<B>Procédé pour la</B> préparation <B>d'une</B> substance antibiotique La présente invention est relative à un pro cédé pour la préparation de tétracycline. Le procédé suivant la présente invention, dans lequel on inocule et fait fermenter sous des conditions d'aérobiose un milieu nutritif aqueux renfermant des substances minérales et des matières qui fournissent, sous forme assi- miIable, du carbone et de l'azote, avec un microorganisme du genre Streptomyces capa ble de produire de la chlortétracycline dans un milieu nutritif contenant des ions chlorure,
est caractérisé en ce qu'on règle la teneur du mi lieu nutritif en ions chlorure de manière à obtenir un produit antibiotique renfermant en majeure partie de la tétracycline.
La substance antibiotique obtenue est ef fectivement la tétracycline, car on a constaté qu'elle possède la structure du composé hypo thétique de base de la chlortétracycline et de l'oxytétracycline (Brunings et al; J.A.C.S., 74 ; 4976-4977, 5 octobre 1952).
L'absence de chlore différencie cette substance de la chlor- tétracycline et son comportement à d'autres égards démontre qu'elle est différente de l'oxy- tétracycline. La tétracycline a pour formule brute C22H24N.O8 <B>et</B> la formule de structure sui vante

La tétracycline, sous forme libre, est am photère et forme des sels tant avec des acides qu'avec des bases.
Un milieu de fermentation convenant pour la mise en oeuvre du procédé suivant l'inven tion contient une source de carbone, telle qu'un hydrate de carbone assimilable, une source d'azoie assimilable, des sels inorganiques, tels que des phosphates, des sels de magné sium, etc., et des sources des éléments mineurs habituels. Des tampons sont ordinairement in clus dans le milieu de fermentation.
La source de carbone peut être soit un hy drate de carbone soluble, tel que le saccharose, soit un hydrate de carbone insoluble, tel qu'un amidon, comme l'amidon de maïs. On peut également utiliser de la dextrine. Alors que le saccharose, qui constitue un sucre soluble, est très satisfaisant, le lactose et le glucose, qui constituent deux autres sucres solubles, sont relativement médiocres. La quantité de source de carbone peut varier dans de larges limites, soit de 0,5 à 10,0,1/o en poids environ, par rap port au -poids total du milieu de fermentation.
Comme source d'azote assimilable, on peut utiliser les sels d'ammonium inorganiques. Parmi ces sels, on peut citer le sulfate d'am monium, le phosphate d'ammonium, etc. Il est également possible d'utiliser-. des sources d'azote organiques telles que les aminoacides et diverses matières protéiniques naturelles.
Parmi les sels inorganiques qui se sont ré vélés avantageux, on peut citer les phosphates, qui peuvent être présents sous forme de phos phate d'ammonium ou de phosphate métalli que, notamment sous forme de phosphate de potassium, de phosphate de magnésium, le phosphate de potassium étant présent, si une autre source de potassium n'est pas présente.
L'addition de faibles quantités de métaux lourds est désirable, à moins que de tels mé taux soient présents dans les autres ingrédients. Parmi ces éléments, qui peuvent être présents sous forme de traces, par exemple, comme im puretés, ou être ajoutés, on peut citer le cui vre, le zinc, le manganèse, le fer et le chrome.
Parmi les agents tampons, on peut citer le carbonate de calcium et les sels d'acides orga niques, tels que citrates, -acétates et lactates, qui sont utiles pour maintenir le pH dans les limites voulues. En outre, les acides organiques peuvent servir comme sources de carbone dans le métabolisme du microorganisme. L'emploi d'un agent démoussant est souhaitable dans les fermentateurs de grande capacité. Comme agent démoussant, on peut utiliser, par exem ple, l'octadécanol dans de l'huile de lard.
Il est souhaitable de cultiver le microorga nisme dans des conditions d'aérobiose submer- gée, en agitant et en aérant convenablement les cultures, par exemple dans un flacon sur une machine à agiter ou dans un fermentateur agité, équipé d'un dispositif permettant d'y in troduire, de manière continue, un courant d'air. La température ne parait pas être criti que dans la gamme de température de 25 à 35,) C, bien qu'on préfère opérer à une tempé rature comprise entre 30 et 33o C.
Le<I>pH</I> initial du milieu doit, de préférence, être voisin de la neutralité, bien qu'une certaine quantité d'antibiotique soit également produite dans les milieux présentant des valeurs de<I>pH</I> initiales de 5,0 seulement ou atteignant 8,5.
Pour la mise en oeuvre du procédé suivant l'invention, on utilise, de préférence, des micro organismes de l'espèce Streptomyces auréofa- ciens.
Les souches, variantes ou mutantes de Streptomyces auréofaciens, auxquelles on fait appel, de préférence, pour la mise en #uvre de la présente invention, sont celles qui produisent des rendements relativement élevés en tétra cycline dans le milieu de fermentation, en par ticulier celles qui produisent plus de 500 mcg/cc. Un exemple est la souche UV-8 qui, lorsqu'on la fait pousser sur l'agar de Waks man (Journal Bacteriology :
7 ;<B>339-341,</B> 1952) produit une forte croissance mycélienne, qui est d'abord de couleur blanche sale et de vient ensuite jaune, et qui est graduellement recouverte d'un mycélium aérien poudreux et blanc, subissant ultérieurement une sporula- tion. De vieilles cultures présentent une colo ration noir jais avec de petites pièces de mycé lium blanc. Cette croissance noire, qui n'a pas été révélée jusqu'ici dans les cultures de Strep tomyces auréofaciens et n'est pas une caracté ristique de l'espèce, consiste en une masse de corps en courtes chaînes, qui sont aisément brisées. Ces corps présentent des formes et dimensions variables.
Les dimensions vont de 0,5 à 4,5 m[., tandis que la forme des corps en question va de celle d'un ovale à celle de sphérules légèrement angulaires. Les dimen sions moyennes sont quelque peu supérieures à celles obtenues dans le Streptomyces auréo- faciens (HRRL <I>- 2209)</I> et les variations de dimensions et de forme sont plus grandes.
Les mutantes naturelles ou induites de Streptomyces auréofaciens peuvent produire de la tétracycline en quantités variables, en plus d'autres antibiotiques. En choisissant une mutante naturelle ou induite, qui produit un rapport relativement élevé de tétracycline à d'autres antibiotiques, et en propageant le microorganisme dans des conditions con-trô- lées, on peut obtenir un rendement relative ment élevé en tétracycline. Il existe plusieurs manières de régler la teneur en ions chlorure du milieu de fermen tation. Une de ces manières consiste à séques trer les ions chlorure dans un complexe inactif.
Des résultats satisfaisants ont été ob tenus par utilisation d'eau distillée et par uti lisation dans le milieu de constituants ne con tenant pas d'ions chlorure. De nombreuses ma tières naturelles, souvent utilisées dans les mi lieux de fermentation, contiennent des ions chlorure. On peut réduire leur teneur en ions chlorure par traitement avec des échangeurs d'ions ou d'autres agents convenant pour l'éli mination des ions chlorure. La titulaire a mis au point un milieu syn thétique qui est sensiblement exempt de ions chlorure, qui donne des rendements supérieurs, tout en étant facile à préparer, de coût mo dique et de composition constante.
Ce milieu contient du saccharose comme hydrate de car bone, du sulfate d'ammonium comme source d'azote, du phosphate de potassium comme sel inorganique, du sulfate de magnésium, du carbonate de calcium, du citrate de sodium et de l'acide acétique comme agents tampons, ainsi que du sulfate de cuivre, du sulfate de zinc et du sulfate de manganèse en petites quantités, comme sources d'éléments mineurs. Des qualités sensiblement exemptes d'ions chlorure de ces matières sont choisies et de l'eau distillée est utilisée, de façon que le mi lieu de fermentation ne contienne pas plus de 1 partie d'ions chlorure pour 1 million de par ties de milieu de fermentation.

<SEP> 20 <SEP> g/1 La culture peut être transférée dans des flacons à agiter, qui peuvent être utilisés comme fermentateurs de laboratoire ou pour la production d'inoculum pour fermentation à plus grande échelle. Pour la fermentation en réservoir à grande échelle, la culture d'inoculum est utilisée pour ensemencer un milieu liquide approprié dans un flacon à agiter, qui est agité sur une ma chine à agiter à mouvement de va-et-vient à des températures comprises entre 26 et 350 C, de préférence à 30 - 330 C.
Une - seconde culture en flacon à agiter est ordinairement appliquée, dans les cas de fermentations en immersion dans un réservoir, afin d'augmenter le volume de l'inoculum jusqu'à 1,5 - 2,5 % du volume du liquide dans le réservoir.
Le<I>pH</I> du milieu contenu dans le réservoir à agiter est initialement de 6,5 - 7,5 et, pendant la croissance, il se produit une diminution con- troue du<I>pH,</I> des valeurs de l'ordre de 3,8 étant enregistrées. Toutefois, un âge physiologique, tel qu'indiqué par une valeur de<I>pH</I> de 5,0, est apparemment la période la plus favorable pour le transfert de l'inoculum.
L'inoculum, dont la quantité correspond à 1,5 - 2,5 % du volume de liquide dans le ré- servoir, est transféré aseptiquement au fermen- tateur et cultivé pendant environ 2 à 3 jours, en agitant et en aérant de manière continue. Selon les dimensions du fermentateur,
on peut utiliser de 0,5 à 1,5 volume d'air par volume de milieu liquide. La formation de mousse dans le milieu peut être contrôlée par l'addi tion stérile d'un agent antimousse, tel que de l'huile de lard contenant 2'% d'octadécanol.
Pour certaines applications, il n'est pas nécessaire d'isoler la tétracycline du milieu de fermentation. Ainsi, le produit de fermentation peut être ajouté directement à des aliments pour animaux, en vue d'obtenir un produit convenant particulièrement pour stimuler la croissance et/ou prévenir ou traiter certaines maladies des animaux. Lorsqu'on veut élimi ner le mycélium et d'autres matières insolu bles, le milieu de fermentation peut être filtré avec ou sans acidification et la liqueur claire peut être ajoutée à l'eau de breuvage ou à des aliments pour animaux.
Si on le désire, la li queur contenant la tétracycline peut être con centrée sous pression réduite ou par séchage en pulvérisation et l'antibiotique peut être uti lisé sous forme brute pour stimuler la crois sance ou prévenir ou traiter certaines maladies chez l'homme, les animaux et la volaille.
Pour isoler la tétracycline sous forme plus pure, à partir du milieu de fermentation, on peut opérer de différentes manières compor tant une ou plusieurs opérations, exécutées dans des ordres variés, d'adsorption, d'élution, d'extraction par solvant avec ou sans supports, de cristallisation, de précipitation sous forme de sels insolubles, tels que les sels de calcium, baryum, strontium ou magnésium à des<I>pH</I> su périeurs à environ 6,5, ou sous forme de sels insolubles avec des sulfates ou des acides sul- foniques du type agent mouillant ou colorant azoïque ;
ou bien la tétracycline peut être transférée dans des solvants, par extraction, en utilisant, comme agents auxiliaires, par exem ple des sulfates ou des sulfonates du type agent mouillant ou colorant azoïque. La tétracycline peut être séparée à partir du milieu ou moût de fermentation brut, par acidification à un <I>pH</I> d'approximativement 3 ou moins, et la té tracycline passera dans la couche aqueuse, les matières solides insolubles pouvant être sépa rées et éliminées.
La tétracycline peut être précipitée dans des solutions aqueuses sous forme de sels de métaux alcalino-terreux à un<I>pH</I> supérieur à environ 6,5. Ce précipité peut être extrait au moyen d'eau acidifiée ou au moyen de sol vants organiques, tels que des alcools, alcoxy- alcanols ou cétones.
L'extraction peut se faire en milieu acide, neutre ou basique. Similaire- ment, la solution aqueuse contenant de la té tracycline peut être extraite par des solvants organiques non miscibles à la couche aqueuse, tels que des alcools inférieurs, des alcoxy- alcanols, des esters et des cétones non misci bles à la couche aqueuse. Parmi les solvants connus, tels que le butanol, le chloroforme et l'acétate d'éthyle, on préfère le butanol pour l'extraction de la tétracycline. Certains alcools, tels que l'isopropanol, qui sont miscibles à l'eau, mais ne sont pas miscibles à une solu tion saline, sont également utiles.
L'addition d'un sel soluble dans l'eau et insoluble dans la couche organique, tel qu'un halogénure d'am monium, un hydrohalogénure d'amine, un ha- logénure de métal alcalin ou les sulfates cor respondants, facilite l'extraction.
La tétracycline peut être précipitée dans des solutions aqueuses par addition de sels hydrosolubles, tels que des halogénures ou sulfates d'ammonium, d'amines ou de métaux alcalins. La formation de sels peut être favo risée par emploi d'une petite fraction d'un solvant organique.
Si le moût de fermentation contient de la chlortétracycline, celle-ci est récupérée avec la tétracycline dans les procédés décrits ci-dessus. On peut permettre à la chlortétracycline de se dégrader, en maintenant le mélange dans des conditions telles que la tétracycline soit plus stable, ou bien on peut réduire la chlortétra- cycline en tétracycline.
La titulaire a également mis au point un nouveau procédé de récupération hautement efficace tant au point de vue de la récupéra tion d'un produit exempt d'impuretés inactives qu'au point de vue de la récupération d'un produit exempt d'autres substances antibioti ques formées en même temps dans le milieu. Ce procédé de récupération permet d'obtenir un produit cristallin de haute pureté aux deux points de vue en question. Ce procédé impli que essentiellement l'utilisation de sels d'am monium quaternaire, qui précipitent sélective ment la tétracycline dans le milieu de fermen tation à un p" alcalin<I>(pH 8</I> à 11) sous forme de sels d'ammonium quaternaire de tétracy cline.
Après filtration, un tel sel est empâté avec une faible quantité d'eau et une quantité relativement importante de chloroforme, en sorte que le sel d'ammonium quaternaire de tétracycline est dissous dans la phase chloro- formique de la pâte. Les phases aqueuse et chloroformique sont alors, de préférence, sé parées de manière à éliminer les impuretés dissoutes dans l'eau.
La tétracycline est alors extraite à l'aide d'une solution aqueuse acide à un pf, d'environ 1 à 2,5, ce qui donne lieu à la formation du sel d'acide de tétracycline, qui passe en solution dans la phase aqueuse, dans laquelle ce sel précipite sous forme d'un pro duit cristallin, par augmentation du<I>pH</I> de 3 à 7, la précipitation commençant à p1, 3.
Les sels d'ammonium quaternaire, qui sont particulièrement utiles dans ce procédé de ré cupération, sont les chlorures d'alcoyl-tri- méthylammonium et les chlorures de dialcoyl- diméthylammonium, dans lesquels le groupe alcoyle contient 8 à 18 (inclusivement) atomes de carbone. La tétracycline peut être différenciée des antibiotiques connus antérieurement, par exem ple, par analyse chimique ou par ses propriétés optiques.
Le trihydrate de la tétracycline, le quel est obtenu en recristallisant la tétracycline à partir d'un mélange méthanol-eau à tempé rature ambiante, présente les indices de réfrac tion suivants a = 1,500 0,003 ; (3 = 1,646 0,003 ; y =<B>1</B>,798 0,003. L'angle optique 2V est de. 80 - 850 et le signe optique est positif. La rotation optique de la tétracycline cris talline de grande pureté dans le méthanol est (a] D C = - 2340 (0,82 @/o dans méthanol).
La tétracycline est soluble dans le métha nol et le (3-méthoxyéthanol (environ 200 mg par cc), quelque peu soluble dans l'éthanol et le n-butanol, légèrement soluble dans l'eau et relativement insoluble dans des solvants tels que l'éther, le benzène et l'éther de pétrole.
<I>Exemple 1</I> Un milieu formé d'une liqueur de macéra tion du maïs est préparé de la manière sui vante : une liqueur de macération de maïs, présentant une teneur en matières solides d'en viron 50 % (poids/volume) est diluée à l'aide d'eau distillée, jusqu'à obtention d'une con centration finale de 2 %.
On fait passer la ma tière diluée résultante dans une colonne con tenant 30 litres d'un mélange comportant deux volumes de produit marque Amberlite IR 45 , une résine d'échange anionique, et un volume de produit marque Amberlite IRC 50 , une résine d'échange cationique. On ne commence à recueillir l'effluent que lors que la résistivité a été amenée à 5000 ohm- centimètres et l'on poursuit ce recueillement jusqu'à ce que la résistivité soit tombée à moins de 3000 ohm-centimètres.
A 1000 cc de liqueur de macération de maïs désionisée, on ajoute les composés suivants, pour la pré paration d'un milieu convenant pour la pro pagation de l'inoculum et la production de tétracycline, de manière à produire un milieu contenant environ 17 ppm de chlorure, bien qu'on préfère une teneur moindre en chlo rure

Une masse sèche de spores prélevée à la surface d'une culture de microorganismes producteurs de tétracycline sur milieu oblique semi-solide, est utilisée pour ensemencer les fractions de 50 cc, qui sont agi tées à 320 C pendant 72 heures sur une ma chine à agiter à mouvement alternatif (82 courses par minute avec un déplacement de 7,3 cm). A la fin de la période d'agitation, 1,0 cc de la suspension mycélienne résultante est utilisé pour inoculer 400 cc de milieu, qui est agité pendant encore 28 heures.
Quatre fractions de 400 cc sont recueillies aseptique- ment et utilisées pour ensemencer le fermen- tateur.
Le milieu de fermentation est préparé dans des fermentateurs d'une capacité de 94,6 litres comme suit : les constituants, aux concentra tions susindiquées, sont dissous dans 43,5 li tres de liqueur de macération de maïs désioni- sée, en sorte qu'on obtient un volume total final de 49,8 litres. Le milieu est stérilisé à 121 C, en injectant de la vapeur sous une pression de 1,05 kg/cm2 directement dans le milieu, pendant 3 minutes. Le gain de volume fourni par le condensai de vapeur produit la différence requise entre les 43,5 litres initiaux de la solution de liqueur de macération de maïs, de sucre et de sels et les 49,8 litres fina lement requis.
Après refroidissement à 300 C, le mélange est ensemencé à l'aide des quatre fractions de 400 cc d'inoculum décrites plus haut, agité et aéré à raison de 1,6 volume d'air par volume de milieu, pendant 38 heu res. A ce moment, le p1, qui était initialement de 5,9, est tombé à 4,1. Le bouillon titre 20 mcg/ml de produit d'antibiotique, lorsqu'on utilise de l'oxytétracycline comme étalon, et contient en majeure partie de la tétracycline. <I>Exemple II</I> 189 litres de l'inoculum décrit dans l'exemple 1 sont introduits dans un réservoir en acier inoxydable d'une capacité de 473 li tres et stérilisés pendant 30 minutes à 1210 C.
La température est ramenée à 33o C et l'on ensemence avec 400 cc d'un inoculum, obtenu dans le même milieu et provenant d'un flacon à agiter, vieux de 72 heures. Le réservoir est aéré à l'aide de 1,0 volume d'air par volume de milieu et agité pendant 20 heures. A ce moment, le pli du milieu atteint 5,05 et le bouillon est utilisé pour ensemencer un fermen- tateur.

2271 litres du milieu de la composition indiquée ci-dessus sont introduits dans un ré servoir en acier inoxydable d'une capacité de 3785 litres et stérilisés pendant 25 minutes à 121o C. Après refroidissement à 300 C, les 189 litres du contenu précédent du réservoir à inoculum sont introduits aseptiquement et le tout est agité et aéré à l'aide de 0,68 volume d'air par volume de milieu et par minute, pen dant 71 heures, après quoi le milieu titre 92 mcg/cc de produit antibiotique.

Pour isoler la tétracycline à partir du mi lieu de fermentation et la purifier, on peut pro céder selon l'un ou l'autre des modes opéra toires suivants A. Le pï est ajusté à 3,5 à l'aide de 4600 cm3 d'hydroxyde d'ammonium concentré et le mélange est filtré avec 6,8 kg d'un adjuvant de filtra tion,
de manière à séparer le mycélium et l'oxalate de calcium précipité. 113,5 litres d'une solution à 5 fl/o de chlorure de N-(lauroyl-colamino-formyl-méthyl)- pyridinium dans de l'acétate d'éthyle sont ajoutés au milieu filtré et le pIl du mé lange obtenu est ajusté à 8,5 à l'aide de 2200 cc d'hydroxyde de sodium à 50 %. Le mélange est agité pendant 20 minutes et ensuite décanté.
La couche d'acétate d'éthyle (82 li tres) contenant l'antibiotique est séparée et extraite à 3 reprises, en utilisant 1500 cc d'eau et en ajustant chaque fois le<I>pH à</I> 1,7 au moyen d'acide chlorhydri que. On peut aussi utiliser de l'acide sul furique. Les extraits sont réunis et leur pH est ajusté à 7,8 au moyen de 6,5 cc d'hydroxyde d'ammonium 4N, en sorte qu'on obtient la tétracycline libre relati- vement insoluble. Le mélange est filtré et le gâteau de filtration est séché dans un dessiccateur à vide.
Le poids de matière brute atteint 21 gr ; cette matière titre 417 mcg/mg de produit actif. Une répar tition à contre-courant de la matière mon- tre qu'environ 75 % de l'antibiotique pré- sent sont constitués par de la tétracycline.
Un milieu de fermentation contenant plus de 130 mcg/ml de tétracycline est acidi fié à un pl, de 3,5 au moyen d'acide sul furique concentré, de manière à solubi liser l'antibiotique, après quoi le milieu est filtré après y avoir ajouté un adjuvant de filtration. Le volume du milieu filtré est de 4050 litres. A ce milieu filtré, on ajoute 2140 cc de produit marque Ar- quad 16 et l'on ajuste le pl, <I>à</I> 10,0, par addition de 6100 cc d'hydroxyde sodique aqueux à 50 0/0.

. Après l'addition de l' Arquad , le<I>pH</I> est ajusté, de manière à former les sels d'ammonium quaternaire de tétra cycline, qui précipitent dans la gamme de pl, allant de 8 à 11. Après addition de 9 kg d'adjuvant de filtration, le milieu est filtré et les matières solides ainsi obte nues sont lavées à l'eau. Les 4/s environ du gâteau de filtration lavé sont empâtés avec 7,5 litres de méthanol et une quan tité suffisante d'acide chlorhydrique con centré est ajoutée, pour amener le<I>pH à</I> 1,8, ce qui accroît la solubilité de la tétra cycline.
Après agitation pendant 15 mi nutes, le mélange est filtré et les matières solides sont lavées au méthanol, jusqu'à être sensiblement exemptes de toute co loration. Le volume total de l'extrait et des eaux de lavage est de 92 litres. A l'extrait et aux liqueurs de lavage réunis, on ajoute suffisamment d'hydroxyde de sodium aqueux 10N pour porter le<I>pH à</I> 6,8, ce qui donne lieu à la formation de la base libre relativement insoluble, qui précipite. Les matières solides sont fil trées et séchées à l'air. On obtient ainsi 1545 gr de tétracycline trihydratée, libre, amorphe et brute, titrant 296 mcg/mg de produit actif.
C. 2140 ce d' Arquad 16 sont ajoutés à 4050 litres de moût de fermentation filtré à un<I>pH</I> de 3,5. Le pl, est amené à 10. 9 kg d'adjuvant de filtration sont ajoutés et le mélange est filtré. Les matières so lides recueillies sont lavées à l'eau. Un kilogramme du gâteau de filtration lavé est empâté avec 3300 cc de chloroforme et le<I>pH</I> est ajusté à 10,5. Ce<I>pH</I> peut être compris entre 8 et 11. Après agitation pendant 15 minutes, les matières solides sont filtrées et empâtées successivement à deux reprises avec un mélange de 150 cc d'eau et de 3300 cc de chloro forme.
Les extraits chloroformiques re cueillis contenant l'antibiotique et dont la phase aqueuse a été séparée sont alors agités avec 600 cc d'acide sulfurique 0,4N et le pl, est ajusté à 2 au moyen d'acide sulfurique 4N. Après 5 minutes d'agitation, la phase aqueuse est séparée et filtrée. Une fraction de 340 cc du concentrat acide est ajustée à un<I>pH</I> de 5,3 et laissée au repos pendant une nuit dans un réfrigérateur. Les cristaux obtenus sont filtrés, lavés à l'eau glacée et séchés sous vide sur du chlorure de calcium anhydre. 19,5 gr de produit titrant 629 mcg/mg sont obtenus.
On ne constate pas la présence de chlor- tétracycline en quantité détectable. Ce procédé de récupération est pré féré, car il permet une récupération effi cace et produit une matière cristalline. Le gâteau de filtration constitué par des sels d'ammonium quaternaire précipités de tétracycline est extrait à l'aide de chloroforme, ce dernier étant humide en raison de l'eau qui y est présente ou qui est présente dans le gâteau de filtration ou en raison de l'addition d'eau. Si l'on emploie du chloroforme dans des con ditions anhydres, on obtient des rende ments moins bons.
Le gâteau de filtra tion est, de préférence, extrait à plusieurs reprises et la phase aqueuse est préféra- blement jetée. L'extrait chloroformique est acidifié à l'aide d'un acide inorgani que jusqu'à un<I>pH</I> compris, de préfé rence entre 1 et 2,5 environ, de manière à convertir les sels de tétracycline en sels acides correspondants de tétracycline, ces derniers sels étant hydrosolubles. Avant ou pendant l'addition de l'acide, on ajoute de l'eau pour former les deux phases avec les sels acides de tétracycline dans la phase aqueuse.
La phase aqueuse est séparée de la phase chloroformique et le pll est élevé jusqu'à environ 3-7, de manière à obtenir l'antibiotique libre re lativement insoluble. Des valeurs .de pll relativement faibles sont préférées, car on obtient des cristaux plus purs. L'anti biotique libre se sépare en laissant la phase aqueuse au repos, sous forme de cristaux trihydratés, qui peuvent être ai sément séparés par filtration et lavés si on le désire.
Deux colonnes contenant chacune 50 gr de résine Amberlite IRC 50 dans le cycle hydrogène sont tamponnées à pll <I>7,</I> en faisant passer, à un débit de 10 cc par minute, un litre de tampon constitué par une solution normale d'acé tate de sodium et d'acide acétique. La résine Amberlite IRC <B>50 </B> est une ré sine d'échange cationique carboxylique fabriquée par la société Robin & Hass Co. à Philadelphie, USA. 10 litres de bouil lon filtré titrant 41 mcg/ml de matière active sont ajustés à un pll de 7 à l'aide d'une solution aqueuse 4N d'hydroxyde de sodium.
On fait passer ces dix litres à travers une des colonnes d' Amberlite IRC 50 tamponnées, à un débit de 5 cc par minute. Les fractions suivantes d'ef fluents sont recueillies 0-2 litres titrant 13,3 mcg/ml 2-5 litres titrant 8,7 mcg/ml 5-7 litres titrant<B>15,1</B> mcg/ml 7-10 litres titrant 25,0 mcg/ml On fait passer l'effluent de la pre mière colonne à travers la seconde co lonne tamponnée également à un débit de 5 cc par minute.
L'effluent recueilli à la sortie de la seconde colonne se com pose des fractions suivantes 0-2 litres titrant 16,9 mcg/ml 2-4 litres titrant 12,4 mcg/ml 4-6 litres titrant 7,5 mcg/ml 6-10 litres titrant 7,
2 mcg/ml Un litre d'acide chlorhydrique normal dans 60 % de méthanol est amené à pas- ser à travers la première colonne, à un débit de 10 cc par minute.
L'éluat, qui contient 74% de produit actif dans le moût filtré est concentré sous vide jus qu'à un tiers environ de son volume, le pll étant alors abaissé jusqu'à 2,5. Le pll est amené à 1,8 par addition d'un adju vant de filtration, en vue d'éliminer les matières solides qui se sont séparées pen dant la concentration. Les matières soli des filtrées sont lavées à l'eau et le filtrat, auquel on ajoute les liqueurs de lavage, est amené à un<I>pH</I> de 7,8 par addition d'hydroxyde de sodium 4N.
Les matières solides qui se séparent sont successive ment lavées et séchées sous vide. On ob tient ainsi 1375 gr de produit titrant 78 mcg/mg de matière active. E. On peut précipiter de la tétracycline dans des solutions aqueuses de tétracycline à un<I>pH</I> alcalin, par addition d'un mélange de chlorure de baryum et de chlorure de magnésium.
Ainsi, lorsque 15 gr de chlo rure de baryum dihydraté et 2 gr de chlo rure de magnésium hexahydraté sont ajoutés à une solution de 3 gr de tétra cycline brute dans un litre d'eau à un py de 2,5 et lorsque de l'hydroxyde de so dium aqueux est ajouté pour porter le <I>pH</I> à 8,5, la totalité du produit actif se retrouve dans le précipité, qui est séparé par filtration de la liqueur contenant les impuretés.
Lorsque ce précipité est em pâté avec de l'eau et que le<I>pH</I> est ramené à 1,5 au moyen d'acide sulfurique, et lorsque le mélange est filtré pour séparer le sulfate de baryum insoluble, l'extrait aqueux résultant contient pratiquement la totalité de l'activité microbiologique ori ginellement présente. Cette matière brute titre approximativement 427 mcg/ mg. La solution aqueuse est ajustée à un <I>pH</I> de 3 par l'addition d'acide chlorhydri que. La solution est alors saturée avec du n-butanol et soumise à une distribution à contre-courant dans 9 entonnoirs de sépa ration en utilisant des volumes égaux de n-butanol saturé au moyen d'eau dont le <I>pH</I> a été ajusté à 3 au moyen d'acide chlorhydrique dilué.
Après distribution, les tubes médians 4, 5, 6 et 7 sont com binés et le<I>pH</I> est amené à 7 par addition d'une solution diluée d'hydroxyde de so dium. On concentre ce mélange sous vide à 40-50 C.
