Verfahren zur Herstellung neuer 7-Aza-benzimidazole
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 1 -Aminoalkyl-7-aza-benzimidazolen der Formel I

worin R1 ein Wasserstoffatom, eine niedere Alkyl-, Alkoxy- oder Alkylmerkaptogruppe oder eine Hydroxygruppe bedeutet, R2 für ein Wasserstoffatom oder die Nitrogruppe steht und R3 und R4 niedere Alkylgruppen darstellen oder zusammen mit dem Stickstoffatom einen heterocyclischen Ring bilden, der auch weitere Heteroatome enthalten kann.
Die neuen Verbindungen und ihre Salze besitzen zentralerregende Eigenschaften, insbesondere sind sie analeptisch wirksam, fördern die Atmung und zeigen Antagonismus gegenüber Narkotika. Sie können dementsprechend als Arzneimittel Verwendung finden.
Von besonderem Interesse wegen ihren therapeutischen Eigenschaften sind in 2-Stellung unsubstituierte Verbindungen, vor allem 1-(ss-Diäthylamino-äthyl)-5- nitro-7-aza-benzimidazol und seine Salze.
Die neuen Azabenzimidazole werden erfindungsgemäss erhalten, wenn man ein Azabenzimidazol der Formel II

mit einem reaktionsfähigen Ester eines Alkohols der Formel HO-CH2-CH2-NR R umsetzt. 3 4
Reaktionsfähige Ester sind insbesondere solche starker anorganischer oder organischer Säuren, wie der Halogenwasserstoffsäuren, oder organischer Sulfonsäuren, wie p-Toluolsulfonsäure. Die Einführung wird vorzugsweise vorgenommen in Gegenwart von Kondensationsmitteln, insbesondere solcher, die mit den Azabenzimidazolen Metallsalze zu bilden vermögen, wie Alkali- und Erdalkalimetalle, beispielsweise Natrium, Lithium, Calcium, ihre Amide, Hydride, Kohlenwasserstoffverbindungen, Alkoholate, Oxyde oder Hydroxyde, z. B.
Natriumamid, Natriumhydrid, Butyllithium, Phenylkalium, Phenyllithium, Kalium-tert.-butylat, Kalium-tert.-amylat, Natrium äthylat, Natriumoxyd oder Natriumhydroxyd, oder unter Verwendung der vorgebildeten Metallsalze der Benzimidazole. Erhält man bei dieser Umsetzung ein Gemisch von stellungsisomeren Verbindungen, so kann dieses in die einzelnen Komponenten getrennt werden, beispielsweise durch Kristallisation der Basen oder ihrer Salze.
Die verfahrensgemässen Umsetzungen können in An- oder Abwesenheit von Verdünnungs-undloder Kondensationsmitteln, wenn nötig, bei erhöhter Temperatur, im offenen oder im geschlossenen Gefäss unter Druck ausgeführt werden.
Je nach der Arbeitsweise erhält man die neuen Verbindungen in Form der freien Basen oder ihrer Salze. Aus den Salzen können in an sich bekannter Weise die freien Basen gewonnen werden. Von letzteren lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen, wie z. B. der
Halogenwasserstoffsäuren, Schwefelsäure,
Salpetersäure, Phosphorsäure,
Rhodanwasserstoffsäure, Essigsäure,
Propionsäure, Oxalsäure, Malonsäure,
Bernsteinsäure, Äpfelsäure, Methansulfonsäure, Äthansulfonsäure, Oxyäthansulfonsäure,
Benzol- oder Toluolsulfonsäure oder von therapeutisch wirksamen Säuren.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.
Die genannten Verbindungen können als Arzneimittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten, pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Im folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
0,95 g 5 (6)-Nitro-7(4)-aza-benzimidazol werden in 50 ml Äthanol suspendiert und mit einer Lösung von 0,15 g Natrium in 30 ml Äthanol versetzt. Die Nitroverbindung geht dabei als orangefarbenes Salz in Lösung. Nun wird zu dieser Lösung innerhalb 10 Minuten eine Lösung von 0,8 g ss-Diäthylamino äthylchlorid in 20 ml Äthanol unter Rühren zugetropft, wobei die Reaktionslösung von orange in gelb umschlägt. Nach beendeter Zugabe wird das Reaktionsgemisch 1 Stunde bei 600 gerührt und danach im Vakuum eingedampft. Der Rückstand wird mit einem Gemisch aus 10 ml 2n Natronlauge und 100 ml Äther durchgeschüttelt, der Ätherextrakt zweimal mit gesättigter Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet und eingedampft.
Das zurückbleibende öl wird mit einem Äquivalent alkoholischer Salzsäure versetzt und das Hydrochlorid durch vorsichtige Zugabe von Äther zur Kristallisation gebracht. Das so gewonnene Hydrochlorid schmilzt unscharf bei 170-180 und besteht aus zwei Isomeren, die sich durch fraktionierte Kristallisation aus Alkohol weitgehend trennen lassen.
Aus den so vorgereinigten Hydrochloriden kann man durch Freisetzen der Basen und Umkristallisation der letzteren aus Äther-Pentan das l-(ss-Diäthylamino-äthyl)-5- nitro-7-aza-benzimidazol F. 66-670 (Schmelzpunkt des Hydrochlorids 206208G) der Formel

isolieren. Die beiden Isomeren haben im UV-Absorptionsspektrum zwei ausgeprägte Banden bei 235 und 305 mp. u. Sie unterscheiden sich aber deutlich durch das Verhältnis der Extinktionen dieser zwei Banden.
Während bei der zuerstgenannten Verbindung 305f 235 = 0,43 beträgt, ist dieser Faktor bei dem Isomeren 6-Nitro-7-azaverbindung 051o 235 = 0,66.
Das als Ausgangsmaterial verwendete 5(6)-Nitro7(4)-aza-benzimidazol der Formeln

lässt sich gemäss nachstehender Reaktionsfolge herstellen:
11 g 2-Chlor-3,5-dinitro-pyridin werden in 80 ml Alkohol gelöst und unter Rühren mit einem Gemisch aus 30 ml konz. Ammoniak und 80 ml Alkohol tropfenweise versetzt, wobei sich die Lösung orangegelb verfärbt und bald ein orangegelbes Produkt auskristallisiert. Nach beendeter Zugabe wird 15 Minuten weitergerührt, auf 0O abgekühlt und filtriert. Das so gewonnene 2-Amino-3 , 5-dinitro-pyridin schmilzt bei 190-192 .
8,8 g von dieser Verbindung werden mit einem Gemisch aus 94 ml Alkohol und 27 ml konz. Ammoniak versetzt und die Suspension wird unter Rühren gleichzeitig mit einer Lösung von 16,2 g Natriumhydrosulfid in 47 ml Wasser und einer Lösung von 16 g Ammoniumchlorid in ebenfalls 47 ml Wasser tropfenweise innerhalb 15 Minuten versetzt. Das Reaktionsgemisch färbt sich tief rot und bald kristallisiert ein dunkelrotes Produkt aus. Nach 3 Stunden wird dieses abgesaugt und mit einem Gemisch aus gleichen Teilen Alkohol und Äther gewaschen. Das so gewonnene 2,3-Diamino-5-nitro-pyridin schmilzt bei 260O.
5 g dieses Nitrodiamins werden mit 10 ml Ameisensäure 20 Minuten am Rückfluss erwärmt, das Reaktionsgemisch wird im Vakuum eingedampft, der Rückstand mit Wasser verrieben, mit Ammoniak auf pH 6 gestellt und abgenutscht. Aus diesem Rohprodukt gewinnt man nach Auskochen mit 200 ml Alkohol in Gegenwart von weniger Norit und Einengen der orangegelben, filtrierten Lösung auf 80 ml ein kristallisiertes, orangegelb gefärbtes Produkt. Es handelt sich um das 5(6)-Nitro-7(4)-aza-benzimidazol, das sich beim Erwärmen oberhalb 2600 unter Sintern allmählich zersetzt.

The new compounds and their salts have central excitatory properties, in particular they are analeptically effective, promote breathing and show antagonism to narcotics. Accordingly, they can be used as medicinal products.
Of particular interest because of their therapeutic properties are compounds unsubstituted in the 2-position, especially 1- (ß-diethylamino-ethyl) -5-nitro-7-aza-benzimidazole and its salts.

The nitro compound goes into solution as an orange salt. A solution of 0.8 g of ss-diethylamino ethyl chloride in 20 ml of ethanol is then added dropwise to this solution within 10 minutes, with stirring, the reaction solution turning from orange to yellow. After the addition has ended, the reaction mixture is stirred at 600 for 1 hour and then evaporated in vacuo. 5 g of this nitrodiamine are refluxed with 10 ml of formic acid for 20 minutes, the reaction mixture is evaporated in vacuo, the residue is triturated with water, adjusted to pH 6 with ammonia and suction filtered. A crystallized, orange-yellow colored product is obtained from this crude product after boiling with 200 ml of alcohol in the presence of a little Norit and concentration of the orange-yellow, filtered solution to 80 ml.
