Von den gemäss der Erfindung erhältlichen Ver bindungen sind besonders das 2,5,7-Trimethyl-4,6- dioxo - 4,5,6,7 - tetrahydro - pyrazolo[3,4-d]pyrimidin und das 2,5-Dimethyl-6-methylmercapto-4-oxo-4,5- dihydro-pyrazolo[3,4-d]pyrimidin von Interesse.
Die neuen Verbindungen wie auch ihre Salze zeigen wertvolle pharmakologische Eigenschaften, insbeson dere koffeinartige Wirkungen, und können als Stimu lantia oder Diuretica sowie als Zwischenprodukte zur Herstellung von Heilmitteln verwendet werden.
Die obengenannten Verbindungen werden erfin- dungsgemäss erhalten, indem man 3-Amino-pyrazol- 4-carbonsäuren oder ihre funktionellen Säurederivate der Teilformel

In erhaltenen Verbindungen mit freier oder verätherter Mercaptogruppe in 6-Stel- lung kann diese zur Oxygruppe hydrolysiert werden.
Zur Umsetzung mit den Aminopyrazolen verwen det man in erster Linie Harnstoff, Thioharnstoff oder, sofern die funktionell abgewandelte Carboxylgruppe der Pyrazolverbindung eine Carbonamidgruppe ist, z. B. einen Chlorameisensäureester oder Phosgen.
Die Kondensation der Aminopyrazole zu den Pyrazolopyrimidinen erfolgt vorzugsweise bei Tem peraturen über 100 , gegebenenfalls in Anwesenheit von Verdünnungs- und/oder Kondensationsmitteln im offenen oder geschlossenen Gefäss.
Je nach den vorhandenen Substituenten in den Verfahrensprodukten lassen sich verschiedene Salze herstellen. Besitzen sie freie Oxy- oder Mercapto- gruppen, so können Metallsalze gewonnen werden, z. B. durch Lösen in Alkalilaugen. Verbindungen von basischem Charakter, wie mit basischen Substituen- ten, bilden Salze mit anorganischen oder organischen Säuren.
Als salzbildende Säuren kommen beispiels weise in Frage: Halogenwasserstoffsäuren, Schwefel säuren, Phosphorsäuren, Salpetersäure, Perchlor- säure; aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Gly- kol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-,
Oxymalein-, Dioxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Oxybenzoe-, Salicyl- oder p-Aminosalicylsäure; Me- thansulfon-, Äthansulfon-, Oxyäthansulfon-, Athylen- sulfonsäure;
Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Die als Ausgangsstoffe zu verwendenden, in 4- Stellung eine freie oder funktionell abgewandelte Carboxylgruppe aufweisenden 3-Amino-pyrazole wer den z.
B. durch Umsetzung von unsubstituierten oder substituierten a-Cyan-ss-oxo-propionsäuren oder ihren funktionellen Derivaten, wie ihren Estern oder Ami- den und/oder ihren Enoläthern, Acetalen oder Mer- captalen mit Hydrazinen erhalten. Letztere können unsubstituiert oder auch mono- oder N,N'-disubsti- tuiert sein, wie z.
B. durch Alkylreste. Als funktio nelle Derivate einer a-Cyan-ss-oxo-propionsäure wer den vorzugsweise Enoläther der a-Cyan-ss-oxo-pro- pionsäureester, wie z. B. der Äthoxymethylencyan- essigsäureäthylester, verwendet. Die Kondensation zu den Pyrazolen verläuft unter milden Bedingungen, zum Teil bei Zimmertemperatur und exotherm. Man kann auch bei höherer Temperatur und in Anwesen heit von Kondensationsmitteln arbeiten, wie z.
B. in Gegenwart von Säuren. Vorteilhaft setzt man die Reaktionsteilnehmer in Anwesenheit eines Verdün nungsmittels, wie eines Alkohols, Toluol oder Chloro form, um.
<I>Beispiel 1</I> 50 g 3-Amino-4-carbäthoxy-pyrazol und 100 g Harnstoff werden gut vermischt und während 40 Mi nuten in einem Bade von 2000- erhitzt. Man zieht dann die Schmelze mit 400 cm3 heisser 1-n. Natron lauge aus, filtriert von. wenig nicht Gelöstem ab und stellt das Filtrat mit Eisessig sauer, worauf ein weisser Niederschlag ausfällt, von dem abgenutscht wird. Man erhält so 4,6-Dioxy-pyrazolo[3,4-d]pyrimidin in weissen Kristallen, die bei 300 noch nicht geschmol zen sind.
Eine Lösung von 15,2 g 4,6-Dioxy-pyrazolo- [3,4-d]pyrimidin in 200 cm3 2-n. Natronlauge wird langsam unter Rühren mit 42g Dimethylsulfat ver setzt. Man lässt dann während 20 Stunden bei Zim mertemperatur -weiterrühren und zieht die Lösung mit viel Chloroform aus. Der Chloroformrückstand wird aus siedendem Wasser umkristallisiert. 2,5,7- Trimethyl-4,6-dioxo - 4,5,6,7 - tetrahydro-pyrazolo- [3,4-d]pyrimidin der Formel

The chloroform residue is recrystallized from alcohol to give 1-isopropyl-4,6-di oxo-5,7-dimethyl-4,5,6,7-tetrahydropyrazolo [3,4-d] pyrimidine of the formula

The 2-isopropyl-3-amino-4-carbamyl-pyrazole used as starting material can be obtained as follows: 30 g of isopropylhydrazine are added to a solution of 48.8 g of ethoxymethylene malonitrile in 500 cm 3 of alcohol. The mixture is then heated to boiling for 10 hours, evaporated to dryness in vacuo and crystallized from a lot of isopropyl ether. 2-Isopropyl-3-amino-4-cyano-pyrazole is obtained in this way in white crystals with a melting point of 94-95. 10 g of the compound produced in this way are 2-n with 200 cm3.
Sodium hydroxide solution and 100 cubic meters of alcohol are added and the solution is heated to boiling for 3 hours. The alcohol is evaporated off in vacuo, left to cool and the precipitate which has separated out is popped off. The latter is recrystallized from alcohol. This gives 2-isopropyl-3-amino-4-carbamyl-pyrazole of the formula
