen verabreicht. Ein rascher Fall der Tem peratur mit einer Auflösung des Entzündungspro zesses nach nicht mehr als 3-4 Tagen nach Beginn der Behandlung wurde immer beobachtet. Auch die Oberflächenbehandlung von infizierten Wunden mit wässrigen Lösungen von Rifamycin war ebenfalls erfolgreich.
Eiterung und Sekretion im allgemeinen hörten sofort auf und die Wundheilung erfolgte nach her rasch.
Diese Resultate wurden oft erreicht, nachdem vorher vergeblich andere Antibiotica angewendet worden waren.
<I>Beispiel 1</I> Eine Kultur von Streptomyces mediterranei wird während 6-8 Tagen auf Bennetagar bei 28 gezüch tet. Der Organismus wird vom Agar durch Spülen befreit und aseptisch in Flaschen von 500 cm3 ein geimpft, welche 100 cm3 eines Nährmediums folgen der Zusammensetzung aufweisen:

Die höchste Menge an antibiotisch wirksamen Substanzen wird nach 45 bis 50 Stunden erhalten (300-400 gamma/cm3 der anti biotischen Mischung) und die Brühe wird dann auf gearbeitet.

The inventive method is characterized in that a strain of a rifamycin-producing microorganism, for. B. Streptomyces mediterranei or a naturally occurring or artificially produced mutant or variant thereof, is cultivated in a medium which contains assimilable carbohydrates, nitrogen compounds and organic salts. The rifamycin or its components can then be obtained from the culture medium.
The strain Streptomyces mediterranei was isolated from a soil sample taken from St. Raphael (France). It has the properties described below, which have been observed on inclined cultures at 28 during a test duration of up to 20 days on various culture media. Penassay Agar (Difcoy>): Poor growth. Yeast extract molasses agar: (contains 7.5 g glycerine, 7.5 g molasses, 5 g yeast extract, 4 g NaCl, 30 g agaragar, 1 liter distilled water. PH corrected to 6.0.) Abundant rough growth, colorless to yellow lich.
Whitish above-ground mycelium. Dark amber soluble pigment (12 / H / 10). Csapek-Dox sucrose agar: poor growth, thin and colorless to pale melon colored (11 / A / 8). Traces of a reddish white mycelium above ground. No soluble pigment. Potato agar: poor growth, thin and colorless. Traces of a whitish aboveground mycelium. No soluble pigment.
Mashed potatoes: poor development, glassy, color of the mash not changed.

Hickey and Tresner's Cobalt Agar: Moderate growth, glassy to light pink orange. Little mycelium above ground. Little yellowish soluble pigment.
Tyrosine Agar: Poor Growth. Calcium maleate agar: good growth, colorless. Above-ground mycelium whitish with a pink tint. No soluble pigment. Partial digestion of calcium maleate. Dorsets Albumin Agar: Good growth, pink. No mycelium above ground and no pigmentation of the medium.
Gelatin: No pigmentation. Slow and incomplete liquefaction.
Nitrates: Surface growth with pink above-ground mycelium. No reduction to nitrite. The extract turns yellow.
Litmus milk: No peptization or coagulation. Slightly alkaline reaction. Cellulose: No growth on filter paper discs placed on carbohydrate-free mineral agar.
Temperature: optimum 28; 37 is still growing, but the properties are less pronounced.
Colon morphology: On Benett agar, this strain forms small colonies (usually not more than 2 mm in diameter) with a well-defined circumference and a dense but rough surface, covered with whitish above-ground mycelium. A large number of colonies grown on agar do not form above-ground but only an orange-red vegetative mycelium, which grows to a considerable thickness until the colonies have a rock-like appearance.
Spore formation: ME / 83 usually does not form spores on the above culture media. If sporulation occurs, the spores appear in long, somewhat twisted chains; Spirals are rare. The spores are ellipsoidal to elongated (0.8-1.0 X 3.0-5.0, u).

All other carbon sources tried showed moderate to good growth: xylose, arabinose, glucose, rhamnose, fructose, mannose, galactose, lactose, sucrose, maltose, glycerin, mannitol, inositol, succinate and salicin.
Streptomyces mediterranei can be characterized as follows: 1. It does not belong to the chromogenic group of Streptomycetes.
2. On most media, vegetative growth results in a yellowish to slightly pinkish orange, but never an intense orange to red color.
3. The mycelium above ground is reddish white, but never has an orange or lavender-like color. Sporulation is weak on most media. 4. Acetate, citrate, raffinose and glycine are not used as carbon sources.
A strain of Streptomyces mediterranei has been deposited with the American Type Culture Collection under number 13,685. Variants or mutants of a rifamycin-producing microorganism can be treated by treating spore suspensions with mutagenic substances, such as. B. UV rays and mustard gas are generated.
To produce rifamycin, the procedure can be that Streptomyces mediterranei or, as said, any other rifamycin-producing microorganism is grown in aerated cultures in an aqueous culture medium at temperatures of 25-37, preferably 28.
As assimilable, well usable nitrogen sources, for example, amino acids individually or mixed, peptides and proteins and their hydrolysates such as peptones, meat extract, yeast extract, soybean meal, cereal porridge extract, fish meal and fish extract, aqueous extracts of seeds and distillation residues come into consideration.
The following substances, for example, can be used as the carbon source: glucose, xylose, lactose, sucrose, maltose, glycerol, mannitol, inositol, succinate starch and dextrins. Fermentation can be carried out for 24-120 hours. The initial pH, usually set at 7-8, drops to 5-7 during fermentation. Usually, the best results are obtained with a fermentation time of 48 to 72 hours. After this time, an excellent yield of antibiotic is obtained.
After fermentation has ended, rifamycin can be isolated as follows: The fermentation medium is filtered at a final pH of 5-7 or, after the pH has been adjusted to 7.5-8.5, at 26-28 or a lower temperature. The filtrate is immediately acidified in order to achieve the best stabilization of the antibiotic substances. The active substances are extracted with water-insoluble solvents such as butanol, ethyl, propyl, butyl or amyl acetate or chloro form.
The ratio between the volume of the fermentation medium and the volume of the solvent is chosen differently depending on the solvent, it can be 2: 1 to 10: 1.
Part of the antibiotic substance mixture is insoluble in an acidic medium and can be obtained therefrom directly, for example by filtration. In any case, the filtrate must be extracted with organic solvents in order to gain as much of the antibiotic active substances as possible. The water-insoluble part can be dissolved in the same solvent and combined with the organic extract from the extraction of the filtrate. After the extraction, the aqueous medium still contains antibiotically active substances which, however, cannot be extracted with conventional organic solvents.
However, these substances can be adsorbed by animal charcoal in a slightly acidic environment and eluted therefrom by water-soluble alcohols such as methanol or ethanol.
The mycelium retains some microbiologically active substances. These can be extracted therefrom using the solvents mentioned or by washing several times with water. As soon as most of the antibiotically active substances are dissolved in the organic solvent, this is evaporated to a small volume under 30 in a nitrogen stream in a vacuum.
After adding petroleum ether or another hydrocarbon mixture, a brown substance precipitates which has a high activity against mycobacteria and gram-positive bacteria. This raw product is a light to dark brown amorphous powder, which sometimes has a greenish nuance. It is a mixture of substances with an antibiotic effect.
Chromatography on Whatman # 1 paper. (Solvent water with 3% ammonium chloride and 1% ascorbic acid, development on an agar plate inoculated with Sarcina lutea) results in at least five spots, which have been designated with the letters A-E, according to the increase in the Rf value from the starting point. Spots C and D are usually larger than the rest. The presence of other parliamentary groups cannot be ruled out.
Instead of precipitating the antibiotic mixture from the organic extract, it can also be extracted with buffer solutions initially at pH 7.0-7.5 and then at pH 10.0-10.5. The choice of suitable pH ranges makes it possible to achieve an approximate separation of the various fractions of the raw powder. At pH 7.0-7.5 the rifamycin B, which has a strongly acidic character, is then selectively extracted. The other fractions are extracted at higher pH values.
Since these components are not very stable in neutral and alkaline solutions, it is necessary to use buffer solutions with 0.5 to 3% ascorbic acid or other substances with a similar stabilizing effect. The volume ratio between organic solution and alkaline buffer solution fluctuates within wide limits, depending on the organic solvents used and the pH of the buffer solution.
From an ethyl acetate solution, which contains 1% active substance, the fraction B at pH 7.0-7.5 in three portions with 1 / 1o of the volume of the buffer solution and at pH 10.0-10.5 with three portions with 1/5 of the volume of the buffer solution, which contains 1 ascorbic acid, the remaining fractions are brought into the aqueous phase.
The antibiotics can be obtained in semi-pure form from the aqueous phase by acidifying the aqueous phase and treating the substances with a solvent, such as. B. ethyl acetate or chloroform, extracted, the resulting solution evaporated and the substances precipitated with petroleum ether.
The antibiotics can also be precipitated from the aqueous solution by acidification and filtered off.
The purification of the crude mixture can be followed by chromatography on various adsorbents, e.g. B. Florisils>, which has been pretreated with methanolic hydrochloric acid. The raw Mi mixture is for this purpose in a solvent such. B. ethyl acetate, dissolved and placed on a column of Florosil. Elution takes place with the same solvent or with solvents or mixtures of increasing polarity.
The eluates with the highest microbiological values are those that contain rifamycin C and D.
A convenient procedure for isolating the five main components of the crude antibiotic mixture is described below, and some of their chemical and physical data are given to enable their identification.
Rifamycin <I> A. </I> It has an Rf value of 0.00 to 0.10 in the paper chromatography described above. Rifamycin A can be obtained from the crude mixture by countercurrent distribution in the two-phase system methanol + 0.01N hydrochloric acid - benzene + petroleum ether (10: 5: 15: 5). After 160 transfers, rifamycin A can be obtained directly from the light phases of the vessels 150-160.
After evaporation and precipitation in petroleum ether, rifamycin A is obtained in the form of a dark brown amorphous powder. Rifarnycin A is insoluble in water, soluble in aqueous alkalis. Dissolved in ethyl acetate, it shows an absorption maximum of 460 mg in the visible area.
After shaking the ethyl acetate solution with an acidic aqueous solution of <I> 1 </I> 2ö stannous chloride, the maximum is shifted to <I> 422 </I> my. The same effect can also be achieved by shaking with a one percent aqueous ascorbic acid solution. Rifamycin A presumably has a quinone structure which is reversibly reducible and oxidizable.
Rifamycin B. It has an Rf value of 0.20 to 0.30 in the paper chromatography described above. Rifamycin B is strongly acidic. It can be isolated by soaking the crude mixture in a solvent such as e.g. B. ethyl acetate or chloroform, and the substance extracted at pH 6.5-7.5 with aqueous phosphate buffer.
Rifamycin B can be obtained from the aqueous phase either by acidification or by extracting the acidified aqueous phase again with ethyl acetate. When the ethyl acetate solution is evaporated, fraction B precipitates in a semi-pure state. The pure product is obtained by recrystallizing from ethyl acetate or benzene.
Rifamycin B forms yellow, shiny crystals. It gets dark at 165 and does not yet melt below 300. Its saturated aqueous solution has a pH of 3.6. It is very sparingly soluble in water and dilute mineral acids, but soluble in bicarbonate solutions with the development of carbonic acid.
It dissolves in concentrated sulfuric acid and turns red. It is poorly soluble in methanol, little in ethanol, chloroform, ethyl acetate and acetone, very poorly in benzene and practically insoluble in petroleum ether, carbon tetrachloride, ethyl ether and hydrocarbons. It decolorizes potassium permanganate, reduces Fehling's solution and Tollens reagent, absorbs bromine and is catalytically reduced with hydrogen.
When treated with nitrite in an alkaline medium, the skin turns red. It does not react with ninhydrin, but it follows the reaction after a strong acidic hydrolysis. It cannot be diazotized and does not react with aromatic diazo bodies. It does not give a Molish reaction and does not reduce triphenyltetrazolium. However, it reacts with ferric chloride to turn red.
The elemental analysis gave the following main values: C 61.75%; H 6.72%; N 1.88 δ; O 29.22 7o. The general formula C. "H47_, 53NO14 can be derived from this. Rifamycin B is optically active: [a] 589 = - 11, c = 1, methanol, photoelectric polarimeter.
A potentiometric titration in water shows two acidic groups: pK, about 2.8 (equivalent weight 780), pK2 about 6.7 (equivalent weight 765).

The IR spectrum of rifamycin B has absorption maxima at the following values (in cm- '): 3465, 3330, 1742, 1730 (shoulder), 1705 (shoulder), 1673, 1602, 1575, 1550, 1445, 1342 , 1300, 1267, 1240 (shoulder), 1205, 1180, 1165, 1102, 1073, 1048, 1020, 975, 948, 915, 905, 850, 795, 760, 730, 680.
Since rifamycin B is acidic, it forms salts with both inorganic and organic bases. The mono- and disodium salts have been shown, the first of which is slightly soluble in water and the second is well soluble. The solutions show a pH of 5.0 and about 7.0, respectively. These solutions are very stable.
Salts have been formed with the following organic bases: dibenzylamine, quinine, 1- (p-chlorobenzyl) -2-pyrrolidylmethylbenzoimidazole, dibenzylethylenediamine, 2-methyl-6-aminoheptane and others. All of these salts are sparingly soluble in water and promise to be useful for therapeutic purposes.
Rifamycin C and D. These two fractions show almost the same Rf values in the paper chromatography described above (Rf = 0.45 to 0.55). The spots are connected to one another on the chromatogram by a wide neck. Rifamycin C and D can be obtained on the raw mixture by countercurrent distribution in a two-phase system consisting of methanol + O, Oln hydrochloric acid, benzene + petroleum ether (10: 5: <B> <I> 15: </I> </B> 5) will.
With 85 transfers, the main amount of rifamycin C between vessels 55-65 and the main amount of rifamycin D between vessels 35-45 are accumulated. By extracting these vessels with benzene, ethyl acetate, chloroform or similar solvents, concentrating the extract and precipitating it with petroleum ether or other hydrocarbon mixtures, the two rifamycins C and D can be obtained as amorphous powders of high purity.
Rifamycin C and D appear as dark brown powders with a green shade. They are soluble in the usual organic solvents and practically insoluble in water. An aqueous solution in a buffer of pH 8.0-10.0 can be prepared by adding ascorbic acid as a stabilizer. The UV and visible spectra of the two rifamycins C and D are similar. The maxima at 385-390 my and at 460 mlc (in methanol and ethyl acetate) are very characteristic.
The maximum at 460 microns is shifted to 422 microns if the ethyl acetate solution of rifamycin C or D is shaken with a 0.1% aqueous stannous chloride solution or with a 1% aqueous ascorbic acid solution.
After further shaking with a 0.1% aqueous solution of potassium ferricyanide, the maximum of 460 my appears again. This shows that rifamycin C and D are reversibly oxidizable and reducible substances, such as. B. the quinones are.
Properties of rifamycin C: Elemental analysis: C 61.51%; H 6.73%; N 4.21%; O 27.55 (difference).
