Verfahren zur Herstellung von alkyfierten Pyrazolopyriniidinen Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von in 3-Stellung unsub- stitui,:
rten, an wenigstens einem Ringstickstoffatom einen gesättigten aliphatischen Rest tragenden 4,6- Dioxo-4,5,6,7 - tetrahydro-py razolo[3,4-d] pyrimidinen oder 4-Oxo-4,5-dihydro-pyrazolo[3,4-d]pyrimidinen, die in 6-Stellung unsubstituiert oder durch eine freie oder verätherte Mercaptogruppe oder durch eine Aminogruppe substituiert sind, oder 6-Oxo-6,
7-di- hydro-pyrazolo[3,4-djpyrimidinen, die in 4-Stellung durch eine freie oder verätherte Mercaptogruppe oder durch eine Aminogruppe substituiert sind, das da durch gekennzeichnet ist, dass man entsprechende Verbindungen, die an dem betreffenden Ringstick- stoffatom unsubstituiert sind, mit einer einen Rest der genannten Art abgehenden Verbindung behandelt.

Die neuen Verbindungen zeigen wertvolle phar makologische Eigenschaften, insbesondere koffein artige Wirkungen, und können als Stimulantia oder Diuretica sowie als Zwischenprodukte zur Herstel lung von Heilmitteln verwendet werden. Sie beein flussen ferner den Stoffwechsel. Besonders wertvoll sind an wenigstens 2 Ringstickstoffatomen niederalky- lierte Verbindungen der genannten Art, in denen sich einer der niederen Alkylreste in 1- oder 2-Stellung befindet, und ihre Salze.
Die in Frage stehenden Verbindungen können natürlich - wie dies bei derartigen Stoffen bekannt ist - auch ganz oder teilweise in tautomeren Formen vorliegen (z. B. 4,6-Dioxy-pyrazolo[3,4-d]pyrimidin anstatt 4,6-Dioxo-4,5,6,7-tetrahydro-pyrazolo[3,4-d]- pyrimidin). Verbindungen, die in 4- oder 6-Stellung eine Mercaptogruppe aufweisen, können nachträglich zu den Oxyverbindungen hydrolysiert werden.
Als gesättigte aliphatische Reste abgebende Mittel können z. B. reaktionsfähige Ester von Alkanolen, insbesondere des Methanols, oder auch von Amino- alkanolen verwendet werden, z. B. Alkyl- oder Aminoalkylhalogenide oder Dialkylsulfate, wobei je nach den angewendeten Reaktionsbedingungen eines oder mehrere der Ringstickstoffatome alkyliert wer den. Dabei können auch andere vorhandene substi- tuierbare Gruppen substituiert werden, wie z.
B. eine Mercaptogruppe.
Je nach den vorhandenen Substituenten in den Verfahrensprodukten lassen sich verschiedene Salze herstellen. Besitzen sie freie Oxy- oder Mercaptogrup- pen, so können Metallsalze gewonnen werden, z. B. durch Lösen in Alkalilaugen. Verbindungen von basischem Charakter, wie mit basischen Substituen- ten, bilden Salze mit anorganischen oder organischen Säuren.
Als salzbildende Säuren kommen beispiels weise in Frage: Halogenwasserstoffsäuren, Schwefel säuren, Phosphorsäuren, Salpetersäure, Perchlorsäure; aliphatische, alicyclische, aromatische oder hetero- cyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-, Dioxymalein- oder Brenztraubensäure;
Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Oxybenzoe-, Salicyl- oder p-Aminosalicylsäure; Methansulfon-, Athansulfon-, Oxyäthansulfon-, Äthylensulfonsäure; Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanil- säure;
Methionin, Tryptophan, Lysin oder Arginin. Die Ausgangsstoffe können gemäss den in den Schweizer Patentschriften Nr. 377834 oder Nr. 380151 beschriebenen Verfahren oder analogen Verfahren hergestellt werden.
Die oben beschriebenen Pyrazolopyrimidine, ihre Salze oder entsprechende Gemische können wie ge sagt als Arzneimittel Verwendung finden. Diese ent halten die genannten Verbindungen gewöhnlich in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial. Für dasselbe kommen solche Stoffe in Frage, die mit den beschriebenen Verbindungen nicht reagieren, wie z. B.
Wasser, Gelatine, Milchzucker, Stärke, Magne- siumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Polyalkylenglykole, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterili siert und bzw. oder enthalten Hilfsstoffe, wie Konser- vierungs-, Stabilisierungs-, Netz- oder Emulgiermit- tel. Sie können auch noch andere therapeutisch wert volle Stoffe enthalten.
<I>Beispiel 1</I> Eine Lösung von 15,2 g 4,6-Dioxy-pyrazolo[3,4- d]pyrimidin in 200 em=3 2n Natronlauge wird lang sam unter Rühren mit 42 g Dimethylsulfat versetzt. Man lässt dann während 20 Stunden bei Zimmertem peratur weiterrühren und zieht die Lösung mit viel Chloroform aus. Der Chloroformrückstand wird aus siedendem Wasser umkristallisiert. 2,5,7-Trimethyl- 4,6-dioxo-4,5,6,7-tetrahydro-pyrazolo[3,4-d]pyrimidin der Formel

<I>Beispiel 3</I> 14 g 4-Oxy-pyrazolo[3,4-d]pyrimidin werden in 150 cm, 2n Natronlauge gebracht; die Lösung wird unter Rühren langsam mit 30 g Dimethylsulfat ver setzt und dann während 10 Stunden bei Zimmertem peratur weitergerührt.
