Procédé de préparation d'un dicarbamate
La présente invention a pour objet un procédé de préparation du dicarbamate de 2-phényl-1, 3-pro- pane-diol. Ce nouveau composé organique possède une activité antispasmodique exceptionnellement intense dans la prévention des attaques consécutives à l'électrochoc.
Ce composé est un solide cristallisé blanc soluble dans la plupart des solvants organiques, mais seulement faiblement soluble dans l'eau. Ses solutions aqueuses et dans les solvants organiques sont stables.
Par chauffage ou ébullition avec un acide ou une base, ce composé s'hydrolyse avec formNation de 1,3-propane-diol substitué en 2 correspondant, d'ammoniaque et d'anhydride carbonique.
Ce composé nouveau est préparé par réaction du 2-phényl-1, 3-propanbdiol avec du phosgène de manière à former le dichlorocarbonate correspondant puis par réaction de celui-ci avec de l'ammoniaque.
La première étape du procédé peut tre effectuée en présence d'un agent de fixation d'acide comme l'hydroxyde de sodium, l'antipyrine ou une dialcoyl- aniline. L'ammoniaque employé dans la deuxième étape du procédé selon l'invention, peut tre anhydre ou aqueux.
Le 2-phényl-1, 3-propane-diol utilisé comme matière première peut tre préparé par un procédé quel- conque connu tel que, par exemple, la réduction de l'ester malonique substitué en 2 correspondant.
Dans un ballon de 500 oc contenant 150 cc de toluène sec refroidi à-10 C, on dispose de 33 g de phosgène (0,3 mol. + 10 < '/o). On ajoute 23 g de 2- phényl-1, 3-propane-diol dissous dans 100 ce de toluène contenant 31 g (0,3 mol.) de N, N-diméthyl- aniline. On maintient la température interne entre-5 et + 5"C. L'addition se fait en vingt minutes environ. On lave le mélange réactionnel froid à l'aide
d'acide chlorhydrique à 5 /o, on sèche la couche toluénique sur sulfate de sodium et on la sature de gaz ammoniac, opération au cours de laquelle la température augmente. On chauffe le mélange au reflux et on le filtre.
Après refroidissement du filtrat, on
recueille le dicarbamate de 2-phényl-1, 3-propane diol solide, formé et on le fait recristalliser au sein d'un mélange 1/1 de benzène et d'éther de pétrole.
Les essais du composé effectués sur des animaux d'expérience convenables, tels que la souris, quant à sa faculté de prévenir les attaques consécutives aux électrochocs, montrent qu'il possède une activité anti- spasmodique remarquable. En revanche, il est relativement inefficace quant à la protection des animaux contre les convulsions produites par le pentylène tetrazal. Ce composé nouveau est exempt d'action hypnotique et ne provoque pas la paralysie des muscles volontaires, sauf aux très fortes doses. Les doses produisant le sommeil ou la paralysie chez la souris sont supérieures à 2000 mg/kg alors que les doses protégeant contre les attaques, provoquées par l'élec- trochoc, sont de l'ordre de 60 à 80 mg/kg.
Le composé est remarquable en raison de sa très faible toxicité. La dose moyenne léthale chez la souris par administration par voie intrapéritonéale est d'environ 4000 mg/kg et la dose moyenne léthale chez la souris après administration par voie orale est d'environ 8000 mg/kilogramme.
Le composé préparé selon le présent procédé est destiné principalement à l'administration par voie orale et mis sous forme de pilules, comprimés ou capsules par les moyens bien connus. Il peut tre dissous dans un solvant approprié comme un mélange d'eau et de polyéthylène-glycol avec formation d'une solu- tion propre aux injections ou à l'administration en n lavements.
Le produit de départ peut tre préparé comme suit :
On réduit 50 g de phényl-malonate de diéthyle de la manière usuelle à l'aide de 12 g d'hydrure d'alu- minium et de lithium, la réduction étant effectuée dans 500 cc d'éther anhydre. On utnise l'excès d'agent réducteur avec un faible volume d'acétate d ? éthyle. On traite le mélange réactionnel de la manière usuelle au moyen, d'eau et d'acide sulfurique étendu, les composants, organiques étant épuisés à l'aide d'éther. On obtient 16 g, soit environ 50 ID/o du rendement théorique, de produit purifié fondant à 49-52 C.

Process for preparing a dicarbamate
The present invention relates to a process for the preparation of 2-phenyl-1, 3-propan-diol dicarbamate. This new organic compound has exceptionally intense antispasmodic activity in the prevention of attacks following electroshock.
This compound is a white crystalline solid soluble in most organic solvents, but only slightly soluble in water. Its aqueous solutions and in organic solvents are stable.
Upon heating or boiling with an acid or a base, this compound hydrolyzes to form the corresponding 2-substituted 1,3-propanediol, ammonia and carbon dioxide.
This new compound is prepared by reacting 2-phenyl-1, 3-propanbdiol with phosgene so as to form the corresponding dichlorocarbonate and then by reacting the latter with ammonia.
The first step of the process can be carried out in the presence of an acid binding agent such as sodium hydroxide, antipyrine or a dialkylaniline. The ammonia used in the second step of the process according to the invention can be anhydrous or aqueous.
The 2-phenyl-1,3-propanediol used as raw material can be prepared by any known process such as, for example, the reduction of the corresponding 2-substituted malonic ester.
In a 500 oc flask containing 150 cc of dry toluene cooled to -10 ° C., 33 g of phosgene (0.3 mol. + 10 <'/ o) are available. 23 g of 2-phenyl-1,3-propanediol dissolved in 100 cc of toluene containing 31 g (0.3 mol) of N, N-dimethyl-aniline are added. The internal temperature is maintained between -5 and + 5 ° C. The addition takes place over about twenty minutes. The cold reaction mixture is washed with
5% hydrochloric acid, the toluene layer is dried over sodium sulfate and saturated with ammonia gas, an operation during which the temperature increases. The mixture is heated to reflux and filtered.
After cooling the filtrate,
collects the solid 2-phenyl-1,3-propane diol dicarbamate formed and it is recrystallized from a 1/1 mixture of benzene and petroleum ether.
Tests of the compound carried out on suitable experimental animals, such as mice, as to its ability to prevent attacks subsequent to electroshock, show that it possesses remarkable anti-spasmodic activity. On the other hand, it is relatively ineffective in protecting animals against seizures produced by pentylene tetrazal. This new compound is devoid of hypnotic action and does not cause paralysis of voluntary muscles, except at very high doses. The doses producing sleep or paralysis in mice are greater than 2000 mg / kg, while the doses protecting against attacks, caused by electroshock, are of the order of 60 to 80 mg / kg.
The compound is remarkable because of its very low toxicity. The mean lethal dose in mice by intraperitoneal administration is approximately 4000 mg / kg and the mean lethal dose in mice after oral administration is approximately 8000 mg / kg.
The compound prepared according to the present process is intended primarily for oral administration and formed into pills, tablets or capsules by well known means. It can be dissolved in an appropriate solvent such as a mixture of water and polyethylene glycol with formation of a solution suitable for injections or administration in n enemas.
The starting product can be prepared as follows:
50 g of diethyl phenyl malonate are reduced in the usual manner with 12 g of lithium aluminum hydride, the reduction being carried out in 500 cc of anhydrous ether. The excess reducing agent is used with a small volume of acetate? ethyl. The reaction mixture is worked up in the usual manner with water and extended sulfuric acid, the organic components being exhausted with ether. 16 g, or approximately 50 ID / o of the theoretical yield, of purified product, melting point 49-52 C.
