Procédé de préparation d'un sel d'ammonium quaternaire.
La présente invention se rapporte à un procédé de préparation de sels d'ammonium quaternaires correspondant à la formule générale suivante :

dans laquelle X est un radical alcoylène infé- rieur ; NRRt est un radical aminotertiaire ; R2 est un radical-alcoyle inférieur ; R3 est un radical alcoyle inférieur ou un radical benzyle ; enfin, An est un anion non toxique. Ces composés obtenus par le procédé conforme à l'invention ont des propriétés pharmacologi- ques utiles ; par exemple ils arrêtent l'évolu- tion des ganglions.
Dans la formule générale donnée ci-dessus. le radical aleoylène inférieur désigné par X possède, de préférence, de deux à quatre atomes de carbone et ses deux valences libres sont fixées sur des atomes de carbone différents. Ainsi X comprend des radicaux tels que -CH2CH2-,

Le radical amino tertiaire représenté ci-dessus par N.lRi peut être notamment un radical dialcoyTamino dans lequel R et Ri sont des groupes alcoyle inférieur, identiques ou différents, chaque groupe alcoyle ayant de 1 à 6 atomes de carbone ; des radicaux dialcoylamino de ce genre comprennent les radicaux diméthylamino, diéthylamino, éthylméthyl- amino, diisopropylamino, éthyl-n-propylamino, di-n-butylamino, di-n-hexylamino, etc.
En outre, le radical amino tertiaire désigné par NRRi peut être un radical saturé N-hétéro- monocyclique ayant 5 ou 6 atomes dans le noyau ; à titre d'exemples de ces radicaux, on peut citer le 1-pipéridyle et ses dérivés substitués par un groupe alcoyle inférieur, tels que le 2-méthyl-1-pipéridyle, le 3-éthyl-1 pipéridyle, le 4-méthyl-1.-pipéridyle, le 2, 6-di méthyll-pipéridyle ; le 1-pyrrolidyle et ses dérivés substitués par un groupe alcoyle inférieur tels que le 2-méthyl-1-pyrrolidyle, le 2, 5-diméthyll-pyrrolidyle, le 4-morpholinyle, etc.

An peut être un anion provenant d'un acide fort. Ces anions comprennent ceux provenant t d'wn acide minéral fort, par exemple le chlorure, le bromure, l'iodure ou le sulfate, ou ceux provenant d'un acide sulfonique organique, par exemple le benzène-sulfonate, le para toluène-sulfonate, etc. L'anion non toxique An n'a pas d'activité pharmacologique appréciable par lui-même dans les dilutions élevées sous la forme desquelles l'ensemble des sels d'ammonium quaternaires sont efficaces. En particulier, les anions ne contribuent en rien à l'activité de blocage des ganglions qui ne réside que dans le reste de la molécule.
Conformément au procédé de l'invention, on prépare les sels d'ammonium quaternaires correspondant à la formule ci-dessus en traitant un 4-nitro-2- (aminoalcoxy-tertiaire)- benzoate d'alcoyle inférieur correspondant avec un ester de formule R3-An et en traitant le sel d'ammonium quaternaire résultant avec un agent réducteur.
On peut préparer les produits de départ de ce procédé en faisant réagir un 4-nitro-2 hydroxy-benzoate d'alcoyie inférieur, sous forme d'un sel métallique avec un halogénure d'aminoalcoyle tertiaire. Dans une variante, on peut utiliser un 4-nitro-2-hydroxy-benzoate d'alcoyle inférieur à l'état libre avec toutefois une diminution résultante du rendement en 4-nitro-2- (aminoalcoxy-tertiaire)-benzoate d'alcoyle inférieur.
La réduction est effectuée soit par des procédés chimiques, soit par hydrogénation catalytique. Des agents de réduction chimique appropriés comprennent le fer et 1'acide chlorhydrique, le sulfate ferreux et l'ammoniac, l'étain et 1'acide chlorhydrique, l'hypo- sulfite de soude, etc. Lors de la mise en oeuvre de la présente invention, la titulaire utilise de préférence du fer et de l'acide chlorhydrique. Les catalyseurs appropriés lorsqu'on utilise l'hydrogénation catalytique sont le nickel de Raney, le platine, le palladium ou d'autres catalyseurs généralement efficaces pour catalyser l'hydrogénation des groupes nitro en groupes amino.
Le présent brevet a pour objet un procédé de préparation du méthoiodure de 4-amino-2 (-diéthylaminoéthoxy)-benzoate d'éthyle, ca- ractérisé en ce qu'on fait réagir le 4-nitro-2 ( B-diéthylamino-éthoxy)-benzoate d'éthyle avec de l'iodure de méthyle et qu'on transforme le groupe nitro du méthoiodure de 4-nitro-2 (ssdiéthylamino-éthoxy)-benzoate d'éthyle ainsi obtenu en groupe amino par traitement avec un agent réducteur.
Le méthoiodure de 4-amino-2- (jg-diéthyl- amino-éthoxy)-benzoate d'éthyle ainsi obtenu
est mie substance nouvelle, cristallisée et qui
fond à 139 2-141 1 C. Il sert en thérapeu tique.
Exemple:
Le 4-nitro-2-(¯-diÚthylamino-Úthoxy)-benzoate d'éthyle, utilisé comme produit de départ, peut être préparé de la manière suivante :
Dans une solution, chauffée à reflux et soumise à l'agitation, de 42, 2 g de 4-nitro-2oxy-benzoate d'éthyle dans 1000 em3 d'alcool alsolu, on ajoute lentement une solution de 4, 6 g de sodium dans 500 em3 d'éthanol ab solu. On ajoute à la solution rouge foncé obtenue, tout en agitant pendant environ 20 mi- nutes, 27, 1 g de chlorure de B-diéthylamino- éthyle, puis on chauffe le mélange résultant à reflux pendant environ 3 heures.
On ajoute ensuite encore 3 g de chlorure de ss-diéthyl- amino-éthyle et on continue de chauffer à reflux pendant 30 minutes. On refroidit le mélange dans de la glace, on le filtre et on évapore le filtrat sous vide. On reprend le résidu dans 500 em3 3 d'acétate d'éthyle, on filtre la solution résultante et on évapore le filtrat à sec ; on obtient ainsi le 4-nitro-2-(ss- diéthylamino-éthoxy)-benzoate d'éthyle brut.
Le chlorhydrate purifié de ce composé fond à 144 4145 2 C (corrigé) et donne les résul- tats suivants à l'analyse :

On peut également préparer le 4-nitro-2
(ss-diéthyL3mino-éthoxy)-benzoate d'éthyle de la manière suivante : On fait chauffer à reflux un mélange de 84, 4 g de 4-nitro-2-oxy-benzoate d'éthyle, 60, 8 g de carbonate de potassium anhydre en poudre et 2000 em3 de métaxylène, tout en agitant, sous un collecteur d'eau, jusqu'à ce que l'on ne recueille plus d'eau (3 heures). On enlève le collecteur d'eau et on ajoute en une seule fois 112, 7 g de para-toluène-sulfonate de -chloro-ethyle.
On continue à chauffer à reflux et à agiter pendant 19 heures. On filtre le mélange tandis qu'il est encore chaud et on lave le gâteau de filtration avec du toluène chaud. On éva- pore sous vide le filtrat et les liquides de ravage jusqu'à siccité et on obtient comme résidu un solide couleur crème. Plusieurs recristallisations de ce solide dans le méthanol et dans le n-pentane donnent avec un rendement de 601/o du 4-nitro-2-(,-ehloro-éthoxy)- benzoate d'éthyle sous forme de petites plaques jaune pâle dont le point de fusion est compris entre 56 6 et 57 2 C (corrigé).

On chauffe alors un mélange du 4-nitro-2- (ss-ehloro-éthoxy)-benzoate d'éthyle ainsi obtenu, de diéthylamine, d'iodure de sodium et d'Úthanol absolu et on obtient, avee un rendement acceptable, du 4-nitro-2-(ss-diéthylamino- éthoxy)-benzoate d'éthyle.
La réaction avec l'iodure de méthyle est effectuée de la manière suivante :
A une solution de 6 g de 4-nitro-2-(¯-di éthylamino-éthoxy)-benzoate d'éthyle dans 50 em3 d'acétate d'éthyle, on ajoute 15 cm3 d'iodure de méthyle. Après environ 5 minutes à la température ambiante, le mélange devient trouble. On le chauffe alors à reflux pendant 1 heure et demi pour assurer l'achèvement de la réaction. Après environ 5 minutes de chauffage à reflux, il se forme des cristaux et la couleur du mélange de réaction passe de l'orange foncé au jaune très pâle. On refroidit le mélange de réaction dans de la glace, on le filtre et on lave le produit avec de l'acétate d'éthyle.
On recristallise ce produit une fois dans un mélange d'isopropanol et d'acétate d'éthyle et une fois dans l'isopropanol seul ; on obtient le méthoiodure du 4-nitro-2- (/ ?-diéthyl- amino-éthoxy)-benzoate d'éthyle, point de fusion : 143 1-144 6 C (corrigé).

La réduction du groupe nitro de ce composé est effectuée de la manière suivante :
On mélange 18, 5 g de méthoiodure de 4-nitro- (fl- di6thylamino-6thoxy)-benzoate d'éthyle avec 400 mg d'oxyde de platine monohydrate et avec assez d'éthanol absolu pour porter le volume total à 150 em3, on traite ensuite ce mélange avec de l'hydrogène, sous pression et à température ambiante. La réaction est terminée après environ 15 minutes.
On filtre le catalyseur et on le lave avec de l'éthanol absolu. On évapore sous vide le filtrat et les liquides de lavage jusqu'à siccité, on traite le résidu avec de l'acétate d'éthyle et on chasse ce dernier par distillation sous vide. On traite à nouveau le résidu avee de l'acétate d'éthyle et on évapore à nouveau le solvant sous vide ; on obtient ainsi un résidu cristallin. On le filtre et on le lave soigneusement avec de l'acétate d'éthyle. On recristallise deux fois en décolorant avec du charbon, une fois dans un mélange l'isopro- panol et d'acétate d'éthyle et une fois dans l'acétone.
Le produit obtenu, qui est le méthoiodure de 4-amino-2- (ss-diéthylamino- éthoxy)-benzoate d'éthyle, fond à 139 2 à 14l"10 (corrigé).
