Procédé de préparation d'un dérivé de la tryptamine
La présente invention a pour objet un procédé de préparation de la 6-fluoro-tryptamine, de formule I:

Le procédé de l'invention, résumé sur le dessin ci-joint, consiste essentiellement à préparer le fluoborate de 3-nitro-4-méthyl- benzène- diazonium au départ de 2-nitro-4-aminotoluène qu'on diazote, puis soumet à l'action de l'acide fluoborique. Le composé fluoré obtenu fournit sous l'effet de la chaleur le 2-nitro-4-fluoro-toluène de formule II, dont la condensation avec l'oxalate d'éthyle en présence d'un éthylate alcalin conduit au sel alcalin du 4'-fluoro2'-nitro-phényl-pyruvate d'éthyle. On réduit et cyclise ce dernier composé par action d'un agent réducteur en milieu acide comme par exemple le zinc dans l'acide acétique, ou le fer dans l'acide chlorhydrique et obtient le 6-fluoro-indole-2-carboxylate d'éthyle de formule III.
La saponification de ce dérivé indolique fournit l'acide correspondant de formule IV, qu'on décarboxyle en 6-fluoro-indole de formule V.
Par condensation avec la diméthylamine et le formol, on transforme le 6-fluoro-indole en 6-fluorogramine de formule VI, dont le sulfométhylate par condensation avec un cyanure alcalin fournit le 6fluoro-3-indolyl-acétonitrile de formule VII. L'hydrogénation catalytique de celui-ci conduit à la 6fluoro-tryptamine de formule I cherchée.
Le nouveau composé trouve son application en tant qu'intermédiaire dans la synthèse de composés physiologiquement actifs de la famille de la réserpine. La 6-fluoro-tryptamine est aussi un antimétabolite puissant du tryptophane; on peut l'utiliser chaque fois que la transformation microbiologique du tryptophane en indole est à éviter, comme c'est le cas, par exemple, dans certaines fermentations intestinales pathologiques.
Dans les exemples suivants, les températures sont exprimées en degrés centigrades. Les points de fusion sont des points de fusion instantanée, déterminés sur bloc de Maquenne.
Exemple
A. Préparation du 2-nitro-4-fluoro-toluène de for
mule II a) Formation du fluoborate de 3-nitro-4-méthyl
benzène-diazonium
On ajoute à 137 g de 2-nitro-4-amino-toluène, 90 cm3 d'eau et 270 cm3 d'acide chlorhydrique concentré, agite jusqu'à disparition de la coloration orangée, puis refroidit à 00 C. Le chlorhydrate formé est diazoté par addition de 67,5 g de nitrite de sodium dans 180 cm3 d'eau. On poursuit l'agitation jusqu'à l'obtention d'une solution homogène en ayant soin de maintenir le mélange à basse température.
On ajoute ensuite 210 cm3 d'acide fluoborique à 40 % et abandonne sous agitation à 00 C pendant une demi-heure. Après essorage, lavage à l'eau et au méthanol on sèche et recueille 182 g (soit 77 %) de cristaux rosés de fluoborate de 3-nitro-4-méthylbenzène-diazonium. b) Sa décomposition
Les 182 g de fluoborate obtenus selon a) sont intimement mélangés avec 325 g de sable sec et chauffés directement à la flamme. Le chauffage est maintenu une demi-heure, la température intérieure atteint 1800 C. Après refroidissement, on reprend la masse charbonneuse par l'eau et entraîne à la vapeur. Les fractions entraînées sont extraites à l'éther et les extraits séchés, puis filtrés et évaporés à sec. Le résidu, 60 g (soit 44 %) d'un liquide jaune brun, est redistillé à 77O C sous 3 mm de mercure.
Le produit passe à point fixe. L'indice de réfraction de l'huile jaune obtenue de formule II est 1,522 à 250 C, en accord avec les valeurs de la littérature.
B. Préparation du 6-fluoro - indole -2 - carboxylate
d'éthyle de formule III
On recouvre par 100 cm3 d'éther l'éthylate de potassium sec, correspondant à 7,9 g de potassium, ajoute en refroidissant et sous agitation 27,2 cm3 d'oxalate d'éthyle, puis très lentement 31 g de composé de formule II, obtenu selon le paragraphe pré cédent. On abandonne le mélange réactionnel à la température ambiante pendant 40 heures, essore ensuite, lave à l'éther anhydre et sèche sous vide pour recueillir 44g de cristaux violets de sel de potassium du 4'- fluoro -2'- nitro - phényl - pyruvate d'éthyle.
On dissout le produit dans 280 cm3 d'alcool et 28 cm3 d'acide acétique et ajoute la solution goutte à goutte à un mélange porté à 900 C de 100 g de limaille de fer, 400 cm3 d'eau et 14 ces d'acide chlorhydrique concentré. L'alcool distille au cours de cette addition qui dure une demi-heure. Le chauffage avec agitation est maintenu pendant une heure encore, on glace et essore. On lave le fer et le composé cyclisé à l'eau et extrait ensuite par de l'éthanol chaud. Les extraits, évaporés sous vide, fournissent 20g (soit 50 % de rendement par rapport au 2-nitro-4-fluoro-toluène) de composé de formule III, F. = 1400 C.
Pour l'analyse on recristallise dans l'éthanol,
F. = 1420 C. Ce nouveau composé se présente sous forme d'aiguilles blanches, solubles dans l'éther et le chloroforme, moins solubles dans l'acétone, l'alcool ou le benzène, insolubles dans l'eau et les acides ou alcalis dilués aqueux.

C. Préparation de l'acide 6-fluora-indole-2-cnrboxy-
lique de formule IV
20 g de composé de formule III, obtenu selon le paragraphe B, sont mélangés avec 120 cm3 d'éthanol, 10g de potasse en pastilles et 100 cm3 d'eau.
On chauffe au reflux pendant une heure. On filtre, ajoute au filtrat 150cm3 d'eau, puis l'acidifie par addition d'acide chlorhydrique. Par refroidissement, on obtient l'acide de formule IV sous forme d'un précipité beige qu'on essore et lave à l'eau. On le redissout ensuite à chaud dans 200cm3 d'eau et 20 cm3 d'ammoniaque concentrée, ajoute du noir animal, filtre et réacidifie par l'acide chlorhydrique.
Après essorage et nouveau lavage à l'eau, on sèche à 800 C et recueille 10,9 g (soit 63 %) de composé de formule IV, F. supérieur à 2200 C. I1 est soluble dans l'alcool, l'éther et l'acétone, insoluble dans l'eau, le benzène et le chloroforme. On le recristallise pour l'analyse dans le méthanol aqueux.

D. Préparation du 6-fluoro-indole de formule V
On introduit 17,1 g de composé de formule IV, obtenu selon le paragraphe C, dans 85 cm3 de quinoléine contenant 1,7 g de chromite de cuivre et chauffe à la température intérieure de 210-220 C pendant une heure et quart. Après refroidissement, on verse dans 200 cm3 d'eau et extrait à l'éther. On épuise la solution éthérée à l'acide chlorhydrique concentré jusqu'à acidité nette, puis lave à l'eau et à l'ammoniaque diluée et encore à l'eau jusqu'à neutralité. La solution éthérée après séchage sur sulfate de sodium est évaporée à sec. On triture le résidu avec l'éther de pétrole, essore et sèche sous vide.
Rendement: 11 g de cristaux du composé de formule V, qu'on purifie par entraînement à la vapeur surchauffée à 1700 C. L'entraînement dure une demi-heure, on glace le distillat, essore, sèche sous vide et recueille 10,3 g (soit 80 %) de composé de formule V, F. = 730 C. Ce composé non décrit dans la littérature se présente sous forme de petites aiguilles blanches, solubles en alcool, acétone, benzène, éther et chloroforme, insolubles dans l'eau et les acides ou alcalis dilués aqueux.

E. Préparation de la 6-fluoro-gramine de formule VI
On prépare à une température inférieure à 50 C le mélange de 26 cm3 de diméthylamine en solution aqueuse à 23,4 %, 18 cm3 d'acide acétique et 1 1 cm3 de formol à 30 %, le laisse ensuite se réchauffer à la température ambiante et l'additionne de 13,5 g de 6-fluoro-indole de formule V, obtenu selon le paragraphe D. Le mélange réactionnel est agité pendant une heure, on le dilue ensuite avec 300 cm3 d'eau et alcalinise par la lessive de soude diluée.
On essore un précipité blanc qu'on lave à l'eau jusqu'à neutralité et sèche sous vide pour recueillir 18,1 g (soit 94 %) de composé de formule VI, F. = 136,50C.
Pour l'analyse on le recristallise dans le toluène, le point de fusion reste constant. Ce composé qui n'est pas décrit dans la littérature est soluble en alcool, éther, acétone, chloroforme et dans les acides dilués aqueux, peu soluble dans le benzène à froid, insoluble dans l'eau et les alcalis dilués.

F. Préparation du 6-fluoro-3-indolyl-acétonitrile de
formule Vll a) Sulfométhylate de 6-fluoro-gramine
Au mélange de 11,1 cm3 de sulfate de méthyle, 11,1 cm3 de tétrahydrofuran et 0,2 cm3 d'acide acétique glacial, on ajoute goutte à goutte et en agitant, une solution de 6-fluoro-gramine, obtenue selon le paragraphe E, dans 20 cm3 de tétrahydrofuran et 0,1 cm3 d'acide acétique glacial. On maintient 1' agi- tation pendant 45 minutes.
Le sulfométhylate de 6-fluoro-gramine est essoré, on l'empâte avec de l'éther et l'utilise aussitôt pour la réaction suivante. b) Formation du nitrile
Au sulfométhylate, obtenu selon b), dissous dans 100 cm3 d'eau, on ajoute 4,5 g de cyanure de potassium et chauffe le mélange réactionnel à 60-650 C sous agitation pendant une heure. On glace, essore, lave le précipité obtenu à l'eau, à l'acide chlorhydrique dilué puis de nouveau à l'eau et sèche sous vide pour recueillir 4,6 g (soit 85 %) de composé de formule VII brut. I1 est directement utilisable pour la suite de la synthèse. Pour l'analyse, on le purifie par sublimation à 1100 C sous 0,1 mm de mercure.
F. = 51-520 C.

Ce composé, qui n'est pas décrit dans la littérature, est soluble en alcool, éther, acétone et chloroforme, assez soluble dans le benzène et insoluble dans l'eau, les alcalis et acides dilués aqueux.
G. Préparation de la 6-fluoro-tryptamine de for
mule I
4 g de composé de formule VII, obtenu selon le paragraphe F, en solution dans 40 cm3 de méthanol saturé d'ammoniac sont hydrogénés en présence de 4,5 g de nickel de Raney. Après 2 heures d'hydrogénation, le volume consommé est de 850cm3. On filtre le catalyseur et évapore le méthanol sous vide.
Le résidu est dissous dans l'acide acétique à 10 %, on alcalinise jusqu'à p, 8 par la soude à 10 %, en refroidissant, traite au noir animal et filtre. On rajoute de la soude au filtrat jusqu'à alcalinité nette à la phénolphtaléine. ll se sépare une huile qui cristallise par refroidissement et amorçage. La tryptamine I est essorée, lavée à l'eau et séchée sous vide.
Rendement: 2 g (soit 50 %), F. = 800 C. Ce composé, qui n'est pas décrit dans la littérature, est peu stable et jaunit rapidement à l'air.

Pour former le picrate du composé de formule I, on dissout 5,47 g de cette tryptamine dans 25 cm3 d'alcool absolu et l'additionne de 7,7 g d'acide picrique dans 54ces d'alcool. Le picrate cristallise en aiguilles orangées qu'on essore après refroidissement et lave avec un peu d'alcool glacé. On sèche et obtient 11,3 g (soit 90 %) de sel de la tryptamine de formule I, F. = 2640 C. La recristallisation dans le méthanol ne change pas le point de fusion. Ce composé nouveau est soluble en acétone, à chaud dans l'alcool, peu soluble dans le chloroforme, le benzène et l'éther. I1 se décompose dans les acides ou alcalis dilués aqueux.

The process of the invention, summarized in the accompanying drawing, consists essentially in preparing 3-nitro-4-methyl-benzene-diazonium fluoborate from 2-nitro-4-aminotoluene which is dinitrogenated, then subjected to to the action of fluoboric acid. The fluorinated compound obtained provides under the effect of heat the 2-nitro-4-fluoro-toluene of formula II, the condensation of which with ethyl oxalate in the presence of an alkali ethoxide leads to the alkali salt of 4 ' Ethyl -fluoro2'-nitro-phenyl-pyruvate. The latter compound is reduced and cyclized by the action of a reducing agent in an acidic medium, such as for example zinc in acetic acid, or iron in hydrochloric acid, and the 6-fluoro-indole-2-carboxylate of ethyl of formula III.
Saponification of this indole derivative provides the corresponding acid of formula IV, which is decarboxylated to 6-fluoro-indole of formula V.
By condensation with dimethylamine and formalin, the 6-fluoro-indole is converted into 6-fluorogramine of formula VI, the sulfomethoxide of which by condensation with an alkali cyanide gives the 6-fluoro-3-indolyl-acetonitrile of formula VII. The catalytic hydrogenation of the latter leads to the desired 6fluoro-tryptamine of formula I.
The new compound finds its application as an intermediate in the synthesis of physiologically active compounds of the reserpine family. 6-Fluoro-tryptamine is also a potent tryptophan antimetabolite; it can be used whenever the microbiological transformation of tryptophan into indole is to be avoided, as is the case, for example, in certain pathological intestinal fermentations.
In the following examples, temperatures are expressed in degrees centigrade. The melting points are instantaneous melting points, determined on a Maquenne block.
Example
A. Preparation of 2-nitro-4-fluoro-toluene from for
mule II a) Formation of 3-nitro-4-methyl fluoborate
benzene-diazonium
Is added to 137 g of 2-nitro-4-amino-toluene, 90 cm3 of water and 270 cm3 of concentrated hydrochloric acid, stirred until the orange color disappears, then cooled to 00 C. The 182 g of fluoborate obtained according to a) are intimately mixed with 325 g of dry sand and heated directly over a flame. The product goes to fixed point. The refractive index of the yellow oil obtained of formula II is 1.522 at 250 C, in agreement with the values in the literature.
B. Preparation of 6-fluoro - indole -2 - carboxylate
ethyl of formula III
Dry potassium ethoxide, corresponding to 7.9 g of potassium, is covered with 100 cm3 of ether, corresponding to 7.9 g of potassium, added while cooling and with stirring 27.2 cm3 of ethyl oxalate, then very slowly 31 g of compound of formula II, obtained according to the previous paragraph. The reaction mixture is left at room temperature for 40 hours, then filtered off, washed with anhydrous ether and dried under vacuum to collect 44 g of purple crystals of the potassium salt of 4'-fluoro -2'-nitro - phenyl - pyruvate. ethyl.

C. Preparation of 6-fluora-indole-2-cnrboxy- acid

20 g of compound of formula III, obtained according to paragraph B, are mixed with 120 cm3 of ethanol, 10 g of potassium hydroxide pellets and 100 cm3 of water.
It is heated under reflux for one hour. After filtering, 150 cm3 of water are added to the filtrate, then acidified by adding hydrochloric acid. On cooling, the acid of formula IV is obtained in the form of a beige precipitate which is filtered off and washed with water. It is then redissolved hot in 200 cm3 of water and 20 cm3 of concentrated ammonia, added animal charcoal, filtered and reacidified with hydrochloric acid.
After draining and washing again with water, the mixture is dried at 800 ° C. and 10.9 g (ie 63%) of compound of formula IV, F. greater than 2200 C. I1 is soluble in alcohol, ether collected. and acetone, insoluble in water, benzene and chloroform. It is recrystallized for analysis from aqueous methanol.

D. Preparation of 6-fluoro-indole of formula V
17.1 g of compound of formula IV, obtained according to paragraph C, are introduced into 85 cm3 of quinoline containing 1.7 g of copper chromite and heated to the internal temperature of 210-220 C for one and a quarter hours. After cooling, poured into 200 cm3 of water and extracted with ether. The ethereal solution is exhausted with concentrated hydrochloric acid until clear acidity, then washed with water and dilute ammonia and again with water until neutral. The ethereal solution after drying over sodium sulphate is evaporated to dryness. The residue is triturated with petroleum ether, filtered off and dried under vacuum.
Yield: 11 g of crystals of the compound of formula V, which are purified by stripping with superheated steam at 1700 C. The entrainment lasts half an hour, the distillate is ice-cold, drained, dried under vacuum and collected 10.3 g (ie 80%) of compound of formula V, F. = 730 C. This compound, not described in the literature, is in the form of small white needles, soluble in alcohol, acetone, benzene, ether and chloroform, insoluble in water and dilute aqueous acids or alkalis.

E. Preparation of 6-fluoro-gramine of formula VI
The mixture of 26 cm3 of dimethylamine in 23.4% aqueous solution, 18 cm3 of acetic acid and 11 cm3 of 30% formalin is prepared at a temperature below 50 ° C., then allowed to warm to room temperature. and the addition of 13.5 g of 6-fluoro-indole of formula V, obtained according to paragraph D. The reaction mixture is stirred for one hour, then diluted with 300 cm 3 of water and basified with lye. diluted soda.
A white precipitate is filtered off and washed with water until neutral and dried under vacuum to collect 18.1 g (ie 94%) of compound of formula VI, F. = 136.50C.
For analysis it is recrystallized from toluene, the melting point remains constant. This compound which is not described in the literature is soluble in alcohol, ether, acetone, chloroform and in dilute aqueous acids, sparingly soluble in cold benzene, insoluble in water and dilute alkalis.

F. Preparation of 6-fluoro-3-indolyl-acetonitrile from
formula VII a) 6-fluoro-gramine sulfomethoxide
To the mixture of 11.1 cm3 of methyl sulfate, 11.1 cm3 of tetrahydrofuran and 0.2 cm3 of glacial acetic acid is added dropwise and with stirring, a solution of 6-fluoro-gramine, obtained according to paragraph E, in 20 cm3 of tetrahydrofuran and 0.1 cm3 of glacial acetic acid. Stirring is continued for 45 minutes.
The 6-fluoro-gramine sulfomethoxide is filtered off, impasted with ether and used immediately for the next reaction. b) Formation of nitrile
To the sulfomethoxide, obtained according to b), dissolved in 100 cm3 of water, 4.5 g of potassium cyanide are added and the reaction mixture is heated at 60-650 C with stirring for one hour. Ice, filtered off, washed the precipitate obtained with water, dilute hydrochloric acid and then again with water and dried under vacuum to collect 4.6 g (ie 85%) of crude compound of formula VII. It can be used directly for the rest of the synthesis. For analysis, it is purified by sublimation at 1100 ° C. under 0.1 mm of mercury.

G. Preparation of 6-fluoro-tryptamine from for
mule I
4 g of compound of formula VII, obtained according to paragraph F, dissolved in 40 cm3 of methanol saturated with ammonia, are hydrogenated in the presence of 4.5 g of Raney nickel. The residue is dissolved in 10% acetic acid, basified to p.8 with 10% sodium hydroxide, while cooling, treated with animal charcoal and filtered. Soda is added to the filtrate until clear alkalinity with phenolphthalein. An oil separates which crystallizes on cooling and priming. The tryptamine I is drained, washed with water and dried under vacuum.
Yield: 2 g (ie 50%), F. = 800 C. This compound, which is not described in the literature, is not very stable and quickly turns yellow in air.

To form the picrate of the compound of formula I, 5.47 g of this tryptamine is dissolved in 25 cm3 of absolute alcohol and the addition of 7.7 g of picric acid in 54ces of alcohol. The picrate crystallizes in orange needles which are filtered off after cooling and washed with a little iced alcohol. Dried and obtained 11.3 g (or 90%) of tryptamine salt of formula I, F. = 2640 C. Recrystallization from methanol does not change the melting point. This new compound is soluble in acetone, hot in alcohol, sparingly soluble in chloroform, benzene and ether. It decomposes in dilute aqueous acids or alkalis.
