die in einer der Stellungen 4 und 6 eine Oxogruppe und in der anderen eine gegebenenfalls substituierte Aminogruppe tragen.
Die Alkylreste in den neuen Verbindungen sind vorzugsweise niedermolekular; besonders zu nennen sind Methyl- und Isopropylreste. Als substituierte Aminogruppen sind vor allem Alkyl- aminogruppen zu nennen.
Die neuen Verbindungen zeigen wertvolle phar makologische Eigenschaften, insbesondere koffein artige Wirkungen, und können als Stimulantia oder Diuretica sowie als Zwischenprodukte zur Herstellung von Heilmitteln verwendet werden. Sie beeinflussen ferner den Stoffwechsel. Besonders wertvoll sind an wenigstens 2 Ringstickstoffatomen niederalkylierte Verbindungen dieser Art, in denen sich einer der niederen Alkylreste in 1- oder 2-Stellung befindet, und ihre Salze.
Das erfindungsgemässe Verfahren besteht darin, dass man entsprechende in 3-Stellung unsubstituierte, an wenigstens einem Ringstickstoffatom alkylierte Hydro-pyrazolo[3,4-d]pyrimidine, die in einer der Stellungen 4 und 6 eine Oxogruppe und in der anderen eine verätherte Mercaptogruppe tragen, mit Ammoniak oder Aminen umsetzt. Als verätherte Mercaptogruppe kommt dabei besonders eine Alkyl- mercaptogruppe in Frage.
Je nach den vorhandenen Substituenten in den Verfahrensprodukten lassen sich verschiedene Salze herstellen. Besitzen sie freie Oxy-, Mercapto- oder Carboxylgruppen, so können Metallsalze gewonnen werden, z. B. durch Lösen in Alkalilaugen. Verbin dungen von basischem Charakter, wie mit basischen Substituenten, bilden Salze mit anorganischen oder organischen Säuren.
Als salzbildende Säuren kommen beispielsweise in Frage: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Per chlorsäure; aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Amei sen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Oxy- malein-, Dioxymalein- oder Brenztraubensäure;
Phe-- nylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p- Oxybenzoe-, Salicyl- oder p-Aminosalicylsäure; Me- thansulfon-, Äthansulfon-, Oxyäthansulfon-, Äthylen- sulfonsäure; Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure;
Methionin, Tryptrophan, Lysin oder Arginin.
Die Ausgangsstoffe können z. B. gemäss den in den Schweizer Patentschriften Nrn.380150 und 377834 beschriebenen Verfahren oder analogen Ver fahren gewonnen werden.
Die oben beschriebenen Pyrazolopyrimidine, ihre Salze oder entsprechende Gemische können, wie ge sagt, als Arzneimittel Verwendung finden. Diese ent halten die genannten Verbindungen gewöhnlich in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial. Für dasselbe kommen solche Stoffe in Frage, die mit den beschriebenen Verbindungen nicht reagieren, wie z. B.
Wasser, Gelatine, Milchzucker, Stärke, Magnesium- stearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Polyalkylenglykole, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und bzw.
oder enthalten Hilfsstoffe, wie Konservierungs-, Sta- bilisierungs-, Netz- oder Emulgiermittel. Sie können auch noch andere therapeutisch wertvolle Stoffe ent halten.
<I>Beispiel 1</I> 10 g 2,7-Dimethyl-4-methylmercapto-6-oxo-6,7- dihydro-pyrazolo[3,4-d]pyrimidin und 100 cm33 flüs siger Ammoniak werden während sechs Stunden im geschlossenen Rohr auf 100 erhitzt. Die nach dem Abdampfen des Ammoniaks zurückgebliebenen grauen Kristalle werden aus Alkohol umkristallisiert; so wird 2,7-Dimethyl-4-amino-6-oxo-6,7-dihydro- pyrazolo[3,4-d]pyrimidin der Formel

Versetzt man dieses mit alkoholischer Salzsäure, so erhält man dessen Monochlorhydrat, das bei 312 unter Zersetzung schmilzt.
