Verfahren zur Herstellung eines neuen Esters.
Die vorliegende Erfindimg bezieht sich auf ein Verfahren zur Herstellung eines neuen Esters und ist dadurch gekennzeichnet, das man Nonaäthylenglykolmonomethyläther mit den p-Butylaminobenzoesäurerest abgebenden Verbindungen umsetzt. Der Endstoff ist ein farbloses 61, welches in den meisten organischen Lösungsmitteln, mit Ausnahme von aliphatischen Kohlenwasserstoffen, leicht löslich ist.

CH3 O (CH2 CH2 ) 8 CH2 CH2 OH . \ls den p-Butylaminobenzoylrest abgebende Verbindungen sind beispielsweise ge beignet :
1. p-Butylaminobenzoesäure, wobei es zweckmässig ist, die Veresterung unter Erhitzung bei einem niedrigen pH-Wert durch- zuführen.
Gute Resultate werden erhalten, wenn man pro Mol p-Butylaminobenzoesäure etwas mehr als 1 Mol p-Toluolsulfonsäure zugibt und dann zusammen mit übersehüssigen Nonaäthylenglykolmonomethyläther erhitzt, unter Entfernung des bei der Veresterung entstehenden Wassers durch Einleiten eines inerten Gases oder im Vakuum.
2. Ein Ester aus p-Butylaminobenzoesäure und einem niedrigmolekularen Alkohol, wobei zweckmässig in der im Beispiel angegebenen Weise verfahren wird.
Der neue Ester soll als Heilmittel verwen det werden.
Beispiel : 4, 42 Gewichtsteile p-Butylaminobenzoe- säureäthylester werden mit 17, 2 Gewichtstei- len Nonaäthylenglykolmonomethyläther, wel- cher bei 0, 01 mm Hg bei 190 siedet, in ein versehlossenes Reaktionsgefäss gebracht, welches mit einem regulierbaren Einlauf f r Lösungsmittel und mit einem Anschluss zum Abdestillieren im Vakuum verbunden ist. Zur vollständigen Trocknung wird wÏhrend einer Stunde auf 100 bis 105 erhitzt und bei einem Vakuum von 12 mm Hg absolutes Xylol unter die Oberfläehe der Mischung eingeleitet.
Dadurch entsteht ein ständiger Strom von Xyloldampf durch die ganze Apparatur, welcher letzte Spuren von Feuchtigkeit und andere flüchtige Verunreinigungen mitreisst.
Das Xylol wird in einem Kühler kondensiert.
Man kühlt auf 20 bis 30 ab und gibt 0, 06 Ge wichtsteile Natriummethylat, gel¯st in 0, 6 Ge wichtsteilen Methanol, zu. Darauf wird, wiederum im Vakuum bei 100 bis 105¯, Xylol durchgeleitet, wobei alles Methanol und das sieh bei der Umsetzung bildende ¯thanol verdampfen. Man lässt die Umestemng unter diesen Bedingungen vor sich gehen, bis eine Probe der Reaktionsmasse in kaltem Wasser klar löslich ist, was ungefähr nach 2 bis 3 Stunden der Fall ist. Man erhält so in fast quantitativer Ausbeute den Ester von folgen- der Formel :

der noch überschüssigen NonaÏthylenglykotmonomethylÏther enthÏlt. Zur Reinigung wird in Benzol gelöst und mehrmals mit 5prozentiger Sodal¯sung gewaschen.
Es ist zweckmässig, alle Wasch]ösungen mit frischem Benzol auszuschüt. teln. Der neue Ester bleibt bei dieser Verteilung zwischen Benzol und Sodalösung im Benzol, der überschüssige NonaÏthylenglykolmonomethylÏther und wenig braune Verunreinigungen werden von der verdünnten Sodalösung aufgenommen. Durch Eindampfen der getrockneten und filtrierten Benzollösung erhält man den neuen Ester in Form eines farblosen bis ganz schwach gelben Öls, das in den meisten organisehen Lösungsmitteln, mit Ausnahme von aliphatisehen Kohlenwasserstoffen, leicht löslich ist.
Aus einer l0prozentigen wässerigen Listing wird der neue Ester beim ErwÏrmen auf etwa 45 ausgefällt, er geht jedoeh beim Ab- kühlen wieder leieht in Losung. Er ist für die therapeutische Anästhesie hervorragend geeignet. Wässerige Losungen sind haltbar und können sterilisiert werden.
